. We identified potential targets for repurposing of licensed medications: using Mendelian randomization, we found evidence that low expression of
IFNAR2
, or high expression of

, are associated with life-threatening disease; and transcriptome-wide association in lung tissue revealed that high expression of the monocyte–macrophage chemotactic receptor
CCR2
is associated with severe COVID-19. Our results identify robust genetic signals relating to key host antiviral defence mechanisms and mediators of inflammatory organ damage in COVID-19. Both mechanisms may be amenable to targeted treatment with existing drugs. However, large-scale randomized clinical trials will be essential before any change to clinical practice.
A genome-wide association study of critically ill patients with COVID-19 identifies genetic signals that relate to important host antiviral defence mechanisms and mediators of inflammatory organ damage that may be targeted by repurposing drug treatments.

Mosaic chromosomal anomalies arising in the product of conception and the final fetal chromosomal arrangement are expression of complex biological mechanisms. The rescue of unbalanced chromosome with selection of the most viable cell line/s in the embryo and the unfavourable imbalances in placental tissues was documented in our previous paper and in the literature. We report four additional cases with mosaic derivative chromosomes in different feto-placental tissues, further showing the instability of an intermediate gross imbalance as a frequent mechanism of de novo cryptic deletions and duplications. In conclusion we underline how the extensive remodeling of unbalanced chromosomes in placental tissues represents the ‘backstage’ of de novo structural rearrangements, as the early phases of a long selection process that the genome undergo during embryogenesis.

A Questionnaire Integrated with the Digital Medical Record Improved the Coverage of a Control Program for Congenital Chagas Disease in Tuscany, Italy

The leading route of Chagas disease transmission in nonendemic countries is congenital. However, policies concerning screening, prevention, and management of congenital Chagas disease are rare in these settings. Since 2012, serological screening for Chagas disease should be provided for pregnant women at risk in Tuscany, Italy according to a Regional resolution. Due to difficulties in the implementation, in November 2019, a checklist aimed at identifying pregnant women at risk for Chagas disease was introduced in digital clinical records at Careggi University Hospital, Florence, Italy. In order to evaluate the effectiveness of the “Chagas checklist”, data about the number of deliveries by women at risk and their screening coverage between 2012 and June 2022 were collected. Out of 1348 deliveries by women at risk, 626 (47%) Trypanosoma cruzi serology tests were performed during the study period. The annual screening coverage increased from an average of 40.3% between 2012 and 2019 to 75.7% between 2020 and June 2022, underlining the big impact of the checklist. Four Chagas disease serological tests out of 626 (0.6%) resulted positive, corresponding to 2 affected women. No cases of congenital transmission occurred. The study showed that a simple digital tool led to a tangible improvement in the coverage of the screening program; its application in a setting where digital charts are available will contribute to the control and elimination of congenital Chagas disease.

WoT Store: a Thing and Application Management Ecosystem for the W3C Web of Things

In the next few years, the W3C Web of Things (WoT) platform will represent a reference solution toward the deployment of fully interoperable systems, hence unlocking the potential of the IoT paradigm on several use-cases characterized by the current fragmentation of devices and technologies. At the same time, the worlwide adoption of the W3C WoT architecture depends on many factors, including also the availability of support tools that might facilitate the deployment of novel WoT applications or the integration with traditional IoT systems. To this purpose, the paper presents the WoT Store, a complete software platform enabling the discovery and management of W3C Things, the monitoring of its properties and events, and the invoking of actions, all within the same dashboard. In addition, the platform leverages on the semantic description of each Thing with the goal of easing and automatizing the installation and execution of WoT applications, e.g. defining the behaviour of a Thing or implementing mash-up operations from multiple Things. We sketch the main features, the architecture and the prototypal implementation of the WoT Store. Moreover, we discuss the WoT Store capabilities on three IoT use-cases, i.e. industry 4.0, smart agriculture and home automation.
