Environmental monitoring in the Mar Grande basin (Ionian Sea, Southern Italy)

Hydrodynamic and water quality data has been recorded since February 2014 by a meteo-oceanographic station installed in the inner part of the Gulf of Taranto, in the northeastern part of the Ionian Sea (Southern Italy). This monitoring action, managed by the research unit of the Technical University of Bari, DICATECh Department, could play a pivotal role in a vulnerable and sensitive area, affected by massive chemical and biological pollutant discharges due to the presence of heavy industry and intense maritime traffic. Monthly trends of winds, waves, currents, and biochemical parameters, such as dissolved oxygen, chlorophyll, and turbidity, are analyzed and discussed. The analysis exhibits that the wave regime is slightly controlled by wind forcing; rather, topography strongly affects the wave propagation direction. Surface currents appear wind induced in the measuring station, while near the bottom a quasi-steady current directed towards southwest is formed. The selected water quality indicators show monthly trends consistent with the typical seasonal convective fluxes and mixing.

Machine learning methods to predict outcomes of pharmacological treatment in psychosis

In recent years, machine learning (ML) has been a promising approach in the research of treatment outcome prediction in psychosis. In this study, we reviewed ML studies using different neuroimaging, neurophysiological, genetic, and clinical features to predict antipsychotic treatment outcomes in patients at different stages of schizophrenia. Literature available on PubMed until March 2022 was reviewed. Overall, 28 studies were included, among them 23 using a single-modality approach and 5 combining data from multiple modalities. The majority of included studies considered structural and functional neuroimaging biomarkers as predictive features used in ML models. Specifically, functional magnetic resonance imaging (fMRI) features contributed to antipsychotic treatment response prediction of psychosis with good accuracies. Additionally, several studies found that ML models based on clinical features might present adequate predictive ability. Importantly, by examining the additive effects of combining features, the predictive value might be improved by applying multimodal ML approaches. However, most of the included studies presented several limitations, such as small sample sizes and a lack of replication tests. Moreover, considerable clinical and analytical heterogeneity among included studies posed a challenge in synthesizing findings and generating robust overall conclusions. Despite the complexity and heterogeneity of methodology, prognostic features, clinical presentation, and treatment approaches, studies included in this review suggest that ML tools may have the potential to predict treatment outcomes of psychosis accurately. Future studies need to focus on refining feature characterization, validating prediction models, and evaluate their translation in real-world clinical practice.

Monitoring Systems and Numerical Models to Study Coastal Sites

The present work aims at illustrating how the joint use of monitoring data and numerical models can be beneficial in understanding coastal processes. In the first part, we show and discuss an annual dataset provided by a monitoring system installed in a vulnerable coastal basin located in Southern Italy, subjected to human and industrial pressures. The collected data have been processed and analysed to detect the temporal evolution of the most representative parameters of the inspected site and have been compared with recordings from previous years to investigate recursive trends. In the second part, to demonstrate to what extent such type of monitoring actions is necessary and useful, the same data have been used to calibrate and run a 3D hydrodynamic model. After this, a reliable circulation pattern in the basin has been reproduced. Successively, an oil pollution transport model has been added to the hydrodynamic model, with the aim to present the response of the basin to some hypothetical cases of oil spills, caused by a ship failure. It is evident that the profitable prediction of the hydrodynamic processes and the transport and dispersion of contaminants strictly depends on the quality and reliability of the input data as well as on the calibration made.

MUC1 Expression Affects the Immunoflogosis in Renal Cell Carcinoma Microenvironment through Complement System Activation and Immune Infiltrate Modulation

Mucin1 (MUC1), a glycoprotein associated with an aggressive cancer phenotype and chemoresistance, is aberrantly overexpressed in a subset of clear cell renal cell carcinoma (ccRCC). Recent studies suggest that MUC1 plays a role in modulating cancer cell metabolism, but its role in regulating immunoflogosis in the tumor microenvironment remains poorly understood. In a previous study, we showed that pentraxin-3 (PTX3) can affect the immunoflogosis in the ccRCC microenvironment by activating the classical pathway of the complement system (C1q) and releasing proangiogenic factors (C3a, C5a). In this scenario, we evaluated the PTX3 expression and analyzed the potential role of complement system activation on tumor site and immune microenvironment modulation, stratifying samples in tumors with high (MUC1H) versus tumors with low MUC1 expression (MUC1L). We found that PTX3 tissue expression was significantly higher in MUC1H ccRCC. In addition, C1q deposition and the expressions of CD59, C3aR, and C5aR were extensively present in MUC1H ccRCC tissue samples and colocalized with PTX3. Finally, MUC1 expression was associated with an increased number of infiltrating mast cells, M2-macrophage, and IDO1+ cells, and a reduced number of CD8+ T cells. Taken together, our results suggest that expression of MUC1 can modulate the immunoflogosis in the ccRCC microenvironment by activating the classical pathway of the complement system and regulating the immune infiltrate, promoting an immune-silent microenvironment.

Correlation between Inflammatory Markers of Atherosclerosis and Carotid Intima-Media Thickness in Obstructive Sleep Apnea

Obstructive Sleep Apnea (OSA) is a sleep-related breathing disorder associated with the development of cardiovascular diseases and atherosclerosis. Systemic inflammation plays an important role in the development of cardiovascular complications in OSA patients. The aim of the study was to evaluate the relationship between carotid intima-media thickness (cIMT) and inflammatory markers plasma levels in OSA patients. We enrolled 80 OSA patients and 40 controls matched for age and body mass index (BMI). The presence and severity of sleep apnea was determined by in-laboratory portable monitoring (PM). Demographic data, blood pressure, heart rate, and cIMT were measured. High-sensitive C-Reactive Protein (hsCRP), interleukin (IL)-6, tumor necrosis factor (TNF)-α and pentraxin (PTX)-3 serum concentrations were detected. cIMT was higher in OSA patients than controls (0.89 ± 0.13 mm vs. 0.65 ± 0.1 mm, p < 0.01). Moderate-severe OSA patients (0.95 ± 0.09 mm) had significantly increased cIMT than mild OSA (0.76 ± 0.1 mm; p < 0.01) and control (0.65 ± 0.1 mm; p < 0.01). hsCRP, IL-6, TNF-α, and PTX-3 in patients with OSA (1.67 ± 0.66 mg/L, 2.86 ± 1.39 pg/mL, 20.09 ± 5.39 pg/mL, 2.1 ± 0.59 ng/mL, respectively) were significantly higher than in controls (1.08 ± 0.53 mg/L, p < 0.01; 1.5 ± 0.67 pg/mL, p < 0.01; 12.53 ± 3.48 pg/mL, p < 0.01; 1.45 ± 0.41 ng/mL, p < 0.01, respectively). Carotid IMT was significantly correlated to CRP (r = 0.44; p < 0.01), IL-6 (r = 0.42; p < 0.01), TNF-α (r = 0.53; p < 0.01), and PTX-3 (r = 0.49; p < 0.01). OSA patients showed increased cIMT, CRP, IL-6, TNF-α, and PTX-3 levels. Inflammatory markers levels are correlated to cIMT in OSA patients.

Childhood leukemia survivors are at risk of long-term complications. Data on bone remodeling in childhood acute lymphoblastic leukemia (ALL) are limited. This 2-year prospective longitudinal study investigated bone remodeling and bone turnover markers at diagnosis, during treatment, and until stopping treatment, in ALL patients < 18 years, to clarify the influence of leukemia itself and/or chemotherapy on bone. Methods: A total of 22 ALL children (12 males, age 5.5 ± 3.6 years) underwent blood sampling at the 5 time point (T0−T4). Osteoprotegerin (OPG), receptor-activator-NF-B-ligand (RANKL), osteocalcin (OC), C-terminal-telopeptide-type-I-collagen (CTX), bone-alkaline-phosphatase (bALP), tartrate-resistant acid-phosphatase-5b (TRACP5b), procollagen-type-I-N-terminal-propeptide (P1NP), Dickkopf-1 (DKK-1), and sclerostin were assessed. Data from patients at T0 were compared to a control group of healthy children. We used the principal component analysis (PCA) for statistics. Results: Levels of CTX, OC, P1NP, and bALP resulted lower in ALL children than controls (p = 0.009 for CTX and p < 0.001 for the others), also DKK1 and sclerostin (p < 0.0001 and p = 0.023). RANKL ed OPG were higher in patients. During T0−T4, CTX, OC, P1NP, TRACP5b, and bALP showed a significant increase, in particular at T0−T1 (end-of-induction). Less evident changes were detected onwards. Conclusions: The onset of leukemia has been revealed as a key point in determining a slowing of bone remodeling in ALL children.

Characteristics of breaking vorticity in spilling and plunging waves investigated numerically by SPH

The present paper, places emphasis on the vorticity induced by wave breaking, which greatly contributes to sediments pick up and suspension as well as to air–water exchange at the wave interface, thus deserving a thorough study. A weakly-compressible smoothed particle (WCSPH) model, coupled with a two-equation model for turbulent stresses, has been employed for this scope. A careful calibration of the SPH’s numerical parameters has been first performed, based on experiments carried out in a sloped wave channel, specifically using wave elevation and velocity data. Once proved the reliable performance of the model, the characteristics of vorticity induced just prior and post breaking for both the cases of a spilling and a plunging wave have been numerically studied. The main and detailed results indicate that for both the types of breakers there is a cause-effect relation observed between the stream wise flow deceleration and the vorticity generation.

Prevalence of autistic traits and their relationships with other psychopathological domains in young adults seeking psychiatric attention: a cluster analysis

Nearly two-thirds of individuals with a mental disorder start experiencing symptoms during adolescence or early adulthood, and the onset of a mental disorder during this critical life stage strongly predicts adverse socioeconomic and health outcomes. Subthreshold manifestations of autism spectrum disorders (ASDs), also called autistic traits (ATs), are known to be associated with a higher vulnerability to the development of other psychiatric disorders. This study aimed to assess the presence of ATs in a population of young adults seeking specialist assistance and to evaluate the study population across various psychopathological domains in order to determine their links with ATs.
We recruited a sample of 263 adolescents and young adults referring to a specialized outpatient clinic, and we administered them several self-report questionnaires for the evaluation of various psychopathological domains. We conducted a cluster analysis based on the prevalence of ATs, empathy, and sensory sensitivity scores.
The cluster analysis identified three distinct groups in the sample: an AT cluster (22.43%), an intermediate cluster (45.25%), and a no-AT cluster (32.32%). Moreover, subjects with higher ATs exhibited greater symptomatology across multiple domains, including mood, anxiety, eating disorder severity, psychotic symptoms, and personality traits such as detachment and vulnerable narcissism.
This study highlights the importance of identifying ATs in young individuals struggling with mental health concerns. Additionally, our findings underscore the necessity of adopting a dimensional approach to psychopathology to better understand the complex interplay of symptoms and facilitate tailored interventions.

How vegetation in flows modifies the turbulent mixing and spreading of jets

While studies on vegetated channel flows have been developed in many research centers, studies on jets interacting with vegetation are still rare. This study presents and analyzes turbulent jets issued into an obstructed cross-flow, with emergent vegetation simulated with a regular array of cylinders. The paper presents estimates of the turbulence diffusion coefficients and the main turbulence variables of jets issued into a vegetated channel flow. The experimental results are compared with jets issued into unobstructed cross-flow. In the presence of the cylinder array, the turbulence length-scales in the streamwise and transverse directions were reduced, relative to the unobstructed crossflow. This contributed to a reduction in streamwise turbulent diffusion, relative to the unobstructed conditions. In contrast, the transverse turbulent diffusion was enhanced, despite the reduction in length-scale, due to enhanced turbulent intensity and the transverse deflection of flow around individual cylinders. Importantly, in the obstructed condition, the streamwise and transverse turbulent diffusion coefficients are of the same order of magnitude.

Hydrodynamic Structure with Scour Hole Downstream of Bed Sills

Experimental turbulence measurements of scour hole downstream of bed sills in alluvial channels with non-cohesive sediments are investigated. Using an Acoustic Doppler Velocimeter (ADV), the flow velocity-field within the equilibrium scour hole was comprehensively measured. In this study, we especially focus on the flow hydrodynamic structure in the scour hole at equilibrium. In addition to the flow velocity distribution in the equilibrium scour hole, the turbulence intensities, the Reynolds shear stresses, the turbulent kinetic energy, and the turbulent length scales are analyzed. Since the prediction of equilibrium scour features is always very uncertain, in this study and based on laboratory turbulence measurements, we apply the phenomenological theory of turbulence to predict the maximum equilibrium scour depth. With this approach, we obtain a new scaling of the maximum scour depth at equilibrium, which is validated using experimental data, satisfying the validity of a spectral exponent equal to −5/3. The proposed scaling shows a quite reasonable accuracy in predicting the equilibrium scour depth in different hydraulic structures.
