Mechanically robust lattices inspired by deep-sea glass sponges
The predominantly deep-sea hexactinellid sponges are known for their ability to construct remarkably complex skeletons from amorphous hydrated silica. The skeletal system of one such species of sponge, Euplectella aspergillum , consists of a square-grid-like architecture overlaid with a double set of diagonal bracings, creating a chequerboard-like pattern of open and closed cells. Here, using a combination of finite element simulations and mechanical tests on 3D-printed specimens of different lattice geometries, we show that the sponge’s diagonal reinforcement strategy achieves the highest buckling resistance for a given amount of material. Furthermore, using an evolutionary optimization algorithm, we show that our sponge-inspired lattice geometry approaches the optimum material distribution for the design space considered. Our results demonstrate that lessons learned from the study of sponge skeletal systems can be exploited for the realization of square lattice geometries that are geometrically optimized to avoid global structural buckling, with implications for improved material use in modern infrastructural applications. Computational analysis and mechanical testing demonstrate that the skeletal system of a marine sponge has, through the course of evolution, achieved a near-optimal resistance to buckling.
Bone marrow–on–a–chip replicates hematopoietic niche physiology in vitro
Bone marrow formed in a cylindrical PDMS device implanted in a mouse can be surgically removed and cultured for a week in vitro without losing any of the hallmarks of in vivo bone marrow niches. Current in vitro hematopoiesis models fail to demonstrate the cellular diversity and complex functions of living bone marrow; hence, most translational studies relevant to the hematologic system are conducted in live animals. Here we describe a method for fabricating 'bone marrow–on–a–chip' that permits culture of living marrow with a functional hematopoietic niche in vitro by first engineering new bone in vivo , removing it whole and perfusing it with culture medium in a microfluidic device. The engineered bone marrow (eBM) retains hematopoietic stem and progenitor cells in normal in vivo –like proportions for at least 1 week in culture. eBM models organ-level marrow toxicity responses and protective effects of radiation countermeasure drugs, whereas conventional bone marrow culture methods do not. This biomimetic microdevice offers a new approach for analysis of drug responses and toxicities in bone marrow as well as for study of hematopoiesis and hematologic diseases in vitro .
Human Gut-On-A-Chip Supports Polarized Infection of Coxsackie B1 Virus In Vitro
Analysis of enterovirus infection is difficult in animals because they express different virus receptors than humans, and static cell culture systems do not reproduce the physical complexity of the human intestinal epithelium. Here, using coxsackievirus B1 (CVB1) as a prototype enterovirus strain, we demonstrate that human enterovirus infection, replication and infectious virus production can be analyzed in vitro in a human Gut-on-a-Chip microfluidic device that supports culture of highly differentiated human villus intestinal epithelium under conditions of fluid flow and peristalsis-like motions. When CVB1 was introduced into the epithelium-lined intestinal lumen of the device, virions entered the epithelium, replicated inside the cells producing detectable cytopathic effects (CPEs), and both infectious virions and inflammatory cytokines were released in a polarized manner from the cell apex, as they could be detected in the effluent from the epithelial microchannel. When the virus was introduced via a basal route of infection (by inoculating virus into fluid flowing through a parallel lower 'vascular' channel separated from the epithelial channel by a porous membrane), significantly lower viral titers, decreased CPEs, and delayed caspase-3 activation were observed; however, cytokines continued to be secreted apically. The presence of continuous fluid flow through the epithelial lumen also resulted in production of a gradient of CPEs consistent with the flow direction. Thus, the human Gut-on-a-Chip may provide a suitable in vitro model for enteric virus infection and for investigating mechanisms of enterovirus pathogenesis.
A 3D-printed, functionally graded soft robot powered by combustion
Roboticists have begun to design biologically inspired robots with soft or partially soft bodies, which have the potential to be more robust and adaptable, and safer for human interaction, than traditional rigid robots. However, key challenges in the design and manufacture of soft robots include the complex fabrication processes and the interfacing of soft and rigid components. We used multimaterial three-dimensional (3D) printing to manufacture a combustion-powered robot whose body transitions from a rigid core to a soft exterior. This stiffness gradient, spanning three orders of magnitude in modulus, enables reliable interfacing between rigid driving components (controller, battery, etc.) and the primarily soft body, and also enhances performance. Powered by the combustion of butane and oxygen, this robot is able to perform untethered jumping.
A three-dimensional actuated origami-inspired transformable metamaterial with multiple degrees of freedom
Reconfigurable devices, whose shape can be drastically altered, are central to expandable shelters, deployable space structures, reversible encapsulation systems and medical tools and robots. All these applications require structures whose shape can be actively controlled, both for deployment and to conform to the surrounding environment. While most current reconfigurable designs are application specific, here we present a mechanical metamaterial with tunable shape, volume and stiffness. Our approach exploits a simple modular origami-like design consisting of rigid faces and hinges, which are connected to form a periodic structure consisting of extruded cubes. We show both analytically and experimentally that the transformable metamaterial has three degrees of freedom, which can be actively deformed into numerous specific shapes through embedded actuation. The proposed metamaterial can be used to realize transformable structures with arbitrary architectures, highlighting a robust strategy for the design of reconfigurable devices over a wide range of length scales. Typically, most structures and devices that can be reconfigured are designed with application specific requirements. Inspired by modular origami ideas, Overvelde et al . present a mechanical metamaterial enabling the design of three-dimensional structures of arbitrary architecture with tunable shape, volume and stiffness.
A mid-Cambrian tunicate and the deep origin of the ascidiacean body plan
Tunicates are an evolutionarily significant subphylum of marine chordates, with their phylogenetic position as the sister-group to Vertebrata making them key to unraveling our own deep time origin. Tunicates greatly vary with regards to morphology, ecology, and life cycle, but little is known about the early evolution of the group, e.g. whether their last common ancestor lived freely in the water column or attached to the seafloor. Additionally, tunicates have a poor fossil record, which includes only one taxon with preserved soft-tissues. Here we describe Megasiphon thylakos nov., a 500-million-year-old tunicate from the Marjum Formation of Utah, which features a barrel-shaped body with two long siphons and prominent longitudinal muscles. The ascidiacean-like body of this new species suggests two alternative hypotheses for early tunicate evolution. The most likely scenario posits M. thylakos belongs to stem-group Tunicata, suggesting that a biphasic life cycle, with a planktonic larva and a sessile epibenthic adult, is ancestral for this entire subphylum. Alternatively, a position within the crown-group indicates that the divergence between appendicularians and all other tunicates occurred 50 million years earlier than currently estimated based on molecular clocks. Ultimately, M. thylakos demonstrates that fundamental components of the modern tunicate body plan were already established shortly after the Cambrian Explosion. Our understanding of the origins of tunicates, an important group of filter-feeding marine invertebrate chordates, is limited due to a poor fossil record. Here, the authors present a 500 million year old tunicate fossil, demonstrating that the modern tunicate body plan was established shortly after the Cambrian Explosion.
Bioinspired design of flexible armor based on chiton scales
Man-made armors often rely on rigid structures for mechanical protection, which typically results in a trade-off with flexibility and maneuverability. Chitons, a group of marine mollusks, evolved scaled armors that address similar challenges. Many chiton species possess hundreds of small, mineralized scales arrayed on the soft girdle that surrounds their overlapping shell plates. Ensuring both flexibility for locomotion and protection of the underlying soft body, the scaled girdle is an excellent model for multifunctional armor design. Here we conduct a systematic study of the material composition, nanomechanical properties, three-dimensional geometry, and interspecific structural diversity of chiton girdle scales. Moreover, inspired by the tessellated organization of chiton scales, we fabricate a synthetic flexible scaled armor analogue using parametric computational modeling and multi-material 3D printing. This approach allows us to conduct a quantitative evaluation of our chiton-inspired armor to assess its orientation-dependent flexibility and protection capabilities. Biology has often served as the inspiration for the design of body armor; one common limitation is the flexibility of the resultant armor. Here, the authors examine the armour of chiton and use the observed design principles to 3D print flexible armor.
The Stomatopod Dactyl Club: A Formidable Damage-Tolerant Biological Hammer
Nature has evolved efficient strategies to synthesize complex mineralized structures that exhibit exceptional damage tolerance. One such example is found in the hypermineralized hammer-like dactyl clubs of the stomatopods, a group of highly aggressive marine crustaceans. The dactyl clubs from one species, Odontodactylus scyllarus, exhibit an impressive set of characteristics adapted for surviving high-velocity impacts on the heavily mineralized prey on which they feed. Consisting of a multiphase composite of oriented crystalline hydroxyapatite and amorphous calcium phosphate and carbonate, in conjunction with a highly expanded helicoidal organization of the fibrillar chitinous organic matrix, these structures display several effective lines of defense against catastrophic failure during repetitive high-energy loading events.
A framework for handweaving robotic textiles with liquid crystal elastomer fibers
Textile production methods present a rich set of strategies for developing materials with both form and function encoded at the fiber scale. Beyond simply acting as a static flexible barrier, the ability to incorporate environmentally responsible materials into fabric architectures significantly expands the textile design space by adding on-demand and programmable 3D structural morphing. To this end, liquid crystal elastomers (LCEs) are a promising candidate for enabling these reversible actuation behaviors in fabric-based constructs. Drawing on traditional textile manufacturing techniques and through a detailed exploration of the vast woven textile design space, we have demonstrated programmable and reversible curling, puffing, and in-plane shrinkage behaviors by embedding the functionality of LCE fibers into single and multi-layered woven structures. Predictable shifts in fabric structure directly influence the mechanical properties and the resulting form factor of the actuated textiles, which can in turn be effectively leveraged for the generation of multi-functional devices, enabling new directions for the engineering of flexible stimuli-responsive materials.
Hierarchical structural design for fracture resistance in the shell of the pteropod Clio pyramidata
The thecosomes are a group of planktonic pteropods with thin, 1 mm-sized aragonitic shells, which are known to possess a unique helical microstructure consisting of interlocking nanofibres. Here we investigate the detailed hierarchical structural and mechanical design of the pteropod Clio pyramidata . We quantify and elucidate the macroscopic distribution of the helical structure over the entire shell (~1 mm), the structural characteristics of the helical assembly (~10–100 μm), the anisotropic cross-sectional geometry of the fibrous building blocks (~0.5–10 μm) and the heterogeneous distribution of intracrystalline organic inclusions within individual fibres (<0.5 μm). A global fibre-like crystallographic texture is observed with local in-plane rotations. Microindentation and electron microscopy studies reveal that the helical organization of the fibrous building blocks effectively constrains mechanical damages through tortuous crack propagation. Uniaxial micropillar compression and cross-sectional transmission electron microscopy directly reveal that the interlocking fibrous building blocks further retard crack propagation at the nanometre scale. The hierarchical structural motifs of biomaterials can lead to advantageous mechanical properties. Here, the authors reveal that a fibre-like helical structure across the shell of a planktonic pteropod suppresses crack propagation and is responsible for a high fracture resistance.
