Norovirus Vaccines: Current Clinical Development and Challenges

Noroviruses are the major viral pathogens causing epidemic and endemic acute gastroenteritis with significant morbidity and mortality. While vaccines against norovirus diseases have been shown to be of high significance, the development of a broadly effective norovirus vaccine remains difficult, owing to the wide genetic and antigenic diversity of noroviruses with multiple co-circulated variants of various genotypes. In addition, the absence of a robust cell culture system, an efficient animal model, and reliable immune markers of norovirus protection for vaccine evaluation further hinders the developmental process. Among the vaccine candidates that are currently under clinical studies, recombinant VP1-based virus-like particles (VLPs) that mimic major antigenic features of noroviruses are the common ones, with proven safety, immunogenicity, and protective efficacy, supporting a high success likelihood of a useful norovirus vaccine. This short article reviews the recent progress in norovirus vaccine development, focusing on those from recent clinical studies, as well as summarizes the barriers that are being encountered in this developmental process and discusses issues of future perspective.

Circulation background of climate patterns in the past millennium: Uncertainty analysis and re-reconstruction of ENSO-like state

The question of whether or not global warming has paused since more than ten years ago, namely "warming hia- tus", has attracted the attention of climate science community including the IPCC. Some authors have attributed the "warming hiatus" to the internal changes in the climate system, i.e., the recombination of ocean-atmosphere circulations. Therefore, it is necessary to propose higher requirements on reconstructing circulation background of climate change for the past millennium. However, the analyses of changes in atmospheric circulation over the last millennium as well as the conclusions of related re- gional climate patterns are so widely different and contradictory, bringing uncertainties to our understanding of regional even global climate change to a great extent. On the other hand, in the last 10 years the high-precision U/Th-dated stalagmite oxygen isotope ratio (δ^18O) sequences provided an accurate chronological frame for the paleoclimate study of the middle and late Pleistocene, in which all authors from China took the Chinese stalagmite δ^18O as the summer monsoon index without excep- tion. However, this point of view misleads the climate scientists into thinking that the stalagmite δ^18O can be as the proxy of precipitation amount. Nevertheless, it is well known that all of these records have a lot in common in the low frequency trend. However, most sequences cannot be calibrated by instrumental precipitation records, and thus the uncertainty of the climate research framework of China and even of the world has increased. Therefore, it is imperative for climatology to clarify the origin of contradiction and to reduce the uncertainty as early as possible. On the basis of analyzing the significance of stalag- mite ~180 in the monsoon regions of China, the author tries to propose a new circulation proxy in this paper: integrating the Chinese stalagmite oxygen isotope sequence to reconstruct the tropical Pacific sea surface temperature gradient, i.e., the large-scale ENSO-like state over the past millennium. Furthermore, the author speculates that it was warm in the modern times and the Medieval Period, but the circulation recombination was different in both periods. And this inference could be support- ed by the longer record since Last Glacial Maximum. In other words, the attribution analysis of the identical low-frequency trends of Chinese stalagmite t~180 on a large scale shows that the ENSO-like state controls the climate change in the monsoon regions of China at different time scales (from interannual to century or even longer time scales). Wherein the important connection of circulations is the western Pacific subtropical high (WPSH), that is to say, besides the interannual and decadal time scales, the WPSH would possess the circulation mode on longer timescales. For example, we may discuss the change of the WPSH in the whole Holocene epoch, i.e., the half precession period. These discussions could make sense to the study of not only the paleoclimate but also the modern climate.

New frontiers in immune checkpoint B7-H3 (CD276) research and drug development

B7-H3 (CD276), a member of the B7 family of proteins, is a key player in cancer progression. This immune checkpoint molecule is selectively expressed in both tumor cells and immune cells within the tumor microenvironment. In addition to its immune checkpoint function, B7-H3 has been linked to tumor cell proliferation, metastasis, and therapeutic resistance. Furthermore, its drastic difference in protein expression levels between normal and tumor tissues suggests that targeting B7-H3 with drugs would lead to cancer-specific toxicity, minimizing harm to healthy cells. These properties make B7-H3 a promising target for cancer therapy.
Recently, important advances in B7-H3 research and drug development have been reported, and these new findings, including its involvement in cellular metabolic reprograming, cancer stem cell enrichment, senescence and obesity, have expanded our knowledge and understanding of this molecule, which is important in guiding future strategies for targeting B7-H3. In this review, we briefly discuss the biology and function of B7-H3 in cancer development. We emphasize more on the latest findings and their underlying mechanisms to reflect the new advances in B7-H3 research. In addition, we discuss the new improvements of B-H3 inhibitors in cancer drug development.

Trust in blockchain-enabled exchanges: Future directions in blockchain marketing

Prior research typically positions blockchain technology as enabling a trustless exchange environment without specifically investigating how blockchain technology provides trust and what makes the data in a blockchain “tamperproof” and “immutable.” This article serves to address these research gaps by conducting semi-structured interviews with 18 informants who have had at least three years of project experience with blockchain-enabled exchanges. Our findings uncover three unique aspects of blockchain that enable trust in exchange vs. a traditional exchange: (1) trust in exchange actors: mathematics and cryptography vs. human guardians within institutions, (2) trust in exchange actions: information transparency enabling tamperproof and immutable data vs. information asymmetry, and (3) trust in exchange assets: digital vs. manual escrows for verifying ownership of valuable goods. This research is vital for marketing scholars and practitioners who seek to understand the rise of threats to trust regarding online advertising, customer trust, privacy, and digital rights.

Ethical Marketing in the Blockchain-Based Sharing Economy: Theoretical Integration and Guiding Insights

Since the introduction of Ethereum in 2015, blockchain technology (BT) has been evolving, and BT has been associated with the concept of the sharing economy by business academics. Despite the marketing research on the sharing economy that has been extensively conducted in the last decade, the linkage between BT and ethical marketing in the sharing economy remains unclear. Through a systematic literature review of 163 articles and a co-citation analysis, this study identifies the key elements of blockchain capabilities, blockchain attributes, and the underlying economic theories of blockchain. It also synthesizes and proposes a shift of ethical marketing logic in the blockchain-based sharing economy that delineates the principles of stakeholder capitalism. The article concludes with a list of future research directions that underline three approaches of stakeholder theory (i.e., the descriptive, instrument, and normative approaches). These directions aim to guide marketing scholars concerning how BT enables an institutionally embedded view of ethical marketing activities and practices that enhance collaborative marketing and subsequently innovate value chains and create sustainable business models in the sharing economy, as well as to the metaverse.

Purpose
Three-dimensional printing of concrete (3DPC) has a potential for the rapid industrialization of the housing sector, with benefits of reduced construction time due to no formwork requirement, ease of construction of complex geometries, potential high construction quality and reduced waste. Required materials adaption for 3DPC is within reach, as concrete materials technology has reached the point where performance-based specification is possible by specialists. This paper aims to present an overview of the current status of 3DPC for construction, including existing printing methods and material properties required for robustness of 3DPC structures or structural elements.
Design/methodology/approach
This paper has presented an overview of three categories of 3DPC systems, namely, gantry, robotic and crane systems. Material compositions as well as fresh and hardened properties of mixes currently used for 3DPC have been elaborated.
Findings
This paper presents an overview of the state of the art of 3DPC systems and materials. Research needs, including reinforcement in the form of bars or fibres in the 3D printable cement-based materials, are also addressed.
Originality/value
The critical analysis of the 3D concrete printing system and materials described in this review paper is original.

Pembrolizumab in patients with thymic carcinoma: a single-arm, single-centre, phase 2 study

Treatment options are limited for patients with thymic carcinoma. These aggressive tumours are not typically associated with paraneoplastic autoimmune disorders, and strong PD-L1 expression has been reported in thymic epithelial tumours. We aimed to assess the activity of pembrolizumab, a monoclonal antibody that targets PD-1, in patients with advanced thymic carcinoma.
We completed a single-arm phase 2 study of pembrolizumab in patients with recurrent thymic carcinoma who had progressed after at least one line of chemotherapy. This was a single-centre study performed at Lombardi Comprehensive Cancer Center, Georgetown University, Washington, DC, USA. Key inclusion criteria were an Eastern Cooperative Oncology Group performance status of 0–2, no history of autoimmune disease or other malignancy requiring treatment or laboratory abnormality, and adequate organ function. Patients received 200 mg of pembrolizumab every 3 weeks for up to 2 years. The primary objective of the study was the proportion of patients who had achieved a response assessed with Response Evaluation Criteria in Solid Tumors version 1.1. Analysis was per protocol, in all eligible patients. The study is registered with ClinicalTrials.gov, number NCT02364076, and is closed to accrual; we report the final analysis.
41 patients were enrolled from March 12, 2015, to Dec 16, 2016, of whom 40 were eligible and evaluable and one was excluded because of elevated liver enzymes at screening. The median follow-up was 20 months (IQR 14–26). The proportion of patients who achieved a response was 22·5% (95% CI 10·8–38·5); one (3%) patient achieved a complete response, eight (20%) patients achieved partial responses, and 21 (53%) patients achieved stable disease. The most common grade 3 or 4 adverse events were increased aspartate aminotransferase and alanine aminotransferase (five [13%] patients each). Six (15%) patients developed severe autoimmune toxicity, including two (5%) patients with myocarditis. There were 17 deaths at the time of analysis, but no deaths due to toxicity.
Pembrolizumab is a promising treatment option in patients with thymic carcinoma. Because severe autoimmune disorders are more frequent in thymic carcinoma than in other tumour types, careful monitoring is essential.

Cancer is a leading cause of death, affecting people in both developed and developing countries. It is a challenging disease due to its complicated pathophysiological mechanism. Many anti-cancer drugs are used to treat cancer and reduce mortality rates, but their toxicity limits their administration. Drugs made from natural products, which act as multi-targeted therapy, have the ability to target critical signaling proteins in different pathways. Natural compounds possess pharmacological activities such as anti-cancer activity, low toxicity, and minimum side effects. Panax notoginseng is a medicinal plant whose extracts and phytochemicals are used to treat cancer, cardiovascular disorders, blood stasis, easing inflammation, edema, and pain. P. notoginseng ’s secondary metabolites target cancer’s dysregulated pathways, causing cancer cell death. In this review, we focused on several ginsenosides extracted from P. notoginseng that have been evaluated against various cancer cell lines, with the aim of cancer treatment. Furthermore, an in vivo investigation of these ginsenosides should be conducted to gain insight into the dysregulation of several pathways, followed by clinical trials for the potential and effective treatment of cancer.
