Preliminary Insights into the Fluorescence and Oxidative Characteristics of Flavin - DNA Systems on PVP - Coated Silver Nanoparticles
Emissive features of flavins (Riboflavin/RF, Flavin MonoNucleotide/FMN and Flavin Adenine Dinucleotide/FAD) labeled native Deoxyribonucleic Acid (DNA) on Polyvinylpyrrolidone (PVP)-coated silver nanoparticles (SNPs), have been studied. The dual emission of flavins in DNA−PVP-coated SNPs systems is strongly influenced by the reaction time and temperature. Changes in the RF emissive features occur as a side effect when DNA is covalently linked hence, the RF destruction depends on DNA damage. Even if in an oxidation process, the FAD-DNA – PVP-coated SNPs system acts as a weak scavenger of reactive oxygen species, its antioxidant activity is approx. five times higher than that of RF-DNA−PVP-coated SNPs system. Destruction of RF by a riboflavin-mediated DNA photo-oxidation process that occurs on PVP-coated SNPs is suggested. Results have relevance in the redox process of riboflavin and provide valuable information for the further development of novel flavin-based SNPs systems as fluorescent antioxidant markers to solve several biological barriers in humans, such as protein-DNA interaction, cell binding.
Cerium-Containing Mesoporous Bioactive Glasses (MBGs)-Derived Scaffolds with Drug Delivery Capability for Potential Tissue Engineering Applications
Finding innovative solutions to improve the lives of people affected by trauma, bone disease, or aging continues to be a challenge worldwide. Tissue engineering is the most rapidly growing area in the domain of biomaterials. Cerium-containing MBG-derived biomaterials scaffolds were synthesized using polymethyl methacrylate (PMMA) as a sacrificial template. The obtained scaffolds were characterized by X-ray powder diffraction (XRPD), infrared spectroscopy (FTIR), and scanning electron microscopy (SEM). The Ce4+/Ce3+ ratio in the scaffolds was estimated. In vitro testing revealed good cytocompatibility of the investigated scaffolds in mouse fibroblast cell line (NCTC clone L929). The results obtained regarding bioactivity, antibacterial activity, and controlled drug delivery functions recommend these scaffolds as potential candidates for bone tissue engineering applications.
Fluorescent Flavin/PVP-Coated Silver Nanoparticles: Design and Biological Performance
A red-emitting fluorescent Riboflavin (RF)/Polyvinylpyrrolidone (PVP)-coated silver nanoparticles system, λem = 527 nm, Φ = 0.242, with a diameter of the metallic core of 27.33 nm and a zeta potential of − 25.05 mV was prepared and investigated regarding its biological activity. We found that PVP has a key role in RF adsorption around the SNPs surface leading to an enhancement of antioxidant properties (∼70%), low cytotoxicity (> 90% cell viability, at 50 µL/mL, after 48 h of incubation) as well as to an efficient process of its cellular uptake (∼ 60%, after 24 h of incubation) in L929 cells. The results are relevant concerning the involvement of RF and its coenzymes forms in SNPs - based systems, in cellular respiration as well as for future studies as antioxidant marker system on tumoral cells for viewing and monitoring them, by cellular imaging.
A curcumin-loaded silica carrier with NH3 sensitivity and antimicrobial properties
In this study curcumin was immobilized in a silica matrix to overcome its poor aqueous solubility and rapid degradation profile. Two kinds of sol–gel powders based on colloidal silica were synthesized and characterized, one prepared with an ethanolic solution of curcumin and the other using cethyltrimethylammonium bromide (CTAB) to solubilize the dye. The successful integration of curcumin in the matrix as well as the interaction with the components of the composite powders was validated by different analysis methods (DSC, FTIR). The samples morphology was evaluated by SEM. UV–Vis and fluorescence studies showed that curcumin was stable, in its enol configuration in both matrices. The sensitivity to ammonia was proved by exposing the powders to NH3 vapors in a sealed vessel. Color changes were measured with a colorimeter. UV–Vis spectra showed that the enol form of the dye in EtOH/CTAB based SiO2 matrices was energetically stable under NH3 atmosphere, even after being kept a few days at low temperature. A fluorescence quenching of both samples was noticed due to interaction curcumin-ammonia. Both samples have antimicrobial properties, the CTAB containing one being more efficient. The EtOH/CTAB based SiO2 powders may represent a novel signaling and antimicrobial system, useful for food or pharmaceutical applications.Graphical Abstract
Preliminary Biological Assessments of Some Algae Basis Biomaterials
Four types of algae—Porphyra umbilicalis, Undaria pinnatifida, Cystoseira barbata, and Chlorella sp.—were used to obtain crude bioproducts enriched in polysaccharides (four bioproducts) and to create formulations enriched with gold cations (four bioproducts). The bioproducts obtained through aqueous extraction from Cystoseira barbata exhibited significant antioxidant activities and a total polyphenol content of (714.17 ± 1.26) mg GAE/L. In the bioproducts derived from the aqueous extract of Porphyra umbilicalis and Undaria pinnatifida, combined with gold ions, gold nanoparticles with sizes of less than 34 nm were formed. In vitro tests performed on the Caco-2 tumour cell line with each of the eight bioproducts, after 24 h of exposure, showed that the crude bioproducts containing polysaccharides derived from Porphyra umbilicalis, Undaria pinnatifida, and Chlorella sp. exhibited cytotoxicity against the Caco-2 cell line. In the case of the HepG2 cell line, after 24 h of exposure, the tests indicated that only the crude polysaccharides derived from Cystoseira barbata exhibited cytotoxic effects. These results indicate the protective effect of the algal polysaccharides against the tumourigenesis processes that may occur in the human digestive system. Regarding the bioproducts containing gold, no cytotoxic effect was observed. However, in the case of the two algal bioproducts containing gold nanoparticles with a size of less than 34 nm, they may represent potential raw materials for electrochemical sensors.
Photophysical Properties of Some Flavones Probes in Homogeneous Media
Photophysical properties of five hydroxyflavones (HF) (some typical models of flavonols), (3 - HF, 6 - HF, 7-HF, 3, 6 - diHF and 3, 7 - diHF) were studied in homogeneous media by means of UV–vis and steady-state and time resolved fluorescence spectroscopies. Their absorption and fluorescence characteristics based on the flavonols structure are presented and discussed. It was found that the fluorescence of the flavonols depends on the nature of the solvent and on their molecular structure, especially on the position and the number of the -OH groups of the substituted phenyl ring. Attention is paid to the number of the -OH groups that influence the excited-state intramolecular proton transfer (ESIPT) process. The fluorescence quantum yield and the lifetime of the flavonols in heterogeneous media have been also determined. The results are discussed with relevance to the flavonols as sensitive fluorescence probe and to their microenvironments in the systems of biological interest and especially in a typical protein environment.
Studies Regarding the Pharmaceutical Potential of Derivative Products from Plantain
In this study, three types of extracts isolated from leaves of Plantain (Plantago lanceolata) were tested for their chemical content and biological activities. The three bioproducts are combinations of polysaccharides and polyphenols (flavonoids and iridoidic compounds), and they were tested for antioxidant, antifungal, antitumor, and prebiotic activity (particularly for polysaccharides fraction). Briefly, the iridoid-enriched fraction has revealed a pro-oxidant activity, while the flavonoid-enriched fraction had a high antioxidant potency; the polysaccharide fraction also indicated a pro-oxidant activity, explained by the co-presence of iridoid glycosides. All three bioproducts demonstrated moderate antifungal effects against Aspergillus sp., Penicillium sp., and dermatophytes, too. Studies in vitro proved inhibitory activity of the three fractions on the leukemic tumor cell line THP-1, the main mechanism being apoptosis stimulation, while the polysaccharide fraction indicated a clear prebiotic activity, in the concentration range between 1 and 1000 µg/mL, evaluated as higher than that of the reference products used, inulin and dextrose, respectively.
Synthesis and properties of fluorescent 4′-azulenyl-functionalized 2,2′:6′,2″-terpyridines
4′-Azulenyl-substituted terpyridines were efficiently synthesized following the Kröhnke methodology via azulenylchalcone intermediates. These azulenyl-containing terpyridines showed fluorescent emission with a fluorescence quantum yield varying from 0.14, in the case of parent terpyridine, to 0.64 when methyl groups are grafted on the azulenyl seven-membered ring. According to the crystal structures and TDDFT calculations, different twisting of the aromatic constituents is responsible for the observed fluorescent behavior. The electrochemical profile contains one-electron oxidation/reduction steps, which can only be explained on the basis of the redox behavior of the azulene unit. The ability of the 4′-azulenyl 2,2′:6′,2″-terpyridine to bind poisoning metal cations was studied by UV–vis titrations using aqueous solutions of Hg(II) and Cd(II) chlorides as illustrative examples.
Ecological formulation for improving resveratrol stability and release in aqueous environment
AbstractResveratrol (RES) is a naturally occurring product with numerous biological activities. Despite its potential benefits, its use is limited due to its low aqueous stability and solubility in its native form. The porous sol–gel silica materials which are able to entrap different organic molecules represent new studied release carriers. The aim of this work was to generate a solid matrix to encapsulate RES ensuring protection, increased solubility and release in solutions. A non-toxic ingredient, namely β-cyclodextrin (β-CD), able to form inclusion complexes (ICs) with RES has been used. Ecological formulations have been processed by entrapping the RES containing ICs in silica matrices obtained from a silica colloidal sol by the aqueous route of the sol–gel method. Characterization methods (DSC, FTIR, UV–Vis, fluorescence studies, SEM) have evidenced the presence of RES–β-CD inclusion complex in the silica powder, RES stability in the matrix and its release in aqueous and organic solutions, and the morphology of the carrier. An evaluation of the antioxidant activity of RES in the present formulation was performed by the chemiluminescence assay and RES release profile in aqueous solutions was obtained by HPLC–MS. Preparation and Biocompatibility of Poly Methyl Methacrylate (PMMA)-Mesoporous Bioactive Glass (MBG) Composite Scaffolds
In recent years, the rising number of bone diseases which affect millions of people worldwide has led to an increased demand for materials with restoring and augmentation properties that can be used in therapies for bone pathologies. In this work, PMMA- MBG composite scaffolds containing ceria (0, 1, 3 mol%) were obtained by the phase separation method. The obtained composite scaffolds were characterized by X-ray diffraction, infrared spectroscopy, and scanning electron microscopy. UV–Vis measurement and EDX analysis confirmed the presence of cerium ions in the composite scaffolds. Evaluation of the in-vitro biocompatibility using MTT assay showed that composite scaffold containing 1 mol% of ceria presented higher viability than control cells (100%) for concentrations ranging between 5 and 50% after 96 h of incubation.
