SLIDE—Novel Approach to Apocrine Sweat Sampling for Lipid Profiling in Healthy Individuals

We designed a concept of 3D-printed attachment with porous glass filter disks—SLIDE (Sweat sampLIng DevicE) for easy sampling of apocrine sweat. By applying advanced mass spectrometry coupled with the liquid chromatography technique, the complex lipid profiles were measured to evaluate the reproducibility and robustness of this novel approach. Moreover, our in-depth statistical evaluation of the data provided an insight into the potential use of apocrine sweat as a novel and diagnostically relevant biofluid for clinical analyses. Data transformation using probabilistic quotient normalization (PQN) significantly improved the analytical characteristics and overcame the ‘sample dilution issue’ of the sampling. The lipidomic content of apocrine sweat from healthy subjects was described in terms of identification and quantitation. A total of 240 lipids across 15 classes were identified. The lipid concentrations varied from 10−10 to 10−4 mol/L. The most numerous class of lipids were ceramides (n = 61), while the free fatty acids were the most abundant ones (average concentrations of 10−5 mol/L). The main advantages of apocrine sweat microsampling include: (a) the non-invasiveness of the procedure and (b) the unique feature of apocrine sweat, reflecting metabolome and lipidome of the intracellular space and plasmatic membranes. The SLIDE application as a sampling technique of apocrine sweat brings a promising alternative, including various possibilities in modern clinical practice.

The Impact of Glucose-Based or Lipid-Based Total Parenteral Nutrition on the Free Fatty Acids Profile in Critically Ill Patients

Introduction: Our study aim was to assess how the macronutrient intake during total parenteral nutrition (TPN) modulates plasma total free fatty acids (FFAs) levels and individual fatty acids in critically ill patients. Method: Adult patients aged 18–80, admitted to the intensive care unit (ICU), who were indicated for TPN, with an expected duration of more than three days, were included in the study. Isoenergetic and isonitrogenous TPN solutions were given with a major non-protein energy source, which was glucose (group G) or glucose and lipid emulsions (Smof lipid; group L). Blood samples were collected on days 0, 1, 3, 6, 9, 14, and 28. Results: A significant decrease (p < 0.001) in total FFAs occurred in both groups with a bigger decrease in group G (p < 0.001) from day 0 (0.41 ± 0.19 mmol∙L−1) to day 28 (0.10 ± 0.07 mmol∙L−1). Increased palmitooleic acid and decreased linoleic and docosahexaenoic acids, with a trend of increased mead acid to arachidonic acid ratio, on day 28 were observed in group G in comparison with group L. Group G had an insignificant increase in leptin with no differences in the concentrations of vitamin E, triacylglycerides, and plasminogen activator inhibitor-1. Conclusion: Decreased plasma FFA in critically ill patients who receive TPN may result from increased insulin sensitivity with a better effect in group G, owing to higher insulin and glucose dosing and no lipid emulsions. It is advisable to include a lipid emulsion at the latest from three weeks of TPN to prevent essential fatty acid deficiency.

Despite the significant interest in molecular hydrogen as an antioxidant in the last eight years, its quantitative metabolic parameters in vivo are still lacking, as is an appropriate method for determination of hydrogen effectivity in the mammalian organism under various conditions.
Intraperitoneally-applied deuterium gas was used as a metabolic tracer and deuterium enrichment was determined in the body water pool. Also, in vitro experiments were performed using bovine heart submitochondrial particles to evaluate superoxide formation in Complex I of the respiratory chain.
A significant oxidation of about 10% of the applied dose was found under physiological conditions in rats, proving its antioxidant properties. Hypoxia or endotoxin application did not exert any effect, whilst pure oxygen inhalation reduced deuterium oxidation. During in vitro experiments, a significant reduction of superoxide formation by Complex I of the respiratory chain was found under the influence of hydrogen. The possible molecular mechanisms of the beneficial effects of hydrogen are discussed, with an emphasis on the role of iron sulphur clusters in reactive oxygen species generation and on iron species-dihydrogen interaction.
According to our findings, hydrogen may be an efficient, non-toxic, highly bioavailable and low-cost antioxidant supplement for patients with pathological conditions involving ROS-induced oxidative stress.

Housing Market Trends and Affordability in Central Europe: Insights from the Czech Republic, Slovakia, Austria, and Poland

This study examines housing affordability trends in Central Europe, focusing on the Czech Republic, Slovakia, Austria, and Poland, in the wake of recent global disruptions including the COVID-19 pandemic, the 2021–2022 energy crisis, and the war in Ukraine. These events have intensified housing affordability challenges by driving up property prices, rental costs, and energy expenses. Using data from December 2022 to March 2023, the paper analyzes wage levels relative to housing costs in major cities—Prague, Brno, Bratislava, Vienna, Graz, Warsaw, and Kraków—through price-to-income and rent-to-income ratios. The findings reveal that affordability is most strained in Czech cities, particularly Prague, where property prices outpace wages, while Vienna demonstrates better affordability due to higher average incomes. The study integrates real estate platform data with official statistics and employs spatial mapping and exploratory econometric testing to identify affordability patterns and disparities. It concludes that affordability outcomes are shaped by wage dynamics, housing supply constraints, migration pressures, and policy responses. The study underscores the importance of targeted housing policies and wage interventions to address these challenges and highlights the need for cross-country policy learning and regional coordination to improve housing affordability and market resilience across Central Europe.

Global Pandemic as an Innovative Impulse for the Labour Market

Research background: The global history of mankind in an international context is permeated by the painful events include epidemics, wars, natural disasters and crises, including economic ones. Purpose of the article: According to research, it will be the generation of hatcheries from digital talents. In the work environment, however, centennials behave differently than all previous generations. Millenials is a fully technological generation with great self-confidence, but at the same time an autonomous willingness to work hard, assertive and much more demanding operating with completely different and international communication tools. Methods: The article will analyses the surveys of international research about the international crisis caused by the pandemic by methods of mathematical statistics. It has paradoxically accelerated digitization in fields where it has been difficult to assert itself so far. Findings & Value added :.Along with the changing behavior of the next generation and the advancing digitization and automation of work, many professions are at risk. The article deals with the possible development of the labor market in connection with the complete disappearance of some professions, but also the emergence of new professions, which now cannot even be imagined. Several weeks of quarantine, caused by a pandemic of a new type of coronavirus, has changed the lives of many companies and employees. For some companies, it meant their definitive end. For others, however, it foreshadowed the acceleration of innovation in the company itself.

Different Forms of Ursolic Acid and Their Effect on Liver Regeneration

The aim of this study was to determine the effect of natural and encapsulated sources of ursolic acid on liver regeneration. Four ursolate sources were tested. Two forms of ursolic acid encapsulates were combined with cyclodextrins, i.e., gamma-CD (gCD) and beta-CD, and two natural sources were adjusted by homogenization (HAP) and micronization of apple peel using Jonagold apples. All ursolate forms were applied intragastrically in daily doses of 20 mg for 7 days. Laboratory rats were fed with standard laboratory diet. Further, gCD and MAP were also tested with a high-fat diet (6 weeks). Partial hepatectomy (PH) was performed 24 hours before the end of the experiment. The concentration of plasma hepatocyte growth factor (HGF) was determined with an immunoassay; simultaneously, the expression of HGF and CYP7A1 in the liver was quantified through qPCR. HGF expression and plasma levels were significantly increased 24 hours after PH in both the HAP (p=0.038) and HFgCD groups (p=0.036), respectively. The correlation between HGF expression and plasma values was significant (p=0.04). The positive effects on liver regeneration were found in both the gCD and HAP forms of ursolic acid, whose effects were confirmed through the upregulation of HGF.

Effective Method of Purification of Betulin from Birch Bark: The Importance of Its Purity for Scientific and Medicinal Use

A new and relatively simple method for purification of betulin from birch bark extract was developed in this study. Its five purification steps are based on the differential solubility of extract components in various solvents and their crystallization and/or precipitation, on their affinity for Ca(OH)2 in ethanol, and on the affinity of some impurities for silica gel in chloroform. In addition, all used solvents can be simply recycled. Betulin of more than 99% purity can be prepared by this method with minimal costs. Various observations including crystallization of betulin, changes in crystals during heating, and attempt of localization of betulin in outer birch bark are also described in this work. The original extract, fraction without betulinic acid and lupeol, amorphous fraction of pure betulin, final crystalline fraction of pure betulin and commercial betulin as a standard were employed to determine the antiproliferative/cytotoxic effect. We used WST-1 tetrazolium-based assays with triple negative breast cancer cell line BT-549. The decrease in cell survival showed clear relationship with the purity of the samples, being most pronounced using our final product of pure crystalline betulin. WST-1 proliferation/cytotoxicity test using triple negative breast cancer cell line BT-549 clearly showed the importance of purity of betulin for biological experiments and, apparently, for its medicinal use.

Differences in the Cost Calculation for Construction Work, Road Transport and Water Supply

This paper presents a method of cost and price calculation of construction work, road transport and water and sewerage charges in the Czech Republic. Costs represent an important economic category that is closely related to the price. Each company must perfectly manage costing, especially absorption costing, which consists of budgeting overhead costs on the production (performance, products and services). Absorption costing is characterized by including indirect (overhead) costs together with direct costs of production into the calculation, i.e. it must absorb them completely into the costs. The absorption method is represented by the surcharge calculation. However, creating the calculation formula for absorption costing and the procedure of cost and price calculation have their specifics in various branches of the economy. Differences can be seen in examples of selected case studies. They deal with the representative cost and price calculation of construction work on a particular construction, cost and price calculation of the road transport and one-component and two-component cost and price calculation of the water and sewerage charges. All types of calculation must reflect the economic situation of the company marketing of a new product. The diversity of the product types mentioned above indicates the need for other inputs in the calculation and different calculation procedures. Without mastering absorption costing, it is not possible to introduce more accurate calculation methods, such as the Dynamic Calculation with Contribution Margin or in the next step implement the Balanced Scorecard. Correct absorption costing can be checked by the fact that the sum of all company production costs must correspond to the sum of the financial accounting cost accounts. However, cost control is not the subject of this article.

Promising Protocol for In Vivo Experiments with Betulin

Background/Objectives: Betulin is a promising agent in many areas of medicine and is being investigated, particularly in the field of cancer. However, in in vivo experiments, its water insolubility becomes a significant obstacle. This study describes a promising method for the administration of betulin in in vivo experiments and the determination of betulin levels in organ samples. Methods: Betulin was first dissolved in ethanol, and this solution was then mixed with acylglycerols, followed by evaporation of the ethanol. Olive oil and food-grade lard were determined to be suitable lipids for noninvasive application per os. A method for processing the organs of experimental animals for betulin determination was developed. Determination of betulin levels in blood is also likely the only viable option for use in future clinical studies and practice. Results: The maximum amount of betulin usable (i.e., absorbable by organisms) in olive oil (10 mg/mL), suppository mass (6 mg/mL), food lard (4 mg/mL), and cocoa butter (2 mg/mL) carriers was found microscopically. A specific distribution of betulin concentration in the organs of experimental animals (Wistar rats) after a weekly diet containing betulin was discovered. The blood was shown to be particularly advantageous, as it allows continuous monitoring of betulin levels in the body. In these pilot experiments, a statistically significant (p < 0.001) synergistic effect of betulin on solid Ehrlich adenocarcinoma tumors was observed when betulin was combined with cytostatic Namitecan (NMRI mice). The high-purity betulin used in this study is very stable even under fluctuating storage conditions. Conclusions: Our study suggests that both the method of betulin administration and the proposed analytical procedure could greatly increase the reliability and reproducibility of in vivo studies, as well as future preclinical and clinical studies on the effects of betulin and potentially other similar water-insoluble triterpenoids on living organisms.

Hydraulic sizing of forest road pipe culverts

This article presents guidelines for assessing the optimal dimensions of forest road pipe culverts, based on input of actual and experimental data to standard engineering techniques. In doing so, we assess the need for (i) changes in the parametrisation of inputs (i.e. culvert micro-catchment dimensions, rainfall and resultant culvert flow, and culvert flow rates during culvert hydraulic dimensioning), and (ii) the need to redesign culvert outlets in relation to flow speed. Our results demonstrate that values for most inputs presently used under current technical practice for forest road pipe culvert sizing are significantly higher than those achieved under experimental conditions. The data on outlet flow velocities strongly suggests that strengthening of culvert outlet aprons will be crucial for their future operation.
