Exosome-derived ENO1 regulates integrin α6β4 expression and promotes hepatocellular carcinoma growth and metastasis

Alpha-enolase (ENO1) has been found to be dysregulated in several human malignancies, including hepatocellular carcinoma (HCC). Although the role of ENO1 as a glycolytic enzyme in HCC cells has been well characterized, little is known about the other roles of ENO1, especially exosome-derived ENO1, in regulating HCC progression. Here, we demonstrated that ENO1 is frequently upregulated in HCC cells or tissues, with even higher expression in highly metastatic HCC cells or metastatic tissues as well as in exosomes derived from highly metastatic sources. Moreover, ENO1 expression is associated with the tumor-node-metastasis (TNM) stage, differentiation grade and poor prognosis in HCC patients. Surprisingly, ENO1 can be transferred between HCC cells via exosome-mediated crosstalk, exhibiting an effect similar to that of ENO1 overexpression in HCC cells, which promoted the growth and metastasis of HCC cells with low ENO1 expression by upregulating integrin α6β4 expression and activating the FAK/Src-p38MAPK pathway. In summary, our data suggest that exosome-derived ENO1 is essential to promoting HCC growth, metastasis, and further patient deterioration. The findings from this study implicate a novel biomarker for the clinical evaluation of HCC progression, especially the prediction of HCC metastatic risk.

Three-dimensional electrodes enhance electricity generation and nitrogen removal of microbial fuel cells
One of the critical problems for practical application of microbial fuel cells (MFCs) is the poor electron transfer between microbial cells and anode. Hence, good biocompatibility and high specific surface area of electrodes are indispensable for MFC scale-up. In this study, three-dimensional electrode MFC (3DEMFC) was developed by filling biochar between anode and cathode. Three types of biochar electrodes (biochar, biochar and zeolite mixture, and MgO-modified biochar) were employed, and the performance of 3DEMFCs treating nitrogen in wastewater was investigated. The results showed that the highest power density of MFCs was 4.45 ± 0.21 W m−3 achieved by 3DEMFC filled with MgO-modified biochar, and the overall power generation of 3DEMFCs (2.40 ± 0.28 ~ 4.45 ± 0.21 W m−3) was higher than that of MFC without biochar (1.31 ± 0.24 W m−3). The linear sweep voltammetry (LSV) results also demonstrated biochar addition to MFC was conducive to electron transfer between microbes and anode and MgO-modified biochar presented the highest coulombs transfer ability. Moreover, the highest removal efficiencies of ammonium, total nitrogen, and COD (93.6 ± 3.2%, 84.8 ± 2%, and 91.6 ± 1.3%, respectively) were achieved by 3DEMFC containing MgO-modified biochar, and simultaneous short-cut nitrification and denitrification were observed in MFCs. Furthermore, the SEM images displayed the bacteria adhesion on biochar and the biofilm dry weights of MgO-modified biochar after experiment was the highest of 103 ± 4 mg g−1 among three kinds of biochar electrodes. Therefore, the power generation and nitrogen removal conspicuously enhanced in 3DEMFCs and biochar exhibited excellent biocompatibility and distinct electrochemical performance for MFC practical applications in wastewater treatment.

Operative ubiquitin-specific protease 22 deubiquitination confers a more invasive phenotype to cholangiocarcinoma

Oncogenic ubiquitin-specific protease 22 (USP22) is implicated in a variety of tumours; however, evidence of its role and underlying molecular mechanisms in cholangiocarcinoma (CCA) development remains unknown. We collected paired tumour and adjacent non-tumour tissues from 57 intrahepatic CCA (iCCA) patients and evaluated levels of the USP22 gene and protein by qPCR and immunohistochemistry. Both the mRNA and protein were significantly upregulated, correlated with the malignant invasion and worse OS of iCCA. In cell cultures, USP22 overexpression increased CCA cell proliferation and mobility, and induced epithelial-to-mesenchymal transition (EMT). Upon an interaction, USP22 deubiquitinated and stabilized sirtuin-1 (SIRT1), in conjunction with Akt/ERK activation. In implantation xenografts, USP22 overexpression stimulated tumour growth and metastasis to the lungs of mice. Conversely, the knockdown by USP22 shRNA attenuated the tumour growth and invasiveness in vitro and in vivo. Furthermore, SIRT1 overexpression reversed the USP22 functional deficiency, while the knockdown acetylated TGF-β-activated kinase 1 (TAK1) and Akt. Our present study defines USP22 as a poor prognostic predictor in iCCA that cooperates with SIRT1 and facilitates tumour development.

SIRT5 Promotes Hepatocellular Carcinoma Progression by Regulating Mitochondrial Apoptosis

SIRT5 belongs to a family of NAD
-dependent lysine deacetylases called sirtuins. Although accumulating evidence indicates SIRT5 upregulation in cancers, including liver cancer, the detailed roles and mechanisms remain to be revealed. Hepatocellular carcinoma (HCC) is the second leading cause of cancer-related deaths among men worldwide, and finding effective targets for HCC treatment and prevention is urgently needed. In the present study, we confirmed that mitochondrial sirtuins, particularly SIRT5, are more highly expressed in HCC cell lines than in normal liver cell lines. Moreover, SIRT5 knockdown suppresses HCC cell proliferation and SIRT5 overexpression promotes HCC cell proliferation. Furthermore, we verified that SIRT5 knockdown increases HCC cell apoptosis via the mitochondrial pathway. By co-IP and western blotting, we illustrated that SIRT5 deacetylates cytochrome c thus regulating HCC cell apoptosis. Taken together, our findings suggest that SIRT5 may function as a prognostic factor and drug target for HCC treatment.

Mitochondrial Genome Comparison and Phylogenetic Analysis of Four Species of Dung Beetles (Coleoptera: Scarabaeidae: Scarabaeinae)

Dung beetles, despite their well‐documented ecological significance, remain surprisingly underrepresented in molecular biology research, with a notable paucity of genomic and molecular data available for this ecologically crucial insect group. We sequenced and analyzed the whole mitochondrial genomes of four species of dung beetles, including Catharsius molossus, Liatongus bucerus, Copris magicus, and Onitis falcatus from the Scarabaeinae subfamily. The results showed that the mitochondrial genome sizes of the four dung beetle species ranged from 14,977 bp to 18,425 bp. Among them, the mitochondrial genome structure, base composition, codon usage, and gene arrangement of L. bucerus and C. magicus were relatively conserved, consistent with other Coleoptera species. However, C. magicus exhibited a unique rearrangement, with the trnT and trnP genes undergoing positional swapping, resulting in the formation of a trnP‐trnT tRNA gene block. In L. bucerus, a long‐distance translocation of the trnI gene and rearrangement between the trnS2 and nad1 genes occurred, leading to the formation of a trnS2‐trnI tRNA gene block. This represents the first identification of such rearrangements in Scarabaeidae. The phylogenetic reconstruction of mitochondrial protein‐coding genes in 63 species of Scarabaeinae revealed significant insights into their evolutionary relationships. It supported multilineage for the tribes Coprini and Onthophagini, and monophyly for the tribes Onitini and Oniticellini. Coprini maintained close affinities with Deltochilini, Dichotomiini, Phanaeini, Ateuchini, and Eurysternini, with Phanaeini as a sister group to each other, and the phylogenetic relationships among the tribes supported the existing findings. It clarified the position of four dung beetles in their respective clades and genera. This study could enrich the genetic data of Scarabaeidae and establishes a foundation for further understanding of the evolutionary relationships of dung beetles. Dung beetles have important ecological and economical values; however, the molecular research of the taxonomic group is very limited. We sequenced mitochondrial genomes of four Scarabaeinae species, with sizes from 14,977 to 18,425 bp, and identified novel gene rearrangements. Phylogenetic analysis of 63 Scarabaeinae species supported multilineage origins for Coprini and Onthophagini tribes, monophyly for Onitini and Oniticellini, and clarified evolutionary ties between tribes, aligning with existing classifications. This work enriched Scarabaeidae genetic data and advanced understanding of dung beetle evolution, and provided a foundation for future molecular and ecological studies.

To explore the whitening components of the EBR, we employed UHPLC-Q-TOF MS to identify the compounds present in the EBR and utilized network pharmacology to compare them with the genes involved in whitening in the database, thereby obtaining the intersection of compound targets and whitening targets. Then, protein-protein interaction network analysis, gene ontology enrichment and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis were carried out to explore the main components of the whitening effect of the EBR. Further, molecular docking and molecular dynamics simulation were used to verify the correlation between the main components of the EBR and the essential target molecules.

studies have found that the EBR exhibits anti-oxidation, TYR-inhibiting activity, and the elimination of melanin deposition. Subsequently, 1538 metabolites were identified by UHPLC-Q-TOF MS analysis, indicating that EBR is a rich source of bioactive compounds. Among them, 82 kinds of phenols and flavonoids may be derived from external enrichment or self-synthesis. Network pharmacology revealed 41 genes related to pigmentation and TYR inhibition. Molecular docking and molecular dynamics simulation confirmed that myricetin, luteolin, apigenin and quercetin have a high binding affinity with TYR, which may regulate melanin production by inhibiting TYR activity.

This work reveals that insect extraction may have broad application prospects in the preparation of cosmetics. To our knowledge, this is the first report identifying the flavonoids quercetin, luteolin, myricetin, and apigenin as whitening-active components in insects.

Research on Meshing Stiffness and Vibration Response of Pitting Fault Gears with Different Degrees

In order to study the influence of pitting on meshing stiffness, the normal distribution function is used to simulate the pitting location of pitting gear, and the potential energy method is used to analyze the influence of pitting on meshing stiffness. At the same time, the meshing stiffness of pitting gears with different degrees is analyzed by finite element method, and the validity of the calculation results with potential energy method is verified. On the basis of meshing stiffness, the dynamic model of gear system is established, and the vibration response of pitting gear system with different degrees is analyzed. The results show that with the increase of pitting area, the meshing stiffness decreases; the closer the meshing area of the driving wheel is to the pitting line, the more the meshing stiffness decreases, resulting in the intensification of vibration response and periodic impact; and in the time history diagram, there is a small spurious frequencies near the meshing frequency; in the phase diagrams and the Poincare diagram, trajectory and discrete point aggregation area is gradually increased.

The Dual Effect of Selenium Application in Reducing Fusarium Wilt Disease Incidence in Banana and Producing Se-Enriched Fruits

Fusarium wilt disease severely constrains the global banana industry. The highly destructive disease is caused by Fusarium oxysporum f. sp. cubense, especially its virulent tropical race 4 (Foc TR4). Selenium (Se), a non-essential mineral nutrient in higher plants, is known to enhance plant resistance against several fungal pathogens. The experiments we conducted showed that selenium (≥10 mg/L) dramatically inhibited the growth of Foc TR4 mycelia and promoted plant growth. The further study we performed recorded a substantial reduction in the disease index (DI) of banana plants suffering from Foc TR4 when treated with selenium. The selenium treatments (20~160 mg/L) demonstrated significant control levels, with recorded symptom reductions ranging from 42.4% to 65.7% in both greenhouse and field trials. The DI was significantly negatively correlated with the total selenium content (TSe) in roots. Furthermore, selenium treatments enhanced the antioxidant enzyme activities of peroxidase (POD), polyphenol oxidase (PPO), and glutathione peroxidase (GSH-Px) in banana. After two applications of selenium (100 and 200 mg/plant) in the field, the TSe in banana pulps increased 23.7 to 25.9-fold and achieved the Se enrichment standard for food. The results demonstrate that selenium applications can safely augment root TSe levels, both reducing Fusarium wilt disease incidence and producing Se-enriched banana fruits. For the first time, this study has revealed that selenium can significantly reduce the damage caused by soil-borne pathogens in banana by increasing the activities of antioxidant enzymes and inhibiting fungal growth.

Improving Effect of the Policosanol from Ericerus pela Wax on Learning and Memory Impairment Caused by Scopolamine in Mice

Policosanol (PC) is a mixture of long-chain fatty alcohols that exhibits multiple biological activities, such as reducing blood lipid and cholesterol levels, lowering blood pressure, and extenuating liver inflammation. To assess PC’s impact on cognitive behavior and function, PC was prepared from Ericerus pela wax using a reduction method and analyzed using gas chromatography (GC). A total of 60 mice were randomly divided into six groups of 10 animals each: control (0.5% CMC-Na solution, i.g.), model (0.5% CMC-Na solution, i.g.), donepezil (3 mg/kg, i.g.), PC low- (2 g/kg, i.g.), medium (4 g/kg, i.g.), and high- (6 g/kg, i.g.) dose groups. All the groups were administered daily for 28 consecutive days. There were four parameters—escape latency, crossings of platform, swimming distance, and time spent in the target quadrant—that were recorded to evaluate the cognitive performance of mice in the Morris Water Maze (MWM). After MWM testing, the levels of acetylcholine (ACh), acetylcholinesterase (AChE), superoxide dismutase (SOD), malondialdehyde (MDA), and glutathione (GSH) that were present in brain tissue were determined using assay kits. The GC data showed that PC consisted of four major components: tetracosanol (14.40%), hexacosanol (48.97%), octacosanol (25.40%), and triacontanol (4.80%). In the MWM test, PC significantly decreased the escape latency (p < 0.05) and increased the crossings of the platform (p < 0.05) and swimming distance (p < 0.05) and time in the target quadrant (p < 0.05) in rodents compared to that in the model group. Moreover, PC increased the levels of ACh, SOD, and GSH; inhibited AChE; and reduced MDA in the brain tissue of the tested animals. This is the first report to evaluate the efficacy of PC for cognitive behavior and function in animals. Our findings demonstrate that PC from E. pela wax is likely to exert an enhancing effect on learning and memory by promoting the cholinergic system and attenuating oxidative stress, which will provide a new insight into the efficacy of PC and expand its application in the food, nutraceutical, and beverage industries.
