Covid-19: implications for insurer risk management and the insurability of pandemic risk
This paper analyzes the insurability of pandemic risk and outlines how underwriting policies and scenario analysis are used to build resilience upfront and plan contingency actions for crisis scenarios. It then summarizes the unique “lessons learned” from the Covid-19 crisis by baselining actual developments against a reasonable, pre-Covid-19 pandemic scenario based on the 2002 SARS epidemic and 1918 Spanish influenza pandemic. Actual developments support the pre-Covid-19 hypothesis that financial market developments dominate claims losses due to the demographics of pandemics and other factors. However, Covid-19 “surprised” relative to the pre-Covid-19 scenario in terms of its impact on the real economy as well as on the property and casualty segment as business interruption property triggers and exclusions are challenged, something that may adversely impact the insurability of pandemics as well as the perception of the industry for some time to come. The unique lessons of Covid-19 reinforce the need for resilience upfront in solvency and liquidity, the need to improve business interruption wordings and re-underwrite the book, and the recognition that business interruption caused by pandemics may not be an insurable risk due to its large accumulation potential and the threat of external moral hazard. These insurability limitations lead to a discussion about the structure and financing of protection against the impact of future pandemics.

Fragile sites in cancer: more than meets the eye
This Opinion article discusses recent studies that have provided new insights into the mechanisms of common fragile site instability and the resulting genomic effects, which include the generation of focal copy number alterations that affect the genomic landscape of many cancers. Ever since initial suggestions that instability at common fragile sites (CFSs) could be responsible for chromosome rearrangements in cancers, CFSs and associated genes have been the subject of numerous studies, leading to questions and controversies about their role and importance in cancer. It is now clear that CFSs are not frequently involved in translocations or other cancer-associated recurrent gross chromosome rearrangements. However, recent studies have provided new insights into the mechanisms of CFS instability, their effect on genome instability, and their role in generating focal copy number alterations that affect the genomic landscape of many cancers.

This memoir tells the story of one aspiring pilgrim, Clive Wilson, who fell in love with New Orleans jazz in his early teens while in boarding school in his native England. Replication stress induces POLQ-mediated structural variant formation throughout common fragile sites after entry into mitosis
Genomic structural variants (SVs) greatly impact human health, but much is unknown about the mechanisms that generate the largest class of nonrecurrent alterations. Common fragile sites (CFSs) are unstable loci that provide a model for SV formation, especially large deletions, under replication stress. We study SV junction formation as it occurs in human cell lines by applying error-minimized capture sequencing to CFS DNA harvested after low-dose aphidicolin treatment. SV junctions form throughout CFS genes at a 5-fold higher rate after cells pass from G2 into M-phase. Neither SV formation nor CFS expression depend on mitotic DNA synthesis (MiDAS), an error-prone form of replication active at CFSs. Instead, analysis of tens of thousands of de novo SV junctions combined with DNA repair pathway inhibition reveal a primary role for DNA polymerase theta (POLQ)-mediated end-joining (TMEJ). We propose an important role for mitotic TMEJ in nonrecurrent SV formation genome wide. The mechanisms of nonrecurrent structural variant (SV) formation are poorly understood. Here, the authors sequenced thousands of SV junctions as they formed at common fragile sites in human cell lines to reveal a primary role for DNA polymerase theta-mediated end joining activated during mitosis.
Titanium Corrosion Mechanisms in the Oral Environment: A Retrieval Study
Corrosion of titanium dental implants has been associated with implant failure and is considered one of the triggering factors for peri-implantitis. This corrosion is concerning, because a large amount of metal ions and debris are generated in this process, the accumulation of which may lead to adverse tissue reactions in vivo. The goal of this study is to investigate the mechanisms for implant degradation by evaluating the surface of five titanium dental implants retrieved due to peri-implantitis. The results demonstrated that all the implants were subjected to very acidic environments, which, in combination with normal implant loading, led to cases of severe implant discoloration, pitting attack, cracking and fretting-crevice corrosion. The results suggest that acidic environments induced by bacterial biofilms and/or inflammatory processes may trigger oxidation of the surface of titanium dental implants. The corrosive process can lead to permanent breakdown of the oxide film, which, besides releasing metal ions and debris in vivo, may also hinder re-integration of the implant surface with surrounding bone.
Detection of Ongoing Mass Loss from HD 63433c, a Young Mini-Neptune
We detect Lyα absorption from the escaping atmosphere of HD 63433c, a R = 2.67R ⊕, P = 20.5 day mini-Neptune orbiting a young (440 Myr) solar analog in the Ursa Major Moving Group. Using Hubble Space Telescope (HST)/Space Telescope Imaging Spectrograph, we measure a transit depth of 11.1 ± 1.5% in the blue wing and 8 ± 3% in the red. This signal is unlikely to be due to stellar variability, but should be confirmed by an upcoming second transit observation with HST. We do not detect Lyα absorption from the inner planet, a smaller R = 2.15R ⊕ mini-Neptune on a 7.1 day orbit. We use Keck/NIRSPEC to place an upper limit of 0.5% on helium absorption for both planets. We measure the host star’s X-ray spectrum and mid-ultraviolet flux with XMM-Newton, and model the outflow from both planets using a 3D hydrodynamic code. This model provides a reasonable match to the light curve in the blue wing of the Lyα line and the helium nondetection for planet c, although it does not explain the tentative red wing absorption or reproduce the excess absorption spectrum in detail. Its predictions of strong Lyα and helium absorption from b are ruled out by the observations. This model predicts a much shorter mass-loss timescale for planet b, suggesting that b and c are fundamentally different: while the latter still retains its hydrogen/helium envelope, the former has likely lost its primordial atmosphere.
