The C-type lectin Dectin-1 is expressed predominantly on the surface of myeloid lineage cells. We examined the induction, regulation, and functions of Dectin-1 in pulmonary epithelial cells by challenging human bronchial epithelial (HBE) cells with
A. fumigatus
. Inflammatory, antimicrobial peptide genes and reactive oxygen species (ROS) were quantified, with and without knockdown of Dectin-1. We found that
A. fumigatus
induced the expression of Dectin-1 mRNA and protein in HBE cells in a toll-like receptor (TLR) 2-dependent manner. In addition,
A. fumigatus
-mediated generation of ROS was dependent on the upregulation of Dectin-1. Moreover,
A. fumigatus
actively induced the expression of TNFα, GM-CSF, IL8, HBD2, and HBD9. Knockdown of Dectin-1 inhibited TNFα, IL8, HBD2, and HBD9 expression. Hence, Dectin-1 was required for the upregulation of pro-inflammatory cytokines and antimicrobial peptides. Finally, knockdown of TLR2 significantly inhibited Dectin-1 upregulation. Our results demonstrate the novel induction of Dectin-1 in human bronchial epithelial cells and its critical role in the innate immune response against
A. fumigatus
in non-phagocytic cells.

First Observational Perspectives of “Millipede Clouds” Over the Eastern Pacific Ocean

The fundamental features of one kind of rarely known stratocumulus, which was termed as “Millipede Cloud,” occurred over the Eastern Pacific Ocean in 2017 were first documented by using Moderate Resolution Imaging Spectroradiometer (MODIS) satellite imagery. These clouds had long and meandering “central axes” extending from several hundreds to thousands kilometers, and a number of “radical cloud arms” extending several tens of kilometers in its two sides. Total 59 “Millipede Clouds,” 4 and 55 of them, were formed over the Northern and the Southern Hemispheres, respectively. Their environmental backgrounds were analyzed by using ERA5 reanalysis data and MODIS sensor Level‐2 data. The cloud top pressures of these “Millipede Clouds” were between 850 and 800 hPa, and their top heights were about 1–2 km. There existed “inversion layer” of air temperature near the cloud tops at 800 hPa, which strongly suggested that these clouds were lower stratocumulus in essence. Plain Language Summary “Millipede Cloud,” one kind of rarely known stratocumulus which looks like “Millipede” shape, is termed for the first time in this paper. It has an obvious “central axis” and a number of well‐organized “radial cloud arms” in two sides of the “central axis” extending in several tens of kilometers length. This paper introduces the fundamental features of “Millipede Clouds” occurred over the Eastern Pacific Ocean in 2017 from the perspective of satellite image. Totally, 59 “Millipede Clouds” were found to occur over the Eastern Pacific Ocean. Their geographic distribution, cloud top features and vertical structure of one typical case on 16 July 2017 were documented. Protein Functionalized Nanodiamond Arrays

Various nanoscale elements are currently being explored for bio-applications, such as in bio-images, bio-detection, and bio-sensors. Among them, nanodiamonds possess remarkable features such as low bio-cytotoxicity, good optical property in fluorescent and Raman spectra, and good photostability for bio-applications. In this work, we devise techniques to position functionalized nanodiamonds on self-assembled monolayer (SAMs) arrays adsorbed on silicon and ITO substrates surface using electron beam lithography techniques. The nanodiamond arrays were functionalized with lysozyme to target a certain biomolecule or protein specifically. The optical properties of the nanodiamond-protein complex arrays were characterized by a high throughput confocal microscope. The synthesized nanodiamond-lysozyme complex arrays were found to still retain their functionality in interacting with E. coli.

The Role of Proline Rich Tyrosine Kinase 2 (Pyk2) on Cisplatin Resistance in Hepatocellular Carcinoma

We previously demonstrated Proline rich tyrosine kinase 2 (Pyk2) plays important roles in regulating tumor progression, migration and invasion in hepatocellular carcinoma (HCC). In this study, we aimed to examine the role of proline rich tyrosine kinase 2 (Pyk2) on cisplatin resistance in HCC and to explore its underlying molecular mechanism.
Stable transfectants either overexpressing or suppressing Pyk2 were established in different HCC cell lines. MTT, colony formation and Annexin-V assays were employed to examine their in vitro responses to cisplatin. Xenograft ectopic and orthotopic nude mice models were generated to investigate the in vivo responses of them to cisplatin treatment. cDNA microarray was performed to identify Pyk2-induced genes which were further validated by quantitative real-time RT-PCR using clinical HCC samples. In vitro functional study demonstrated that Pyk2-overexpressing HCC transfectants exhibited relatively lower cytotoxicity, higher colony-forming ability and lower apoptosis to cisplatin compared with the control transfectants. Moreover, Pyk2 overexpressing HCC transfectants had a higher survival rate under cisplatin treatment by up-regulation of AKT phosphorylation. In vivo xenograft nude mice model demonstrated that Pyk2-overexpressing transfectants developed higher tolerance to cisplatin treatment together with less tumor necrosis and apoptosis. cDNA microarray analysis revealed that there were more than 4,000 genes differentially expressed upon overexpression of Pyk2. Several upregulated genes were found to be involved in drug resistance and invasion in cancers. Among them, the expression profiles of MDR1, GAGE1, STAT1 and MAP7 were significantly associated with the expression of Pyk2 in clinical HCC samples.
Our results may suggest a new evidence of Pyk2 on promoting cisplatin resistance of HCC cells through preventing cell apoptosis, activation of AKT pathway and upregulation of drug resistant genes.

Identification of some heat-induced genes of Trichinella spiralis
2001
Three heat-induced genes of the infective-stage larvae of Trichinella spiralis were successfully identified by the suppression subtractive hybridization (SSH) technique. As indicated by reverse Northern blotting, 19 of 25 clones were scored as differentially transcribed in the heat-shocked infective-stage larvae. The sequencing data showed the presence of 12 different genes. Three were homologous to histone H3, histone H2B and translationally controlled tumour protein (TCTP). A 0.6 kb cDNA of histone H3 was generated by the RACE method and sequenced. It contained an open reading frame of 136 amino acids that demonstrated 94% identity with genes from Drosophila hydei. Semi-quantitative RT-PCR indicated that after heat-shock treatment, the expression levels of histone H3, histone H2B and TCTP increased 4.8, 27 and 5.7-fold, respectively. Northern analysis confirmed the upregulation of histone H3, histone H2B and TCTP transcripts. The upregulation of these genes during stress conditions has not been reported in parasitic organisms. The stress proteins may play an active role to sustain the parasite after exposure to hostile host factors.

Residual torticollis in patients after strabismus surgery for congenital superior oblique palsy

Aim:To study postoperative residual vertical deviation and abnormal head posture (AHP) after surgical treatment for congenital superior oblique palsy (SOP).Method:Children with both SOP and AHP who underwent extraocular muscle surgery for correction of AHP were recruited. The patients received complete ophthalmic and orthopaedic examinations. Residual AHP was classified according to severity of face turn, head tilt and chin elevation.Results:Thirty-two children with mean age at operation of 82.6 months were recruited, with mean follow-up of 37.9 months. Of these children, 65.6% had a postoperative vertical deviation of less than three prism dioptres. In addition, 34.4% patients had resolved (0°), 34.4% had mild (1°–10°), and 31.3% had significant residual torticollis (>10°). Of the patients with significant residual torticollis, 33.3% had ocular causes. The mean age at operation for the patients with residual torticollis (95.9 months) was older than those without torticollis (79.9 months) (p = 0.018). Residual torticollis was found to be related to sternocleidomastoid muscle tightness (p = 0.013).Conclusion:The success rate for eliminating significant AHP after strabismus surgery for patients with congenital SOP was 68.8%. Early surgery was associated with a better outcome. Association was also found between sternocleidomastoid tightness and AHP. A multidisciplinary approach is recommended in the management of torticollis as ophthalmic and orthopaedic comorbidities can coexist.

This study examined carrier transport in single cobalt-doped zinc oxide (Co:ZnO) nanorods in temperatures ranging from 80 to 405 K. Measurements were taken on single nanorods deposited on a Si template, where two- and four-point metallic contacts were previously made using e-beam lithography, dielectrophoresis, and focused ion beam. In both two- and four-point probe measurements, the current–voltage curves were clearly linear and symmetrical with respect to both axes. The electrical measurements were carried out according to three different measuring methods to accurately determine the resistivity of the single Co:ZnO nanorods. The Co-doped nanorods exhibited ferromagnetic behavior at room temperature. The remanence permanent magnet of the nanorods increased with increasing Co concentration, however, this was accompanied by the decrease in electrical resistivity. The transport properties were dominated by the thermal activation of electrons from the Fermi level to the conduction band for a temperature above 140–160 K, and to the impurity band at a lower temperature. The electronic transport of the nanorods was influenced by the surface states when exposed in the air.

The article reports techniques that we have devised for immobilizing and allocating a single nanodiamond on the electron beam (E-beam) lithography patterned semiconductor substrate. By combining the E-beam patterned smart substrate with the high throughput of a confocal microscope, we are able to overcome the limitation of the spatial resolution of optical techniques (~1 μm) to obtain the data on individual nano-object with a size range between 100 and 35 nm. We have observed a broad photoluminescence centered at about 700 nm from a single nanodiamond which is due to the defects, vacancies in the nanodiamonds, and the disordered carbon layer covered on the nanodiamond surface. We also observe red-shift in energy and broadening in linewidth of the sp
3
bonding Raman peak when the size of the single nanodiamond is reduced due to the phonon-confinement effects.
