Automatic design of fiber-reinforced soft actuators for trajectory matching
Soft actuators are the components responsible for producing motion in soft robots. Although soft actuators have allowed for a variety of innovative applications, there is a need for design tools that can help to efficiently and systematically design actuators for particular functions. Mathematical modeling of soft actuators is an area that is still in its infancy but has the potential to provide quantitative insights into the response of the actuators. These insights can be used to guide actuator design, thus accelerating the design process. Here, we study fluid-powered fiber-reinforced actuators, because these have previously been shown to be capable of producing a wide range of motions. We present a design strategy that takes a kinematic trajectory as its input and uses analytical modeling based on nonlinear elasticity and optimization to identify the optimal design parameters for an actuator that will follow this trajectory upon pressurization. We experimentally verify our modeling approach, and finally we demonstrate how the strategy works, by designing actuators that replicate the motion of the index finger and thumb.

Immune checkpoint inhibitor combinations—current and emerging strategies
In an attempt to overcome resistance to immune checkpoint inhibitors (ICI), an ever-increasing number of trials are exploring combination treatment approaches. Outcomes of a novel ICI doublet presented by Desai and colleagues are discussed along with emerging novel strategies and a view to future ongoing rational trial design maximising patient benefit.
Human-in-the-loop Bayesian optimization of wearable device parameters
The increasing capabilities of exoskeletons and powered prosthetics for walking assistance have paved the way for more sophisticated and individualized control strategies. In response to this opportunity, recent work on human-in-the-loop optimization has considered the problem of automatically tuning control parameters based on realtime physiological measurements. However, the common use of metabolic cost as a performance metric creates significant experimental challenges due to its long measurement times and low signal-to-noise ratio. We evaluate the use of Bayesian optimization-a family of sample-efficient, noise-tolerant, and global optimization methods-for quickly identifying near-optimal control parameters. To manage experimental complexity and provide comparisons against related work, we consider the task of minimizing metabolic cost by optimizing walking step frequencies in unaided human subjects. Compared to an existing approach based on gradient descent, Bayesian optimization identified a near-optimal step frequency with a faster time to convergence (12 minutes, p < 0.01), smaller inter-subject variability in convergence time (± 2 minutes, p < 0.01), and lower overall energy expenditure (p < 0.01).
Extensive estuarine sedimentary storage of plastics from city to sea: Narragansett Bay, Rhode Island, USA
Plastics are an important new component of the global sedimentary system, and much concern exists about their transport, fate and impact. This study presents the first system-scale assessment of sedimentary storage of microplastic for an estuary, Narragansett Bay, RI (USA), and the measurements of shoreline and seabed sediments add to the growing body of literature demonstrating high coastal concentrations. Microplastic concentrations in sediments ranged from 396 to over 13,000 MP particles kg −1 dry sediment (DW), comparable to other shoreline and seafloor sites located near urban centers. As previously reported for fine sediment and other pollutants, estuarine plastic storage is extensive in Narragansett Bay, especially within the upper urbanized reaches. Over 16 trillion pieces of plastic weighing near 1000 tonnes is calculated to be stored in surface sediments of the Bay based on a power-law fit. This work highlights that estuaries may serve as a significant filter for plastic pollution, and this trapping may have negative consequences for these valuable, productive ecosystems but offer potential for efficient removal.
Declining tropical cyclone frequency under global warming
Assessing the role of anthropogenic warming from temporally inhomogeneous historical data in the presence of large natural variability is difficult and has caused conflicting conclusions on detection and attribution of tropical cyclone (TC) trends. Here, using a reconstructed long-term proxy of annual TC numbers together with high-resolution climate model experiments, we show robust declining trends in the annual number of TCs at global and regional scales during the twentieth century. The Twentieth Century Reanalysis (20CR) dataset is used for reconstruction because, compared with other reanalyses, it assimilates only sea-level pressure fields rather than utilize all available observations in the troposphere, making it less sensitive to temporal inhomogeneities in the observations. It can also capture TC signatures from the pre-satellite era reasonably well. The declining trends found are consistent with the twentieth century weakening of the Hadley and Walker circulations, which make conditions for TC formation less favourable.Detecting change in tropical cyclones is difficult from observational records. Here a reconstruction using reanalysis data of annual cyclone numbers shows they have declined globally and regionally over the twentieth century.
Recent poleward shift of tropical cyclone formation linked to Hadley cell expansion
Recent research indicates that the annual-mean locations of tropical cyclones have migrated toward higher latitudes. Concurrently, an anthropogenically forced tropical expansion has been observed, yet the connection between the two processes remains little-explored. Here, using observational and reanalysis data, we investigate how large-scale dynamical effects, combined with coherent changes in the regional Hadley circulation, explain recent changes in regional tropical cyclone genesis over 1980–2014. We show that the recent anomalous upper-level weakening of the rising branch of the Hadley circulation in the deep tropics, possibly induced by the increased vertical stability, has likely suppressed the low-latitude tropical cyclone genesis in most ocean basins via anomalous large-scale subsidence. Regional Hadley circulation variations have also favoured a poleward displacement of tropical-cyclone-favourable climate conditions through poleward shift of the Hadley circulation’s meridional extent. With projections indicating continued tropical expansion, these results indicate that tropical cyclone genesis will also continue to shift poleward, potentially increasing tropical-cyclone-related hazards in higher-latitude regions.
Multidonor intensive faecal microbiota transplantation for active ulcerative colitis: a randomised placebo-controlled trial
The intestinal microbiota is implicated in the pathogenesis of ulcerative colitis. Faecal microbiota transplantation is a novel form of therapeutic microbial manipulation, but its efficacy in ulcerative colitis is uncertain. We aimed to establish the efficacy of intensive-dosing, multidonor, faecal microbiota transplantation in active ulcerative colitis. We conducted a multicentre, double-blind, randomised, placebo-controlled trial at three hospitals in Australia. We randomly allocated patients with active ulcerative colitis (Mayo score 4–10) in a 1:1 ratio, using a pre-established randomisation list, to either faecal microbiota transplantation or placebo colonoscopic infusion, followed by enemas 5 days per week for 8 weeks. Patients, treating clinicians, and other study staff were unaware of the assigned treatment. Faecal microbiota transplantation enemas were each derived from between three and seven unrelated donors. The primary outcome was steroid-free clinical remission with endoscopic remission or response (Mayo score ≤2, all subscores ≤1, and ≥1 point reduction in endoscopy subscore) at week 8. Analysis was by modified intention-to-treat and included all patients receiving one study dose. We performed 16S rRNA stool analysis to assess associated microbial changes. This trial is registered with ClinicalTrials.gov, number NCT01896635. The trial has ended; this report presents the final analysis. From November, 2013, to May, 2015, 85 patients were enrolled to our trial, of whom 42 were randomly assigned faecal microbiota transplantation and 43 were allocated placebo. One patient assigned faecal microbiota transplantation and three allocated placebo did not receive study treatment and were excluded from the analysis. The primary outcome was achieved in 11 (27%) of 41 patients allocated faecal microbiota transplantation versus three (8%) of 40 who were assigned placebo (risk ratio 3·6, 95% CI 1·1–11·9; p=0·021). Adverse events were reported by 32 (78%) of 41 patients allocated faecal microbiota transplantation and 33 (83%) of 40 who were assigned placebo; most were self-limiting gastrointestinal complaints, with no significant difference in number or type of adverse events between treatment groups. Serious adverse events occurred in two patients assigned faecal microbiota transplantation and in one allocated placebo. Microbial diversity increased with and persisted after faecal microbiota transplantation. Several bacterial taxa were associated with clinical outcome; in particular, the presence of Fusobacterium spp was associated with lack of remission. Intensive-dosing, multidonor, faecal microbiota transplantation induces clinical remission and endoscopic improvement in active ulcerative colitis and is associated with distinct microbial changes that relate to outcome. Faecal microbiota transplantation is, thus, a promising new therapeutic option for ulcerative colitis. Future work should focus on precisely defining the optimum treatment intensity and the role of donor–recipient matching based on microbial profiles.
