Tetrahymena RIB72A and RIB72B are Microtubule Inner Proteins in the ciliary doublet microtubules
Doublet and triplet microtubules are essential and highly stable core structures of centrioles, basal bodies, cilia and flagella. In contrast to dynamic cytoplasmic microtubules, their luminal surface is coated with regularly arranged Microtubule Inner Proteins (MIPs). However, the protein composition and biological function(s) of MIPs remain poorly understood. Using genetic, biochemical and imaging techniques we identified Tetrahymena RIB72A and RIB72B proteins as ciliary MIPs. Fluorescence imaging of tagged RIB72A and RIB72B showed that both proteins co-localize to Tetrahymena cilia and basal bodies, but assemble independently. Cryo-electron tomography of RIB72A and/or RIB72B knockout strains revealed major structural defects in the ciliary A-tubule involving MIP1, MIP4 and MIP6 structures. The defects of individual mutants were complementary in the double mutant. All mutants had reduced swimming speed and ciliary beat frequencies, and high-speed video imaging revealed abnormal highly curved cilia during power stroke. Our results show that RIB72A and RIB72B are crucial for the structural assembly of ciliary A-tubule MIPs and are important for proper ciliary motility.

Expression of p63 in tooth germ, dentigerous cyst and ameloblastoma
Context: To assess the role of p63, a p53 homolog, in the cytodifferentiation (odontogenesis) and oncogenesis of odontogenic epithelium. Aim: The present study aimed to compare the expression pattern of p63 in the epithelium of tooth germ, dentigerous cyst (DC) and ameloblastoma (AB). Materials and Methods: Tissue specimens of thirty tooth germs, thirty ABs and thirty DCs were examined by immunohistochemistry for the expression of p63. Results: p63 labeling index (LI) was observed in descending order in epithelial cells of ABs, tooth germs and DCs. p63 LI was statistically nonsignificant among all the three groups. ABs revealed the highest p63 expression, but, surprisingly, tooth germs showed higher expression than DCs. Conclusion: p63 plays a role in the cytodifferentiation and proliferation of odontogenic epithelial cells irrespective of the tissue (normal developing or lesional tissue). This implies that p63 cannot be used as a diagnostic marker. However, our results indicate p63 overexpression as a mark of increased proliferation. Thus, it can be stipulated that p63 can be used as a prognostic marker in odontogenic lesions with more aggressive and invasive phenotype. Our results also suggest the differential function of p63 in both developing and lesional odontogenic tissues, which, however, depends on p63 isoform predominantly being expressed. Therefore, identification of p63-predominant isoform in a particular lesion is more important than the presence or absence of p63. Consequently, we suggest the performance of polymerase chain reaction analysis along with immunohistochemical evaluation in further studies.

Development of Press-Coated, Floating Pulsatile Drug Delivery of Lisinopril
Lisinopril is an angiotensin-converting enzyme (ACE) inhibitor, primarily used for the treatment of hypertension, congestive heart failure, and heart attack. It belongs to BCS class III having a half-life of 12 hrs and 25% bioavailability. The purpose of the present work was to develop a press-coated, floating-pulsatile drug delivery system. The core tablet was formulated using the super-disintegrants crosprovidone and croscarmellose sodium. A press-coated tablet (barrier layer) contained the polymer carrageenan, xanthan gum, HPMC K4M, and HPMC K15M. The buoyant layer was optimized with HPMC K100M, sodium bicarbonate, and citric acid. The tablets were evaluated for physical characteristics, floating lag time, swelling index, FTIR, DSC, and in vitro and in vivo behavior. The 5% superdisintgrant showed good results. The FTIR and DSC study predicted no chemical interactions between the drug and excipients. The formulation containing xanthan gum showed drug retaining abilities, but failed to float. The tablet containing HPMC K15M showed a high swelling index. The lag time for the tablet coated with 200 mg carrageenan was 3±0.1 hrs with 99.99±1.5% drug release; with 140 mg HPMC K4M, the lag time was 3±0.1 hrs with 99.71±1.2% drug release; and with 120 mg HPMC K15M, the lag time was 3±0.2 hrs with 99.98±1.7% drug release. The release mechanism of the tablet followed the Korsmeyer-Peppas equation and a first-order release pattern. Floating and lag time behavior have shown good in vitro and in vivo correlations.

A study to assess the effectiveness of planned teaching programme on knowledge regarding electroconvulsive therapy among third year basic B.Sc nursing students in selected nursing college. Objectives: of the study, To assess the level of knowledge regarding Electroconvulsive therapy among third year Basic B.Sc. nursing students, assess the effectiveness of planned teaching on level of knowledge regarding electroconvulsive therapy among third year Basic B.Sc. nursing students and also to determine the association between the level of knowledge regarding Electroconvulsive therapy among third year Basic B.Sc nursing students with their selected demographic variables. This study was based on quantitative approach. Sample: The population was third year B. Sc nursing students in selected nursing college. The sample consisted of 30 students. The inclusion criteria were: (a) Third year basic B.Sc nursing students in selected nursing college. (b) Third year basic BSC nursing students who are willing to participate in the study. (c) Student who knows English. The tool was self-structured knowledge questionnaire. Based on the objectives and the hypothesis the data were analyzed and by using various statistical tests i.e. percentage, mean, chi square and standard deviations‘t’ test. The level of significance set for testing the hypothesis was at 0.05. Results: The findings show that the calculated‘t’ value was 4.29 for knowledge regarding electroconvulsive therapy. The calculated‘t’ value was much higher than the tabulated value. Hence, it was strongly interpreted that the planned teaching regarding electroconvulsive therapy was effective. So, the H1 was accepted.
