We report a fatal case of Crimean-Congo hemorrhagic fever in Portugal. An 83-year-old man, initially suspected of having Mediterranean spotted fever, was later confirmed to have Crimean-Congo hemorrhagic fever by the detection of viral genome in the patient's serum and the presence of specific IgM antibodies.

Rhipicephalus bursa Sialotranscriptomic Response to Blood Feeding and Babesia ovis Infection: Identification of Candidate Protective Antigens

Ticks are among the most prevalent blood-feeding arthropods, and they act as vectors and reservoirs for numerous pathogens. Sialotranscriptomic characterizations of tick responses to blood feeding and pathogen infections can offer new insights into the molecular interplay occurring at the tick-host-pathogen interface. In the present study, we aimed to identify and characterize
salivary gland (SG) genes that were differentially expressed in response to blood feeding and
infection. Our experimental approach consisted of RNA sequencing of SG from three different tick samples, fed-infected, fed-uninfected, and unfed-uninfected, for characterization and inter-comparison. Overall, 7,272 expressed sequence tags (ESTs) were constructed from unfed-uninfected, 13,819 ESTs from fed-uninfected, and 15,292 ESTs from fed-infected ticks. Two catalogs of transcripts that were differentially expressed in response to blood feeding and
infection were produced. Four genes coding for a putative vitellogenin-3, lachesin, a glycine rich protein, and a secreted cement protein were selected for RNA interference functional studies. A reduction of 92, 65, and 51% was observed in
, and
mRNA levels in SG, respectively. The
knockdown led to increased tick mortality, with 77% of ticks dying post-infestation. The reduction of the secreted cement protein-mRNA levels resulted in 46% of ticks being incapable of correctly attaching to the host and significantly lower female weights post-feeding in comparison to the control group. The
knockdown resulted in a 70% reduction of the levels associated with
infection in
SG and 70% mortality. These results improved our understanding of the role of tick SG genes in
infection/proliferation and tick feeding. Moreover, lachesin, vitellogenin-3, and secreted cement proteins were validated as candidate protective antigens for the development of novel tick and tick-borne disease control measures.

Trichoderma Species from Soil of Pernambuco State, Brazil

Trichoderma is an important fungal genus, known mainly for its potential for the biological control of phytopathogens. Accurate identification of these fungi is essential for research and applications involving them, to be addressed correctly. The objectives of this study were to isolate, identify, and report the species richness of Trichoderma species that occur in the soil of different regions of Pernambuco, Brazil. DNA sequences of portions of the translation elongation factor 1-α (TEF1) gene region were generated for 56 isolates of Trichoderma, obtained from the Zona da Mata, Agreste, and Sertão regions of Pernambuco. According to the phylogenetic analysis based on these sequences, these fungi belong to two Sections—Trichoderma (35 isolates) and Pachybasidium (21 isolates). These fungi have been resolved in nine species, including Trichoderma afroharzianum, Trichoderma asperelloides, Trichoderma asperellum, Trichoderma koningiopsis, and five possible new species to be confirmed in further studies. This study shows that the soils of Pernambuco host a diversity of Trichoderma species and consequently of biological resources with potential for application in agriculture.

Brain–Computer Interfaces (BCIs) have been regarded as potential tools for individuals with severe motor disabilities, such as those with amyotrophic lateral sclerosis, that render interfaces that rely on movement unusable. This study aims to develop a dependent BCI system for manual end-point control of a robotic arm. A proof-of-concept system was devised using parieto-occipital alpha wave modulation and a cyclic menu with auditory cues. Users choose a movement to be executed and asynchronously stop said action when necessary. Tolerance intervals allowed users to cancel or confirm actions. Eight able-bodied subjects used the system to perform a pick-and-place task. To investigate the potential learning effects, the experiment was conducted twice over the course of two consecutive days. Subjects obtained satisfactory completion rates (84.0 ± 15.0% and 74.4 ± 34.5% for the first and second day, respectively) and high path efficiency (88.9 ± 11.7% and 92.2 ± 9.6%). Subjects took on average 439.7 ± 203.3 s to complete each task, but the robot was only in motion 10% of the time. There was no significant difference in performance between both days. The developed control scheme provided users with intuitive control, but a considerable amount of time is spent waiting for the right target (auditory cue). Implementing other brain signals may increase its speed.

Immobilization of Low-Cost Alternative Vegetable Peroxidase (Raphanus sativus L. peroxidase): Choice of Support/Technique and Characterization

In the present work the radish (Raphanus sativus L.) was used as the low-cost alternative source of peroxidase. The enzyme was immobilized in different supports: coconut fiber (CF), calcium alginate microspheres (CAMs) and silica SBA-15/albumin hybrid (HB). Physical adsorption (PA) and covalent binding (CB) as immobilization techniques were evaluated. Immobilized biocatalysts (IBs) obtained were physicochemical and morphologically characterized by SEM, FTIR and TGA. Also, optimum pH/temperature and operational stability were determined. For all supports, the immobilization by covalent binding provided the higher immobilization efficiencies—immobilization yield (IY%) of 89.99 ± 0.38% and 77.74 ± 0.42% for HB and CF, respectively. For CAMs the activity recovery (AR) was of 11.83 ± 0.68%. All IBs showed optimum pH at 6.0. Regarding optimum temperature of the biocatalysts, HB-CB and CAM-CB maintained the original optimum temperature of the free enzyme (40 °C). HB-CB showed higher operational stability, maintaining around 65% of the initial activity after four consecutive cycles. SEM, FTIR and TGA results suggest the enzyme presence on the IBs. Radish peroxidase immobilized on HB support by covalent binding is promising in future biotechnological applications.

Thousands of people suffer from severe malaria every year. The innate immune response plays a determinant role in host’s defence to malaria. Transient receptor potential vanilloid 1 (TRPV1) modulates macrophage-mediated responses in sepsis, but its role in other pathogenic diseases has never been addressed. We investigated the effects of capsazepine, a TRPV1 antagonist, in malaria. C57BL/6 mice received 105 red blood cells infected with Plasmodium berghei ANKA intraperitoneally. Noninfected mice were used as controls. Capsazepine or vehicle was given intraperitoneally for 6 days. Mice were culled on day 7 after infection and blood and spleen cell phenotype and activation were evaluated. Capsazepine decreased circulating but not spleen F4/80+Ly6G+ cell numbers as well as activation of both F4/80+and F4/80+Ly6G+ cells in infected animals. In addition, capsazepine increased circulating but not spleen GR1+ and natural killer (NK) population, without interfering with natural killer T (NKT) cell numbers and blood NK and NKT activation. However, capsazepine diminished CD69 expression in spleen NKT but not NK cells. Infection increased lipid peroxidation and the release of TNFα and IFNγ, although capsazepine-treated group exhibited lower levels of lipid peroxidation and TNFα. Capsazepine treatment did not affect parasitaemia. Overall, TRPV1 antagonism modulates the innate immune response to malaria.

Etiology and severity of diarrheal diseases in infants at the semiarid region of Brazil: A case-control study

Diarrheal diseases are an important cause of morbidity and mortality among children in developing countries. We aimed to study the etiology and severity of diarrhea in children living in the low-income semiarid region of Brazil.
This is a cross-sectional, age-matched case-control study of diarrhea in children aged 2-36 months from six cities in Brazil's semiarid region. Clinical, epidemiological, and anthropometric data were matched with fecal samples collected for the identification of enteropathogens.
We enrolled 1,200 children, 596 cases and 604 controls. By univariate analysis, eight enteropathogens were associated with diarrhea: Norovirus GII (OR 5.08, 95% CI 2.10, 12.30), Adenovirus (OR 3.79, 95% CI 1.41, 10.23), typical enteropathogenic Escherichia coli (tEPEC), (OR 3.28, 95% CI 1.39, 7.73), enterotoxigenic E. coli (ETEC LT and ST producing toxins), (OR 2.58, 95% CI 0.99, 6.69), rotavirus (OR 1.91, 95% CI 1.20, 3.02), shiga toxin-producing E. coli (STEC; OR 1.77, 95% CI 1.16, 2.69), enteroaggregative E. coli (EAEC), (OR 1.45, 95% CI 1.16, 1.83) and Giardia spp. (OR 1.39, 95% CI 1.05, 1.84). By logistic regression of all enteropathogens, the best predictors of diarrhea were norovirus, adenovirus, rotavirus, STEC, Giardia spp. and EAEC. A high diarrhea severity score was associated with EAEC.
Six enteropathogens: Norovirus, Adenovirus, Rotavirus, STEC, Giardia spp., and EAEC were associated with diarrhea in children from Brazil's semiarid region. EAEC was associated with increased diarrhea severity.

Visceral leishmaniasis (VL) is mainly caused by Leishmania (Leishmania) donovani and Leishmania (L.) infantum; however, other Leishmania species have been associated with VL. We report a case of a patient simultaneously diagnosed with VL caused by Leishmania (L.) amazonensis and Hodgkin’s lymphoma. After treatment with liposomal amphotericin B and chemotherapy, the patient presented a clinical cure. This case report reinforces the hypothesis that other Leishmania species can cause visceral lesions mainly related to immunosuppression.

Campylobacter jejuni virulence genes and immune-inflammatory biomarkers association with growth impairment in children from Northeastern Brazil

Campylobacter spp. have been associated with anthropometric Z-score decrements, but the role of specific virulence genes associated with these outcomes has not been explored. This study aimed to investigate whether specific Campylobacter jejuni virulence-related gene and immune-inflammatory biomarkers are associated with malnutrition in children from Northeastern Brazil. A case-control study was performed in Fortaleza, Brazil. Children aging 6–24 months were characterized as malnourished (cases) if weight-for-age Z-score (WAZ) = 2 and as nourished (controls) if WAZ ≥ 1. DNA samples were extracted from stools and screened for C. jejuni/coli by real-time PCR. A subsequent C. jejuni-specific PCR was employed and positive samples were evaluated for 18 C. jejuni virulence genes by using four multiplex PCRs. C. jejuni was detected in 9.71% (33/340) of the children’s samples, being 63.63% (21/33) from nourished and 37.37% (12/33) from malnourished children. The cadF, iamA, cheW, and sodB genes were the most frequent genes (100%, 90.9%, 87.9%, and 75.8%, respectively), while some others (ceuE, jlpA, pldA, and pVir) showed low rates (all below 6%). Malnourished children were significantly associated with infection with C. jejuni strains lacking cdtB gene (active subunit of cytolethal distending toxin) and harboring flgE gene (flagellar hook protein). These strains were also associated with children presenting increased serum SAA and sCD-14, but decreased IgG anti-LPS. These data reinforce the impact of Campylobacter jejuni infection on children without diarrhea and highlight the contribution of a specific virulence gene profile, cdtB(−)flgE(+) and increased systemic response in malnutrition children.

One of the main constraints hindering the increase of animal production in semi-arid regions of Africa is the inadequate supply of nutrients during the dry season. Incorporation of alternative feed resources in ruminant diets during this period could be a viable approach to overcome these limitations. The objective of this study was to evaluate the nutritive value of muiumba (
Baikiaea plurijuga
) tree seeds as an alternative nutrient source for ruminants. Muiumba seeds were compared to other eight feedstuffs including two cereal grains (corn and oat), two wheat by-products (wheat bran and distilled wheat) and four protein meals (coconut meal, sunflower meal, soybean meal and rapeseed meal) as to its chemical composition,
in vitro
organic matter digestibility (IVOMD) and
in vitro
gas production. The moderate crude protein concentrations (145 g/kg DM) of muiumba seeds indicate that this feedstuff could not be used as a protein supplement, contrarily to the majority of multipurpose tree seeds. Although the starch content was scarce (15 g/kg DM), the low neutral detergent fibre (235 g/kg DM), low molecular weight sugar (76.1 g/kg DM) and non-starch polysaccharide (510.5 g/kg DM) contents indicate that this feedstuff has potential feeding value. This was confirmed by the IVOMD (0.770) and by the data provided by the
in vitro
gas production showing that muiumba seeds had high (
P
< 0.05) maximum gas production and fractional fermentation rates, suggesting that these seeds are characterized by a highly fermentable fraction.
