Engineering a HEK-293T exosome-based delivery platform for efficient tumor-targeting chemotherapy/internal irradiation combination therapy
Exosomes are nanoscale monolayer membrane vesicles that are actively endogenously secreted by mammalian cells. Currently, multifunctional exosomes with tumor-targeted imaging and therapeutic potential have aroused widespread interest in cancer research. Herein, we developed a multifunctional HEK-293T exosome-based targeted delivery platform by engineering HEK-293T cells to express a well-characterized exosomal membrane protein (Lamp2b) fused to the αv integrin-specific iRGD peptide and tyrosine fragments. This platform was loaded with doxorubicin (Dox) and labeled with radioiodine-131 ( 131 I) using the chloramine-T method. iRGD exosomes showed highly efficient targeting and Dox delivery to integrin αvβ3-positive anaplastic thyroid carcinoma (ATC) cells as demonstrated by confocal imaging and flow cytometry in vitro and an excellent tumor-targeting capacity confirmed by single-photon emission computed tomography-computed tomography after labeling with 131 I in vivo. In addition, intravenous injection of this vehicle delivered Dox and 131 I specifically to tumor tissues, leading to significant tumor growth inhibition in an 8505C xenograft mouse model, while showing biosafety and no side effects. These as-developed multifunctional exosomes (denoted as Dox@iRGD-Exos- 131 I) provide novel insight into the current treatment of ATC and hold great potential for improving therapeutic efficacy against a wide range of integrin αvβ3-overexpressing tumors. Time-Series Cross-Radiometric Calibration and Validation of GF-6/WFV Using Multi-Site
The GaoFen6 (GF-6) satellite, equipped with a wide full-swath (WFV) sensor, offers high spatial resolution and extensive coverage, making it widely utilized in agricultural and forestry classification, land resource monitoring, and other fields. Accurate on-orbit radiometric calibration of GF-6/WFV is crucial for these quantitative applications. Currently, the absolute radiometric calibration of GF-6/WFV relies primarily on vicarious calibration conducted by the China Center for Resources Satellite Data and Application (CRESDA). However, annual vicarious calibration may not adequately capture the radiometric performance of GF-6/WFV due to performance degradation. Therefore, increasing the frequency of on-orbit radiometric calibration throughout the lifetime of GF-6/WFV is essential. This study proposes a method for conducting long-term cross-radiometric calibrations of GF-6/WFV by taking the multispectral imager (MSI) onboard the Sentinel-2 satellite as a reliable reference sensor and the sites from RadCalNet as reference ground targets. Firstly, we conducted 62 on-orbit cross-radiometric calibrations of GF-6/WFV since its launch by tracking with the Sentinel-2/MSI sensor after correcting the discrepancy spectrum and solar zenith angle. Then, validation of cross-radiometric calibration results against RadCalNet products indicated an average absolute relative error between 3.55% and 4.64%. Cross-validation with additional reference sensors, including Landsat-8/OLI and MODIS, confirmed the reliability of calibration, demonstrating relative differences from GF-6/WFV of less than 5%. Furthermore, the overall uncertainty of the cross-radiometric calibration was estimated to be from 4.08% to 4.89%. Finally, trend analysis of the time-series radiometric performance was also conducted and revealed an annual degradation rate ranging from 0.57% to 2.31%. This degradation affects surface reflectance retrieval, introducing a bias of approximately 0.0073 to 0.0084. Our findings highlight the operational effectiveness of the proposed method in achieving long-time-series on-orbit radiometric calibration and degradation monitoring of GF-6/WFV. The study also demonstrates that the radiometric performance of GF-6/WFV is relatively stable and suitable for further quantitative applications, especially for long-term monitoring applications.
Bio-based lubricants: Progress in research
Biomass is widely distributed, inexpensive, and easily obtainable. As a transportable and storable organic carbon source, biomass has significant advantages in terms of sustainability, environmental friendliness, resource richness, and versatility. The utilization of biomass resources instead of traditional non-renewable energy is advantageous for addressing issues related to energy, the environment, and product diversity. The conversion of biomass into energy chemicals serves as an effective complement to current energy products. In addition to their role as a protective agent, lubricants play a crucial role in machinery operation. This paper provides a comprehensive review of various plant-based lubricant-based oil synthesis technologies with promising applications, such as olefin metathesis, cross-lactone, plant sugar fermentation, and C-C coupling. It also covers modification techniques including additive, chemical, and biological modifications. Technical process of each synthesis and modification method is summarized, as well as the physical and chemical properties of the obtained lubricating oil products. This technology also overcomes limitations of traditional vegetable oil modification methods. A thorough analysis is provided on the performance and process economics of plant-based lubricant base oil synthesis technology to guide industrial development in this area. Additionally, it includes an analysis of future trends in plant-based lubricant synthesis technology.
Comparison of clinical efficacy between autologous partially demineralized dentin matrix and deproteinized bovine bone mineral for bone augmentation in orthodontic patients with alveolar bone deficiency: a randomized controlled clinical trial
Background It is common to see patients who need orthodontic treatment but with insufficient alveolar bone volume. However, safe and effective tooth movement requires sufficient alveolar bone width and height. The aim of this study is to compare the bone augmentation efficacy of Autologous Partially Demineralized Dentin Matrix (APDDM) and Deproteinized Bovine Bone Mineral (DBBM) in orthodontic patients with insufficient bone by using a randomized controlled clinical trial approach. Materials and methods Twenty-seven orthodontic patients involving 40 posterior teeth alveolar sites ( n  = 40) with insufficient alveolar bone volume were randomly divided into a control group ( n  = 20) and an experimental group ( n  = 20). The patients in the experimental group were treated with APDDM, and those in the control group were treated with DBBM. After surgery, the adjacent teeth are moved toward the bone grafting sites according to the orthodontic treatment plan. Patients completed a postoperative response questionnaire by the Visual Analogue Scale (VAS) score to indicate pain and swelling in the bone grafted area at the time of suture removal; and CBCT scans were conducted before surgery, 6 months and 2 years after surgery to assess changes in buccal and central alveolar heights, as well as widths at the alveolar ridge apex and 3 mm, 5 mm below the apex, respectively. The CBCT image sequences were imported into Mimics 21.0 software in DICOM format. The data of the patients in both groups were collected and analyzed by SPSS 25.0. Results The VAS scores were significantly lower in the APDDM group than in the DBBM group ( p  < 0.05). Significant increases were observed in alveolar bone height and width at 6 months and 2 years postoperative ( p  < 0.05); At 2 years, the APDDM group exhibited a reduction in buccal crest height and in 3 mm, 5 mm width below alveolar ridge apex, relative to 6 months ( p  < 0.05), while the DBBM group showed a decrease only in the central height of the alveolar bone ( p  < 0.05). There was a significant bone augmentation increase found only 3 mm below the alveolar ridge apex in the APDDM group compared with the DBBM group among all 6 months group comparison ( p  < 0.05). At 2 years, the augmentation effects were similar across both groups ( p  > 0.05). Conclusion Radiomics analysis indicates that APDDM serves as a viable bone augmentation material for orthodontic patients with insufficient alveolar bone volume, achieving comparable clinical efficacy to DBBM. Additionally, APDDM is associated with a milder postoperative response than DBBM. The registration number (TRN) ChiCTR2400084607.
The Spatiotemporal Evolution and Driving Factors of Surface Urban Heat Islands: A Comparative Study of Beijing and Dalian (2003–2023)
The urban heat island (UHI) effect significantly impacts urban environments and quality of life, yet research comparing coastal and inland cities is relatively lacking. This study, using the MYD11A2 dataset, analyzes the spatiotemporal evolution of land surface temperature (LST) and the surface urban heat island intensity index (SUHIII) in Beijing (inland) and Dalian (coastal) from 2003 to 2023, exploring the driving factors from 2003 to 2018 and proposing mitigation strategies for similar cities. Key findings: (1) Beijing’s SUHIII is 0.45 °C higher than Dalian’s during summer days, while Dalian’s SUHIII is 0.24 °C stronger than Beijing’s during winter nights, likely due to Dalian’s maritime climate, which raises nighttime LSTs and intensifies the winter SUHIII. (2) Both cities show similar trends in LST and SUHIII, with fluctuations until 2010, an increase after 2011, and a peak in 2023, with the expansion of heat island areas occurring mainly in suburban regions. (3) From 2003 to 2018, TEMP is the primary factor promoting SUHIII, followed by ET and POP, with EVI as the main mitigating factor. Beijing’s PREP weakens SUHI, while Dalian’s PREP promotes it. Coastal cities should focus on water bodies and wetland planning to mitigate heat islands, especially in areas like Dalian which are affected by PREP.
Retrospective study of the influence of hypothyroidism on liver function before radioiodine therapy in China: a comparison analysis based on patients with differentiated thyroid cancer
PurposeThe aim of the present study is to investigate the risk factors for hepatic dysfunction before radioiodine therapy in patients with differentiated thyroid cancer (DTC).Methods996 patients (314 men, 682 women; age of 45.07±12.98 years) with postoperative DTC were recruited and divided into two groups including patients with and without hepatic dysfunction. The changes in baseline data and traced liver function levels, together with other metabolic profiles, were compared between the two groups.ResultOverall, 31.6% of patients had hepatic dysfunction. Higher aspartate aminotransferase and/or alanine aminotransferase was the most common abnormality (the prevalence rate was 47.5%). The percentages of mild and moderate hepatic dysfunction were 80.0% and 20.0%, respectively. Univariate analyses demonstrated that the most prominent risk factors for hepatic dysfunction (OR=0.324–3.171, p<0.01) were male sex with levothyroxine discontinuation and free triiodothyronine <2.01 pmol/L, free thyroxine (FT4) <4.78 pmol/L, thyroid-stimulating hormone >78.195 µIU/mL, total cholesterol >5.17 mmol/L, triglycerides (TG) >1.71 mmol/L and more than 21 days of thyroid hormone withdrawal. Multivariate analyses demonstrated that for men, FT4 <3.80 pmol/L and TG ≥1.28 mmol/L were the most prominent risk factors.ConclusionsPatients with minor hepatic dysfunction and ortholiposis are more likely to recover to normal liver function. Patients with moderate hepatic dysfunction should be treated with hepatoprotective drugs. For men, FT4 and TG levels tended to be associated with hepatic dysfunction, and the prognosis of hepatic dysfunction was closely related to the TG level.
Implant restoration of hypodontia resulting from ectodermal dysplasia: a case report
Background Ectodermal dysplasia is a congenital genetic disorder with a prevalence of 1:10,000 to 1:100,000. The clinical features of ectodermal dysplasia include sparse hair, missing teeth, and abnormal development of the skin, sweat glands, and other tissues and organs. Since 1985, dentists have used implants to correct tooth defects in patients with ectodermal dysplasia with reasonable success rates. However, there is still no widely accepted treatment for the oral defects caused by this disease. Case summary: This report describes a 19-year-old female patient with congenital ectodermal dysplasia, congenital absence of most of her teeth, and mandibular and maxillary dysplasia. The patient and her family were concerned about the patient’s growth and development, so she came to our hospital to restore her missing teeth. During several months of treatment, the patient underwent bone augmentation surgery for the maxilla and mandible, implant placement, a root repositioning flap, and free gingival grafting, which was completed in five stages. The patient completed the permanent implant restoration, and the desired results were achieved. Conclusion The patient’s mouth was restored through the use of bone augmentation and soft tissue grafting techniques, enabling appropriate function and appearance.
Analysis of risk factors of rapid thyroidal radioiodine-131 turnover in Graves’ disease patients
Rapid iodine-131( 131 I) turnover in the thyroid gland is an important feature of Graves’ disease (GD) and also a strong predictor of radioiodine therapy failure. The aim of this study was to explore the predictors of rapid 131 I turnover. The clinical data on 2543 patients were retrospectively reviewed. Patients were divided into 2 groups depending on present or absent with rapid 131 I turnover defined as a 4-hour to 24-hour 131 I uptake ratio of ≥1. Overall, 590 cases (23.2%) had a rapid 131 I turnover. In the univariate analysis, gender, age, FT 3 /FT 4 concentration, disease duration, with or without antithyroid drugs (ATD), time of ATD, thyroid weight and thyroid textures displayed significant differences. Cutoff values of age, FT 3 and thyroid weight to predict rapid 131 I turnover were 38 years, 35 pmol/l and 56 g by receiver operating characteristic curves. Binary logistic regression analysis further revealed higher probability of rapid 131 I turnover in patients with thyroid weight ≥56 g (odds ratio [OR]:3.7, 95% confidence interval [CI]: 3.032–4.559), age <38 years (OR:2.3, 95%CI: 1.906–2.856), FT 3 concentration ≥35 pmol/l (OR:7.6, 95%CI: 5.857–8.563) and females (OR:2.2, 95%CI: 1.757–2.791). In conclusion, larger goiters, younger age, higher FT 3 concentration and females are independently associated with rapid 131 I turnover in GD patients.
A new confuciusornithid bird with a secondary epiphyseal ossification reveals phylogenetic changes in confuciusornithid flight mode
The confuciusornithids are the earliest known beaked birds, and constitute the only species-rich clade of Early Cretaceous pygostylian birds that existed prior to the cladogenesis of Ornithothoraces. Here, we report a new confuciusornithid species from the Lower Cretaceous of western Liaoning, northeastern China. Compared to other confuciusornithids, this new species and the recently reported Yangavis confucii both show evidence of stronger flight capability, although the wings of the two taxa differ from one another in many respects. Our aerodynamic analyses under phylogeny indicate that varying modes of flight adaptation emerged across the diversity of confuciusornithids, and to a lesser degree over the course of their ontogeny, and specifically suggest that both a trend towards improved flight capability and a change in flight strategy occurred in confuciusornithid evolution. The new confuciusornithid differs most saliently from other Mesozoic birds in having an extra cushion-like bone in the first digit of the wing, a highly unusual feature that may have helped to meet the functional demands of flight at a stage when skeletal growth was still incomplete. The new find strikingly exemplifies the morphological, developmental and functional diversity of the first beaked birds. A new species of Confuciusornis , a genus of basal birds from the Early Cretaceous, reveals that these early birds had stronger flight capability and more diverse flight modes than previously expected.
Sleep Quality of Patients with Differentiated Thyroid Cancer
We aimed to measure prevalence of sleep disturbance in patients with differentiated thyroid cancer (DTC) by calculating Pittsburgh Sleep Quality Index (PSQI), and compare these data with patients with benign thyroid nodules or normal participants. Three groups of patients participated in this cross-sectional study. In the first group, 162 patients with DTC received total thyroidectomy, and then 131I therapy. The second group consisted of 84 patients with benign thyroid nodules, who received partial thyroidectomy. The third group was 78 normal healthy control cases. PSQI was used to assess the sleep quality. Inter-group differences were analyzed by Kruskal-Wallis test or independent samples T test. χ2 test was also used to check prevalence differences of poor sleep quality among the groups. Differences of PSQI score and poor sleep quality prevalence before and after 131I therapy in the same group of DTC participants were analyzed by paired T test and Mcnemar's test. Higher PSQI score (7.59 ± 4.21) and higher rate of poor sleep quality (54.32%) were shown in DTC patients than in any other group. After 131I therapy, PSQI score and prevalence of poor sleep quality in DTC patients increased significantly to 8.78 ± 4.72 and 70.99%. Then DTC patients were divided into two subgroups based on their metastatic status. DTC patients with metastasis (87/162 cases, 53.70%) had significantly higher PSQI score (10.87 ± 5.18) and higher prevalence of poor sleep quality (79.31%). DTC patients suffer from sleep disturbance, 131I therapy and awareness of metastatic status could worsen sleep problem. Psychological fear of cancer, nuclear medicine therapy and metastasis could be one major underlying reason. Longitude and interventional studies are necessary for further investigations.
