Parturition is a painful event experienced by cows at the onset of lactation. This pain could lead to a reduced feed intake, altered metabolic and immunological status, and a host of other diseases that could seriously limit her productive herd lifespan. The objective of the current study was to assess the effect of administration of a single dose of oral meloxicam, a non-steroidal anti-inflammatory drug (NSAID), on the production and health status of cows during their lactation. A total of 2,653 (1,009 meloxicam-treated and 1,644 untreated control) cows were enrolled across 20 herds in the provinces of Ontario and Quebec, Canada. Relative to untreated controls, meloxicam-treated cows produced 0.64 kg/day (SE = 0.29. P = 0.03) more milk over the first 3 test days (90-120 days in lactation), had 0.75 times the odds of subclinical mastitis at first test (SE = 0.08, P = 0.01), and were culled or died at 0.46 times the rate (SE = 0.16, P = 0.03) before 60 days in milk. These results are consistent with previous research and lend support to the hypothesis that parturition is a painful event in cattle. Attempts to ameliorate such pain with analgesics is associated with a variety of positive health and production outcomes.

The initiation of lactation challenges the ability of the modern lactating cow to maintain calcium homeostasis, and typically results in a drop in blood calcium levels, leading to mobilization of calcium reserves from skeletal stores. As such, the recommendation to provide supplemental calcium at parturition to older cows has become an industry-standard practice.
Mature cows were treated at calving and 12 hrs later with either the novel calcium bolus (NB) or a commercially available calcium bolus (CB). Blood was collected from animals at 0, 1, 6, 12, 13, and 24 hrs following calving, and the resulting serum samples were analyzed.
Overall, there was no statistical difference between the NB and CB groups for blood calcium levels within the first 24 hrs following parturition (P = 0.50). Cows in both groups experienced a significant increase in serum calcium by 1 hr after parturition; however, this increase was not sustained through subsequent sampling times.
This pilot study demonstrates that both boluses have a similar effect in the elevation of blood calcium.

Geobiological feedbacks and the evolution of thermoacidophiles

Oxygen-dependent microbial oxidation of sulfur compounds leads to the acidification of natural waters. How acidophiles and their acidic habitats evolved, however, is largely unknown. Using 16S rRNA gene abundance and composition data from 72 hot springs in Yellowstone National Park, Wyoming, we show that hyperacidic (pH<3.0) hydrothermal ecosystems are dominated by a limited number of archaeal lineages with an inferred ability to respire O
2
. Phylogenomic analyses of 584 existing archaeal genomes revealed that hyperacidophiles evolved independently multiple times within the Archaea, each coincident with the emergence of the ability to respire O
2
, and that these events likely occurred in the recent evolutionary past. Comparative genomic analyses indicated that archaeal thermoacidophiles from independent lineages are enriched in similar protein-coding genes, consistent with convergent evolution aided by horizontal gene transfer. Because the generation of acidic environments and their successful habitation characteristically require O
2
, these results suggest that thermoacidophilic Archaea and the acidity of their habitats co-evolved after the evolution of oxygenic photosynthesis. Moreover, it is likely that dissolved O
2
concentrations in thermal waters likely did not reach levels capable of sustaining aerobic thermoacidophiles and their acidifying activity until ~0.8 Ga, when present day atmospheric levels were reached, a time period that is supported by our estimation of divergence times for archaeal thermoacidophilic clades.

Extracorporeal life support (ECLS) is increasingly used in the treatment of infarct-related cardiogenic shock despite a lack of evidence regarding its effect on mortality.
In this multicenter trial, patients with acute myocardial infarction complicated by cardiogenic shock for whom early revascularization was planned were randomly assigned to receive early ECLS plus usual medical treatment (ECLS group) or usual medical treatment alone (control group). The primary outcome was death from any cause at 30 days. Safety outcomes included bleeding, stroke, and peripheral vascular complications warranting interventional or surgical therapy.
A total of 420 patients underwent randomization, and 417 patients were included in final analyses. At 30 days, death from any cause had occurred in 100 of 209 patients (47.8%) in the ECLS group and in 102 of 208 patients (49.0%) in the control group (relative risk, 0.98; 95% confidence interval [CI], 0.80 to 1.19; P = 0.81). The median duration of mechanical ventilation was 7 days (interquartile range, 4 to 12) in the ECLS group and 5 days (interquartile range, 3 to 9) in the control group (median difference, 1 day; 95% CI, 0 to 2). The safety outcome consisting of moderate or severe bleeding occurred in 23.4% of the patients in the ECLS group and in 9.6% of those in the control group (relative risk, 2.44; 95% CI, 1.50 to 3.95); peripheral vascular complications warranting intervention occurred in 11.0% and 3.8%, respectively (relative risk, 2.86; 95% CI, 1.31 to 6.25).
In patients with acute myocardial infarction complicated by cardiogenic shock with planned early revascularization, the risk of death from any cause at the 30-day follow-up was not lower among the patients who received ECLS therapy than among those who received medical therapy alone. Chemosynthetic sediment and planktonic community composition and sizes, aqueous geochemistry and sediment mineralogy were determined in 15 non-photosynthetic hot springs in Yellowstone National Park (YNP). These data were used to evaluate the hypothesis that differences in the availability of dissolved or mineral substrates in the bulk fluids or sediments within springs coincides with ecologically differentiated microbial communities and their populations. Planktonic and sediment-associated communities exhibited differing ecological characteristics including community sizes, evenness and richness. pH and temperature influenced microbial community composition among springs, but within-spring partitioning of taxa into sediment or planktonic communities was widespread, statistically supported (P < 0.05) and could be best explained by the inferred metabolic strategies of the partitioned taxa. Microaerophilic genera of the Aquificales predominated in many of the planktonic communities. In contrast, taxa capable of mineral-based metabolism such as So oxidation/reduction or Fe-oxide reduction predominated in sediment communities. These results indicate that ecological differentiation within thermal spring habitats is common across a range of spring geochemistry and is influenced by the availability of dissolved nutrients and minerals that can be used in metabolism.
The presence of minerals, such as elemental sulfur, that can support microbial metabolism promotes the ecological differentiation of sediment- and planktonic-associated microbial populations within Yellowstone National Park hot springs.

A Comparative Pharmacokinetic Analysis of Oral and Subcutaneous Meloxicam Administered to Postpartum Dairy Cows

The dairy industry needs evidence-based solutions to mitigate painful procedures and conditions in dairy cattle. The objective of this study was to compare the pharmacokinetic properties of orally versus subcutaneously administered meloxicam in early-lactation dairy cattle. The study was conducted at a commercial dairy herd in southwestern Ontario, Canada. Twelve postpartum cows were enrolled in the study, receiving either subcutaneous meloxicam (MET) at 0.5 mg/kg body weight (n = 6) or oral meloxicam (MOS) at a higher dose of 1.0 mg/kg body weight (n = 6) immediately following parturition. The predicted half-life (12.5 ± 2.0 vs. 28.5 ± 2.0 h), Cmax (1.59 ± 0.15 vs. 1.95 ± 0.16 μg/mL), Tmax (5.33 vs. 11.7 h), and AUC0→∞ (39.6 ± 7.4 vs. 115.6 ± 19 h * µg/mL) differed significantly between MET and MOS cows, respectively. After controlling for the treatment group, first lactation cows had a significantly higher half-life (4.1 ± 2.1 h), Cmax (0.56 ± 0.2 µg/mL), and AUC0→∞ (21.6 ± h * µg/mL) relative to second lactation or greater cows, respectively. Administration of meloxicam through the subcutaneous or oral route results in appreciable, dose-dependent systemic levels.

Recent advances in the archaeology of lowland South America are furthering our understanding of the Holocene development of plant cultivation and domestication, cultural niche construction, and relationships between environmental changes and cultural strategies of food production. This article offers new data on plant and landscape management and mobility in Southwestern Amazonia during a period of environmental change at the Middle to Late Holocene transition, based on archaeobotanical analysis of the Monte Castelo shellmound, occupied between 6000 and 650 yr BP and located in a modern, seasonally flooded savanna–forest mosaic. Through diachronic comparisons of carbonized plant remains, phytoliths, and starch grains, we construct an ecology of resource use and explore its implications for the long-term history of landscape formation, resource management practices, and mobility. We show how, despite important changes visible in the archaeological record of the shellmound during this period, there persisted an ancient, local, and resilient pattern of plant management which implies a degree of stability in both subsistence and settlement patterns over the last 6000 years. This pattern is characterized by management practices that relied on increasingly diversified, rather than intensive, food production systems. Our findings have important implications in debates regarding the history of settlement permanence, population growth, and carrying capacity in the Amazon basin.
