Assessment of permafrost distribution maps in the Hindu Kush Himalayan region using rock glaciers mapped in Google Earth

The extent and distribution of permafrost in the mountainous parts of the Hindu Kush Himalayan (HKH) region are largely unknown. A long tradition of permafrost research, predominantly on rather gentle relief, exists only on the Tibetan Plateau. Two permafrost maps are available digitally that cover the HKH and provide estimates of permafrost extent, i.e., the areal proportion of permafrost: the manually delineated Circum-Arctic Map of Permafrost and Ground Ice Conditions (Brown et al., 1998) and the Global Permafrost Zonation Index, based on a computer model (Gruber, 2012). This article provides a first-order assessment of these permafrost maps in the HKH region based on the mapping of rock glaciers. Rock glaciers were used as a proxy, because they are visual indicators of permafrost, can occur near the lowermost regional occurrence of permafrost in mountains, and can be delineated based on high-resolution remote sensing imagery freely available on Google Earth. For the mapping, 4000 square samples (~ 30 km2) were randomly distributed over the HKH region. Every sample was investigated and rock glaciers were mapped by two independent researchers following precise mapping instructions. Samples with insufficient image quality were recorded but not mapped. We use the mapping of rock glaciers in Google Earth as first-order evidence for permafrost in mountain areas with severely limited ground truth. The minimum elevation of rock glaciers varies between 3500 and 5500 m a.s.l. within the region. The Circum-Arctic Map of Permafrost and Ground Ice Conditions does not reproduce mapped conditions in the HKH region adequately, whereas the Global Permafrost Zonation Index does so with more success. Based on this study, the Permafrost Zonation Index is inferred to be a reasonable first-order prediction of permafrost in the HKH. In the central part of the region a considerable deviation exists that needs further investigations.

Inferring snowpack ripening and melt-out from distributed measurements of near-surface ground temperatures

Seasonal snow cover and its melt regime are heterogeneous both in time and space. Describing and modelling this variability is important because it affects diverse phenomena such as runoff, ground temperatures or slope movements. This study presents the derivation of melting characteristics based on spatial clusters of ground surface temperature (GST) measurements. Results are based on data from Switzerland where ground surface temperatures were measured with miniature loggers (iButtons) at 40 locations referred to as footprints. At each footprint, up to ten iButtons have been distributed randomly over an area of 10 m × 10 m, placed a few cm below the ground surface. Footprints span elevations of 2100–3300 m a.s.l. and slope angles of 0–55°, as well as diverse slope expositions and types of surface cover and ground material. Based on two years of temperature data, the basal ripening date and the melt-out date are determined for each iButton, aggregated to the footprint level and further analysed. The melt-out date could be derived for nearly all iButtons; the ripening date could be extracted for only approximately half of them because its detection based on GST requires ground freezing below the snowpack. The variability within a footprint is often considerable and one to three weeks difference between melting or ripening of the points in one footprint is not uncommon. The correlation of mean annual ground surface temperatures, ripening date and melt-out date is moderate, suggesting that these metrics are useful for model evaluation.

Vacuum mattress or long spine board: which method of spinal stabilisation in trauma patients is more time consuming? A simulation study

Background
Spinal stabilisation is recommended for prehospital trauma treatment. In Germany, vacuum mattresses are traditionally used for spinal stabilisation, whereas in anglo-american countries, long spine boards are preferred. While it is recommended that the on-scene time is as short as possible, even less than 10 minutes for unstable patients, spinal stabilisation is a time-consuming procedure. For this reason, the time needed for spinal stabilisation may prevent the on-scene time from being brief. The aim of this simulation study was to compare the time required for spinal stabilisation between a scoop stretcher in conjunction with a vacuum mattress and a long spine board.
Methods
Medical personnel of different professions were asked to perform spinal immobilizations with both methods. A total of 172 volunteers were immobilized under ideal conditions as well as under realistic conditions. A vacuum mattress was used for 78 spinal stabilisations, and a long spinal board was used for 94. The duration of the procedures were measured by video analysis.
Results
Under ideal conditions, spinal stabilisation on a vacuum mattress and a spine board required 254.4 s (95 % CI 235.6–273.2 s) and 83.4 s (95 % CI 77.5–89.3 s), respectively (
p
< 0.01). Under realistic conditions, the vacuum mattress and spine board required 358.3 s (95 % CI 316.0–400.6 s) and 112.6 s (95 % CI 102.6–122.6 s), respectively (
p
< 0.01).
Conclusions
Spinal stabilisation for trauma patients is significantly more time consuming on a vacuum mattress than on a long spine board. Considering that the prehospital time of EMS should not exceed 60 minutes and the on-scene time should not exceed 30 minutes or even 10 minutes if the patient is in extremis, based on our results, spinal stabilisation on a vacuum mattress may consume more than 20 % of the recommended on-scene time. In contrast, stabilisation on a spine board requires only one third of the time required for that on a vacuum mattress.
We conclude that a long spine board may be feasible for spinal stabilisation for critical trauma patients with timesensitive life threatening ABCDE-problems to ensure the shortest possible on-scene time for prehospital trauma treatment, not least if a patient has to be rescued from an open or inaccessible terrain, especially that with uneven overgrown land.

Water uptake by biomass burning aerosol at sub- and supersaturated conditions: closure studies and implications for the role of organics

We investigate the CCN activity of freshly emitted biomass burning particles and their hygroscopic growth at a relative humidity (RH) of 85%. The particles were produced in the Mainz combustion laboratory by controlled burning of various wood types. The water uptake at sub- and supersaturations is parameterized by the hygroscopicity parameter, κ (c.f. Petters and Kreidenweis, 2007). For the wood burns, κ is low, generally around 0.06. The main emphasis of this study is a comparison of κ derived from measurements at sub- and supersaturated conditions (κG and κCCN), in order to see whether the water uptake at 85% RH can predict the CCN properties of the biomass burning particles. Differences in κGand κCCN can arise through solution non-idealities, the presence of slightly soluble or surface active compounds, or non-spherical particle shape. We find that κG and κCCN agree within experimental uncertainties (of around 30%) for particle sizes of 100 and 150 nm; only for 50 nm particles is κCCN larger than κG by a factor of 2. The magnitude of this difference and its dependence on particle size is consistent with the presence of surface active organic compounds. These compounds mainly facilitate the CCN activation of small particles, which form the most concentrated solution droplets at the point of activation. The 50 nm particles, however, are only activated at supersaturations higher than 1% and are therefore of minor importance as CCN in ambient clouds. By comparison with the actual chemical composition of the biomass burning particles, we estimate that the hygroscopicity of the water-soluble organic carbon (WSOC) fraction can be represented by a κWSOC value of approximately 0.2. The effective hygroscopicity of a typical wood burning particle can therefore be represented by a linear mixture of an inorganic component with κ ≅ 0.6, a WSOC component with κ ≅ 0.2, and an insoluble component with κ = 0.

Aerosol optical properties observed during Campaign of Air Quality Research in Beijing 2006 (CAREBeijing-2006): Characteristic differences between the inflow and outflow of Beijing city air

Ground‐based measurements of aerosol optical properties were conducted during Campaign of Air Quality Research in Beijing 2006 (CAREBeijing‐2006) (11 August to 9 September 2006) at a suburban site ∼30 km south of Beijing. Averaged over the measurement campaign (arithmetic mean ± standard deviation), the total scattering coefficients (σs) were 469 ± 374 Mm−1 (450 nm), 361 ± 295 Mm−1 (550 nm), and 249 ± 206 Mm−1 (700 nm) and the absorption coefficient (σa) was 51.8 ± 36.5 Mm−1 (532 nm). The average Ångström exponent was 1.42 ± 0.19 (450 nm/700 nm) and the average single scattering albedo (ω532) was 0.86 ± 0.07 (532 nm) with minimum values as low as 0.5. Pronounced diurnal cycles were observed in σs, σa, and ω532 and can be explained by boundary layer mixing effects. Additionally, an enhancement of absorbing particles in the early morning (0500–0800 local time) was observed; this may be attributed to soot emissions from traffic activity. When the measured air masses originated in the north and passed over Beijing, the single scattering albedo was generally low (ω532 < 0.8), which indicates that the local emissions of particulate matter in Beijing were dominated by primary particles from combustion sources (soot). The southerly inflow to Beijing had typically very high σs and higher than average ω532 values, suggesting a large amount of secondary aerosol (e.g., sulfate and oxidized organics). Overall, the results suggest that a majority of the particle pollution in Beijing is transported into the city from the south.

Discovering a New Metabolic Pathway. Early Work with My Friend, Viswanathan Natarajan

This article summarizes our early work with Viswanathan Natarajan in the 1980s at the University of Minnesota’s Hormel Institute, when he was at the beginning of his brilliant academic career. At that time most metabolic pathways for the biosynthesis and degradation of phospholipids were well established and known in considerable detail. Hence, it was exciting to discover a novel sequence of biochemical reactions, first in dog heart and later in various other vertebrate cells and tissues that became known as the transacylation-phosphodiesterase pathway of phospholipid metabolism. Because one of the metabolites, N-arachidonoylethanolamine, produced by this reaction sequence, was later found to bind to and activate cannabinoid receptors, investigations of this pathway became part of the rapidly growing field of endocannabinoid research. This is briefly summarized here as well.

The focus on animal welfare in society has increased during the last 50 years. Animal welfare legislation and private standards have developed, and today many farmers within animal production have both governmental legislation and private standards to comply with. In this paper intentions and values are described that were expressed in 14 animal welfare legislation and standards in four European countries; Sweden, United Kingdom, Germany and Spain. It is also discussed if the legislation and standards actually accomplish what they, in their overall description and ethics, claimed to do, i.e. if this is followed up by relevant paragraphs in the actual body of the text in the legislation and standards respectively. The method used was an on-line questionnaire from the EconWelfare research project and text analyses. This study shows that the ethical values within a set of legislation or private standards are not always consistently implemented, and it is not always possible to follow a thread between the intentions mentioned initially and the actual details of the legislation or standard. Since values will differ so will also the animal welfare levels and the implications of similar concepts in the regulations. In general, the regulations described were not based on animal welfare considerations only, but also other aspects, such as food safety, meat quality, environmental aspects and socio-economic aspects were taken into account. This is understandable, but creates a gap between explicit and implicit values, which we argue, can be overcome if such considerations are made more transparent to the citizens/consumers.

Effects of cortisol administration on craving in heroin addicts

Heroin dependence is a severe and chronically relapsing substance use disorder with limited treatment options. Stress is known to increase craving and drug-taking behavior, but it is not known whether the stress hormone cortisol mediates these stress effects or whether cortisol may rather reduce craving, for example, by interfering with addiction memory. The aim of the present study was to determine the effects of cortisol administration on craving in heroin-dependent patients and to determine whether the effects depend on the daily dose of heroin consumption. We used a double-blind, placebo-controlled, cross-over study in 29 heroin-dependent patients in a stable heroin-assisted treatment setting. A single oral dose of 20 mg of cortisol or placebo was administered 105 min before the daily heroin administration. The primary outcome measure was cortisol-induced change in craving. Secondary measures included anxiety, anger and withdrawal symptoms. For the visual analog scale for craving, we found a significant interaction (
P
=0.0027) between study medication and heroin-dose group (that is, daily low, medium or high dose of heroin). Cortisol administration reduced craving in patients receiving a low dose of heroin (before heroin administration:

=0.0019; after heroin administration:

=0.0074), but not in patients receiving a medium or high dose of heroin. In a picture-rating task with drug-related pictures, cortisol administration did not affect the ratings for the picture-characteristic craving in all the three heroin-dose groups. Cortisol also did not significantly affect secondary outcome measures. In conclusion, a single administration of cortisol leads to reduced craving in low-dose heroin addicts. The present findings might have important clinical implications with regard to understanding stress effects and regarding treatment of addiction.

Cancer associated fibroblasts (CAFs) play a critical role for growth, invasion, and metastasis of cancer. Therefore, targeting CAFs with small molecule inhibitors may be an attractive anti-tumor strategy. The current study aims to identify small molecule kinase inhibitors affecting CAF's growth and to characterize the biological effects of active compounds on primary CAFs from lung cancer. We screened two individual CAF strains for their sensitivity to a panel of 160 kinase inhibitors. Five kinase inhibitors were identified inhibiting more than 50% of the growth of both cell lines. Three of them were inhibitors of PDGFR at nanomolar concentrations. Therefore, we further tested the FDA approved PDGFR inhibitors Dasatinib, Nilotinib, Sorafenib, and Imatinib. All 37 CAF strains investigated were highly sensitive to Dasatinib at clinically relevant concentrations. Imatinib was slightly less effective, whereas the inhibitory effects of Nilotinib and Sorafenib were significantly less pronounced.
We investigated the effect of Dasatinib on the CAF transcriptome by microarray analysis of 9 individual CAF strains. 492 genes were identified whose expression was changed at least twofold. 104 of these encoded cell cycle related proteins with 97 of them being downregulated by Dasatinib. The majority of regulated genes, however, were of diverse biological functions not directly related to proliferation. We compared this Dasatinib expression signature to previously described differential signatures of normal tissue associated fibroblasts (NAFs) and CAFs and to a signature of fibroblast serum response. There was a significant overlap between genes regulated by Dasatinib and serum repression genes. More importantly, of the 313 genes downregulated by Dasatinib 64 were also reduced in NAFs compared to CAFs. Furthermore, 26 of 179 genes identified as upregulated by Dasatinib were also found to be elevated in NAFs compared to CAFs. These data demonstrate that Dasatinib partially reverses the phenotype of CAFs to a normal fibroblast like phenotype. This is further supported by the finding that incubation of tumor cells with conditioned medium from CAFs pre-incubated with Dasatinib significantly reduced tumor cell proliferation, suggesting that Dasatinib partially reverses the CAF mediated tumor promoting effect. Therefore, targeting CAFs with Dasatinib represents a promising therapeutic principle.
