miRNA‐encapsulated abiotic materials and biovectors for cutaneous and oral wound healing: Biogenesis, mechanisms, and delivery nanocarriers
MicroRNAs (miRNAs) as therapeutic agents have attracted increasing interest in the past decade owing to their significant effectiveness in treating a wide array of ailments. These polymerases II‐derived noncoding RNAs act through post‐transcriptional controlling of different proteins and their allied pathways. Like other areas of medicine, researchers have utilized miRNAs for managing acute and chronic wounds. The increase in the number of patients suffering from either under‐healing or over‐healing wound demonstrates the limited efficacy of the current wound healing strategies and dictates the demands for simpler approaches with greater efficacy. Various miRNA can be designed to induce pathway beneficial for wound healing. However, the proper design of miRNA and its delivery system for wound healing applications are still challenging due to their limited stability and intracellular delivery. Therefore, new miRNAs are required to be identified and their delivery strategy needs to be optimized. In this review, we discuss the diverse roles of miRNAs in various stages of wound healing and provide an insight on the most recent findings in the nanotechnology and biomaterials field, which might offer opportunities for the development of new strategies for this chronic condition. We also highlight the advances in biomaterials and delivery systems, emphasizing their challenges and resolutions for miRNA‐based wound healing. We further review various biovectors (e.g., adenovirus and lentivirus) and abiotic materials such as organic and inorganic nanomaterials, along with dendrimers and scaffolds, as the delivery systems for miRNA‐based wound healing. Finally, challenges and opportunities for translation of miRNA‐based strategies into clinical applications are discussed.
The use of tear ferning test in cats for evaluation of ocular surface
Background The tear film is a trilaminar fluid composed mainly of lipids, electrolytes, proteins and water. It is responsible for lubrication, nutrition and protection against microbial and toxic agents. Disruption of any these components may weaken the ocular surface, making it more susceptible to disease. Increasing evidence suggests that qualitative tear film deficiencies are an important predisposing factor or cause of some of the most common and challenging ocular diseases in cats, including conjunctivitis, corneal ulcer, spontaneous chronic corneal epithelial defects (SCCED), pigmentary keratitis, corneal sequestrum and dry eye syndrome. The aim of this study was to describe the tear ferning test in healthy cats and to compare the results by using two grading scales for humans. Tear samples were collected using Schirmer tear test (STT) strips from 60 healthy cats, and, after centrifuging the strips to obtain the samples, the aliquot was placed on clean microscope glass until it dried and the tear ferning patterns were observed under a polarized light microscope and classified according to the Rolando and Masmali grading scales. Results Ferning patterns in the lower grades showed full crystallization with high density, without gaps between the ferns and branches, forming several nuclei that were easily distinguished. According to the Rolando scale, 50% (60/120), 46.6% (56/120) and 3.4% (4/120) of eyes showed type I, II and III patterns, respectively. According to the Masmali scale, 15% (18/120), 56.6% (68/120 eyes) and 28.4% (34/120) of eyes showed grade 0, 1 and 2 patterns, respectively. No difference was observed between the right and left eyes for both Rolando (P = 0.225) and Masmali (P = 0.683) scales. Conclusions The tear ferning test is a qualitative test that can be used in cats as a complementary evaluation of the ocular surface. While the Rolando scale showed an increased prevalence of types I and II, the Masmali scale showed an increased prevalence of grades 1 and 2. This can be attributed to the species-specific differences between human and feline tear film. So Masmali grade 2 can be considered a normal tear pattern for the species, because all the cats used in study were clinically healthy. For this reason, future complementary studies are necessary for comparing healthy eyes and eyes with different ocular surface disease in cats. Both scales can be feasible options for grading tear crystallization in cats, but as Rolando scale included 96.6% of the samples in the 2 types that are considered normal for humans, we think that this scale seemed to be more precise to classify crystallization pattern in cats. The crystallization patterns observed in this study can form the basis for standardizing ocular surface parameters in cats.
Molecular Characterization of Gram-Negative Bacilli Isolated from a Neonatal Intensive Care Unit and Phenotypic and Molecular Detection of ESBL and Carbapenemase
Introduction: The increase in the rates of multidrug-resistant bacteria in healthcare environments has been recognized as a global public health problem. In view of the scarcity of data on the neonatal population, this study aimed to provide information on the genotypic and epidemiological characteristics of Gram-negative microorganisms isolated from colonization and infection sites in neonates admitted to a tertiary university center of high complexity. Methods: Enterobacterales and non-fermenting Gram-negative bacilli previously collected in a prospective cohort study were submitted to genotypic identification, detection of extended-spectrum β-lactamases (ESBL), carbapenemases and biofilm production, detection of specific virulence markers in Pseudomonas aeruginosa, and typing by pulsed-field gel electrophoresis. Results: The data found here revealed higher rates of infection by Klebsiella spp. and Serratia marcescens that caused bloodstream infection and pneumonia, respectively. In this study, high biofilm production was observed, with 95.0% of Enterobacterales and 100% of non-fermenting Gram-negative bacilli being producers. Most of the P. aeruginosa isolates carried pathogenicity factors such as alginate, hemolytic phospholipase C, exotoxin A, and rhamnolipids. The phenotypic analysis of ESBL revealed that 16 (5.3%) isolates produced these enzymes. Four of these isolates (66.7%) carried the CTX-M-9 gene, three (50%) carried the TEM gene, and one (16.7%) was positive for the SHV and CMY-2 genes. Univariate and multivariate Cox regression analyses were used to identify risk factors for colonization and infection by Gram-negative microorganisms. The results of multivariate analysis revealed that biofilm production by these microorganisms was associated with the persistence of colonization by the same pathogen in the newborn and increased by 75% the daily probability of the newborn developing infection. The production of ESBL also increased the daily probability of infection by 46.8 times. Conclusions: Enterobacterales showed average biofilm production, while the majority of non-fermenting Gram-negative bacilli were strong producers. The present data increase our knowledge of the molecular epidemiology of important Enterobacterales species, with emphasis on ESBL-producing Enterobacter cloacae and Klebsiella pneumoniae with emerging epidemiological potential in the neonatal intensive care unit of a tertiary university hospital. Furthermore, the results highlight the need for the monitoring and implementation of control measures and for restricting the use of broad-spectrum antibiotics.
Comprehensive insights on treatment modalities with conventional and herbal drugs for the treatment of duodenal ulcers
Areas of the body accessible to gastric secretions, such as the stomach and duodenum, are most commonly damaged by circumscribed lesions of the upper gastrointestinal tract mucosa. Peptic ulcer disease is the term for this illness (PUD). About 80% of peptic ulcers are duodenal ulcers, with stomach ulcers accounting for the remaining 20%. Duodenal ulcers are linked to the two primary results about Helicobacter pylori infection and COX inhibitor users. Additional causes might include drinking, smoking, stress, and coffee consumption. The indications and symptoms of a duodenal ulcer depend on the patient's age and the lesion's location. For duodenal ulcers, proton pump inhibitors (PPIs) are the usual course of treatment. This comprehensive study included an in-depth literature search in the literature and methods section using electronic databases such as PubMed, ScienceDirect, and Google Scholar. The search method included publications published from the inception of the relevant database to the present. Inclusion criteria included studies investigating different treatment options for duodenal ulcer disease, including traditional pharmacotherapy and naturopathic treatments. Data mining includes information on treatment techniques, treatment outcomes, and possible synergies between conventional and herbal treatments. In addition, this review critically examines the available information on the effectiveness, safety, and possible side effects of different treatments. The inclusion of conventional and herbal treatments is intended to provide a comprehensive overview of the many treatment options available for duodenal ulcer disease. A more comprehensive and personalized treatment plan can be achieved by incorporating dietary changes, lifestyle modifications, and, if necessary, herbal therapies to complement other treatments normally. Root growth and carbohydrate partitioning in cotton subjected to shading in the initial phase
Cotton cultivation in Brazil tends to occur mostly in the rainy season, and at the time of developing the crop there are often prolonged periods of low solar radiation. The objective of this work was to evaluate the root development of cotton cultivars subjected to shading in the initial phase. The experiment was carried out in a greenhouse, using rhizotrons. The experimental design was a 2x2 factorial in randomized blocks with five replications. The treatments were composed of cotton cultivars TMG 1WS (early) and TMG 8WS (late) and luminosity conditions: non-shaded and shaded (50% reduction of luminosity). There were no interactions between the factors “shading” and “cultivars” for any parameter evaluated. Shading reduced total and lateral root growth from the 21 days after emergence (DAE) and the main root at 24DAE. Shading caused reductions in length (60%), surface (65%), mean diameter (15%), root volume (69%), root dry matter mass (83%) and shoot (64%), as well as decreasing the content (36%) and the accumulation of carbohydrates in the shoot (73%). Shading at early cotton development reduces the shoot carbohydrates production, affecting dry matter accumulation and root growth, and this response is independent of the cultivar. RESUMO: O cultivo do algodoeiro no Brasil está concentrado no período chuvoso, e no momento do estabelecimento da lavoura frequentemente há períodos prolongados de baixa radiação solar. O objetivo deste trabalho foi avaliar o desenvolvimento radicular de cultivares de algodoeiro submetidos ao sombreamento na fase inicial da cultura. O experimento foi realizado em ambiente controlado, em rizotrons. O delineamento experimental foi um fatorial 2x2 em blocos ao acaso com cinco repetições. Os tratamentos foram as cultivares de algodão TMG 11WS (precoce) e TMG 81WS (tardia) e as condições de sombra: sem sombra e com sombra (50% de redução da luminosidade). Não houve interação entre os fatores “sombra” e “cultivares estudadas” para nenhum parâmetro avaliado. A sombra reduziu o crescimento radicular total e das raízes laterais a partir dos 21 dias de emergência (DAE) e da raiz principal aos 24DAE. O sombreamento reduziu o comprimento (60%), a superfície (65%), o diâmetro médio (15%), o comprimento por volume (60%), o volume radicular (69%), a massa de matéria de seca da raiz (83%) e da parte aérea (64%), o teor (36%) e o acúmulo de carboidratos na parte aérea (73%). Em conclusão, o sombreamento na fase inicial de desenvolvimento do algodoeiro reduz a produção de carboidratos da parte aérea, afetando o acúmulo de matéria seca e o crescimento radicular, sendo que essa resposta independe da cultivar.
Sugarcane Yield Estimation Using Satellite Remote Sensing Data in Empirical or Mechanistic Modeling: A Systematic Review
The sugarcane crop has great socioeconomic relevance because of its use in the production of sugar, bioelectricity, and ethanol. Mainly cultivated in tropical and subtropical countries, such as Brazil, India, and China, this crop presented a global harvested area of 17.4 million hectares (Mha) in 2021. Thus, decision making in this activity needs reliable information. Obtaining accurate sugarcane yield estimates is challenging, and in this sense, it is important to reduce uncertainties. Currently, it can be estimated by empirical or mechanistic approaches. However, the model’s peculiarities vary according to the availability of data and the spatial scale. Here, we present a systematic review to discuss state-of-the-art sugarcane yield estimation approaches using remote sensing and crop simulation models. We consulted 1398 papers, and we focused on 72 of them, published between January 2017 and June 2023 in the main scientific databases (e.g., AGORA-FAO, Google Scholar, Nature, MDPI, among others), using the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) methodology. We observed how the models vary in space and time, presenting the potential, challenges, limitations, and outlooks for enhancing decision making in the sugarcane crop supply chain. We concluded that remote sensing data assimilation both in mechanistic and empirical models is promising and will be enhanced in the coming years, due to the increasing availability of free Earth observation data.
High-resolution map of sugarcane cultivation in Brazil using a phenology-based method
Sugarcane is the most important source of sugar, and its cultivation area has undergone rapid expansion, replacing other crops, pastures, and forests. Brazil is the world's largest sugarcane producer and contributed to approximately 38.6 % of the world's total production in 2019. Sugarcane in Brazil can be harvested from April to December in the south-central area and from September to April in the northeast area. The flexible phenology and harvest conditions of sugarcane in Brazil make it difficult to identify the harvest area at state to country scales. In this study, we developed a phenology-based method to identify the harvest area of sugarcane in Brazil by incorporating the multiple phenology conditions into a time-weighted dynamic time warping method (TWDTW). Then, we produced annual 30 m spatial resolution sugarcane harvest maps (2016–2019) for 14 states in Brazil (over 98 % of the harvest area) based on the proposed method using Landsat-7, Landsat-8, and Sentinel-2 optical data. The proposed method performed well in identifying sugarcane harvest area with limited training sample data. Validations for the 2018 harvest year displayed high accuracy, with the user's, producer's, and overall accuracies of 94.35 %, 87.04 %, and 91.47 % in Brazil, respectively. In addition, the identified harvest area of sugarcane exhibited good correlations with the agricultural statistical data provided by the Brazilian Institute of Geography and Statistics (IBGE) at the municipality, microregion, and mesoregion levels. The relationship between user types and gamification designs
Gamification has been discussed as a standout approach to improve user experience, with different studies showing that users can have different preferences over game elements according to their user types. However, relatively less is known how different kinds of users may react to different types of gamification. Therefore, in this study (N=331) we investigate how user orientation (Achiever, Disruptor, Free Spirit, Philanthropist, Player, and Socializer) is associated with the preference for and perceived sense of accomplishment from different gamification designs. Beyond singular associations between the user orientation and the gamification designs, the findings indicate no comprehensive and consistent patterns of associations. From the six user orientations, five presented significant associations: Socializer orientation was positively associated with Social, Fictional, and Personal designs, while negatively associated with Performance design; Player orientation was positively associated with Social (Accomplishment), Personal, and Ecological designs, while negatively associated with the Social design (Preference); Disruptor orientation was positively associated with Social design; Achiever orientation was positively associated with Performance and Social designs; and Free Spirit orientation was negatively associated with Social design. Based on the results, we provide recommendations on how to personalize gamified systems and set further research trajectories on personalized gamification.
Naringenin Nano-Delivery Systems and Their Therapeutic Applications
Naringenin (NRG) is a polyphenolic phytochemical belonging to the class of flavanones and is widely distributed in citrus fruits and some other fruits such as bergamot, tomatoes, cocoa, and cherries. NRG presents several interesting pharmacological properties, such as anti-cancer, anti-oxidant, and anti-inflammatory activities. However, the therapeutic potential of NRG is hampered due to its hydrophobic nature, which leads to poor bioavailability. Here, we review a wide range of nanocarriers that have been used as delivery systems for NRG, including polymeric nanoparticles, micelles, liposomes, solid lipid nanoparticles (SLNs), nanostructured lipid carriers (NLCs), nanosuspensions, and nanoemulsions. These nanomedicine formulations of NRG have been applied as a potential treatment for several diseases, using a wide range of in vitro, ex vivo, and in vivo models and different routes of administration. From this review, it can be concluded that NRG is a potential therapeutic option for the treatment of various diseases such as cancer, neurological disorders, liver diseases, ocular disorders, inflammatory diseases, skin diseases, and diabetes when formulated in the appropriate nanocarriers.
Psychometric investigation of the gamification Hexad user types scale in Brazilian Portuguese
Gamification has become a significant direction in designing technologies, services, products, organizational structures, and any human activities towards being more game-like and consequently being more engaging and motivating. Albeit its success, research indicates that personal differences exist with regards to susceptibility to gamification at large as well as to different types of gamification designs. As a response, models and measurement instruments of user types when it comes to gamification have been developed. One of the most discussed related instruments is the Hexad user types scale. However, there has been paucity of research related to the validity and reliability of the Hexad instrument in general but also of its different formulations and language versions. To face this gap, our study focused on analyzing the psychometric properties of the Hexad scale in Brazilian Portuguese by conducting two confirmatory factor analyses and two multi-group confirmatory factor analyses. The survey was answered by 421 Brazilian respondents (52% self-reported women, 47% self-reported men, 0.5% preferred not to provide their gender, and 0.5% checked the option “other”), from the five Brazilian regions (23 different states and the Federal District), and aged between 10 and 60 years old. Findings support the structural validity of the scale as an oblique model and indicate opportunities for small improvements. Further research, both at academy and practice, may use this study as the source of measurement of user types related to gamification (in Brazilian Portuguese), as well as, as a theoretical and practical source for further studies discussing personalized gamification.
