Exploring the Mumps Virus Glycoproteins: A Review
The resurgence of mumps in vaccinated adult populations has raised concerns about possible waning vaccine immunity or a potential lack of protection to the circulating strain. A number of individual studies have investigated if there are amino acid variations between the circulating wild-type strains and vaccine strains. In these studies, the HN and F mumps surface glycoproteins have been of interest, because of their role in viral infection, and because the HN protein is the target of neutralizing antibodies. Here, we summarize the single nucleotide variants and their potential effect that have been identified between mumps genotypes in the HN and F proteins.
Acute kidney injury in birth asphyxiated patients: A cross sectional study at Bahawal Victoria Hospital
Background: The presence of perinatal asphyxia and its severity appear to correlate with increasing incidence of Acute kidney injury (AKI). The objective of this study is to determine the frequency of AKI and its outcome in birth asphyxia. Methods: This cross-sectional study was carried out in the Department of Pediatric Medicine from March 2019 to September 2019. A total of 111 newborns with birth asphyxia of gestational age 37-41 weeks were included. Neonates born to mothers having hypertension and diabetes mellitus, patients with congenital kidney anomalies like polycystic kidney disease and renal agenesis, and mothers taking nephrotoxic drugs or any other known cause of AKI like hypovolemic shock were excluded. Urine output (UOP) and final outcome of the patient were also noted. AKI was noted. Results: The mean gestational age was 38.29 ± 1.07 weeks. The mean weight of neonates was 3.08 ± 0.31 kg. The frequency of AKI in birth asphyxia was 20 (18.02%) neonates. Complete recovery in AKI patients was seen in 07 (35.0%) and death in 13 (65.0%) patients. Conclusion: This study has shown that the frequency of AKI in birth asphyxia was found in 18.02% neonates with complete recovery seen in 35.0% and death in 65.0% patients.
Comparative Teratogenic and Skin whitening effect of Herbal extract Glycyrrhiza glabra and Citrus sineus on Zebrafish (Danio rerio)
Objective: To determine acute toxicity and Synergistic effect of the anti-melanogenic activity of methanolic extract Glycyrrhiza glabra and Citrus sineus on zebrafish. Materials and method: Fish embryo toxicity (FET) was studied as per OECD guidelines 236. The LC50 and maximum non-lethal concentration of both the extracts were calculated. The images of zebrafish larvae exposed to different concentrations of methanolic extracts and their combination were taken by using labomed digital microscope. The skin whitening effect of extract was studied by measuring integrated density of images by using imageJ software Result: LC50 Value and maximum non-lethal concentration of methanolic extracts of Glycyrrhiza glabra and Citrus sineus were found to be 56.23ppm and 34.7ppm respectively. The result revealed significant reduction in percentage hatchability (p < 0.0014 for glycyrrhiza glabra and p<0.0011 for Citrus sinenus), dose dependant increased in percentage mortality and different developmental morphological abnormalities in zebrafish embryos exposed to the methanolic extracts Glycyrrhiza glabra and Citrus sineus. The synergistic effect of herbal extracts was found to be more effective in reducing the melanin content. Conclusion: Very High concentration of Glycyrrhiza glabra and Citrus Sineus showed teratogenic effect in zebrafish embryoes. However, based on OECD aquatic toxicity classification, both the extracts were found to be safe. Our study also revealed synergestic skin whitening effect of both the extracts in zebrafish larvae which might be due to reduction of melatonin content by these extracts.
Safety and Outcomes of Permanent and Retrievable Inferior Vena Cava Filters in the Oncology Population
Background. The role for inferior vena cava (IVC) filters in the oncology population is poorly defined. Objectives. Our primary endpoint was to determine the rate of filter placement in cancer patients without an absolute contraindication to anticoagulation and the rate of recurrent VTE after filter placement in both retrievable and permanent filter groups. Patients/Methods. A single-institution, retrospective study of patients with active malignancies and acute VTE who received a retrievable or permanent IVC filter between 2009-2013. Demographics and outcomes were confirmed on independent chart review. Cost data were obtained using Current Procedural Terminology (CPT) codes. Results. 179 patients with retrievable filters and 207 patients with permanent filters were included. Contraindication to anticoagulation was the most cited reason for filter placement; however, only 76% of patients with retrievable filters and 69% of patients with permanent filters had an absolute contraindication to anticoagulation. 20% of patients with retrievable filters and 24% of patients with permanent filters had recurrent VTE. The median time from filter placement to death was 8.9 and 3.2 months in the retrievable and permanent filter groups, respectively. The total cost of retrievable filters and permanent filters was $2,883,389 and $3,722,688, respectively. Conclusions. The role for IVC filters in cancer patients remains unclear as recurrent VTE is common and time from filter placement to death is short. Filter placement is costly and has a clinically significant complication rate, especially for retrievable filters. More data from prospective, randomized trials are needed to determine the utility of IVC filters in cancer patients.
Work-From-Home in the New Normal: A Phenomenological Inquiry into Employees’ Mental Health
The COVID-19 pandemic has forced employees to adapt and adjust to the new normal in an unprecedented way. While some employees have been able to move to work-from-home (WFH) relatively easily, many find it challenging. Notwithstanding the magnitude of change, little is known about the determinants of WFH employees’ mental health during COVID-19. This study therefore aims to explore (1) the salient factors that contribute to the mental health issues of WFH employees and (2) strategies to overcome WFH challenges. A qualitative approach using phenomenological inquiry was adopted. Forty-one employees who worked from home in Pakistan were sampled using the purposive and snowball sampling techniques. Data was collected via semi-structured interviews and analyzed using thematic analysis. Overall, employees believe that organizations offer inadequate support in both work-related and non-work-related matters. Five themes were elicited and coded as factors that contribute to mental health issues among WFH employees. Technical issues and system complexities, the absence of flexible working arrangements, distractions, a lack of communication, and inadequate social support were found to obstruct WFH and cause mental distress. Behavioral and cognitive coping strategies were also determined to tackle these mental issues. This study complements the human resource literature by exploring the factors that obstruct WFH and cause mental health issues in the context of the pandemic crisis. As mental well-being is more intricate than administrative arrangements, the study is useful for organizations to develop a feasible mechanism that facilitates the smooth execution of WFH for employees while ensuring their mental health is preserved. Using a phenomenological inquiry, the present study is one of the few to explore the factors that contribute to the mental health of WFH employees in the context of the pandemic crisis. Apart from its contribution to knowledge on human resource management and organizational behavior, it provides useful implications for managers, policymakers, and practitioners to manage WFH employees more effectively.
Sustainability-Driven Digital Transformation and Customer Experience in Emerging Markets: The Interplay of Business Model Innovation and Value Co-Creation
Service industries are fast turning digital, and they are disrupting the manner in which firms organize and relate to their customers. Nevertheless, mechanisms that allow digital capabilities to be converted to better customer experience remain poorly understood, especially in the emerging economies. This paper presents the dual-mediation model to investigate the effects of Digital Supply Chain Integration (DSCI) and capabilities of digital customer engagement (DCEC) on Customer Experience Outcomes (CXO) based on Sustainable Business Model Innovation (SBMI) and Customer Value Co-Creation (CVC) with references to Dynamic Capabilities Theory and Service-Dominant Logic. The data were collected by a cross-sectional survey of 360 managers of the banking, telecom, healthcare, and hospitality organizations in Pakistan and analyzed with the help of Partial Least Squares Structural Equation Modeling (PLS-SEM). The findings indicate that the customer experience is enhanced well beyond what is feasible alone by means of Sustainable Business Model Innovation due to the Digital Supply Chain Integration, whereas customer experience is enhanced by the Digital Customer Engagement Capabilities through Customer Value Co-Creation. The results also show that the mechanisms of co-creation have the largest impact on the outcome of customer experience. The research is an addition to the body of literature on digital transformation because it illustrates how digital integration, innovation that is focused on sustainability, and relational co-creation can collectively convert digital capabilities into experiential value. The results also provide practical factors that should be considered by service companies when embarking on digital transformation programs to align the programs with sustainability and customer engagement mechanisms.
Liposomes in Cosmetics: Formulation and Skin Delivery
The cosmetic industry is one of the vibrant industrial sectors as from the start of civilization people are using cosmetic products for routine body care and beauty enhancement. Different formulations are available to deliver active pharmaceutical ingredients to the site of action. But in the case of cosmetic products, the skin delivery of a drug is challenging as the skin has a lipid barrier. The evolution of liposomes has been used in the development of the drug carrier concept as a promising delivery system. Liposomes are spherical vesicles composed of phospholipids with an aqueous core that can be used as the delivery vehicle. Liposomes improve and facilitate the bioavailability of carried active components to the site of action. Traditional skincare products are difficult to absorb into the skin. However, Liposomes are used to carry the active ingredients deeper into the skin layers. Moreover, liposomes enhance the stability of active ingredient, increases dermal bioavailability, and improves the hydration of the skin.
Proinflammatory immune cells disrupt angiogenesis and promote germinal matrix hemorrhage in prenatal human brain
Germinal matrix hemorrhage (GMH) is a devastating neurodevelopmental condition affecting preterm infants, but why blood vessels in this brain region are vulnerable to rupture remains unknown. Here we show that microglia in prenatal mouse and human brain interact with nascent vasculature in an age-dependent manner and that ablation of these cells in mice reduces angiogenesis in the ganglionic eminences, which correspond to the human germinal matrix. Consistent with these findings, single-cell transcriptomics and flow cytometry show that distinct subsets of CD45 + cells from control preterm infants employ diverse signaling mechanisms to promote vascular network formation. In contrast, CD45 + cells from infants with GMH harbor activated neutrophils and monocytes that produce proinflammatory factors, including azurocidin 1, elastase and CXCL16, to disrupt vascular integrity and cause hemorrhage in ganglionic eminences. These results underscore the brain’s innate immune cells in region-specific angiogenesis and how aberrant activation of these immune cells promotes GMH in preterm infants. Chen et al. show that subtypes of immune cells in prenatal human brain promote angiogenesis in the germinal matrix. Conversely, in preterm infants, proinflammatory immune cells disrupt angiogenesis and promote germinal matrix hemorrhage.
