Stage specific requirement of platelet-derived growth factor receptor-α in embryonic development
Platelet-derived growth factor receptor alpha (PDGFRα) is a cell-surface receptor tyrosine kinase for platelet-derived growth factors. Correct timing and level of Pdgfra expression is crucial for embryo development, and deletion of Pdgfra caused developmental defects of multiple endoderm and mesoderm derived structures, resulting in a complex phenotypes including orofacial cleft, spina bifida, rib deformities, and omphalocele in mice. However, it is not clear if deletion of Pdgfra at different embryonic stages differentially affects these structures. To address the temporal requirement of Pdgfra in embryonic development. We have deleted the Pdgfra in Pdgfra-expressing tissues at different embryonic stages in mice, examined and quantified the developmental anomalies. Current study showed that (i) conditional deletion of Pdgfra at different embryonic days (between E7.5 and E10.5) resulted in orofacial cleft, spina bifida, rib cage deformities, and omphalocele, and (ii) the day of Pdgfra deletion influenced the combinations, incidence and severities of these anomalies. Deletion of Pdgfra caused apoptosis of Pdgfra-expressing tissues, and developmental defects of their derivatives. Orofacial cleft, spina bifida and omphalocele are among the commonest skeletal and abdominal wall defects of newborns, but their genetic etiologies are largely unknown. The remarkable resemblance of our conditional Pdgfra knockout embryos to theses human congenital anomalies, suggesting that dysregulated PDGFRA expression could cause these anomalies in human. Future work should aim at defining (a) the regulatory elements for the expression of the human PDGFRA during embryonic development, and (b) if mutations / sequence variations of these regulatory elements cause these anomalies.
Real-World Measurement of Hybrid Buses’ Fuel Consumption and Pollutant Emissions in a Metropolitan Urban Road Network
This study investigates pollutant emissions and fuel consumption of six Euro VI hybrid-diesel public transport buses operating on different scheduled routes in a metropolitan urban road network. Portable emission measurement systems (PEMS) are used in measurements and results are compared to those obtained from a paired number of Euro V conventional buses of the same body type used as control over the same routes. The selected routes vary from urban to highway driving and the experimentation was conducted over the first half of 2015. The available emissions data correspond to a wide range of driving, operating, and ambient conditions. Fuel consumption, distance- and energy-based emission levels are derived and presented in a comparative manner. The effect of different factors, including speed, ambient temperature, and road grade on fuel consumption and emissions performance is investigated. Mean fuel consumption of hybrid buses was found 6.1% lower than conventional ones, from 20% lower up to 16% higher, over six routes tested in total. The mean route difference between the two technologies was not statistically significant. Air conditioning decreased consumption benefits of the hybrid buses. Decrease of the mean route speed from 15 km h−1 tο 8 km h−1 increased the hybrid buses consumption by 63%. Nitrogen oxides (NOx) emissions of the Euro VI hybrid buses were 93 ± 5% lower than conventional Euro V ones. Nitrous oxide (N2O) emissions from hybrid Euro VI buses made up 5.9% of total greenhouse gas emissions and largely offset carbon dioxide (CO2) benefits. The results suggest that hybrid urban buses need to be assessed under realistic operation and environmental conditions to assess their true environmental and fuel consumption benefits.
CTGF is a central mediator of tissue remodeling and fibrosis and its inhibition can reverse the process of fibrosis
CTGF is a secreted matricellular protein with very complex biology. It has been shown to modulate many signaling pathways leading to cell adhesion and migration, angiogenesis, myofibroblast activation, and extracellular matrix deposition and remodeling, which together lead to tissue remodeling and fibrosis. It has been reported in the literature that inhibition of CTGF expression by siRNA prevents CCl 4 -induced liver fibrosis and can reverse fibrosis when administered after significant collagen deposition is observed. A monoclonal antibody to CTGF that is currently in clinical development (FG-3019) has demonstrated the ability to reverse vascular stiffening and improve cardiac function in a rat model of diabetic complications. FG-3019 has also exhibited activity in a murine radiation-induced pulmonary fibrosis model. When FG-3019 was administered to mice after a significant radiation-induced increase in lung density could be observed by CT imaging, the density of the lungs was observed to decrease over the period during which the antibody was administered and to remain stable after therapy had ceased. When considered together, these data indicate that inhibition of CTGF can prevent and reverse the process of fibrosis.
A review of long-term outcome and quality of life of patients after Kasai operation surviving with native livers
Biliary atresia (BA) is a rare neonatal cholestatic disease which leads to progressive obliterative cholangiopathy, resulting in biliary obstruction and jaundice. The standard surgical treatment is hepatoportoenterostomy (Kasai operation). Although approximately 50% of the affected infants would require liver transplantation within the first 2 years of life, the other 50% of the patients can live for years with their native liver, despite the progression of cirrhosis and chronic liver disease. Many of these patients will be affected by long-term complications such as repeated cholangitis, portal hypertension, variceal bleeding, growth problems, biochemical abnormalities, and hepatic osteodystrophy. These morbidities impose a huge impact on the quality of life of the patients and their families. Herein, we performed a comprehensive review on the clinical status and quality of life of long-term survivors of biliary atresia with their native livers, to facilitate meticulous longitudinal follow-up of these patients, and alert caregivers the probable complications to be aware of.
“Worse than I anticipated” or “This isn’t so bad”?: The impact of affective forecasting accuracy on self-reported task performance
Various motivational theories emphasize that desired emotional outcomes guide behavioral choices. Although motivational theory and research has emphasized that behavior is affected by desired emotional outcomes, little research has focused on the impact of anticipated feelings about engaging in behavior. The current research seeks to partly fill that void. Specifically, we borrow from affective forecasting research in suggesting that forecasts about engaging in performance-relevant behaviors can be more or less accurate. Furthermore, we suggest that the degree of accuracy has implications for self-reported task performance. To examine these ideas, we conducted two studies in which individuals made affective predictions about engaging in tasks and then later reported how they actually felt during task engagement. We also assessed their self-reported task performance. In Study 1, 214 workers provided affective forecasts about upcoming work tasks, and in Study 2, 185 students made forecasts about studying for an exam. Results based on polynomial regression were largely consistent across the studies. The accuracy of the forecasts did not conform to the pattern of affective forecasting accuracy typically found outside the performance domain. Furthermore, anticipated and experienced affect jointly predicted self-reported task performance in a consistent manner. Collectively, these findings suggest that taking into account anticipated affect, and its relationship with later experienced affect, provides a more comprehensive account of affect's role in task performance.
Childhood intussusception: 17-year experience at a tertiary referral centre in Hong Kong
To review all paediatric patients with intussusception over the last 17 years. Retrospective case series. A tertiary centre in Hong Kong. Children who presented with intussusception from January 1997 to December 2014 were reviewed. The duration of symptoms, successful treatment modalities, complication rate, and length of hospital stay were studied. A total of 173 children (108 male, 65 female) presented to our hospital with intussusception during the study period. Their median age at presentation was 12.5 months (range, 2 months to 16 years) and the mean duration of symptoms was 2.3 (standard deviation, 1.8) days. Vomiting was the most common symptom (76.3%) followed by abdominal pain (46.2%), per rectal bleeding or red currant jelly stool (40.5%), and a palpable abdominal mass (39.3%). Overall, 160 patients proceeded to pneumatic or hydrostatic reduction, among whom 127 (79.4%) were successful. Three (1.9%) patients had bowel perforation during the procedure. Early recurrence of intussusception occurred in four (3.1%) patients with non-operative reduction. No recurrence was reported in the operative group. The presence of a palpable abdominal mass was a risk factor for operative treatment (relative risk=2.0; 95% confidence interval, 1.8-2.2). Analysis of our results suggested that duration of symptoms did not affect the success rate of non-operative reduction. Non-operative reduction has a high success rate and low complication rate, but the presence of a palpable abdominal mass is a risk factor for failure. Operative intervention should not be delayed in those patients who encounter difficult or doubtful non-operative reduction.
On the subjective experience and correlates of downtime at work
In the current paper, we studied downtime at work. Downtime represents a unique aspect of work time, as employees have low workload and more discretion about their activities but are still paid to be at work. Despite its prevalence and potentially significant implications for productivity and well-being, the experience of downtime has not been a focus in the literature. To study this phenomenon, we first used a qualitative method involving 15 focus groups. Results from 95 employees showed that downtime is distinct from formal breaks and withdrawal behavior and is generally a negative subjective experience. Categories of antecedents of experience and downtime activities were classified. Next, to further explore the phenomenon, and based on Study 1 and relevant literature, we proposed several hypotheses and tested them using an experience sampling method. Results from 86 employees (across 5 workdays and 689 data points) indicated that the amount of downtime was negatively related to day level job satisfaction, job performance, and subjective health. The duration of relaxation activities during downtime mitigated the negative effects of downtime on fatigue. The effects of enjoyment level of the activities, job autonomy, work engagement, and boredom proneness were also examined. Implications, limitations, and future research are discussed.
Direct cellular reprogramming and transdifferentiation of fibroblasts on wound healing—Fantasy or reality?
Induced pluripotent stem cell (iPSC) technology is one of the de novo approaches in regeneration medicine and has led to new research applications for wound healing in recent years. Fibroblasts have attracted wide attention as the first cell line used for differentiation into iPSCs. Researchers have found that fibroblasts can be induced into different types of cells in variable mediums or microenvironments. This indicates the potential “stem” characteristics of fibroblasts in terms of direct cellular reprogramming compared with the iPSC detour. In this review, we described the morphology and biological function of fibroblasts. The stem cell characteristics and activities of fibroblasts, including transdifferentiation into myofibroblasts, osteogenic cells, chondrogenic cells, neurons, and vascular tissue, are discussed. The biological values of fibroblasts are then briefly reviewed. Finally, we discussed the potential applications of fibroblasts in clinical practice. Affective Forecasts for the Experience Itself: An Investigation of the Impact Bias during an Affective Experience
Research documents that forecasts about the emotional consequences of decisions are prone to error. However, there is relatively little known about affective forecasts regarding engaging in activities (versus about the consequences of decisions or outcomes). Here, we examined affective forecasting in the context of a trail race, hypothesizing that forecasts about engaging in an activity (i.e., running the race) were more accurate than forecasts about completing it (i.e., finishing the race). We also investigated whether forecasting accuracy differed for the valence and arousal dimensions of affect and among different discrete emotions. Results of the within-person study showed that affective forecasts largely were inaccurate in reference to engaging in the activity and completing it. Accuracy varied between valence and arousal as well as among emotions of a given valence.
