Technology in Language Use, Language Teaching, and Language Learning
This article offers a capacious view of technology to suggest broad principles relating technology and language use, language teaching, and language learning. The first part of the article considers some of the ways that technological media influence contexts and forms of expression and communication. In the second part, a set of heuristic questions is proposed to help guide language teachers and researchers in determining how to incorporate technology into their teaching practice or research agenda and evaluate its suitability and impact. These questions are based primarily on the goal of helping learners to pay critical attention to the culturally encoded connections among forms, contexts, meanings, and ideologies that they will encounter and produce in different mediums, both traditional and new.

Reprogramming normal human epithelial tissues to a common, lethal neuroendocrine cancer lineage
Epithelial cancers develop resistance to targeted therapies in a number of different ways. Several cancer types do so by undergoing phenotypic conversion to a highly aggressive cancer called small cell neuroendocrine carcinoma (SCNC). Whether distinct cancer types accomplish this “reprogramming” through the same mechanism has been unclear. Park et al. show that the same set of oncogenic factors transforms both normal lung and normal prostate epithelial cells into SCNCs that resemble clinical samples (see the Perspective by Kareta and Sage). This convergence of molecular pathways could potentially simplify the development of new therapies for SCNC, which is currently untreatable. Science , this issue p. 91 ; see also p. 30 Prostate and lung cancers convert to a drug-resistant, lethal form of cancer through the same reprogramming mechanism. The use of potent therapies inhibiting critical oncogenic pathways active in epithelial cancers has led to multiple resistance mechanisms, including the development of highly aggressive, small cell neuroendocrine carcinoma (SCNC). SCNC patients have a dismal prognosis due in part to a limited understanding of the molecular mechanisms driving this malignancy and the lack of effective treatments. Here, we demonstrate that a common set of defined oncogenic drivers reproducibly reprograms normal human prostate and lung epithelial cells to small cell prostate cancer (SCPC) and small cell lung cancer (SCLC), respectively. We identify shared active transcription factor binding regions in the reprogrammed prostate and lung SCNCs by integrative analyses of epigenetic and transcriptional landscapes. These results suggest that neuroendocrine cancers arising from distinct epithelial tissues may share common vulnerabilities that could be exploited for the development of drugs targeting SCNCs.
Using Near-Ground Storm Relative Helicity in Supercell Tornado Forecasting
This study examines the possibility that supercell tornado forecasts could be improved by utilizing the storm-relative helicity (SRH) in the lowest few hundred meters of the atmosphere (instead of much deeper layers). This hypothesis emerges from a growing body of literature linking the near-ground wind profile to the organization of the low-level mesocyclone and thus the probability of tornadogenesis. This study further addresses the ramifications of near-ground SRH to the skill of the significant tornado parameter (STP), which is probably the most commonly used environmental indicator for tornadic thunderstorms. Using a sample of 20 194 severe, right-moving supercells spanning a 13-yr period, sounding-derived parameters were compared using forecast verification metrics, emphasizing a high probability of detection for tornadic supercells while minimizing false alarms. This climatology reveals that the kinematic components of environmental profiles are more skillful at discriminating significantly tornadic supercells from severe, nontornadic supercells than the thermodynamic components. The effective-layer SRH has by far the greatest forecast skill among the components of the STP, as it is currently defined. However, using progressively shallower layers for the SRH calculation leads to increasing forecast skill. Replacing the effective-layer SRH with the 0–500 m AGL SRH in the formulation of STP increases the number of correctly predicted events by 8% and decreases the number of missed events and false alarms by 18%. These results provide promising evidence that forecast parameters can still be improved through increased understanding of the environmental controls on the processes that govern tornado formation.
A basal stem cell signature identifies aggressive prostate cancer phenotypes
Evidence from numerous cancers suggests that increased aggressiveness is accompanied by up-regulation of signaling pathways and acquisition of properties common to stem cells. It is unclear if different subtypes of late-stage cancer vary in stemness properties and whether or not these subtypes are transcriptionally similar to normal tissue stem cells. We report a gene signature specific for human prostate basal cells that is differentially enriched in various phenotypes of late-stage metastatic prostate cancer. We FACS-purified and transcriptionally profiled basal and luminal epithelial populations from the benign and cancerous regions of primary human prostates. High-throughput RNA sequencing showed the basal population to be defined by genes associated with stem cell signaling programs and invasiveness. Application of a 91-gene basal signature to gene expression datasets from patients with organ-confined or hormone-refractory metastatic prostate cancer revealed that metastatic small cell neuroendocrine carcinoma was molecularly more stem-like than either metastatic adenocarcinoma or organ-confined adenocarcinoma. Bioinformatic analysis of the basal cell and two human small cell gene signatures identified a set of E2F target genes common between prostate small cell neuroendocrine carcinoma and primary prostate basal cells. Taken together, our data suggest that aggressive prostate cancer shares a conserved transcriptional program with normal adult prostate basal stem cells.

Evidence of Artemisinin-Resistant Malaria in Western Cambodia
