Prevalence and hepatic histopathological findings of fascioliasis in sheep slaughtered in Jeddah, Saudi Arabia
Hepatic fascioliasis is an important parasitic disease responsible for morbidity and mortality in many domestic ruminants, especially sheep, goats, and cattle, due to Fasciola ( F. ) hepatica and F. gigantica . This study aimed to determine the prevalence of fascioliasis in sheep slaughtered in Jeddah, Saudi Arabia, and to describe the morphological and histopathological changes in the liver. A total of 109,253 sheep slaughtered between July 2017 and July 2018 were screened to assess the prevalence of fascioliasis. The livers were grossly investigated for Fasciola infection and morphological changes. Tissue samples were collected for proper histopathological examinations. Livers of local and imported sheep represented infection rates of 0.67% and 2.12%, respectively, and the highest infection rate was in the spring season. Macroscopically, the affected liver showed hepatomegaly, thickened capsule and discoloration with necrosis, fibrosis, dilation of the bile duct, engorgement of the gallbladder and enlargement of the portal lymph nodes. Microscopic examination showed fibrotic thickening, calcification and hyperplasia of the bile ducts filled with debris, as well as massive hemorrhagic foci. Histopathological examinations of the infected liver showed a central vein region with disturbed parenchyma cells, focal lymphocytic infiltration, elongated endothelial cells, blood sinusoids that showed enlarged Kupffer cells, patches of lysed or necrotic hepatocytes, eosinophil infiltration, lymphocytes and proliferating fibroblast, thickening of hepatic artery and arteriolar walls. We concluded that fascioliasis among sheep slaughtered in Jeddah is not uncommon. The identified histopathological changes in the liver of infected sheep reflect tissue damage, which can lead to significant economic losses for the animals.
Kaempferol activity on Cryptosporidium parvum infection in an experimentally infected immunocompromised mouse model: In silico  and in vivo investigations
No vaccine or effective therapy for cryptosporidiosis currently exists, except for nitazoxanide (NTZ), which has limited effectiveness in immunocompromised hosts. Kaempferol (KPF), a naturally occurring flavonoid, has various pharmacological effects and promising antiparasitic properties. The current work aimed to examine the impact of KPF compared with NTZ on ( ) using both molecular docking and studies with an experimentally infected immunocompromised mouse model. The present study utilized KPF and NTZ as multi-target ligands to predict, determine, and evaluate their target-interacted forms, thereby demonstrating their therapeutic properties against . Mice were immunosuppressed and divided into six groups: DEXA, Model, KPF as prophylaxis, treatment with KPF, NTZ, and a combination of KPF and NTZ. Treatment efficacy was assessed through parasitological, histopathological examination, and immunohistochemical analysis of intestinal tissues using NLRP3 protein. The findings supported the use of KPF and NTZ as inhibitors of growth by blocking the action of choline and pyruvate kinases. The study demonstrated that KPF exhibits anti-cryptosporidial efficacy, particularly when combined with NTZ. The KPF+NTZ had the best results, as evidenced by a significant decrease (  < 0.001) in oocyst shedding of 83%, improved intestinal histological damage, and inhibition of the NLRP3 inflammasome pathway. KPF demonstrated the potential to mitigate the consequences of cryptosporidiosis and serve as an adjuvant agent to NTZ by reducing the oocyst burden, alleviating intestinal inflammation, and facilitating mucosal repair.
Toxoplasmosis awareness among women: a cross-sectional study on knowledge, attitudes, perceptions, and risk factors
Toxoplasmosis, a prevalent zoonotic infection caused by Toxoplasma gondii , poses significant risks for pregnant women. Despite its global prevalence, awareness remains inconsistent, particularly in endemic regions like Pakistan. A cross-sectional study evaluated knowledge, attitudes, and perceptions of 612 women in Khyber Pakhtunkhwa, Pakistan, using a validated questionnaire. Descriptive statistics, chi-square tests (χ²), Spearman’s correlation, and logistic regression were employed to examine demographic and behavioral associations. The dependent variable investigated in this study was high-risk status for toxoplasmosis exposure, based on various demographic and behavioral factors such as dietary habits, hand hygiene, and environmental exposure. The study revealed that while 66.8% of participants had a high level of knowledge, significant gaps persisted. Notably, 37.1% were unaware that cat feces can be a source of transmission, 41.8% did not associate the consumption of raw meat with the risk of infection, 28.8% were unaware of the importance of screening during pregnancy, and 25% did not recognize Toxoplasma as a protozoan. Attitudes were moderately positive (56.9%), yet 45.9% underestimated the severity of toxoplasmosis. High-risk practices included the occasional consumption of raw meat (6.93%; χ² = 388.94, p  < 0.001) and poor hand hygiene (4.57% never washed hands after meat handling). Logistic regression identified younger women and unmarried women as being at high risk; education significantly predicted knowledge but not safer practices. Despite moderate awareness, persistent gaps between knowledge and Risk factor exposure demand targeted interventions in high-risk groups.
Zinc oxide nanoparticles produced by Zingiber officinale ameliorates acute toxoplasmosis-induced pathological and biochemical alterations and reduced parasite burden in mice model
Although, approximately 30% of the world's population is estimated to be infected with Toxoplasma gondii (T. gondii) with serious manifestations in immunocompromised patients and pregnant females, the available treatment options for toxoplasmosis are limited with serious side effects. Therefore, it is of great importance to identify novel potent, well tolerated candidates for treatment of toxoplasmosis. The present study aimed to evaluate the effect of Zinc oxide nanoparticles (ZnO NPs) synthesized using Zingiber officinale against acute toxoplasmosis in experimentally infected mice. The ethanolic extract of ginger was used to prepare ZnO NPs. The produced ZnO NPs were characterized in terms of structure and morphology using Fourier Transformed Infrared Spectroscopy (FTIR), X-Ray Diffraction (XRD), UV- spectroscopy and scanning electron microscopy (SEM). The prepared formula was used in treatment of T. gondii RH virulent strain. Forty animals were divided into four groups, with ten mice per group. The first group was the uninfected, control group. The second group was infected but untreated. The third and the fourth groups received ZnO NPs and Spiramycin orally in a dose of 10 mg/kg and 200 mg/kg/day respectively. The effect of the used formulas on the animals survival rate, parasite burden, liver enzymes -including Alanine transaminase (ALT) and aspartate transaminase (AST)-, nitric oxide (NO) and Catalase antioxidant enzyme (CAT) activity was measured. Moreover, the effect of treatment on histopathological alterations associated with toxoplasmosis was examined. Mice treated with ZnO NPs showed the longest survival time with significant reduction in the parasite load in the livers and peritoneal fluids of the same group. Moreover, ZnO NPs treatment was associated with a significant reduction in the level of liver enzymes (ALT, AST) and NO and a significant increase in the antioxidant activity of CAT enzyme. SEM examination of tachyzoites from the peritoneal fluid showed marked distortion of T. gondii tachyzoites isolated from mice treated with ZnO NPs in comparison to untreated group. T. gondii induced histopathological alterations in the liver and brain were reversed by ZnO NPs treatment with restoration of normal tissue morphology. The produced formula showed a good therapeutic potential in treatment of murine toxoplasmosis as demonstrated by prolonged survival rate, reduced parasite burden, improved T. gondii associated liver injury and histopathological alterations. Thus, we assume that the protective effect observed in the current research is attributed to the antioxidant capability of NPs. Based on the results obtained from the current work, we suggest greenly produced ZnO NPs as a chemotherapeutic agent with good therapeutic potential and high levels of safety in the treatment of toxoplasmosis.
Prevalence of intestinal parasites and comparison of detection techniques for soil-transmitted helminths among newly arrived expatriate labors in Jeddah, Saudi Arabia
Diversity in clinical signs and symptoms are associated with soil transmitted diseases (STD), which are spread to humans by intestinal worms and transmitted in a variety of ways. There is a need for the present study, which aimed to investigate the prevalence of intestinal parasites and to compare between the common detection techniques for soil-transmitted helminths (STHs) among newly arrived expatriate labors in Jeddah, Saudi Arabia. A total of 188 stool samples were analyzed by macroscopic examination, and microscopic examination using direct iodine smear and the formal ether sedimentation technique. Trichrome and modified Kinyoun's stains were used to confirm the morphology of any detected protozoa stages and oocyst of , respectively. A chromatographic immunoassay kit was used for , and . In addition, real-time PCR was employed only to identify various STHs. Out of 188, several types of parasites were detected in 35 samples (18.62%), of which some with multiple infections. Nine samples (4.79%) were positive for , seven samples (3.72%) for , six samples (3.19%) for , four samples (2.13%) for , four samples (2.13%) for , four samples (2.13%) for , three samples (1.60%) for and two samples (1.06%) for . In comparison between laboratory techniques for STHs, real-time PCR was able to detect the DNA of 19 samples (10.1%) followed by Ritchie sedimentation technique (18, 9.6%), and direct smear (7, 3.7%) ( > 0.05). The high rate of newly arrived foreign workers infected with intestinal parasites could lead to a risk to society. Continuous and regular surveys are needed to deal with the occurrence of intestinal parasitic infections including STHs. To improve the identification of these infections, we recommend a supporting infrastructure for the application of concentration methods and molecular assays.
Contribution of socio-demographic factors in prevalence of soil-transmitted helminth infections among newly arrived laborers in Jeddah, Saudi Arabia
Soil-transmitted diseases are caused by intestinal worms transmitted various routes into the human body resulting in various clinical manifestations. This study aimed to investigate the socio-demographic factors contributing mainly to the prevalence of soil-transmitted helminths (STHs) among newly arrived laborers in Jeddah, Saudi Arabia. A total of 188 stool specimens were collected and examined macroscopically and microscopically using different techniques. In addition, real-time PCR was used as a molecular tool to detect several STHs. The descriptive analysis was used to evaluate demographic data and categorical variables in association with STHs infection. Of all participants, the mean age was 26.08 ± 2.67 years, including 103 females and 85 males. A total of 86 (45.70%) were illiterate, followed by 60 (31.90%) of participants belonging to elementary level. STHs and other intestinal parasites were detected among 35 newly arrived laborers (18.61%). Nineteen cases (10.11%) were infected with STHs, including 15 cases with one STHs (four four four two one ) and four cases with two STHs (two and ; one and ; one and ). High rates of STHs infection were detected among drivers (11.70%) followed by housemaids (11.20%), but with no significant association to occupation. Higher rates of STHs infection were detected among those used to walk on soil barefoot in their home countries before arriving in Jeddah. This study found that none of the newly arrived expatriate laborers in Jeddah was aware of STHs. About 10% of them had infection with one or two STHs, mainly among newly arrived laborers from Asian countries. There is a need for awareness programs and regular screening for STHs and other intestinal parasites among newly arrived expatriate laborers.
Genetic diversity and population structure of Echinococcus multilocularis: An in-silico global analysis
Objectives: Alveolar echinococcosis is caused by Echinococcus multilocularis, a parasite of zoo¬notic significance with a wide range of intermediate and final hosts, and the parasite survives suc¬cessfully in diversified conditions. Plentiful studies have been done to study the genetic structure of the population of the parasite and the level of intimate kinship using mitochondrial (mt) DNA. The present study was conducted to investigate the population structure, genetic variation, and phylogenetic relationship of various isolates of E. multiocularis submitted to GenBank worldwide. Sequences of mt genes (mt-cytochrome c oxidase (cox1), mt-NADH dehydrogenase (nad1)) of E. multilocularis were analyzed to achieve the set goals. Materials and Methods: A total of 275 and 124 gene sequences of mt-cox1 and mt-nad1 belong¬ing to E. multilocularis, respectively, were retrieved from the National Center for Biotechnology Information GenBank. The retrieved sequences were subjected to alignment with respective reference sequences using MEGA software. The PopArt software was used to establish medi¬an-joining networks, while DnaSp was used to calculate neutrality and diversity indices. MrBayes software was used to investigate the phylogenetic association between haplotypes based on Bayesian phylogeny. Results: Approximately 13 and 20 distinctive haplotypes of nad1 and cox1 genes, respectively, were observed in the present study. In both of the mt genes, diversity indices indicated low haplo¬type (mt-cox1 = 0.140; mt-nad1 = 0.374) and nucleotide (mt-cox1 = 0.00111; mt-nad1 = 0.00287) diversities. The values of Tajima’s D and Fu Fs for a population of both of the genes under study were found to be negative. Conclusion: This study is a maiden attempt to provide insights into the population structure and genetic variation of E. multilocularis on a global scale. However, it is suggested that to better understand the population structure and genetic diversity of E. multilocularis, more geographical locations and amplifications of full-length gene sequences should be considered, which could be helpful in widening the insights into the genetic diversity of E. multilocularis.
Seroprevalence and Potential Risk Factors of Toxoplasma gondii in Dromedary Camels
(1) Background: Toxoplasma gondii (T. gondii) is one of the most prevalent parasites to affect humans and animals; (2) Methods: From January to December 2020, using a commercial enzyme-linked immunosorbent assay (ELISA) kit, a cross-sectional study was conducted to establish the seroprevalence of T. gondii in 390 dromedary camels raised in three governorates in Egypt and to identify the potential risk factors associated with infection; (3) Results: Overall, T. gondii seroprevalence in camels was 46.9%. Moreover, locality, sex, age, contact with small ruminants, history of abortion, and number of parities were found as risk factors for T. gondii infection in univariable analysis. The seropositivity to T. gondii increased significantly in camels living in Marsa Matrouh (OR = 2.02), among camels of more than 8 years old (OR = 5.28). Additionally, the likelihood of acquiring T. gondii infection was increased in camels that had contact with small ruminants (OR = 3.85) and a history of abortion (OR = 3.84) with these having parity more than four times (OR = 17.72); (4) Conclusions: The evaluation of seroprevalence and related risk factors for T. gondii infection is crucial for implementing an effective control programme to minimise and control T. gondii infection in camels and, as a result, transmission to humans.
Hygiene practices and factors influencing intestinal parasites among food handlers in the province of Belgarn, Saudi Arabia
This study aimed to evaluate the association between socio-demographic status, and hygienic habits among food handlers with intestinal parasitic infections. It was a cross-sectional study in which 112 participants were recruited, from Belgarn province of Saudi Arabia. The descriptive analysis was used to evaluate demographic data and categorical variables. The association between sociodemographic characteristics and Relative Risk regression analysis was performed for each investigated factor. -value of <0.05, was assumed to be statistically significant. One hundred and twelve food handlers with a mean age of 33.5 ± 9.2 years were included in this study. The food handlers were from 15 different countries (11 Asians and four Africans). The majority of the food handlers were cooks (87, 77.7%), and waiters and dish washers (24, 21.4%). Among them, 106 (94.6%) used uniforms, and gloves at work. In hand washing practices, 104 (92.9%) wash their hands with soap before handling and preparing food and eight (7.1%) wash without soap, 98 (87.5%) wash with soap before meals and 14 (12.5%) wash without soap, 105 (93.8%) wash with soap after visiting toilets and seven (6.2%) wash without soap. Twenty-five (42.3%) of infected food handlers are not used to trim their fingernails. Intestinal infection was observed in 59 (52.68%) participants with mean age (32.5 ± 8.1 years) for infected participants. In this study, food handlers had a high prevalence of intestinal parasites. Although some of the food handlers had a high level of education, the infection with intestinal parasites was detected. In addition to regular screening for intestinal parasites among food handlers, there is a need for educational programs on proper hygiene habits, modes of transmission and prevention of the infection.
Platelet Rich Plasma as a Potential Therapy for Chronic Toxoplasmosis in Immunocompetent and Immunocompromised Murine Model
: ( ) is one of the most prevalent parasitic zoonoses worldwide, and the host's immunological state significantly influences its clinical manifestations, which can be potentially fatal in immunocompromised hosts. The unavailability of a vaccine, combined with the considerable toxicity of existing medications, necessitates the urgent search for new therapies or adjunctive techniques, including regenerative and immunomodulatory approaches. Hence, the present study investigated, for the first time, the therapeutic potential of syngeneic platelet rich plasma (PRP) against ME49 strain-induced chronic toxoplasmosis in both immunocompetent and immunosuppressed mouse models. : 72 albino mice were divided into two sections, immunocompetent and immunosuppressed. Each section contained six groups: healthy, model, cotrimoxazole (CTZ)-treated, PRP-treated, half-dose of both CTZ and PRP-treated, and full-dose of both CTZ and PRP-treated. Treatment efficacy was assessed via parasitological, histological, immunohistochemical, and immunological analyses. : PRP, especially when coadministered with the CTZ, mitigated the consequences of toxoplasmosis by significantly reducing brain cyst counts ( < 0.0001), restoring brain tissue architecture, modulating apoptotic pathways by restoring caspase-3 expression in the brain, and normalizing systemic IFN-γ, TNF-α, and IL-10 cytokine profiles. : The findings highlight PRP as an adjunct to the reference treatment, CTZ, for controlling toxoplasmosis in both immunocompetent and immunosuppressed conditions via anti-infective, neuroprotective, and immunomodulatory activities.
