Mycotoxins as Emerging Contaminants. Introduction to the Special Issue “Rapid Detection of Mycotoxin Contamination”

Concerns for human and environmental health regarding mycotoxins are predominantly raised in connection with their occurrence in food and feed (especially in grains) [1,2]. Overall, 56 authors contributed 13 articles (12 original research articles and a review) discussing various aspects of mycotoxin research, but with mycotoxin analysis involved in all cases. Sample Preparation and Handling to Support Method Accuracy As seen also from studies on method development in this Special Issue [11,12], sample preparation is a step of key importance in the chemical analysis process; not only due to its required features of applicability and recovery, but also because novel standardized methods, such as the QuEChERS dispersive solid-phase extraction protocol can facilitate standardization of the analytical procedure improving inter-laboratory standard errors. [21] apply an immunochromatographic assay based on digital detection using colloidal gold nanoparticles labeled to monoclonal antibodies to detect ZEN in authentic cereal (corn, wheat, wheat flour, cereal product) and feed samples within a monitoring campaign carried out in China in 2019.

Re-registration Challenges of Glyphosate in the European Union

One of the most controversial societal issues today, regarding pesticide registration in the European Union (EU) may be the case surrounding re-registration of the active herbicide ingredient glyphosate. Shortly before the announcement of the conflicting views regarding the carcinogenicity status of this regulated agrochemical by EU Agencies, the European Food Safety Authority (EFSA) and the European Chemicals Agency (ECHA) on the one hand, and the International Agency for Research on Cancer (IARC) on the other hand, the Cancer Assessment Review Committee of the US Environmental Protection Agency (US EPA) also published re-evaluations. The US EPA assessment classified glyphosate into Group E, “not likely to be carcinogenic to humans”. Similar positions were reached by EFSA and ECHA, assessing glyphosate as “unlikely to pose a carcinogenic hazard to humans” and “not classified as a carcinogen”, respectively. A strongly opposing evaluation has previously been reached by IARC by classifying glyphosate into Group 2A, “probably carcinogenic to humans”. IARC identified potential cancer hazards in this case, but did not estimate the level of risk it may present, which was taken into consideration by opposing agencies. Multiple effects of glyphosate have been reported, of which carcinogenic effects are only one component. Formulated glyphosate products – especially with polyethoxylated tallowamine and related compounds – have been shown to cause stronger cytotoxic or endocrine disrupting effects than the active ingredient glyphosate alone. Questions related to hazards and corresponding risks identified in relation to this active ingredient and its formulated herbicide preparations divide scientific circles and official health and environmental authorities and organizations, and touch upon fundamental aspects of risk assessment and product regulation. The decision has to consider both hazard-based (IARC) and risk-based analysis (EFSA); the former may not be suitable to calculate practical significances, and the latter being challenged if exposure estimations are uncertain in light of new data on residue levels. The results of current analytical surveys on surface water are particularly worrisome. In turn, the precautionary principle appears to be the optimal approach in this case for regulation in the EU.

Application of Electrochemical Biosensors for Determination of Food Spoilage

Food security is significantly affected by the mass production of agricultural produce and goods, the growing number of imported foods, and new eating and consumption habits. These changed circumstances bring food safety issues arising from food spoilage to the fore, making food safety control essential. Simple and fast screening methods have been developed to detect pathogens and biomarkers indicating the freshness of food for safety. In addition to the traditional, sequential, chemical analytical and microbiological methods, fast, highly sensitive, automated methods suitable for serial tests have appeared. At the same time, biosensor research is also developing dynamically worldwide, both in terms of the analytes to be determined and the technical toolkit. Consequently, the rapid development of biosensors, including electrochemical-based biosensors, has led to significant advantages in the quantitative detection and screening of food contaminants. These techniques show great specificity for the biomarkers tested and provide adequate analytical accuracy even in complex food matrices. In our review article, we summarize, in separate chapters, the electrochemical biosensors developed for the most important food groups and the food safety issues they can ensure, with particular respect to meat and fish products, milk and dairy products, as well as alcoholic and non-alcoholic beverages.

Current Trends in Mycotoxin Detection with Various Types of Biosensors

One of the most important tasks in food safety is to properly manage the investigation of mycotoxin contamination in agricultural products and foods made from them, as well as to prevent its occurrence. Monitoring requires a wide range of analytical methods, from expensive analytical procedures with high-tech instrumentation to significantly cheaper biosensor developments or even single-use assays suitable for on-site monitoring. This review provides a summary of the development directions over approximately a decade and a half, grouped according to the biologically sensitive components used. We provide an overview of the use of antibodies, molecularly imprinted polymers, and aptamers, as well as the diversity of biosensors and their applications within the food industry. We also mention the possibility of determining multiple toxins side by side, which would significantly reduce the time required for the analyses.

Assessing surface water quality in Hungary’s Danube basin using geochemical modeling, multivariate analysis, irrigation indices, and Monte Carlo simulation

Evaluation of water quality is crucial for managing surface water effectively, ensuring its suitability for human use, and sustaining the environment. In the lower Danube River basin, various methods were employed to assess surface water quality for irrigation, drinking, human health risk purposes and the main mechanism control the surface water chemistry. These methods included water quality indicators (WQIs), complex statistical analyses, geographic information systems (GIS), Monte Carlo simulation, and geochemical modeling. Physicochemical analyses of surface water samples revealed primarily Ca–Mg–HCO
3
−
is the dominant water types. Principal component analysis (PCA), ionic ratios and piper, chloro alkaline index, Chadha, and Gibbs diagrams identified three distinct water characteristics influenced by water-rocks interaction, evaporation, ions exchange, and human activities. The geochemical modeling showed Danube River water’s strong ability to dissolve gypsum, halite, and anhydrite (SI < 0) and precipitate aragonite, dolomite, and calcite with saturation index (SI) value greater than 0 along its flow path. The irrigation water quality index (IWQI = 99.6–107.6), sodium adsorption ratio (SAR = 0.37–0.68), sodium percentage (Na% = 13.7–18.7), soluble sodium percentage (SSP = 12.5–17.5), Potential Salinity (PS = 0.73–1.6), and Residual Sodium Carbonate (RSC = − 1.27–0.58) values were used, mainly indicating acceptable quality with some limitations. Danube River water was unsuitable for drinking based on WQI value (WQI = 81–104). Oral exposure of children to specific components showed a higher hazard index (HI > 1) compared to adults, indicating a 2.1 times higher overall non-carcinogenic risk hazard index. However, Monte Carlo simulation demonstrated negligible iron, manganese, and nitrate health hazards for both age groups. These findings are valuable for water quality management decisions, contributing to long-term resource sustainability.

Glyphosate (GLY), the most widely used herbicide active ingredient (AI) in the world, is frequently detected in aquatic environments where it can affect non-target organisms. Globally, more than 2000 commercial GLY-based herbicides (GBHs) are used to control weeds. Non-target organisms are exposed to complex pesticide formulations under real environmental conditions, but the co-formulants contained in GBHs are classified as so-called inert and inactive ingredients in terms of their biological effects. The main objective of this comprehensive review is to compile the results of aquatic ecotoxicological studies on the side-effects of GLY, GBHs, and their formulating agents. Based on the results demonstrated for a variety of plant and animal aquatic organisms, oxidative stress appears to be a major trigger for these adverse effects, affecting the integrity of DNA and other biochemical functions. Furthermore, there is evidence of impairment of various physiological and behavioral functions. Adverse effects of GLY and GBHs have been observed even at very low concentrations. There are also differences in the sensitivity of the aquatic organisms tested, even with similar lifestyles, habitats or identical taxa. The studies typically investigate the short-term effects of a single exposure to GLY/GBH on a single species, whilst in reality multiple applications of GBHs together with other pesticides are common during a cropping cycle. Moreover, the interactions between GLY/GBHs and other aquatic contaminants are rarely studied. Higher toxicity of GBHs compared to GLY alone has often been observed, demonstrating that co-formulants can be highly toxic on their own and markedly increase the toxicity of the GBH formulation. The possible impurities in GBHs, such as heavy metals, can cause additional problems for the environment and food safety. The widespread and massive use of GBHs leads to increased exposure and environmental hazards. In addition, the need for a revision of the risk assessment system is emphasized. According to the results of aquatic ecotoxicological studies, the current use and pollution of the aquatic environment by GLY/GBHs is highly problematic and cannot be considered environmentally sustainable. It is, therefore, necessary to at least tighten the permitted forms of use.

Mycotoxin contamination of cereals used for feed can cause intoxication, especially in farm animals; therefore, efficient analytical tools for the qualitative and quantitative analysis of toxic fungal metabolites in feed are required. Current trends in food/feed analysis are focusing on the application of biosensor technologies that offer fast and highly selective and sensitive detection with minimal sample treatment and reagents required. The article presents an overview of the recent progress of the development of biosensors for deoxynivalenol and zearalenone determination in cereals and feed. Novel biosensitive materials and highly sensitive detection methods applied for the sensors and the application of these sensors to food/feed products, the limit, and the time of detection are discussed.

Terrestrial ecotoxicity of glyphosate, its formulations, and co-formulants: evidence from 2010–2023

Glyphosate (GLY), the most widely used herbicide in the world, is frequently detected in various environmental matrices, including soil, the foundation of agriculture. In practice, more than 2000 GLY-based herbicide (GBH) products are used, consisting of one or more active ingredients (AIs) and so-called “inert” co-formulants that increase the efficacy of the AIs. However, the focus of ecotoxicological assessments is mainly on AIs, while organisms are exposed to complex pesticide formulations under real-world conditions. Overall, the effects on non-target organisms indicate a broad range of biochemical and physiological modes of action, which contrasts with the general assumption that herbicides are specific and act only on target plants. Both GLY alone and GBHs have unintended side-effects on many terrestrial organisms, including non-target plants, microorganisms, insects, spiders, or earthworms, as well as vertebrates such as amphibians, reptiles, or mammals. One of the triggering mechanisms for these effects is oxidative stress with consequences on biochemical parameters and DNA damage. In addition, disruptions of various physiological, behavioral and ecological processes have been reported. Most studies have examined the short-term effects of a single application of GLY/GBH to a single species. However, the agricultural practice of applying GBHs two to three times during a cultivation season over an extended period of time, the interactions with other pesticides and agrochemicals applied to the same field, and effects on ecological interactions within the field and landscape are rarely considered. In the vast majority of cases, the toxicity of GBHs exceeds the toxicity of GLY, demonstrating that supposedly inert co-formulants are either toxic in their own right or interact and add to the toxicity of AIs. The chemical diversity of different GBHs and the non-disclosure of the co-formulants make it difficult to attribute effects to specific chemical substances within a GBH. Moreover, impurities in GBHs (e.g., heavy metals such as arsenic, chromium, cobalt) pose additional environment and food safety risks. These impacts are even more critical because GBHs are so widely distributed worldwide and interact with other pollutants and environmental stressors. Based on the available literature on terrestrial ecotoxicity, and given the drastic decline in biodiversity, we conclude that the continued high use of GBHs, resulting in increased exposure and risk, cannot be considered ecologically sustainable.

Advanced approach combines integrated weight water quality index and potential toxic elements for environmental and health risk assessment supported by simulation technique in Oued Souf, Algeria

The current research study evaluated the health and environmental risks issues associated with potentially toxic elements (PTEs) in the complex terminal aquifer located in the Algerian desert. The methods used included principal component and cluster (dendrogram) analysis to estimate source of ions and contamination. Various indices such as the Heavy Metal Pollution Index (HPI), Metal Index, hazard quotient, hazard index (HI), and cancer risk (CR) were applied to assess both environmental and human health risks. Furthermore, the Monte Carlo method was applied for probabilistic assessment of carcinogenic and non-carcinogenic risks through oral and dermal exposure routes in both adults and children. The results revealed that approximately 16% of the samples fell within the low pollution category (HPI < 100), indicating relatively lower levels of heavy metal contamination. However, the remaining 84% of the samples exhibited high pollution levels, indicating a significant presence of heavy metal pollutants in the northeastern part of the investigated area. The calculated average risk index (RI) for the collected samples was 18.99, with a range from 0.03 to 103.21. This indicates that a large portion, 82% of the samples, could cause low ecological risk (RI < 30), whereas the remaining 18% indicate a significant environmental pollution risk. The HI for oral ingestion showed that adults had HI values ranging from 0.231 to 1.54, while children exhibited higher values, ranging from 0.884 to 5.9 (Fig. 5a). For dermal exposure, HI values in adults ranged from 2.71E−07 to 8.74E−06 and in children, from 2.18E−06 to 7.03E−05. These findings highlight the potential non-carcinogenic risks associated with oral exposure to PTEs and underscore the increased vulnerability of children to metals such as Fe, Mn, Pb, and Cr. Most samples showed CR exceeding 1 × 10
−4
for chromium (Cr) and lead (Pb), indicating a significant vulnerability to carcinogenic effects in both children and adults.

Direct and Competitive Optical Grating Immunosensors for Determination of Fusarium Mycotoxin Zearalenone

Novel optical waveguide lightmode spectroscopy (OWLS)-based immunosensor formats were developed for label-free detection of Fusarium mycotoxin zearalenone (ZON). To achieve low limits of detection (LODs), both immobilised antibody-based (direct) and immobilised antigen-based (competitive) assay setups were applied. Immunoreagents were immobilised on epoxy-, amino-, and carboxyl-functionalised sensor surfaces, and by optimising the immobilisation methods, standard sigmoid curves were obtained in both sensor formats. An outstanding LOD of 0.002 pg/mL was obtained for ZON in the competitive immunosensor setup with a dynamic detection range between 0.01 and 1 pg/mL ZON concentrations, depending on the covalent immobilisation method applied. This corresponds to a five orders of magnitude improvement in detectability of ZON relative to the previously developed enzyme-linked immonosorbent assay (ELISA) method. The selectivity of the immunosensor for ZON was demonstrated with structural analogues (α-zearalenol, α-zearalanol, and β-zearalanol) and structurally unrelated mycotoxins. The method was found to be applicable in maize extract using acetonitrile as the organic solvent, upon a dilution rate of 1:10,000 in buffer. Thus, the OWLS immunosensor method developed appears to be suitable for the quantitative determination of ZON in aqueous medium. The new technique can widen the range of sensoric detection methods of ZON for surveys in food and environmental safety assessment.
