Bio-upcycling of plastics is an emerging alternative process that focuses on extracting value from a wide range of plastic waste streams. Such streams are typically too contaminated to be effectively processed using traditional recycling technologies. Medium-chain-length (mcl) diols and dicarboxylates (DCA) are major products of chemically or enzymatically depolymerized plastics, such as polyesters or polyethers. In this study, we enabled the efficient metabolism of mcl-diols and -DCA in engineered
Pseudomonas putida
as a prerequisite for subsequent bio-upcycling. We identified the transcriptional regulator GcdR as target for enabling metabolism of uneven mcl-DCA such as pimelate, and uncovered amino acid substitutions that lead to an increased coupling between the heterologous β-oxidation of mcl-DCA and the native degradation of short-chain-length DCA. Adaptive laboratory evolution and subsequent reverse engineering unravelled two distinct pathways for mcl-diol metabolism in
P. putida
, namely
via
the hydroxy acid and subsequent native β-oxidation or
via
full oxidation to the dicarboxylic acid that is further metabolized by heterologous β-oxidation. Furthermore, we demonstrated the production of polyhydroxyalkanoates from mcl-diols and -DCA by a single strain combining all required metabolic features. Overall, this study provides a powerful platform strain for the bio-upcycling of complex plastic hydrolysates to polyhydroxyalkanoates and leads the path for future yield optimizations.

Reference Architecture for Cross-Company Electronic Collaboration

Cross-organizational electronic collaboration is about to gain significant momentum and facilitates the emergence of a globally networked information service economy. However, existing solutions for the realization of such business relationships still exhibit weaknesses with respect to both managerial and technological aspects. In this work, we propose a service-oriented reference architecture for electronic business media that overcomes the drawbacks of today’s business-to-business (B2B) software products and services. Based on the St. Gallen media reference model, this reference architecture incorporates the design principle of modularity that proved critical for the success of numerous artifacts in other more mature industries. In particular, we investigate and revisit the principle of modularity with respect to its role in the computer industry and transfer it to the context of organizing and implementing electronic cross-company collaboration. On the basis of a case study in the field of public administration in Switzerland, we show its real-world applicability and its improvement potential.

A complete introduction tocar-to-X communications networking Automotive Inter-networking will introduce a range of new network and system technologies for vehicle safety, entertainment and comfort systems currently being researched and developed. C2X networking is not only a matter of technology, but is also very closely related to policy-making about deployment. This book will provide the background on technical developments but will also discuss the potential benefits, costs and risks. Also discussed will be concepts related to application of vehicle-to-vehicle and vehicle-to-infrastructure communication technologies for various purposes such as automobile safety enhancement, vehicle user applications for comfort and convenience and efficiency along with other potential commercial applications. The adoption of advanced integration technologies that enable private and public organizations to seamlessly execute their business transactions electronically is still relatively low, especially among governmental bodies and Small and Medium-sized Enterprises (SMEs). Current solutions often lack a common understanding of the underlying business document semantics and most existing approaches are not able to cope with the huge variety of business document formats, stemming from highly diverse requirements of the different stakeholders. Developed and applied in the course of the EU-funded research project GENESIS, this paper presents a comprehensive core component-based business document modelling approach that builds upon existing standards such as the OASIS Universal Business Language (UBL) and the UN/CEFACT Core Component Technical Specification (CCTS). These standards are extended by introducing the concept of generic business document templates out of which specific documents can be derived according to the actual user's needs. Key principle to achieve this flexibility is the integration of business context information that allows for modelling standard-based but at the same time customized business documents. The resulting modelling framework ranges from (tool-supported) graphical data models to the technical representation of the business documents as XML schema documents designed in compliance with the UN/CEFACT XML schema Naming and Design Rules (NDR).
