Natural and Synthetic Saponins as Vaccine Adjuvants
Saponin adjuvants have been extensively studied for their use in veterinary and human vaccines. Among them, QS-21 stands out owing to its unique profile of immunostimulating activity, inducing a balanced Th1/Th2 immunity, which is valuable to a broad scope of applications in combating various microbial pathogens, cancers, and other diseases. It has recently been approved for use in human vaccines as a key component of combination adjuvants, e.g., AS01b in Shingrix® for herpes zoster. Despite its usefulness in research and clinic, the cellular and molecular mechanisms of QS-21 and other saponin adjuvants are poorly understood. Extensive efforts have been devoted to studies for understanding the mechanisms of QS-21 in different formulations and in different combinations with other adjuvants, and to medicinal chemistry studies for gaining mechanistic insights and development of practical alternatives to QS-21 that can circumvent its inherent drawbacks. In this review, we briefly summarize the current understandings of the mechanism underlying QS-21’s adjuvanticity and the encouraging results from recent structure-activity-relationship (SAR) studies.
Asset Bubbles and Credit Constraints
We provide a theory of rational stock price bubbles in production economies with infinitely-lived agents. Firms meet stochastic investment opportunities and face endogenous credit constraints. They are not fully committed to repaying debt. Credit constraints are derived from incentive constraints in optimal contracts which ensure default never occurs in equilibrium. Stock price bubbles can emerge through a positive feedback loop mechanism and cannot be ruled out by transversality conditions. These bubbles command a liquidity premium and raise investment by raising the debt limit. Their collapse leads to a recession and a stock market crash.
Generating dual-active species by triple-atom sites through peroxymonosulfate activation for treating micropollutants in complex water
The peroxymonosulfate (PMS)-triggered radical and nonradical active species can synergistically guarantee selectively removing micropollutants in complex wastewater; however, realizing this on heterogeneous metal-based catalysts with single active sites remains challenging due to insufficient electron cycle. Herein, we design asymmetric Co–O–Bi triple-atom sites in Co-doped Bi₂O₂CO₃ to facilitate PMS oxidation and reduction simultaneously by enhancing the electron transfer between the active sites. We propose that the asymmetric Co–O–Bi sites result in an electron density increase in the Bi sites and decrease in the Co sites, thereby PMS undergoes a reduction reaction to generate SO₄•- and •OH at the Bi site and an oxidation reaction to generate ¹O₂ at the Co site. We suggest that the synergistic effect of SO₄•-, •OH, and ¹O₂ enables efficient removal and mineralization of micropollutants without interference from organic and inorganic compounds under the environmental background. As a result, the Co-doped Bi₂O₂CO₃ achieves almost 99.3% sulfamethoxazole degradation in 3 min with a k-value as high as 82.95 min−1 M−1, which is superior to the existing catalysts reported so far. This work provides a structural regulation of the active sites approach to control the catalytic function, which will guide the rational design of Fenton-like catalysts.
Broadly neutralizing antibodies to SARS-CoV-2 and other human coronaviruses
Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a recently emerged pathogenic human coronavirus that belongs to the sarbecovirus lineage of the genus Betacoronavirus. The ancestor strain has evolved into a number of variants of concern, with the Omicron variant of concern now having many distinct sublineages. The ongoing COVID-19 pandemic caused by SARS-CoV-2 has caused serious damage to public health and the global economy, and one strategy to combat COVID-19 has been the development of broadly neutralizing antibodies for prophylactic and therapeutic use. Many are in preclinical and clinical development, and a few have been approved for emergency use. Here we summarize neutralizing antibodies that target four key regions within the SARS-CoV-2 spike (S) protein, namely the N-terminal domain and the receptor-binding domain in the S1 subunit, and the stem helix region and the fusion peptide region in the S2 subunit. Understanding the characteristics of these broadly neutralizing antibodies will accelerate the development of new antibody therapeutics and provide guidance for the rational design of next-generation vaccines.The ancestral strain of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has evolved into a number of variants of concern. In this Review, Wang and colleagues discuss progress in the development and characterization of broadly neutralizing antibodies to SARS-CoV-2, which may lead to new antibody therapeutics and inform the design of next-generation vaccines.
Dynamic active-site induced by host-guest interactions boost the Fenton-like reaction for organic wastewater treatment
In heterogeneous catalysis, uncovering the dynamic evolution of active sites in the working conditions is crucial to realizing increased activity and enhanced stability of catalyst in Fenton-like activation. Herein, we capture the dynamic changes in the unit cell of Co/La-SrTiO 3 catalyst during the exemplary peroxymonosulfate activation process using X-ray absorption spectroscopy and in situ Raman spectroscopy, revealing the substrate tuned its structural evolution, which is the reversible stretching vibration of O-Sr-O and Co/Ti-O bonds in different orientations. This process effectively promotes the generation of key SO 5 * intermediates, which is beneficial to the formation of 1 O 2 and SO 4 •− from persulfate on the Co active site. Density functional theory and X-ray absorption spectroscopy show that the optimized structural distortion enhanced the metal-oxygen bond strength by tuning the e g orbitals and increased the number of transferred electrons to peroxymonosulfate by about 3-fold, achieving excellent efficiency and stability in removing organic pollutants. Uncovering the dynamic evolution of active sites in the working conditions is crucial to realizing increased activity and enhanced stability of catalyst in Fenton-like activation. Simultaneous atmospheric water production and 24-hour power generation enabled by moisture-induced energy harvesting
Water and electricity scarcity are two global challenges, especially in arid and remote areas. Harnessing ubiquitous moisture and sunlight for water and power generation is a sustainable route to address these challenges. Herein, we report a moisture-induced energy harvesting strategy to realize efficient sorption-based atmospheric water harvesting (SAWH) and 24-hour thermoelectric power generation (TEPG) by synergistically utilizing moisture-induced sorption/desorption heats of SAWH, solar energy in the daytime and radiative cooling in the nighttime. Notably, the synergistic effects significantly improve all-day thermoelectric power density (~346%) and accelerate atmospheric water harvesting compared with conventional designs. We further demonstrate moisture-induced energy harvesting for a hybrid SAWH-TEPG device, exhibiting high water production of 750 g m −2 , together with impressive thermoelectric power density up to 685 mW m −2 in the daytime and 21 mW m −2 in the nighttime. Our work provides a promising approach to realizing sustainable water production and power generation at anytime and anywhere. Collecting water directly from air provides one sustainable solution to water scarcity. Li et al. combine sorption-based atmospheric water harvesting with a thermoelectric module for efficient harvesting water and electricity in arid environments and show that the two components operate synergistically.
Pauling-type adsorption of O2 induced electrocatalytic singlet oxygen production on N–CuO for organic pollutants degradation
Due to environmentally friendly operation and on-site productivity, electrocatalytic singlet oxygen ( 1 O 2 ) production via O 2 gas is of immense interest in environment purification. However, the side-on configuration of O 2 on the catalysts surface will lead to the formation of H 2 O, which seriously limits the selectivity and activity of 1 O 2 production. Herein, we show a robust N-doped CuO (N–CuO) with Pauling-type (end-on) adsorption of O 2 at the N–Cu–O 3 sites for the selective generation of 1 O 2 under direct-current electric field. We propose that Pauling-type configuration of O 2 not only lowers the overall activation energy barrier, but also alters the reaction pathway to form 1 O 2 instead of H 2 O, which is the key feature determining selectivity for the dissociation of Cu–O bonds rather than the O–O bonds. The proposed N dopant strategy is applicable to a series of transition metal oxides, providing a universal electrocatalysts design scheme for existing high-performance electrocatalytic 1 O 2 production. Side-on configuration of O 2 on the catalysts conventionally leads to reduction of O 2 to water. Here, the authors propose a nitrogen doping strategy with Pauling-type adsorption of O 2 for selective electrocatalytic singlet oxygen production.
Efficient reduction-oxidation coupling degradation of nitroaromatic compounds in continuous flow processes
Nitroaromatic compounds (NACs) with electron-withdrawing nitro (-NO 2 ) groups are typical refractory pollutants. Despite advanced oxidation processes (AOPs) being appealing degradation technologies, inefficient ring-opening oxidation of NACs and practical large-scale applications remain challenges. Here we tackle these challenges by designing a reduction-oxidation coupling (ROC) degradation process in LaFe 0.95 Cu 0.05 O 3 @carbon fiber cloth (LFCO@CFC)/PMS/Vis continuous flow system. Cu doping enhances the photoelectron transfer, thus triggering the -NO 2 photoreduction and breaking the barriers in the ring opening. Also, it modulates surface electronic configuration to generate radicals and non-radicals for subsequent oxidation of reduction products. Based on this, the ROC process can effectively remove and mineralize NACs under the environmental background. More importantly, the LFCO catalyst outperformed most of the recently reported catalysts with lower cost (13.72 CNY/ton) and higher processing capacity (3600 t/month). Furthermore, the high scalability, material durability, and catalytic activity of LFCO@CFC under various realistic environmental conditions prove the potential ability for large-scale applications. Nitroaromatic compounds (NACs) with electron-withdrawing -NO2 groups are typical refractory pollutants. Here the authors designed the LaFe0.95Cu0.05O3@carbon fiber cloth (LFCO@CFC) to realize efficient reduction-oxidation coupling degradation of NACs with lower cost and higher durability.
Study on distribution characteristic of tourism attractions in international cultural tourism demonstration region in South Anhui in China
Taking data of tourism attractions in international cultural tourism demonstration region in South Anhui in China, this study summarized the distribution characteristics of tourism attractions in region by applying GIS spatial analysis method such as nearest neighbor distance index and kernel density estimation method, and explored how natural conditions, urban infrastructure, social and economic development affect the distribution in order to better understand the distribution of regional tourism resources and serve the direction of tourism development. The study found that the tourist attractions in the demonstration area have a significant agglomeration on the whole, with Huangshan City as the main center and other districts and counties as the sub centers, presenting the trend of hierarchical development. In different kinds of tourism attractions, the distribution of natural tourism attractions and rural pastoral tourism attractions highly related to the topography. And the distribution of modern recreational tourism attractions and humanistic tourism attractions is closely related to hydrology. In terms of urban infrastructure, modern recreational tourism attractions hold the best accessibility with transportation. The core area of the demonstration region include Huangshan City, Chizhou city and Xuancheng city has better tourist industry development. Tourist attractions in other areas are relatively scarce, but the hold great potential for development in the future.
Single ion qubit with estimated coherence time exceeding one hour
Realizing a long coherence time quantum memory is a major challenge of current quantum technology. Until now, the longest coherence-time of a single qubit was reported as 660 s in a single 171 Yb + ion-qubit through the technical developments of sympathetic cooling and dynamical decoupling pulses, which addressed heating-induced detection inefficiency and magnetic field fluctuations. However, it was not clear what prohibited further enhancement. Here, we identify and suppress the limiting factors, which are the remaining magnetic-field fluctuations, frequency instability and leakage of the microwave reference-oscillator. Then, we observe the coherence time of around 5500 s for the 171 Yb + ion-qubit, which is the time constant of the exponential decay fit from the measurements up to 960 s. We also systematically study the decoherence process of the quantum memory by using quantum process tomography and analyze the results by applying recently developed resource theories of quantum memory and coherence. Our experimental demonstration will accelerate practical applications of quantum memories for various quantum information processing, especially in the noisy-intermediate-scale quantum regime. Extending qubit coherence times represent one of the key challenges for quantum technologies. Here, after properly suppressing magnetic-field fluctuations, frequency instability and leakage of the microwave reference-oscillator, the authors infer coherence times of 5500 s for an Yb ion qubit.
