Amada's Blessings from the Peyote Gardens of South Texas
\"Amada Cardenas, a Mexican American woman from the borderlands of South Texas, played a pivotal role in the little-known history of the peyote trade. She and her husband were the first federally licensed peyote dealers. They began harvesting and selling the sacramental plant to followers of the Native American Church (NAC) in the 1930s, and after her husband's death in the late 1960s Mrs. Cardenas continued to befriend and help generations of NAC members until her death in 2005, just short of her 101st birthday. Author Stacy B. Schaefer, a close friend of Amada, spent thirteen years doing fieldwork with this remarkable woman. Her book weaves together the geography, biology, history, cultures, and religions that created the unique life of Mrs. Cardenas and the people she knew. Schaefer includes their words to help tell the story of how Mexican Americans, Tejanos, gringos, Native Americans, and others were touched and inspired by Amada Cardenas's embodiment of the core NAC values: faith, hope, love, and charity\"-- Provided by publisher.
The bicuspid aortic valve: an integrated phenotypic classification of leaflet morphology and aortic root shape

Objective:To establish a classification of bicuspid aortic valve (BAV) that includes both leaflet morphology and aortic shape.Setting:Two academic medical centres of the University of Washington, Seattle.Patients:191 adult patients with BAV.Interventions:Review of clinical data and transthoracic echocardiograms.Main outcome measures:Assessment of leaflet morphology; valve function; aortic shape and dimensions.Results:We identified three morphologies: type 1, fusion of right and left coronary cusp (n = 152); type 2, right and non-coronary fusion (n = 39); and type 3, left and non-coronary fusion (n = 1). Comparing type 1 and 2 BAV, there were no significant differences in age, height, weight, blood pressure or aortic valve function. Type 1 was more common in men (69 vs 45%). The aortic sinuses were larger in type 1, while type 2 had larger arch dimensions. Myxomatous mitral valves were more common in type 2 BAV (13% vs 2.6%, p<0.05). Three aortic shapes were defined: normal (N), sinus effacement (E), and ascending dilatation (A). Comparing type 1 to type 2 BAV, shape N was more common in type 1 (60% vs 32%), and type A was more common in type 2 (35% vs 54%,); type E was rare (p<0.01 across all groups).Conclusion:A comprehensive BAV phenotype includes aortic shape. Type 1 BAV is associated with male gender and normal aortic shape but a larger sinus diameter. Type 2 leaflet morphology is associated with ascending aorta dilatation , larger arch dimensions and higher prevalence of myxomatous mitral valve disease.

Earth's anomalous middle-age magmatism driven by plate slowdown

The mid-Proterozoic or \"boring billion\" exhibited extremely stable environmental conditions, with little change in atmospheric oxygen levels, and mildly oxygenated shallow oceans. A limited number of passive margins with extremely long lifespans are observed from this time, suggesting that subdued tectonic activity—a plate slowdown—was the underlying reason for the environmental stability. However, the Proterozoic also has a unique magmatic and metamorphic record; massif-type anorthosites and anorogenic Rapakivi granites are largely confined to this period and the temperature/pressure (thermobaric ratio) of granulite facies metamorphism peaked at over 1500 °C/GPa during the Mesoproterozoic. Here, we develop a method of calculating plate velocities from the passive margin record, benchmarked against Phanerozoic tectonic velocities. We then extend this approach to geological observations from the Proterozoic, and provide the first quantitative constraints on Proterozoic plate velocities that substantiate the postulated slowdown. Using mantle evolution models, we calculate the consequences of this slowdown for mantle temperatures, magmatic regimes and metamorphic conditions in the crust. We show that higher mantle temperatures in the Proterozoic would have resulted in a larger proportion of intrusive magmatism, with mantle-derived melts emplaced at the Moho or into the lower crust, enabling the production of anorthosites and Rapakivi granites, and giving rise to extreme thermobaric ratios of crustal metamorphism when plate velocities were slowest.

Spatial variation and antecedent sea surface temperature conditions influence Hawaiian intertidal community structure

Global sea surface temperatures (SSTs) are increasing, and in Hawaiʻi, rates of ocean warming are projected to double by the end of the 21 st century. However, current nearshore warming trends and their possible impacts on intertidal communities are not well understood. This study represents the first investigation into the possible effects of rising SST on intertidal algal and invertebrate communities across the Main Hawaiian Islands (MHI). By utilizing citizen-science data coupled with high-resolution, daily SST satellite measurements from 12 intertidal sites across the MHI from 2004–2019, the response of intertidal algal and invertebrate abundance and community diversity to changes in SST was investigated across multiple spatial scales. Results show high rates of SST warming (0.40°C Decade -1 ) over this study’s timeframe, similar to predicted rates of warming for Hawaiʻi by the end of the 21 st century. Changes in abundance and diversity in response to SST were variable among intertidal sites, but differences in antecedent SST among intertidal sites were significantly associated with community dissimilarity. In addition, a statistically significant positive relationship was found between SST and Simpson’s diversity index, and a significant relationship was also found between SST and the abundance of six dominant taxa. For five of these six dominant taxa, antecedent SSTs over the 6–12 months preceding sampling were the most influential for describing changes to abundance. The increase in community diversity in response to higher SSTs was best explained by temperatures in the 10 months preceding sampling, and the resultant decreased abundance of dominant turf algae. These results highlight rapidly warming nearshore SSTs in Hawaiʻi and the longer-term effects of antecedent SSTs as significant drivers of change within Hawaiian intertidal communities. Therefore, we suggest that future research and management should consider the possibility of lagging effects of antecedent SST on intertidal communities in Hawaiʻi and elsewhere.

GRB 110213A: A Study of Afterglow Electromagnetic Cascade Radiation

We obtained well-sampled optical photometry of GRB 110213A, including Swift/UVOT and XRT. Combining our data from those of other ground-based telescopes, we present 15 optical multicolor light curves showing similar shapes with two peaks. In contrast, in the X-ray band, only a single peak is observed between the two optical peaks. Temporal and spectral analysis of GRB 110213A shows that the X-rays differ from the optical for Phases I–III (before the second peak of the optical band at ∼5.6 ks). Moreover, they have the same spectral behavior at late times (Phases IV–VI). These data indicate that the optical and X-ray emission are dominated by different components. The synchrotron-supported pair cascade emission is included in the standard external forward-shock model, which is dominated by synchrotron radiation and synchrotron self-Compton (SSC). We find that the optical bands of GRB 110213A are dominated by the cascade emission from synchrotron radiation + SSC at the early stage, while the primary synchrotron + SSC radiation dominates the X-ray band. At late stages, both the X-ray and optical bands are dominated by emission from primary synchrotron + SSC radiation. The cascade component can reasonably explain the first optical peak. In contrast, the primary synchrotron + SSC emission mainly contributes to the second peak.

A screening study of the spatial distribution and cumulative toxicity of agricultural pesticides in the European Union’s waters

Pesticides can be an important stressor to aquatic ecosystems, and their use is strictly regulated in the European Union (EU). However, data on the use of pesticides are rather limited and poorly available, and monitoring is often insufficient to characterize their actual exposure and impact. The aim of the work presented here is to harness the limited data available and assess, for the first time, the distribution of concentrations and toxicity of 148 pesticide active substances (AS) for the whole EU. Starting from available estimates of pesticide use in agriculture and a simple screening-level model of their fate and transport, we quantify pesticide concentrations in soil and water. A comparison with monitoring data shows that predicted water concentrations are in plausible orders of magnitude, hence the model can be regarded as a first-approximation representation of the distribution of pesticides in the environment. The toxicity of individual pesticide active substances (AS) is characterized by their concentrations divided by the respective no observed effect concentrations (NOEC) for aquatic organisms, which represents the “toxic units” (TU) of each AS. The cumulative toxicity of pesticides in soils and streams of the EU is obtained by summing the TU of individual AS. We estimate that the toxicity of individual AS is generally well below 0.1 TU, indicating relatively safe environmental exposure. However, the cumulative toxicity of a mixture of AS can exceed 0.1 toxic units (TU) for more than 27% of the length of the EU’s stream network, and 1 TU for more than 4%. The cumulative toxicity at a given location is driven by only a handful of AS, but these differ from site to site reflecting the variability of pesticide use. Still, we estimate that only about 20 AS out of 148 appear among the top contributors to cumulative toxicity in most cases. While our assessment suggests a relatively widespread risk due to pesticide pollution, it also points to the important limitations concerning knowledge of pesticide use and monitoring of pesticide occurrence in the environment. These limitations need to be addressed in order to evaluate more accurately the effectiveness of EU pesticide policies. The assessment represents a proof-of-concept of a method that can be applied in support of the monitoring of pesticide policies implementation in the EU and elsewhere, once pesticide use can be estimated.

Specific sequences are designated for de novo DNA methylation at CpG dinucleotides in mammalian germ cells. The result is the long-term transcriptional silencing of the methylated sequences, most of which are retrotransposons and CpG-rich sequences associated with imprinted genes. There is profound sexual dimorphism in both the nature of the sequences that undergo de novo methylation in germ cells and in the mechanism by which de novo methylation is regulated. The restriction of future gene expression by the imposition of heritable methylation patterns in germ cell genomes is characteristic of mammals but is rare in other taxa.

Radiological Screening Methods in Deceased Organ Donation: An Overview of Guidelines Worldwide

Organ transplantation is performed worldwide, but policies regarding donor imaging are not uniform. An overview of the policies in different regions is missing. This study aims to investigate the various protocols worldwide on imaging in deceased organ donation. An online survey was created to determine the current policies. Competent authorities were approached to fill out the survey based on their current protocols. In total 32 of the 48 countries approached filled out the questionnaire (response rate 67%). In 16% of the countries no abdominal imaging is required prior to procurement. In 50%, abdominal ultrasound (US) is performed to screen the abdomen and in 19% an enhanced abdominal Computed Tomography (CT). In 15% of the countries both an unenhanced abdominal CT scan and abdominal US are performed. In 38% of the countries a chest radiographic (CXR) is performed to screen the thorax, in 28% only a chest CT, and in 34% both are performed. Policies regarding radiologic screening in deceased organ donors show a great variation between different countries. Consensus on which imaging method should be applied is missing. A uniform approach will contribute to quality and safety, justifying (inter)national exchange of organs.

Detection of minimal residual disease (MRD) is the most sensitive method to evaluate treatment response and one of the strongest predictors of outcome in childhood acute lymphoblastic leukemia (ALL). The 10-year update on the I-BFM-SG MRD study 91 demonstrates stable results (event-free survival), that is, standard risk group (MRD-SR) 93%, intermediate risk group (MRD-IR) 74%, and high risk group (MRD-HR) 16%. In multicenter trial AIEOP-BFM ALL 2000, patients were stratified by MRD detection using quantitative PCR after induction (TP1) and consolidation treatment (TP2). From 1 July 2000 to 31 October 2004, PCR target identification was performed in 3341 patients: 2365 (71%) patients had two or more sensitive targets (⩽10
−4
), 671 (20%) patients revealed only one sensitive target, 217 (6%) patients had targets with lower sensitivity, and 88 (3%) patients had no targets. MRD-based risk group assignment was feasible in 2594 (78%) patients: 40% were classified as MRD-SR (two sensitive targets, MRD negativity at both time points), 8% as MRD-HR (MRD ⩾10
−3
at TP2), and 52% as MRD-IR. The remaining 823 patients were stratified according to clinical risk features: HR (

=108) and IR (

=715). In conclusion, MRD-PCR-based stratification using stringent criteria is feasible in almost 80% of patients in an international multicenter trial.

Organic chemicals jeopardize the health of freshwater ecosystems on the continental scale

Organic chemicals can contribute to local and regional losses of freshwater biodiversity and ecosystem services. However, their overall relevance regarding larger spatial scales remains unknown. Here, we present, to our knowledge, the first risk assessment of organic chemicals on the continental scale comprising 4,000 European monitoring sites. Organic chemicals were likely to exert acute lethal and chronic long-term effects on sensitive fish, invertebrate, or algae species in 14% and 42% of the sites, respectively. Of the 223 chemicals monitored, pesticides, tributyltin, polycyclic aromatic hydrocarbons, and brominated flame retardants were the major contributors to the chemical risk. Their presence was related to agricultural and urban areas in the upstream catchment. The risk of potential acute lethal and chronic long-term effects increased with the number of ecotoxicologically relevant chemicals analyzed at each site. As most monitoring programs considered in this study only included a subset of these chemicals, our assessment likely underestimates the actual risk. Increasing chemical risk was associated with deterioration in the quality status of fish and invertebrate communities. Our results clearly indicate that chemical pollution is a large-scale environmental problem and requires far-reaching, holistic mitigation measures to preserve and restore ecosystem health.
