Hermetia illucens, the black soldier fly, is a common and widespread fly of the family Stratiomyidae. Its ability to grow on contaminated substrates suggests the production of antimicrobial peptides that enable its survival. This study aimed to verify the impact of direct and indirect infection with Salmonella enteritidis on the expression of defensins and cecropins in Hermetia illucens larvae. In addition to an infection with a microorganism, it was interesting to verify if the expression of peptides and the relative action of hemolymph changed in larvae subjected to mechanical stress by abdominal puncture. The peptide fraction of the hemolymph of infected larvae was tested using antibiogram and minimum inhibitory concentration tests against Salmonella enteritidis and Salmonella typhimurium. Both molecular and microbiological tests were carried out at three different time points, on larvae not subjected to any treatment (T-0), four hours after treatment (T-1), and 24 h after treatment (T-2). The results of the microbiological tests showed the antimicrobial action of the peptide fraction of the hemolymph against both S. typhimurium and S. enteritidis; for the latter one, the action was more marked. Interesting results were also found for larvae subjected only to mechanical stress by puncture. Molecular tests on the expression of defensins and cecropins were in full agreement with those obtained in the microbiological tests, with expression more pronounced in larvae infected directly with Salmonella enteritidis. Temporal and condition-specific regulation of defensins and cecropins highlights the complexity of the immune response and suggests sophisticated mechanisms by which the host fine-tunes antimicrobial peptide expression to enhance pathogen defense while preventing excessive immune activation.

Toxoplasmosis is a parasitic disease affecting a wide range of species, including humans, and can be responsible for important clinical manifestations such as abortion and neurological signs. Sheep show a remarkable susceptibility to its causative agent, Toxoplasma gondii , and zoonotic transmission may occur in case of consumption of undercooked meat obtained from infected animals. Toxoplasma gondii seroprevalence in sheep can significantly vary on a geographical basis, as shown by numerous surveys conducted worldwide. To investigate environmental and climate conditions that may affect the likelihood of ovine infection, 405 serum samples from selected sheep raised in 91 farms were collected from two abattoirs, with each abattoir receiving animals from two regions (1/Tuscany-Latium and 2/Campania-Basilicata). The seroprevalence of infection in all examined animals was 53.8%. Young animals ( n = 165) had a lower likelihood of being T. gondii positive compared to the adults (OR = 0.21), and the seropositive rate of animals slaughtered in abattoir 2 was significantly higher than that of animals slaughtered in abattoir 1 (60.5 vs. 43.2%, p < 0.01). The significant bioclimatic variables ( p < 0.05) associated with the presence of T. gondii antibodies were related to areas with a lower range of temperature and higher precipitation. In conclusion, this study expands on the interpretation of serological data, with the inclusion of environmental and climatic variables, as possible risk factors in the spread of toxoplasmosis in the study area. These findings provide novel insights to support public health measures, such as risk-based control plan, and contribute to a “One Health” approach, taking into account the environmental and climatic perspectives.

Comparison of Direct and Indirect Detection of Toxoplasma gondii in Ovine Using Real-Time PCR, Serological and Histological Techniques

Toxoplasma gondii is a zoonotic pathogen and the ingestion of tissue cysts by consumption of lamb or mutton has been identified as a possible cause of infection in humans. Many serological surveys in sheep have been performed, showing relevant serological rates; however, while the detection of antibodies indicates an exposure to T. gondii, this does not necessarily imply the presence of tissue cysts in edible tissue. The current study aims to provide further understanding on the occurrence of T. gondii in sheep muscles and the strength of correlation between serological positivity and presence of the parasite in sheep. From 349 sheep, samples (i.e., blood, heart and diaphragm) were collected and subjected to ELISA tests, real-time PCR and histological tests. Despite the high seroprevalence, T. gondii DNA was detected in the heart and/or the diaphragm from 13 out of the 349 tested sheep (3.7%); all were adults (13/191). Furthermore, the histological tests did not reveal the presence of T. gondii tissue cysts in any of the examined portions of interventricular septum. It should be considered that the likelihood of detecting genetic material of the parasite is probably influenced by the uneven distribution of the tissue cysts in the carcass as well as the methodology applied. The findings of this study support the importance of describing the uncertainty associated with the data used for risk assessment to reduce inaccurate estimation or risk overestimation.

Many studies worldwide reported high antibody levels to T. gondii in sheep, but this does not always indicate that tissues destined for consumption contain infectious cysts. This study aimed to understand T. gondii occurrence in sheep muscles and the correlation between seropositivity and parasite presence. Samples from 349 sheep (blood, heart and diaphragm) underwent ELISA, real-time PCR and histological testing. Despite high seroprevalence rates, T. gondii DNA was found in only 3.7% of tested sheep (13/349), all adults. Histological tests did not detect T. gondii tissue cysts in any samples tested. The results agree with previously published research and might be influenced by the uneven distribution of tissue cysts in carcasses or muscle and by the analytical methods used. Isolation of Mycobacterium avium Subsp. Paratuberculosis in the Feces and Tissue of Small Ruminants Using a Non-Automated Liquid Culture Method

Paratuberculosis is a chronic disease of ruminants caused by Mycobacterium avium subsp. Paratuberculosis (MAP). Since isolation of MAP type I (S) is rarely reported in Italy, our research was aimed at isolating, by an inexpensive liquid culture manual method, this type of MAP isolates. At first, we used an ELISA to point out to serologically positive samples from five flocks. Secondly, we used a fecal direct IS900-qPCR on the ELISA positive samples, in order to detect shedder animals. Feces from IS900-qPCR positive samples were inoculated in solid and liquid culture media. IS900-qPCR was further used to test the growth of MAP isolates in liquid medium, which were further confirmed by f57-qPCR and submitted to typing by specific PCR in order to identify the MAP type. Twenty-eight samples (24 fecal and four tissutal samples) were processed by culture methods, resulting in the isolation of six type I MAP field isolates. Notably, no isolates were recovered by solid media, underlining the utility of this liquid method. Few data about this type of MAP are currently available in Italy, and further analyses should be carried out in order to study the origin and epidemiology of type I strains circulating in Italy.

Rural electrification in remote areas of developing countries has several challenges which hinder energy access to the population. For instance, the extension of the national grid to provide electricity in these areas is largely not viable. The Kenyan Government has put a target to achieve universal energy access by the year 2020. To realize this objective, the focus of the program is being shifted to establishing off-grid power stations in rural areas. Among rural areas to be electrified is Habaswein, which is a settlement in Kenya’s northeastern region without connection to the national power grid, and where Kenya Power installed a stand-alone hybrid mini-grid. Based on field observations, power generation data analysis, evaluation of the potential energy resources and simulations, this research intends to evaluate the performance of the Habaswein mini-grid and optimize the existing hybrid generation system to enhance its reliability and reduce the operation costs. The result will be a suggestion of how Kenyan rural areas could be sustainably electrified by using renewable energy based off-grid power stations. It will contribute to bridge the current research gap in this area, and it will be a vital tool to researchers, implementers and the policy makers in energy sector.

Effectiveness of patients’ involvement in a medical and nursing pain education programme: a protocol for an open-label randomised controlled trial including qualitative data

IntroductionPain is a multidimensional experience that varies among individuals and has a significant impact on their health. A biopsychosocial approach is recommended for effective pain management; however, health professionals’ education is weak on this issue. Patient involvement is a promising didactic methodology in developing a more holistic perspective, however there is a lack of reliable evidence on this topic. The aim of the present study is to evaluate the effectiveness of patient involvement in pain education in undergraduate medicine and nursing students.Methods and analysisAn open-label randomised controlled trial including qualitative data will be conducted. After an introductory lesson, each student will be randomly assigned to the intervention group, which includes an educational session conducted by a patient–partner along with an educator, or to the control group in which the session is exclusively conducted by an educator. Both sessions will be carried out according to the Case-Based Learning approach. Primary outcomes will be students’ knowledge, attitudes, opinions and beliefs about pain management, whereas the secondary outcome will be students’ satisfaction. The Pain Knowledge and Attitudes (PAK) and Chronic Pain Myth Scale (CPMS) will be administered preintervention and postintervention to measure primary outcomes. Students’ satisfaction will be measured by a questionnaire at the end of the session. Two focus groups will be conducted to evaluate non-quantifiable aspects of learning.Ethics and disseminationThe protocol of this study was approved by the independent Area Vasta Emilia Nord ethics committee.Adherence to The Declaration of Helsinki and Good Clinical Practice will ensure that the rights, safety and well-being of the participants in the study are safeguarded, as well as data reliability. The results will be disseminated through scientific publications and used to improve the educational offer. A version of the anonymised data set will be released for public access.Trial registrationTrial was not registered on ClinicalTrials.gov as the interventions being compared only concern educational programmes and the outcomes considered do not refer to any clinical dimension.
