A natural antisense lncRNA controls breast cancer progression by promoting tumor suppressor gene mRNA stability
The human genome encodes thousands of long noncoding RNA (lncRNA) genes; the function of majority of them is poorly understood. Aberrant expression of a significant number of lncRNAs is observed in various diseases, including cancer. To gain insights into the role of lncRNAs in breast cancer progression, we performed genome-wide transcriptome analyses in an isogenic, triple negative breast cancer (TNBC/basal-like) progression cell lines using a 3D cell culture model. We identified significantly altered expression of 1853 lncRNAs, including ~500 natural antisense transcript (NATs) lncRNAs. A significant number of breast cancer-deregulated NATs displayed co-regulated expression with oncogenic and tumor suppressor protein-coding genes in cis. Further studies on one such NAT, PDCD4-AS1 lncRNA reveal that it positively regulates the expression and activity of the tumor suppressor PDCD4 in mammary epithelial cells. Both PDCD4-AS1 and PDCD4 show reduced expression in TNBC cell lines and in patients, and depletion of PDCD4-AS1 compromised the cellular levels and activity of PDCD4. Further, tumorigenic properties of PDCD4-AS1-depleted TNBC cells were rescued by exogenous expression of PDCD4, implying that PDCD4-AS1 acts upstream of PDCD4. Mechanistically, PDCD4-AS1 stabilizes PDCD4 RNA by forming RNA duplex and controls the interaction between PDCD4 RNA and RNA decay promoting factors such as HuR. Our studies demonstrate crucial roles played by NAT lncRNAs in regulating post-transcriptional gene expression of key oncogenic or tumor suppressor genes, thereby contributing to TNBC progression.
Pathologic Antibodies to Platelet Factor 4 after ChAdOx1 nCoV-19 Vaccination
Scully and colleagues report 23 cases of abnormal clotting, primarily involving the cerebral veins, 6 to 24 days after the first dose of the ChAdOx1 nCoV-19 vaccine. The syndrome was incited by antibodies to platelet factor 4, independent of heparin therapy. Early recognition and avoidance of platelet transfusion are key.
Spatio-temporal trends of rainfall across Indian river basins
Daily gridded high-resolution rainfall data of India Meteorological Department at 0.25° spatial resolution (1901–2015) was analyzed to detect the trend in seasonal, annual, and maximum cumulative rainfall for 1, 2, 3, and 5 days. The present study was carried out for 85 river basins of India during 1901–2015 and pre- and post-urbanization era, i.e., 1901–1970 and 1971–2015, respectively. Mann–Kendall (α = 0.05) and Theil–Sen’s tests were employed for detecting the trend and percentage of change over the period of time, respectively. Daily extreme rainfall events, above 95 and 99 percentile threshold, were also analyzed to detect any trend in their magnitude and number of occurrences. The upward trend was found for the majority of the sub-basins for 1-, 2-, 3-, and 5-day maximum cumulative rainfall during the post-urbanization era. The magnitude of extreme threshold events is also found to be increasing in the majority of the river basins during the post-urbanization era. A 30-year moving window analysis further revealed a widespread upward trend in a number of extreme threshold rainfall events possibly due to urbanization and climatic factors. Overall trends studied against intra-basin trend across Ganga basin reveal the mixed pattern of trends due to inherent spatial heterogeneity of rainfall, therefore, highlighting the importance of scale for such studies.
Primary tumor associated macrophages activate programs of invasion and dormancy in disseminating tumor cells
Metastases are initiated by disseminated tumor cells (DTCs) that colonize distant organs. Growing evidence suggests that the microenvironment of the primary tumor primes DTCs for dormant or proliferative fates. However, the manner in which this occurs remains poorly understood. Here, using the Window for High-Resolution Intravital Imaging of the Lung (WHRIL), we study the live lung longitudinally and follow the fate of individual DTCs that spontaneously disseminate from orthotopic breast tumors. We find that spontaneously DTCs have increased levels of retention, increased speed of extravasation, and greater survival after extravasation, compared to experimentally metastasized tumor cells. Detailed analysis reveals that a subset of macrophages within the primary tumor induces a pro-dissemination and pro-dormancy DTC phenotype. Our work provides insight into how specific primary tumor microenvironments prime a subpopulation of cells for expression of proteins associated with dissemination and dormancy. The understanding of the mechanisms underlying the ability of disseminated tumor cells (DTCs) to form metastasis is incomplete. Here, by using high-resolution intravital imaging of the murine lung to track the fate of breast-derived DTCs, the authors show that macrophages within the primary tumor induce a pro-dissemination and pro-dormancy phenotype in tumor cells, favouring their extravasation in the lung.
white-rot fungus Phanerochaete chrysosporium: conditions for the production of lignin-degrading enzymes
Investigating optimal conditions for lignin-degrading peroxidases production by Phanerochaete chrysosporium (P. chrysosporium) has been a topic for numerous researches. The capability of P. chrysosporium for producing lignin peroxidases (LiPs) and manganese peroxidases (MnPs) makes it a model organism of lignin-degrading enzymes production. Focusing on compiling and identifying the factors that affect LiP and MnP production by P. chrysosporium, this critical review summarized the main findings of about 200 related research articles. The major difficulty in using this organism for enzyme production is the instability of its productivity. This is largely due to the poor understanding of the regulatory mechanisms of P. chrysosporium responding to different nutrient sources in the culture medium, such as metal elements, detergents, lignin materials, etc. In addition to presenting the major conclusions and gaps of the current knowledge on lignin-degrading peroxidases production by P. chrysosporium, this review has also suggested further work, such as correlating the overexpression of the intra and extracellular proteins to the nutrients and other culture conditions to discover the regulatory cascade in the lignin-degrading peroxidases production process, which may contribute to the creation of improved P. chrysosporium strains leading to stable enzyme production.
