Regional aroma characteristics of spontaneously fermented Cabernet Sauvignon wines produced from seven sub-regions in Shangri-La of China

Shangri-La is a promising wine region in China, which has great potential for producing high-quality wines with distinctive regional characteristics. In this work, the aroma properties of spontaneously fermented Cabernet Sauvignon wines produced from seven sub-regions of Shangri-La (Adong, Liutongjiang, Sinong, Xidang, and Nitong from Lancang River basin; Benzilan and Dari from Jinsha River Basin) were comprehensively analyzed using the headspace micro-extraction with gas chromatography-mass spectrometry, odor activity values, and olfactory evaluation. Results showed that a total of 59 volatiles belonging to seven groups were identified and quantified in all regional wines, with alcohols constituting the most abundant group, followed by esters and volatile fatty acids. -norisoprenoids, and three volatile fatty acids were identified as key volatiles which significantly contribute to the aroma of these wines. Principal component analysis showed the distinct compositions of these 19 key volatiles among the seven regional wines. Olfactory evaluation revealed certain differences in aroma profiles, particularly “Tropical fruit”, “Dried fruit”, “Vegetal”, and “Sweet” among them. This study enhances our understanding on the unique
terroir
flavors of Shangri-La wines and is helpful for further producing high-quality wines with regional characteristics.

Biodiversity of non-Saccharomyces yeasts associated with spontaneous fermentation of Cabernet Sauvignon wines from Shangri-La wine region, China

Shangri-La is a wine region that has the highest altitude vineyards in China. This is the first study investigated the biodiversity of non-Saccharomyces yeasts associated with spontaneous fermentation of Cabernet Sauvignon wines produced from two sub-regions (Lancang River and Jinsha River) of Shangri-La. The culturable yeasts were preliminarily classified based on their colonial morphology on the Wallerstein Laboratory nutrient agar plates. Yeast species were identified by the sequencing of the 26S rRNA D1/D2 region and the 5.8S rRNA ITS region. Twenty-five non-Saccharomyces yeast species belonging to sixteen genera were isolated and identified in Shangri-La wine region. Candida, Hanseniaspora, Pichia, and Starmerella were found in both sub-regions, but the Lancang River showed more diverse yeast species than the Jinsha River. Shangri-La not only exhibited high diversity of non-Saccharomyces yeasts, and furthermore, seven species of non-Saccharomyces yeasts were exclusively found in this region, including B. bruxellensis, D. hansenii, M. guilliermondii, S. vini, S. diversa, T. delbrueckii and W. anomalus, which might play an important role in distinctive regional wine characteristics. This study provide a relatively comprehensive analysis of indigenous non-Saccharomyces yeasts associated with Cabernet Sauvignon from Shangri-La, and has significance for exploring ‘microbial terroir’ of wine regions in China.

Methods
The clinical significance of NAT10 was explored based on the TCGA and GEO data sets and the 80 CRC patients cohort of our hospital. qRT-PCR, dot blot, WB, and IHC were performed to detect the level of NAT10 and ac
4
C modification in CRC tissues and matched adjacent tissues. CCK-8, colony formation, transwell assay, mouse xenograft, and other in vivo and in vitro experiments were conducted to probe the biological functions of NAT10. The potential mechanisms of NAT10 in CRC were clarified by RNA-seq, RIP-seq, acRIP-seq, luciferase reporter assays, etc.
Results
The levels of NAT10 and ac
4
C modification were significantly upregulated. Also, the high expression of NAT10 had important clinical values like poor prognosis, lymph node metastasis, distant metastasis, etc. Furthermore, the in vitro experiments showed that NAT10 could inhibit apoptosis and enhance the proliferation, migration, and invasion of CRC cells and also arrest them in the G2/M phase. The in vivo experiments discovered that NAT10 could promote tumor growth and liver/lung metastasis. In terms of mechanism, NAT10 could mediate the stability of KIF23 mRNA by binding to its mRNA 3’UTR region and up-regulating its mRNA ac

c modification. And then the protein level of KIF23 was elevated to activate the Wnt/β-catenin pathway and more β-catenin was transported into the nucleus which led to the CRC progression. Besides, the inhibitor of NAT10, remodelin, was applied in vitro and vivo which showed an inhibitory effect on the CRC cells.
Conclusions
NAT10 promotes the CRC progression through the NAT10/KIF23/GSK-3β/β-catenin axis and its expression is mediated by GSK-3β which forms a feedback loop. Our findings provide a potential prognosis or therapeutic target for CRC and remodelin deserves more attention.

A systematic analysis of marine lysogens and proviruses

Viruses are ubiquitous in the oceans, exhibiting high abundance and diversity. Here, we systematically analyze existing genomic sequences of marine prokaryotes to compile a Marine Prokaryotic Genome Dataset (MPGD, consisting of over 12,000 bacterial and archaeal genomes) and a Marine Temperate Viral Genome Dataset (MTVGD). At least 40% of the MPGD genomes contain one or more proviral sequences, indicating that they are lysogens. The MTVGD includes over 12,900 viral contigs or putative proviruses, clustered into 10,897 viral genera. We show that lysogens and proviruses are abundant in marine ecosystems, particularly in the deep sea, and marine lysogens differ from non-lysogens in multiple genomic features and growth properties. We reveal several virus-host interaction networks of potential ecological relevance, and identify proviruses that appear to be able to infect (or to be transferred between) different bacterial classes and phyla. Auxiliary metabolic genes in the MTVGD are enriched in functions related to carbohydrate metabolism. Finally, we experimentally demonstrate the impact of a prophage on the transcriptome of a representative marine
Shewanella
bacterium. Our work contributes to a better understanding of the ecology of marine prokaryotes and their viruses.

Prevalence of carbapenem-resistant Klebsiella pneumoniae (CRKP) has compromised antimicrobial efficacy against severe infections worldwide. To monitor global spread, we conducted a comprehensive genomic epidemiologic study comparing sequences from 21 blaOXA-232–carrying CRKP isolates from China with K. pneumoniae sequence type (ST) 15 strains from 68 countries available in GenBank. Phylogenetic and phylogeographic analyses revealed all blaOXA-232–carrying CRKP isolates belonged to ST15 lineage and exhibited multidrug resistance. Analysis grouped 330 global blaOXA-232–carrying ST15 CRKP strains into 5 clades, indicating clonal transmission with small genetic distances among multiple strains. The lineage originated in the United States, then spread to Europe, Asia, Oceania, and Africa. Most recent common ancestor was traced back to 2000; mutations averaged ≈1.7 per year per genome. Our research helps identify key forces driving global spread of blaOXA-232–carrying CRKP ST15 lineage and emphasizes the importance of ongoing surveillance of epidemic CRKP.

Genomic dissection of the clonal background and global dissemination of hypervirulent Klebsiella pneumoniae CG23-KL57 lineage

represents a growing threat to global public health. Here we investigate the genomic epidemiology and evolutionary dynamics of emerging
K. pneumoniae
clonal group 23 (CG23) lineages, with a particular focus on ST218-KL57 (predominantly found in Asia) and ST23-KL57 (circulating in Europe) to understand how genomic recombination and adaptive evolution drive the convergence of hypervirulence and multidrug resistance in CG23. We analyzed 3,118 clinical isolates from China together with 75,954 publicly available genomes, encompassing 122 ST218-KL57 and 45 ST23-KL57 isolates; 13 ST218-KL57 isolates were newly sequenced. Phylogenetic analyses revealed that ST23-KL57 forms a distinct subclade within CG23, more closely related to ST218-KL57 than to the classical ST23-KL1 linage. Neither ST218-KL57 nor ST23-KL57 harbors the chromosomal virulence loci (GIE492 and
all
island), which are characteristic of hypervirulent ST23-KL1. ST23-KL57 simultaneously harbors dual carbapenemases and key virulence genes (

), reflecting its hybrid origin of approximately 80% Asian hypervirulent ST218-KL57 and 20% European multidrug-resistant ST395 linage. The genomic decay of the

operon in ST23-KL57 suggests an adaptive trade-off between antimicrobial resistance and virulence. These findings underscore the importance of enhanced genomic surveillance and refined risk assessment for the evolving CG23-KL57 lineage.

Research on ecological value realization based on carbon trading—Take blue carbon as an example

This study aims to develop and expand a new perspective on ecological value realization (EVR) and provide policy recommendations for marine ecological value realization (MEVR) based on Carbon Trading. Currently, the immaturity of EVR calculation methods and difficulties in determining the price of ecological products pose significant challenges to ecological value trading. By employing mathematical models and logical reasoning, this study proposes a novel framework for EVR, illustrated through several diagrams. According to this framework, ecological value is not static but fluctuates with factors such as human well‐being (HV) or gross domestic product (GDP). Therefore, ecological value should be determined by an exchange market rather than solely relying on hypothetical calculation methods. Consequently, carbon trading cases are crucial in understanding ecological value. Based on the analysis of blue carbon (BC) trading cases, including the lack of international BC exchanges, challenges in carbon sink projects, and the Free Rider Effect, this paper identifies current issues in MEVR and BC trading in China. To address these challenges, we propose integrating carbon trading databases with evaluations of ecological protection and restoration projects, along with BC trading data, to calculate ecological value. Additionally, we recommend increasing the supply of BC products in both national carbon trading markets and voluntary markets, promoting the internationalization of BC accounting, addressing the Free Rider Effect through government actions and market mechanisms, attracting more foreign investment in BC enhancement projects, and formulating a BC enhancement plan during marine resource development.

Sea Surface pCO2 Response to Typhoon “Wind Pump” and Kuroshio Intrusion in the Northeastern South China Sea

The Luzon Strait (LS) is a key region for estimating carbon sources and sinks in the South China Sea (SCS) and is highly influenced by the Kuroshio Current (KC) and typhoons. Understanding the variations in the sea surface partial pressure of carbon dioxide (pCO2-sw) under the combined effects of typhoons and KC in this region is crucial for estimating local and regional changes in ocean carbon flux. Based on valuable in situ pCO2-sw and remote sensing data, this study aimed to reveal the temporal variations and the physical mechanisms of pCO2-sw variations under the comprehensive effects of both typhoons and Kuroshio Intrusion (KI) in the LS. One week after the passage of the tropical cyclone (TC) Nanmadol, the concentration in the pCO2-sw and the influencing mechanisms varied in three different regions (W1–W3) on Transect A (120°E). In the region dominated by SCS waters (W1), the average pCO2-sw increased by 5.1 μatm after TC, which was mainly due to the TC “Wind Pump” inducing strong vertical mixing, which brought dissolved inorganic carbon (DIC)-rich deeper water up to the surface. In the region affected by KC (W2 and W3), pCO2-sw decreased after the TC (−8.2 μatm and −1.8 μatm, respectively) with TC-enhanced KI because the invasion of lower pCO2-sw of Kuroshio waters inhibited the TC-induced upwelling. More significant TC-induced upwelling (W3) would alleviate the decrease in pCO2-sw caused by the TC-enhanced KI. This study is a rare case providing a better understanding of the variations in pCO2-sw under TC-enhanced KI, which provides support for regional climate change prediction and carbon flux estimation in the western boundary current regions.

Fermented Plant-Based Milks Based on Chestnut and Soybean: Comprehensive Evaluation of Fermentation Characteristics and Aroma Profiles Using Four Lactic Acid Bacteria Strains

In this study, four lactic acid bacteria (LAB) strains, including Lactiplantibacillus plantarum CICC21790, Lacticaseibacillus casei CICC6117, Lacticaseibacillus rhamnosus ATCC7469, and Limosilactobacillus fermentum CICC22704, were used to ferment a plant-based milk composed of chestnut and soybean. The fermentative characteristics of the four LAB strains and the aroma characteristics of the resulting plant-based milks were systematically investigated. The results showed that all four LAB strains successfully fermented the plant-based milk. The viable cell counts ranged from 7.67 to 8.57 lg CFU mL−1, and pH values were between 3.80 and 4.10. Comprehensive analyses performed using E-nose and HS-GC-IMS revealed distinct aroma characteristics in plant-based milks fermented by different LAB strains. Specifically, LAB fermentation, particularly by the CICC22704, significantly reduced the concentrations of aldehydes (e.g., hexanal, heptanal), thereby diminishing Green aroma characteristics. The increased concentrations of alcohols (e.g., 1-pentanol), ketones (e.g., 2, 3-butanedione) and furan compounds (e.g., 2-pentylfuran) in fermented plant-based milks enhanced Pungent, Creamy, and Fruity aroma characteristics, respectively. Fermentation by CICC21790, ATCC7469, and CICC6117 may result in stronger intensities of these three aroma characteristics compared to fermentation by the CICC22704. For the Fatty aroma characteristic, it was enhanced by CICC21790 fermentation but diminished by ATCC7469, CICC6117, and CICC22704 fermentations.
