Application of stress distribution simulation under fracture in predicting drilling leakage in Bozhong area
Faults and fractures are the most important factors that cause serious drilling leakage. Aiming to meet the production demand of improving the the reliability of leakage prediction, this paper uses the forward modeling technique of stress distribution in fault zones, by forward modeling of different fault combination modes, different scale fault stress influence ranges, and different fault zone filling modes, the stress changes, sizes, distribution characteristics, impact ranges, and properties of the fault zones on both sides of the fault were quantitatively displayed, achieving a comprehensive and quantitative characterization of the stress distribution under the fault; In addition, under the guidance of stress distribution, the reliability of leakage prediction can be improved effectively; Farther more, this paper combined the characteristics of the well-seismic data in the actual fault zone with the distribution of drilling leakage, proves that the three-dimensional space stress simulation and the stress intensity can effectively predict the plane and longitudinal distribution position of drilling leakage. The practicability of the method has been effectively applied and tested in the drilling leakage prediction and tracking in Bozhong area.
Simulating and Testing Microvibrations on an Optical Satellite Using Acceleration Sensor-Based Jitter Measurements
The present study uses a method to address microvibrations effects on an optical satellite by combining simulations and experiments based on high-precision acceleration sensors. The displacement and angular displacement of each optical component can be obtained by introducing flywheel perturbation data from a six-component test bench to the finite element model of the optical satellite. Combined with an optical amplification factor inferred from the linear optical model, the pixel offset of the whole optical system is calculated. A high accuracy and broad frequency range for a new microvibration measurement experimental system is established to validate the simulation. The pixel offset of the whole optical system can be measured by testing the acceleration signals of each optical component and calculating optical amplification factors. The results are consistent with optical imaging test results, indicating correctness of the experimental scheme and the effectiveness of the simulation. The results suggest that the effect of microvibrations on a camera can be verified by using mechanical simulators instead of a whole optical camera for the experiment scheme, which is demonstrated to be an effective way for increasing efficiency in jitter measurements.
Calcitonin gene‐related peptide inhibits angiotensin II‐induced NADPH oxidase‐dependent ROS via the Src/STAT3 signalling pathway
We had previously demonstrated that the calcitonin gene‐related peptide (CGRP) suppresses the oxidative stress and vascular smooth muscle cell (VSMC) proliferation induced by vascular injury. A recent study also indicated that CGRP protects against the onset and development of angiotensin II (Ang II)‐induced hypertension, vascular hypertrophy and oxidative stress. However, the mechanism behind the effects of CGRP on Ang II‐induced oxidative stress is unclear. CGRP significantly suppressed the level of reactive oxygen species (ROS) generated by NADPH oxidase in Ang II‐induced VSMCs. The Ang II‐stimulated activation of both Src and the downstream transcription factor, STAT3, was abrogated by CGRP. However, the antioxidative effect of CGRP was lost following the expression of constitutively activated Src or STAT3. Pre‐treatment with H‐89 or CGRP8–37 also blocked the CGRP inhibitory effects against Ang II‐induced oxidative stress. Additionally, both in vitro and in vivo analyses show that CGRP treatment inhibited Ang II‐induced VSMC proliferation and hypertrophy, accompanied by a reduction in ROS generation. Collectively, these results demonstrate that CGRP exhibits its antioxidative effect by blocking the Src/STAT3 signalling pathway that is associated with Ang II‐induced VSMC hypertrophy and hyperplasia.
Predicting Human Age with Bloodstains by sjTREC Quantification
The age-related decline of signal joint T-cell receptor rearrangement excision circles (sjTRECs) in human peripheral blood has been demonstrated in our previous study and other reports. Until now, only a few studies on sjTREC detection in bloodstain samples were reported, which were based on a small sample of subjects of a limited age range, although bloodstains are much more frequently encountered in forensic practice. In this present study, we adopted the sensitive Taqman real-time quantitative polymerase chain reaction (qPCR) method to perform sjTREC quantification in bloodstains from individuals ranging from 0-86 years old (n = 264). The results revealed that sjTREC contents in human bloodstains were declined in an age-dependent manner (r = -0.8712). The formula of age estimation was Age = -7.1815Y-42.458 ± 9.42 (Y dCt(TBP-sjTREC); 9.42 standard error). Furthermore, we tested for the influence of short- or long- storage time by analyzing fresh and stored bloodstains from the same individuals. Remarkably, no statistically significant difference in sjTREC contents was found between the fresh and old DNA samples over a 4-week of storage time. However, significant loss (0.16-1.93 dCt) in sjTREC contents was detected after 1.5 years of storage in 31 samples. Moreover, preliminary sjTREC quantification from up to 20-year-old bloodstains showed that though the sjTREC contents were detectable in all samples and highly correlated with donor age, a time-dependent decrease in the correlation coefficient r was found, suggesting the predicting accuracy of this described assay would be deteriorated in aged samples. Our findings show that sjTREC quantification might be also suitable for age prediction in bloodstains, and future researches into the time-dependent or other potential impacts on sjTREC quantification might allow further improvement of the predicting accuracy.
The Knowledge and Attitude Towards Advance Care Planning Among Chinese Patients with Advanced Cancer
To describe the knowledge and attitude of Chinese patients with advanced cancer towards advanced care planning (ACP), a convenience sample of 275 patients with advanced cancer was recruited from a tertiary cancer hospital in Beijing, China, between February and December 2017. The multi-item questionnaire focused on patients’ demographics, disease characteristics and knowledge about and attitude towards ACP and was administered to eligible patients. Descriptive statistics were performed. Most patients had never heard about ACP (82.2%) and had never talked about ACP (83.0%), but only a few (18.3%) were not willing to talk about ACP. A total of 67.8% patients chose to refuse resuscitation attempts or life-sustaining medical interventions, and 70.8% of patients hoped to have surrogate decision makers when they became unconscious. By binary logistic regression analysis, patients who were of greater age, female and living in urban areas preferred to refuse resuscitation attempts or life-sustaining medical interventions (OR = 1.023, P = 0.042; OR = 2.011, P = 0.020; OR = 0.254, P < 0.01); patients who had very rich or rich family economic status preferred to involve surrogate decision makers compared with patients of very poor family economic status (OR = 0.250, P = 0.011). There is a large gap between the knowledge about ACP and the expectation of implementing ACP in Chinese patients with advanced cancer. To develop culturally appropriate and individualized programmes to promote knowledge and implementation in practice of ACP among Chinese patients with advanced cancer and their relatives is still a significant challenge.
Predicting truncated multiple recursive generators with unknown parameters
Pseudorandom sequences are widely used in cryptography. Multiple recursive generators are an important class of pseudorandom sequence generators. A typical application is to obtain truncated sequences by truncating partial bits of the sequences output by the generators. This paper studies the predictability of truncated multiple recursive generators with unknown parameters. Given a few truncated digits of high-order bits output by a multiple recursive generator, we give a method based on lattice reduction to recover the parameters and the initial state of the generator. Our method is an extension of Stern’s algorithm which was proposed to predict the truncated sequences of linear congruential generators.
Chemokine CXCL3 mediates prostate cancer cells proliferation, migration and gene expression changes in an autocrine/paracrine fashion
IntroductionWe have previously indicated that CXCL3 was upregulated in the tissues of prostate cancer, and exogenous administration of CXCL3 played a predominant role in the tumorigenicity of prostate cancer cells. In the present study, we further explored the role and the underlying mechanism of CXCL3 overexpression in the oncogenic potential of prostate cancer in an autocrine/paracrine fashion.MethodsCXCL3-overexpressing prostate cancer cell line PC-3 and immortalized prostate stromal cell line WPMY-1 were established by gene transfection. CCK-8, transwell assays and growth of tumor xenografts were conducted to characterize the effects of CXCL3 on PC-3 cells’ proliferation and migration. Western blotting was conducted to test whether CXCL3 could affect the expression of tumorigenesis-associated genes.ResultsThe results showed that CXCL3 overexpression in PC-3 cells and the PC-3 cells treated with the supernatants of CXCL3-transfected WPMY-1 cells stimulated the proliferation and migration of PC-3 cells in vitro and in a nude mouse xenograft model. Western blotting revealed higher levels of p-ERK, Akt and Bcl-2 and lower levels of Bax in the tumor xenografts transplanted with CXCL3-transfected PC-3 cells. Moreover, the tumor xenografts derived from the PC-3 cells treated with supernatants of CXCL3-transfected WPMY-1 cells showed higher expression of ERK, Akt and Bcl-2 and lower expression of Bax.ConclusionsThese findings suggest that CXCL3 autocrine/paracrine pathways are involved in the development of prostate cancer by regulating the expression of the target genes that are related to the progression of malignancies.
Digital-Control-Based Approximation of Optimal Wave Disturbances Attenuation for Nonlinear Offshore Platforms
The irregular wave disturbance attenuation problem for jacket-type offshore platforms involving the nonlinear characteristics is studied. The main contribution is that a digital-control-based approximation of optimal wave disturbances attenuation controller (AOWDAC) is proposed based on iteration control theory, which consists of a feedback item of offshore state, a feedforward item of wave force and a nonlinear compensated component with iterative sequences. More specifically, by discussing the discrete model of nonlinear offshore platform subject to wave forces generated from the Joint North Sea Wave Project (JONSWAP) wave spectrum and linearized wave theory, the original wave disturbances attenuation problem is formulated as the nonlinear two-point-boundary-value (TPBV) problem. By introducing two vector sequences of system states and nonlinear compensated item, the solution of introduced nonlinear TPBV problem is obtained. Then, a numerical algorithm is designed to realize the feasibility of AOWDAC based on the deviation of performance index between the adjacent iteration processes. Finally, applied the proposed AOWDAC to a jacket-type offshore platform in Bohai Bay, the vibration amplitudes of the displacement and the velocity, and the required energy consumption can be reduced significantly.
The Spatiotemporal Development of Intercalated Disk in Three-Dimensional Engineered Heart Tissues Based on Collagen/Matrigel Matrix
Intercalated disk (ID), which electromechanically couples cardiomyocytes into a functional syncitium, is closely related to normal morphology and function of engineered heart tissues (EHTs), but the development mode of ID in the three-dimensional (3D) EHTs is still unclear. In this study, we focused on the spatiotemporal development of the ID in the EHTs constructed by mixing neonatal rat cardiomyocytes with collagen/Matrigel, and investigated the effect of 3D microenvironment provided by collagen/Matrigel matrix on the formation of ID. By histological and immmunofluorescent staining, the spatiotemporal distribution of ID-related junctions was detected. Furthermore, the ultra-structures of the ID in different developmental stages were observed under transmission electron microscope. In addition, the expression of the related proteins was quantitatively analyzed. The results indicate that accompanying the re-organization of cardiomyocytes in collagen/Matrigel matrix, the proteins of adherens junctions, desmosomes and gap junctions redistributed from diffused distribution to intercellular regions to form an integrated ID. The adherens junction and desmosome which are related with mechanical connection appeared earlier than gap junction which is essential for electrochemical coupling. These findings suggest that the 3D microenvironment based on collagen/Matrigel matrix could support the ordered assembly of the ID in EHTs and have implications for comprehending the ordered and coordinated development of ID during the functional organization of EHTs.
