Munc18 and Munc13 serve as a functional template to orchestrate neuronal SNARE complex assembly
The transition of the Munc18-1/syntaxin-1 complex to the SNARE complex, a key step involved in exocytosis, is regulated by Munc13-1, SNAP-25 and synaptobrevin-2, but the underlying mechanism remains elusive. Here, we identify an interaction between Munc13-1 and the membrane-proximal linker region of synaptobrevin-2, and reveal its essential role in transition and exocytosis. Upon this interaction, Munc13-1 not only recruits synaptobrevin-2-embedded vesicles to the target membrane but also renders the synaptobrevin-2 SNARE motif more accessible to the Munc18-1/syntaxin-1 complex. Afterward, the entry of SNAP-25 leads to a half-zippered SNARE assembly, which eventually dissociates the Munc18-1/syntaxin-1 complex to complete SNARE complex formation. Our data suggest that Munc18-1 and Munc13-1 together serve as a functional template to orchestrate SNARE complex assembly. Synaptic exocytosis depends on formation of the SNARE complex but its assembly mechanism is still under debate. Here, the authors identify an interaction between Munc13-1 and synaptobrevin-2 that is critical for the transition of the Munc18-1/syntaxin-1 complex to the SNARE complex.

A global meta-analysis on the effects of organic and inorganic fertilization on grasslands and croplands
A central role for nature-based solution is to identify optimal management practices to address environmental challenges, including carbon sequestration and biodiversity conservation. Inorganic fertilization increases plant aboveground biomass but often causes a tradeoff with plant diversity loss. It remains unclear, however, whether organic fertilization, as a potential nature-based solution, could alter this tradeoff by increasing aboveground biomass without plant diversity loss. Here we compile data from 537 experiments on organic and inorganic fertilization across grasslands and croplands worldwide to evaluate the responses of aboveground biomass, plant diversity, and soil organic carbon (SOC). Both organic and inorganic fertilization increase aboveground biomass by 56% and 42% relative to ambient, respectively. However, only inorganic fertilization decreases plant diversity, while organic fertilization increases plant diversity in grasslands with greater soil water content. Moreover, organic fertilization increases SOC in grasslands by 19% and 15% relative to ambient and inorganic fertilization, respectively. The positive effect of organic fertilization on SOC increases with increasing mean annual temperature in grasslands, a pattern not observed in croplands. Collectively, our findings highlight organic fertilization as a potential nature-based solution that can increase two ecosystem services of grasslands, forage production, and soil carbon storage, without a tradeoff in plant diversity loss. Inorganic fertilization reduces plant biodiversity. Here, the authors conduct a global meta-analysis on the use of organic and inorganic fertilizer in croplands and grasslands, showing that while both fertilizers increase plant biomass, only organic fertilizer increases biodiversity.
STC-YOLO: Small Object Detection Network for Traffic Signs in Complex Environments
The detection of traffic signs is easily affected by changes in the weather, partial occlusion, and light intensity, which increases the number of potential safety hazards in practical applications of autonomous driving. To address this issue, a new traffic sign dataset, namely the enhanced Tsinghua-Tencent 100K (TT100K) dataset, was constructed, which includes the number of difficult samples generated using various data augmentation strategies such as fog, snow, noise, occlusion, and blur. Meanwhile, a small traffic sign detection network for complex environments based on the framework of YOLOv5 (STC-YOLO) was constructed to be suitable for complex scenes. In this network, the down-sampling multiple was adjusted, and a small object detection layer was adopted to obtain and transmit richer and more discriminative small object features. Then, a feature extraction module combining a convolutional neural network (CNN) and multi-head attention was designed to break the limitations of ordinary convolution extraction to obtain a larger receptive field. Finally, the normalized Gaussian Wasserstein distance (NWD) metric was introduced to make up for the sensitivity of the intersection over union (IoU) loss to the location deviation of tiny objects in the regression loss function. A more accurate size of the anchor boxes for small objects was achieved using the K-means++ clustering algorithm. Experiments on 45 types of sign detection results on the enhanced TT100K dataset showed that the STC-YOLO algorithm outperformed YOLOv5 by 9.3% in the mean average precision (mAP), and the performance of STC-YOLO was comparable with that of the state-of-the-art methods on the public TT100K dataset and CSUST Chinese Traffic Sign Detection Benchmark (CCTSDB2021) dataset.
High-fidelity photonic quantum logic gate based on near-optimal Rydberg single-photon source
Compared to other types of qubits, photon is one of a kind due to its unparalleled advantages in long-distance quantum information exchange. Therefore, photon is a natural candidate for building a large-scale, modular optical quantum computer operating at room temperature. However, low-fidelity two-photon quantum logic gates and their probabilistic nature result in a large resource overhead for fault tolerant quantum computation. While the probabilistic problem can, in principle, be solved by employing multiplexing and error correction, the fidelity of linear-optical quantum logic gate is limited by the imperfections of single photons. Here, we report the demonstration of a linear-optical quantum logic gate with truth table fidelity of 99.84(3)% and entangling gate fidelity of 99.69(4)% post-selected upon the detection of photons. The achieved high gate fidelities are made possible by our near-optimal Rydberg single-photon source. Our work paves the way for scalable photonic quantum applications based on near-optimal single-photon qubits and photon-photon gates. The current main source of errors for photonic quantum logic gates is the imperfections of the single photons. Here, by using high-quality photons from Rydberg atoms, the authors are able to reach 99.7% entangling gate fidelity in a photonic CNOT gate.
Global pattern of soil priming effect intensity and its environmental drivers
The microbial priming effect—the decomposition of soil organic carbon (SOC) induced by plant inputs—has long been considered an important driver of SOC dynamics, yet we have limited understanding about the direction, intensity, and drivers of priming across ecosystem types and biomes. This gap hinders our ability to predict how shifts in litter inputs under global change can affect climate feedbacks. Here, we synthesized 18,919 observations of CO₂ effluxes in 802 soils across the globe to test the relative effects (i.e., log response ratio [RR]) of litter additions on native SOC decomposition and identified the dominant environmental drivers in natural ecosystems and agricultural lands. Globally, litter additions enhanced native SOC decomposition (RR = 0.35, 95% CI: 0.32–0.38), with greater priming effects occurring with decreasing latitude and more in agricultural soils (RR = 0.43) than in uncultivated soils (RR = 0.28). In natural ecosystems, soil pH and microbial community composition (e.g., bacteria: fungi ratio) were the best predictors of priming, with greater effects occurring in acidic, bacteria-dominated sandy soils. In contrast, the substrate properties of plant litter and soils were the most important drivers of priming in agricultural systems since soils with high C:N ratios and those receiving large inputs of low-quality litter had the highest priming effects. Collectively, our results suggest that, though different factors may control priming effects, the ubiquitous nature of priming means that alterations of litter quality and quantity owing to global changes will likely have consequences for global C cycling and climate forcing.
The impact of spatial distribution of noise pollution from music recreation facilities and residents’ perceptions in Chongqing municipality
With the rapid development of the night economy, the effects of noise pollution and the spatial differentiation characteristics of music leisure and entertainment facilities have become important issues in urban governance. Taking Chongqing city as an example, this study utilized AutoNavi POI data, noise complaint records, and field monitoring data to reveal the spatial distribution patterns and residents’ perceptions of noise pollution from music facilities. The study found that: (1) Music leisure and entertainment facilities in Chongqing exhibit a “multi-core” decentralized pattern, differing from the “core solo diffusion” pattern observed in lowland cities. Due to topographic segmentation, Chongqing presents a unique “valley agglomeration-platform jumping” differentiation. The core areas (Guanyin Bridge, Jiefangbei, etc.) overlap with active night economy zones, while secondary nodes (such as university towns) show local agglomerations due to terrain constraints. (2) The distribution of music leisure and entertainment facilities is significantly correlated with noise complaint locations. The Pearson correlation coefficient peaks at a 250 m grid scale ( r  = 0.75), and the number of complaints increases nonlinearly when facility density exceeds 140 n/km², revealing the critical threshold effect of noise-carrying capacity in mountainous cities. (3) The innovative integration of citizen complaint data to construct a noise perception map verifies the feasibility of using public participation data for identifying noise hotspots. However, this approach is limited by the subjectivity of residents’ complaint behaviour and the spatial coverage bias. The results of this study provide guidance for urban acoustic environment zoning control strategies and offer insights for mountainous cities to balance commercial vitality with acoustic environment governance.
RNA sequencing of Xp11 translocation-associated cancers reveals novel gene fusions and distinctive clinicopathologic correlations
Both Xp11 translocation renal cell carcinomas and the corresponding mesenchymal neoplasms are characterized by a variety of gene fusions involving TFE3 . It has been known that tumors with different gene fusions may have different clinicopathologic features; however, further in-depth investigations of subtyping Xp11 translocation-associated cancers are needed in order to explore more meaningful clinicopathologic correlations. A total of 22 unusual cases of Xp11 translocation-associated cancers were selected for the current study; 20 cases were further analyzed by RNA sequencing to explore their TFE3 gene fusion partners. RNA sequencing identified 17 of 20 cases (85%) with TFE3 -associated gene fusions, including 4 ASPSCR1/ASPL-TFE3 , 3 PRCC-TFE3 , 3 SFPQ/PSF-TFE3 , 1 NONO-TFE3 , 4 MED15-TFE3 , 1 MATR3-TFE3 , and 1 FUBP1-TFE3 . The results have been verified by fusion fluorescence in situ hybridization (FISH) assays or reverse transcriptase polymerase chain reaction (RT-PCR). The remaining 2 cases with specific pathologic features highly suggestive of MED15-TFE3 renal cell carcinoma were identified by fusion FISH assay. We provide the detailed morphologic and immunophenotypic description of the MED15-TFE3 renal cell carcinomas, which frequently demonstrate extensively cystic architecture, similar to multilocular cystic renal neoplasm of low malignant potential, and expressed cathepsin K and melanotic biomarker Melan A. This is the first time to correlate the MED15-TFE3 renal cell carcinoma with specific clinicopathologic features. We also report the first case of the corresponding mesenchymal neoplasm with MED15-TFE3 gene fusion. Additional novel TFE3 gene fusion partners, MATR3 and FUBP1 , were identified. Cases with ASPSCR1-TFE3 , SFPQ-TFE3 , PRCC-TFE3 , and NONO-TFE3 gene fusion showed a wide variability in morphologic features, including invasive tubulopapillary pattern simulating collecting duct carcinoma, extensive calcification and ossification, and overlapping and high columnar cells with nuclear grooves mimicking tall cell variant of papillary thyroid carcinoma. Furthermore, we respectively evaluated the ability of TFE3 immunohistochemistry, TFE3 FISH, RT-PCR, and RNA sequencing to subclassify Xp11 translocation-associated cancers. In summary, our study expands the list of TFE3 gene fusion partners and the clinicopathologic features of Xp11 translocation-associated cancers, and highlights the importance of subtyping Xp11 translocation-associated cancers combining morphology, immunohistochemistry, and multiple molecular techniques.
A Changing Paradigm for the Treatment of Intermediate-Stage Hepatocellular Carcinoma: Asia-Pacific Primary Liver Cancer Expert Consensus Statements
The Asia-Pacific Primary Liver Cancer Expert (APPLE) Consensus Statement on the treatment strategy for patients with intermediate-stage hepatocellular carcinoma (HCC) was established on August 31, 2019, in Sapporo, Hokkaido during the 10th Annual APPLE Meeting. This manuscript summarizes the international consensus statements developed at APPLE 2019. Transarterial chemoembolization (TACE) is the only guideline-recommended global standard of care for intermediate-stage HCC. However, not all patients benefit from TACE because intermediate-stage HCC is a heterogeneous disease in terms of tumor burden and liver function. Ten important clinical questions regarding this stage of HCC were raised, and consensus statements were generated based on high-quality evidence. In intermediate-stage HCC, preservation of liver function is as important as achieving a high objective response (OR) because the treatment goal is to prolong overall survival. Superselective conventional TACE (cTACE) is recommended as the first choice of treatment in patients eligible for effective (curative) TACE, whereas in patients who are not eligible, systemic therapy is recommended as the first choice of treatment. TACE is not indicated as the first-line therapy in TACE-unsuitable patients. Another important statement is that TACE should not be continued in patients who develop TACE failure/refractoriness in order to preserve liver function. Targeted therapy is the recommended first-line treatment for TACE-unsuitable patients. Especially, the drug, which can have higher OR rate, is preferred. Immunotherapy, transarterial radioembolization, TACE + targeted therapy or other modalities may be considered alternative options in TACE-unsuitable patients who are not candidates for targeted therapy. Better liver function, such as albumin-bilirubin grade 1, is an important factor for maximizing the therapeutic effect of systemic therapy.
