Photonic-circuited resonance fluorescence of single molecules with an ultrastable lifetime-limited transition

Resonance fluorescence as the emission of a resonantly-excited two-level quantum system promises indistinguishable single photons and coherent high-fidelity quantum-state manipulation of the matter qubit, which underpin many quantum information processing protocols. Real applications of the protocols demand high degrees of scalability and stability of the experimental platform, and thus favor quantum systems integrated on one chip. However, the on-chip solution confronts several formidable challenges compromising the scalability prospect, such as the randomness, spectral wandering and scattering background of the integrated quantum systems near heterogeneous and nanofabricated material interfaces. Here we report an organic-inorganic hybrid integrated quantum photonic platform that circuits background-free resonance fluorescence of single molecules with an ultrastable lifetime-limited transition. Our platform allows a collective alignment of the dipole orientations of many isolated molecules with the photonic waveguide. We demonstrate on-chip generation, beam splitting and routing of resonance-fluorescence single photons with a signal-to-background ratio over 3000 in the waveguide at the weak excitation limit. Crucially, we show the photonic-circuited single molecules possess a lifetime-limited-linewidth transition and exhibit inhomogeneous spectral broadenings of only about 5% over hours’ measurements. These findings and the versatility of our platform pave the way for scalable quantum photonic networks.
Single molecules can generate high-quality single photons for quantum technologies, but coupling to waveguides is difficult. Here, the authors show on-chip background-free resonance fluorescence generation and routing from single molecules with lifetime-limited transition and waveguide-aligned dipoles.

Influence of Organic Impurities on Fractional Crystallization of NaCl and Na2SO4 from High-Salinity Coal Chemical Wastewater: Thermodynamics and Nucleation Kinetics Analysis

It is a valid path to realize the zero discharge of coal chemical wastewater by using the fractional crystallization method to recycle the miscellaneous salt in high-salinity wastewater. In this study, the thermodynamics and nucleation kinetics of sodium chloride (NaCl) and sodium sulfate (Na2SO4) crystallization in coal chemical wastewater were systematically studied. Through analyses of solubility, metastable zone width, and induction period, it was found that the impurity dimethoxymethane would increase the solid–liquid interface energy and critical crystal size during the nucleation of Na2SO4. Ternary phase diagrams of the pseudo-ternary Na2SO4-NaCl-H2O systems in simulated wastewater were plotted in the temperature range of 303.15 to 333.15 K, indicating that a co-ionization effect existed between NaCl and Na2SO4, and NaCl had a strong salting out effect on Na2SO4. Finally, the nucleation rate and growth rate of Na2SO4 crystals under simulated wastewater conditions were determined by the intermittent dynamic method, and the crystallization kinetic models of Na2SO4 were established. The crystallization nucleation of Na2SO4 crystals was found to be secondary nucleation controlled by surface reactions. The basic theoretical research of crystallization in this study is expected to fundamentally promote the application of fractional crystallization to realize the resource utilization of high-salinity wastewater in the coal chemical industry.

Background
Lung adenocarcinoma (LUAD) presents a considerable danger to human health and has evolved into a major public health concern. Ribosome biogenesis (RiboSis) is a critical process for synthesizing ribosomes, closely associated with cancer initiation, progression, and treatment resistance, potentially serving as a target for future cancer therapies.
Methods
Utilizing single-cell RNA sequencing (scRNA-seq) technology, a single-cell atlas of LUAD was delineated, focusing on the analysis of T cell subpopulations. Cells were scored based on the expression patterns of 331 genes associated with RiboSis across different cell types, and monocle2 was employed to analyze the developmental trajectory of CD4
+
T cells. Employing various machine learning algorithms, a ribosome biogenesis-related signature (RBS) was constructed and compared to 140 published LUAD prognostic models. The relationship between RBS risk scores and various factors in LUAD patients, including prognosis, the tumor immune microenvironment (TIME), responsiveness to immunotherapy, and sensitivity to pharmacological treatments was specifically analyzed. Immunohistochemistry was utilized to validate the expression levels of immune markers in the high- and low- RBS groups, and in vitro experiments were performed to validate the functional role of the pivotal gene KIF23 in the progression of LUAD.
Results
Using single-cell analysis, two distinct T cell subtypes were identified: CD8
+
interferon (IFN) response T cells and CD4
+
stress response T cells. It was observed that CD4
+
naive-like T cells exhibit high expression of RiboSis-related genes, with a gradual decrease in RiboSis activity as CD4
+
T cells develop. Compared to other prognostic models, RBS demonstrated superior performance in prognosis prediction. The low-RBS group exhibited a tumor microenvironment (TME) more favorable for efficient immune monitoring and reaction, higher responsiveness to immunotherapy, and a better prognosis. Immunohistochemistry confirmed higher expression levels of immune markers in the low-RBS group, while in vitro experiments validated the promoting role of KIF23 in LUAD cell proliferation, migration and invasion.
Conclusion
This study delves into the relationship between RiboSis and LUAD cell subpopulations, identifying a potent prognostic biomarker for LUAD. This biomarker aids in assessing immunotherapy efficacy in LUAD patients, ultimately enhancing their prognosis and guiding clinical decision-making.

Exosomal lncRNA PCAT1 Promotes Tumor Circulating Cell-Mediated Colorectal Cancer Liver Metastasis by Regulating the Activity of the miR-329-3p/Netrin-1-CD146 Complex

Objective. This study explored the colorectal cancer exosome lncRNA prostate cancer associated transcript 1– (PCAT1) mediated circulating tumors and the mechanism of cell colorectal cancer liver metastasis. Methods. Exosomes were extracted from the primary colorectal cancer (CRC) cell lines HCT116 and SW480 and cultured with T84 and human umbilical vein endothelial (HUVE) cells. The expression of PCAT1 and miR-329-3p was detected by real-time quantitative polymerase chain reaction (RT-qPCR), the expression of Netrin-1, CD146, and epithelial mesenchymal transition (EMT) related proteins was detected by Western blot, the proliferation activity of T84 cells was detected by cell counting kit 8 (CCK-8), and cell migration was detected by Transwell. The expression of the F-actin signal was detected by immunofluorescence after coculture of exosomes with human umbilical vein endothelial cells (HUVECs). Changes in subcutaneous tumor and liver nodule size after PCAT1 deletion were observed in a mouse model of liver metastasis from rectal cancer. Results. PCAT1 expression was upregulated in primary cell lines and their exosomes. After exosomes were cocultured with colorectal cancer tumor circulating T84 cells, the expression of Netrin-1 and CD146 was upregulated, the expression of miR-329-3p was downregulated, the proliferation and migration ability of T84 cells were enhanced, and EMT occurred. After knocking down PCAT1, the above phenomenon was reversed. Similarly, after exosomes were cocultured with HUVECs, the expression of the F-actin signal increased, and after PCAT1 was knocked down, the F-actin signal also decreased. PCAT1 regulates miR-329-3p/Netrin-1 and affects the biological behavior of T84 and F-actin signal expression in HUVECs. In a mouse model of colorectal cancer liver metastasis, knocking down PCAT1 significantly reduced the nodules formed by liver metastasis in mice. Conclusions. LncRNA PCAT1 derived from colorectal cancer exosomes regulates the activity of the Netrin-1-CD146 complex in circulating tumor cells (CTCs) to promote the occurrence of colorectal cancer EMT and liver metastasis and provides new molecular targets for the treatment of colorectal cancer liver metastasis.

Selective far-field addressing of coupled quantum dots in a plasmonic nanocavity

Plasmon–emitter hybrid nanocavity systems exhibit strong plasmon–exciton interactions at the single-emitter level, showing great potential as testbeds and building blocks for quantum optics and informatics. However, reported experiments involve only one addressable emitting site, which limits their relevance for many fundamental questions and devices involving interactions among emitters. Here we open up this critical degree of freedom by demonstrating selective far-field excitation and detection of two coupled quantum dot emitters in a U-shaped gold nanostructure. The gold nanostructure functions as a nanocavity to enhance emitter interactions and a nanoantenna to make the emitters selectively excitable and detectable. When we selectively excite or detect either emitter, we observe photon emission predominantly from the target emitter with up to 132-fold Purcell-enhanced emission rate, indicating individual addressability and strong plasmon–exciton interactions. Our work represents a step towards a broad class of plasmonic devices that will enable faster, more compact optics, communication and computation.
Plasmonic nanostructures can tailor excitation and emission for quantum emitters, but generally only for a single emitter. In this work, the authors selectively excite and detect one out of two quantum dots coupled to a deep-subwavelength cavity composed of three gold nanorods assembled into a U-shape.

LncRNA GATA6-AS inhibits cancer cell migration and invasion in gallbladder cancer by downregulating miR-421

Long non-coding RNS (lncRNA) GATA6-AS regulates endothelial cell growth, which is involved in many types of human diseases.
Our study was carried out to investigate the involvement of GATA6-AS in gallbladder cancer (GBC).
Sixty-two patients with GBC were recruited in this study. PCR analysis was performed to determine the expression of GATA6-AS, miR-421 and TIMP-2 in tissues or cell lines. Transwell migration and invasion assay was applied.
We found that GATA6-AS was downregulated in tumor tissues than in adjacent healthy tissues of GBC patients, and GATA6-AS expression levels in tumor tissues decreased with the increase of clinical stages. MiR-421 was upregulated in tumor tissues than in adjacent healthy tissues of GBC patients and was inversely correlated with the expression levels of GATA6-AS. MiR-421 overexpression failed to significantly affect GATA6-AS in GBC cells, while GATA6-AS overexpression resulted in inhibited miR-421 expression. GATA6-AS overexpression led to decreased migration and invasion rates of GBC cells. MiR-421 overexpression led to increased migration and invasion rates of GBC. Rescue experiments (co-transfection) showed that miR-421 overexpression led to attenuated effects of GATA6-AS overexpression.
GATA6-AS may inhibit cancer cell migration and invasion in GBC by downregulating miR-421.

Advanced Neural Network-Based Equalization in Intensity-Modulated Direct-Detection Optical Systems: Current Status and Future Trends

Intensity-modulated direct-detection (IM/DD) optical systems are most widely employed in short-reach optical interconnects due to their simple structure and cost-effectiveness. However, IM/DD systems face mixed linear and nonlinear channel impairments, mainly induced by the combination of square-law detection and chromatic dispersion, as well as the utilization of low-cost non-ideal transceivers. To solve this issue, recent years have witnessed a growing trend of introducing machine learning technologies such as neural networks (NNs) into IM/DD systems for channel equalization. NNs usually present better system performance than traditional approaches, and various types of NNs have been investigated. Despite the excellent system performance, the associated high computational complexity is a major drawback that hinders the practical application of NN-based equalizers. This paper focuses on the performance and complexity trade-off of NNs employed in IM/DD systems, presenting a systematic review of the current status of NN-based equalizers as well as a number of effective complexity reduction approaches. The future trends of leveraging advanced NN in IM/DD links are also discussed.

Carbon balance of a primary tropical seasonal rain forest

The role of primary tropical rain forests in the global carbon cycle is under active debate. By combining long‐term forest inventory data with physiological measurement data in a 1 ha permanent ecological research plot beneath an eddy covariance flux tower in a primary tropical seasonal rain forest, the ecosystem carbon balance was investigated and a detailed site‐specific carbon budget was established. The studied ecosystem was a carbon sink as determined by both eddy covariance (1.19 Mg C ha−1 yr−1) and biometric methods (3.59 Mg C ha−1 yr−1). Biometric‐ and eddy covariance‐based net ecosystem production showed no convergence in our investigation period. The large biomass increment, caused by the rapid annual growth rate of large trees, primarily accounted for the large ecosystem carbon sink derived from the biometric method. High leaf respiration in relation to carbon allocation and low ecosystem carbon use efficiency (0.34) were observed at our site.

The triterpenes are structurally diverse group of specialized metabolites with important roles in plant defense and human health. Glycyrrhizin, with a carboxyl group at C-30 of its aglycone moiety, is a valuable triterpene glycoside, the production of which is restricted to legume medicinal plants belonging to the
species. Cytochrome P450 monooxygenases (P450s) are important for generating triterpene chemodiversity by catalyzing site-specific oxidation of the triterpene scaffold. CYP72A154 was previously identified from the glycyrrhizin-producing plant
as a C-30 oxidase in glycyrrhizin biosynthesis, but its regioselectivity is rather low. In contrast, CYP72A63 from
showed superior regioselectivity in C-30 oxidation, improving the production of glycyrrhizin aglycone in engineered yeast. The underlying molecular basis of C-30 product regioselectivity is not well understood. Here, we identified two amino acid residues that control C-30 product regioselectivity and contribute to the chemodiversity of triterpenes accumulated in legumes. Amino acid sequence comparison combined with structural analysis of the protein model identified Leu149 and Leu398 as important amino acid residues for C-30 product regioselectivity. These results were further confirmed by mutagenesis of CYP72A154 homologs from glycyrrhizin-producing species, functional phylogenomics analyses, and comparison of corresponding residues of C-30 oxidase homologs in other legumes. These findings could be combined with metabolic engineering to further enhance the production of high-value triterpene compounds.

Joint SOP-Based and Fading-Suppressed Phase-Based Vibration Sensing Integrated in Short-Reach Optical Interconnects

With the advancement of artificial intelligence (AI) technologies such as large language models and autonomous driving, the data traffic via optical interconnects in data centers has surged significantly. The stability of the optical interconnects relies on intelligent operation and maintenance (O&M). Integrated sensing and communication (ISAC) over fibers enables vibration sensing utilizing existing communication fibers, providing critical support for intelligent O&M in data centers. Compared to sensing in the coherent systems, it is difficult to use phase and state of polarization (SOP) monitoring for vibration detection in intensity-modulation and direct-detection (IM-DD) systems. In this paper, we propose a joint phase-based and SOP-based sensing scheme integrated in IM-DD systems. In the proposed scheme, the received IM-DD communication signals are tapped for sensing with a power ratio of 10%. Then the tapped signals are split for vibration sensing based on SOP and phase, respectively. In the phase-based sensing arm, a circulator, a 3×3 coupler and two Faraday rotating mirrors (FRMs) are used to build an unbalanced Michelson interferometer without phase fading and polarization fading. For the purpose of SOP-based sensing, a polarizer is used to monitor the vibration-induced SOP variations. Experimental results demonstrate that the proposed scheme enables vibration sensing based on both phase and SOP across a frequency range of 200 Hz to 10 kHz. Regarding the communication performance, the integration of the sensing system only induces 0.8 dB received optical power penalty. This vibration-sensing scheme based on both phase and SOP can be integrated into pluggable optical modules, providing an efficient and reliable solution for intelligent optical network O&M.
