SARS-CoV-2 and viral sepsis: observations and hypotheses
Since the outbreak of coronavirus disease 2019 (COVID-19), clinicians have tried every effort to understand the disease, and a brief portrait of its clinical features have been identified. In clinical practice, we noticed that many severe or critically ill COVID-19 patients developed typical clinical manifestations of shock, including cold extremities and weak peripheral pulses, even in the absence of overt hypotension. Understanding the mechanism of viral sepsis in COVID-19 is warranted for exploring better clinical care for these patients. With evidence collected from autopsy studies on COVID-19 and basic science research on severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and SARS-CoV, we have put forward several hypotheses about SARS-CoV-2 pathogenesis after multiple rounds of discussion among basic science researchers, pathologists, and clinicians working on COVID-19. We hypothesise that a process called viral sepsis is crucial to the disease mechanism of COVID-19. Although these ideas might be proven imperfect or even wrong later, we believe they can provide inputs and guide directions for basic research at this moment.
Concise Review: Is Cardiac Cell Therapy Dead? Embarrassing Trial Outcomes and New Directions for the Future
Stem cell therapy is a promising strategy for tissue regeneration. The therapeutic benefits of cell therapy are mediated by both direct and indirect mechanisms. However, the application of stem cell therapy in the clinic is hampered by several limitations. This concise review provides a brief introduction into stem cell therapies for ischemic heart disease. It summarizes cell‐based and cell‐free paradigms, their limitations, and the benefits of using them to target disease. Assessment of the Genetic Diversity of Different Job's Tears (Coix lacryma-jobi L.) Accessions and the Active Composition and Anticancer Effect of Its Seed Oil
Job's tears (Coix lachryma-jobi L.) is an important crop used as food and herbal medicine in Asian countries. A drug made of Job's tears seed oil has been clinically applied to treat multiple cancers. In this study, the genetic diversity of Job's tears accessions and the fatty acid composition, triglyceride composition, and anti-proliferative effect of Job's tears seed oil were analyzed using morphological characteristics and ISSR markers, GC-MS, HPLC-ELSD, and the MTT method. ISSR analysis demonstrated low genetic diversity of Job's tears at the species level (h = 0.21, I = 0.33) and the accession level (h = 0.07, I = 0.10), and strong genetic differentiation (GST = 0.6702) among all accessions. It also clustered the 11 accessions into three cultivated clades corresponding with geographical locations and two evidently divergent wild clades. The grouping patterns based on morphological characteristics and chemical profiles were in accordance with those clustered by ISSR analysis. Significant differences in morphological characteristics, fatty acid composition, triglyceride composition, and inhibition rates of seed oil were detected among different accessions, which showed a highly significant positive correlation with genetic variation. These results suggest that the seed morphological characteristics, fatty acid composition, and triglyceride composition may be mainly attributed to genetic factors. The proportion of palmitic acid and linoleic acid to oleic acid displayed a highly significant positive correlation with the inhibition rates of Job's tears seed oil for T24 cells, and thus can be an important indicator for quality control for Job's tears.
The Hippo pathway links adipocyte plasticity to adipose tissue fibrosis
Fibrosis disrupts adipose tissue (AT) homeostasis and exacerbates metabolic dysfunction upon chronic caloric excess. The molecular mechanisms linking adipocyte plasticity to AT fibrosis are largely unknown. Here we show that the Hippo pathway is coupled with TGFβ signaling to orchestrate a cellular and/or functional shift of adipocytes from energy storage to extracellular matrix (ECM) remodeling in AT fibrosis. We found that Lats1/2 -knockout adipocytes could dedifferentiate into DPP4 + progenitor cells and convert to DPP4 − myofibroblasts upon TGFβ stimulation. On the other hand, Hippo pathway inhibition during obesity impaired adipocyte identity while promoted ECM remodeling activity of adipocytes. Macrophages recruited by CCL2 produced TGFβ to accelerate AT fibrosis. YAP and TAZ, the Hippo downstream effectors, enhanced SMAD2 stability to promote fibrotic responses. Importantly, inhibition of YAP/TAZ activity in obese mice markedly relieved AT fibrosis and improved metabolic homeostasis. Together, our findings identify the Hippo pathway as a molecular switch in the initiation and development of AT fibrosis, implying it as a therapeutic target. Adipose tissue fibrosis is connected to obesity-related metabolic dysfunction. Qiu and colleagues discover that the Hippo pathway acts as a molecular switch in the initiation and development of adipose tissue fibrosis upon TGFβ stimulation.
Ascorbic Acid Sensitizes Colorectal Carcinoma to the Cytotoxicity of Arsenic Trioxide via Promoting Reactive Oxygen Species-Dependent Apoptosis and Pyroptosis
Arsenic trioxide (ATO) is an effective therapeutic agent against acute promyelocytic leukemia (APL); however, its anti-tumor effect on solid tumors such as colorectal cancer (CRC) is still in debate. Ascorbic acid (AA) also produces a selective cytotoxic activity against tumor cells. Here, we exploit the potential benefit of ATO/AA combination in generating cytotoxicity to CRC cells, which may lay the groundwork for the potential combinational chemotherapy of CRCs. According to the results, we found that ATO and AA effectively inhibited the viability of human CRC cells in a synergistic manner. AA and ATO corporately activated caspase-3 to trigger apoptosis and upregulated the expression of caspase-1 and promoted formation of inflammasomes to induce pyroptosis. Furthermore, the stimulation of reactive oxygen species (ROS) overproduction was demonstrated as a subcellular mechanism for apoptosis and pyroptosis induced by ATO/AA combination treatment. Our findings suggest that ATO combination with a conventional dosage of AA offers an advantage for killing CRC cells. The synergistic action of ATO/AA combination might be considered a plausible strategy for the treatment of CRC and perhaps other solid tumors as well.
Insights Into Amentoflavone: A Natural Multifunctional Biflavonoid
Amentoflavone is an active phenolic compound isolated from Selaginella tamariscina over 40 years. Amentoflavone has been extensively recorded as a molecule which displays multifunctional biological activities. Especially, amentoflavone involves in anti-cancer activity by mediating various signaling pathways such as extracellular signal-regulated kinase (ERK), nuclear factor kappa-B (NF-κB) and phosphoinositide 3-kinase/protein kinase B (PI3K/Akt), and emerges anti-SARS-CoV-2 effect via binding towards the main protease (Mpro/3CLpro), spike protein receptor binding domain (RBD) and RNA-dependent RNA polymerase (RdRp) of SARS-CoV-2. Therefore, amentoflavone is considered to be a promising therapeutic agent for clinical research. Considering the multifunction of amentoflavone, the current review comprehensively discuss the chemistry, the progress in its diverse biological activities, including anti-inflammatory, anti-oxidation, anti-microorganism, metabolism regulation, neuroprotection, radioprotection, musculoskeletal protection and antidepressant, specially the fascinating role against various types of cancers. In addition, the bioavailability and drug delivery of amentoflavone, the molecular mechanisms underlying the activities of amentoflavone, the molecular docking simulation of amentoflavone through in silico approach and anti-SARS-CoV-2 effect of amentoflavone are discussed.
Reproductive effects of cadmium on sperm function and early embryonic development in vitro
Cadmium is a major environmental toxicant that is released into the atmosphere, water and soil in the form of cadmium oxide, cadmium chloride, or cadmium sulfide via industrial activities, such as the manufacturing of batteries and pigments, metal smelting and refining and municipal waste incineration. In the present study, we investigated the effects of cadmium exposure on sperm quality parameters, fertilization capacity and early embryonic development. Our study showed that in vitro incubation of human or mouse sperms with cadmium for a long time (up to 24 hours) could significantly decreased sperm motility in a concentration- and time-dependent manner. Exposure to cadmium in the environment for a short term (30 min) did not affect sperm motility but significantly reduced in vitro fertilization rate. We also evaluated the effects of cadmium at concentrations of 0.625 μg/ml, and 1.25 μg/ml on early embryonic development in vitro and observed that the blastocyst formation rate dramatically decreased with increasing cadmium concentration. This finding emphasizes the hazardous effects of cadmium on sperm quality as well as on natural embryo development and raises greater concerns regarding cadmium pollution.
Effect of solution focused approach on women aged 35 or over with in vitro fertilization-embryo transfer: A quasi-experimental trial
We aimed to explore the influence of solution-focused approach (SFA) on anxiety and depression, sleep quality, quality of life and clinical pregnancy rate among women aged 35 or over undergoing in vitro fertilization-embryo transfer (IVF-ET). The study was performed at the reproductive center in a public hospital in Lanzhou city. Totally, 112 women were enrolled in this study, and were divided into group SFA (n = 56) and group control (n = 56). The patients in the group SFA completed five sessions (30 minutes/turn), and patients in the group control received routine care. The intervention group showed a significant decrease in anxiety (t =  11.906, P <  0.001) and depression scores (t =  14.991, P <  0.001), as well as PSQI scores (t =  7.055, P <  0.001), and increased FertiQoL scores (t =  -2.828, P <  0.001). Comparing the two groups after the intervention, the intervention group demonstrated significantly lower SAS scores (t =  -10.348, P <  0.001), SDS scores (t =  -8.416, P <  0.001), and PSQI scores (t =  -5.087, P <  0.001), while FertiQoL scores were higher than the control group (t =  2.389, P =  0.019). The intervention group reported a satisfaction rate of 96.2% to 100% with the SFA. SFA can help relieve anxiety and depression, improve sleep quality and reproductive life quality. Improvement in psychological distress might not contribute to increasing female fecundity. Patients in the group SFA were satisfied with the intervention. Estimation of Sugarcane Yield Using a Machine Learning Approach Based on UAV-LiDAR Data
Sugarcane is a multifunctional crop mainly used for sugar and renewable bioenergy production. Accurate and timely estimation of the sugarcane yield before harvest plays a particularly important role in the management of agroecosystems. The rapid development of remote sensing technologies, especially Light Detecting and Ranging (LiDAR), significantly enhances aboveground fresh weight (AFW) estimations. In our study, we evaluated the capability of LiDAR mounted on an Unmanned Aerial Vehicle (UAV) in estimating the sugarcane AFW in Fusui county, Chongzuo city of Guangxi province, China. We measured the height and the fresh weight of sugarcane plants in 105 sampling plots, and eight variables were extracted from the field-based measurements. Six regression algorithms were used to build the sugarcane AFW model: multiple linear regression (MLR), stepwise multiple regression (SMR), generalized linear model (GLM), generalized boosted model (GBM), kernel-based regularized least squares (KRLS), and random forest regression (RFR). The results demonstrate that RFR (R2 = 0.96, RMSE = 1.27 kg m−2) performs better than other models in terms of prediction accuracy. The final fitted sugarcane AFW distribution maps exhibited good agreement with the observed values (R2 = 0.97, RMSE = 1.33 kg m−2). Canopy cover, the distance to the road, and tillage methods all have an impact on sugarcane AFW. Our study provides guidance for calculating the optimum planting density, reducing the negative impact of human activities, and selecting suitable tillage methods in actual cultivation and production.
