Stimuli-responsive liposomal membranes have attracted growing interest as dynamic soft materials capable of regulating permeability, fusion, and cargo release in response to external or internal triggers. By incorporating functional molecular or nanostructured guests, such as photochromic compounds, plasmonic nanoparticles, or ionizable lipids, bilayers can be endowed with reversible and tunable properties. These modifications often rely on the precise control of lipid packing, phase behaviour, and the formation of transient membrane defects that facilitate molecular transport. This review aims to provide an overview of the molecular design strategies and underlying mechanisms used to engineer such responsive liposomal systems, with particular emphasis on light- and heat-triggered behaviours and on supramolecular approaches that modulate membrane structure and dynamics. Emerging trends, current limitations, and opportunities for future development in functional lipid-based materials and biointerfaces will also be discussed.

Drug delivery systems
2019
Liposomes loaded with drug–cyclodextrin complexes are widely used as drug delivery systems, especially for species with low aqueous solubility and stability. Investigation of the intimate interactions of macrocycles with liposomes are essential for formulation of efficient and stable drug-in-cyclodextrin-in-liposome carriers. In this work, we reported the preparation of unilamellar vesicles of 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) embedded with native β-cyclodextrin and two synthetic derivatives: heptakis(2,3,6-tri-O-methyl)-β-cyclodextrin (TMCD) and heptakis(2,3-di-O-acetyl)-β-cyclodextrin (DACD). We then studied the effect of these macrocycles on the liposomal size, membrane viscosity, and liposomal stability at different temperatures and concentrations. We observed that TMCD and DACD affected vesicle size and the change of size was related to CD concentration. Irrespective of its nature, the macrocycle established interactions with the phospholipidic head groups, preventing cyclodextrins to diffuse into the lipid bilayer, as confirmed by molecular dynamics simulations. Such supramolecular structuring improves liposome stability making these colloid systems promising carriers for biologically active compounds.

Poly(methyl methacrylate) (PMMA) is characterized by high CO2 capture yield under mild pressures and temperatures. A morphological modification of powdery amorphous PMMA (pPMMA) is carried out by electrospinning to increase the surface/volume ratio of the resulting electrospun PMMAs (ePMMAs). This modification improves the kinetics and the capture yields. The rate constants observed for ePMMAs are two to three times higher than those for pPMMA, reaching 90% saturation values within 5–7 s. The amount of sorbed CO2 is up to eleven times higher for ePMMAs at 1 °C, and the highest difference in captured CO2 amount is observed at the lowest tested pressure of 1 MPa. The operating life of the ePMMAs shows a 5% yield loss after ten consecutive runs, indicating good durability. Spent electrospun PMMAs after several cycles of CO2 sorption-desorption can be regenerated by melting and again electrospinning the molten mass, resulting in a CO2 capture performance that is undistinguishable from that observed with fresh ePMMA. Scanning electron and atomic force microscopies show a reduction in surface roughness after gas exposure, possibly due to the plasticization effect of CO2. This study shows the potential of electrospun PMMAs as solid sorbents for carbon capture from natural gas or pre-combustion and oxyfuel combustion processes.

(1) Background: The aim of this study was to optimize, through a cheap and facile protocol, the covalent functionalization of graphene oxide (GO)-decorated cortical membrane (Lamina®) in order to promote the adhesion, the growth and the osteogenic differentiation of DPSCs (Dental Pulp Stem Cells); (2) Methods: GO-coated Laminas were fully characterized by Scannsion Electron Microscopy (SEM) and Atomic Force Microscopy (AFM) analyses. In vitro analyses of viability, membrane integrity and calcium phosphate deposition were performed; (3) Results: The GO-decorated Laminas demonstrated an increase in the roughness of Laminas, a reduction in toxicity and did not affect membrane integrity of DPSCs; and (4) Conclusions: The GO covalent functionalization of Laminas was effective and relatively easy to obtain. The homogeneous GO coating obtained favored the proliferation rate of DPSCs and the deposition of calcium phosphate.

Many biologically active compounds feature low solubility in aqueous media and, thus, poor bioavailability. The formation of the host-guest complex by using calixarene-based macrocycles (i.e., resorcinol-derived cyclic oligomers) with a good solubility profile can improve solubilization of hydrophobic drugs. Herein, we explore the ability of resorc[4]arenes to self-assemble in polar solutions, to form supramolecular aggregates, and to promote water-solubility of an isoflavone endowed with anti-cancer activity, namely Glabrescione B (GlaB). Accordingly, we synthesized several architectures featuring a different pattern of substitution on the upper rim including functional groups able to undergo acid dissociation (i.e., carboxyl and hydroxyl groups). The aggregation phenomenon of the amphiphilic resorc[4]arenes has been investigated in a THF/water solution by UV–visible spectroscopy, at different pH values. Based on their ionization properties, we demonstrated that the supramolecular assembly of resorc[4]arene-based systems can be modulated at given pH values, and thus promoting the solubility of GlaB.

Comparison of extraction methods for active biomolecules using sub-critical dimethyl ether and n-butane
2023
Several extraction methods are used to isolate natural compounds, and recent approaches utilize subcritical or supercritical extraction media. In this paper we compare extraction methods based on subcritical eluents, dimethyl ether (sC-DME) and n-butane (sC-nB), under mild conditions, using coffee beans and powder as an exemplary raw material. The parameters to be controlled to improve the extraction are considered, and the resulting data discussed. The results obtained display higher selectivity of sC-DME for caffeine (1.9%w/w sC-DME vs. 1.7%w/w sC-nB, on dry extract) and a good yield (0.479 mg/g of caffeine from green coffee beans) compared to, e.g., supercritical carbon dioxide (SC-CO2), which shows 0.32 mg/g of caffeine at higher pressure and temperature (25 MPa, 40 °C). We also discuss some technical implementations for optimizing the use of sub-critical eluents through proper combinations of pressure and temperature. We show that extraction processes based on sub-critical eluents are easy to operate and efficient, and can be easily automated.

Regenerative medicine and tissue engineering aim to restore or replace impaired organs and tissues using cell transplantation supported by scaffolds. Recently scientists are focusing on developing new biomaterials that optimize cellular attachment, migration, proliferation, and differentiation. Nanoparticles, such as graphene oxide (GO), have emerged as versatile materials due to their high surface-to-volume ratio and unique chemical properties, such as electrical conductivity and flexibility. However, GO faces challenges such as cytotoxicity at high concentrations, a negative surface charge, and potential inflammatory responses; for these reasons, variations in synthesis have been studied. A GO derivative, Graphene Oxide-Polyethylenimine (GO-PEI), shows controlled porosity and structural definition, potentially offering better support for cell growth. Human amniotic fluid stem cells (hAFSCs) are a promising candidate for regenerative medicine due to their ability to differentiate into mesodermic and ectodermic lineages, their non-immunogenic nature, and ease of isolation. This study investigates the effects of GO and GO-PEI on hAFSCs, focusing on the effects on adhesion, proliferation, and metabolic features. Results indicate that GO-PEI restores cell proliferation and mitochondrial activity to control levels, with respect to GO that appeared less biocompatible. Both materials also influence the miRNA cargo of hAFSC-derived microvesicles, potentially influencing also cell-to-cell communication.

Equilibrium constants for the proton transfer reaction between pyridines and trifluoroacetic acid were measured in room‐temperature ionic liquids (ILs) of different cation–anion compositions. The experimental equilibrium constants for ion‐pair formation were corrected according to the Fuoss equation. The calculated equilibrium constants for the formation of free ions were taken as a quantitative measure of the base strength in IL solutions and compared with the relative constants in water. The effect of IL composition is discussed for a series of fixed IL anions and fixed IL cations. Finally, the sensitivity of the proton transfer reaction to the electronic effects of the substituent groups on the pyridine ring was quantified by applying the Hammett equation. A more marked levelling effect on the base strength was observed in ILs than in water. The Hammett reaction constants ρ were then correlated with solvent parameters according to a multi‐parametric analysis, which showed that both specific hydrogen‐bond donor/acceptor and non‐specific interactions play an important role, with α and permittivity being the main parameters affecting the ability of the IL to differentiate the strength of the base. All about that base: The basicities of some pyridines were investigated in different ionic liquids by direct titration with trifluoroacetic acid. The calculated equilibrium constants increase as the hydrogen‐bond donor character of the ionic liquid cation increases. The application of the Hammett equation shows that the sensitivity to substituent effects also depends on the anion and cation composition of the ionic liquid.
