A phase II, single-center, randomized, double-blind, parallel control clinical study evaluating the immunogenicity and safety of a two-dose schedule of serogroups ACYW meningococcal polysaccharide conjugate vaccine
This was a single-center, randomized, double-blind, parallel control study evaluating the immunogenicity and safety of a two-dose schedule of serogroups ACYW meningococcal polysaccharide conjugate vaccine with tetanus toxoid (TT) conjugate protein, in infants and toddlers of 3–35 months old. 720 participants were stratified according to the age of 3–5 months old, 6–11 months old, and 12–35 months old and randomly assigned with an equal ratio to two different dose groups, i.e., 40- and 20-μg doses. Blood samples were taken from all participants before the first vaccination and 30 days after the full-course vaccination to detect the serogroups ACYW meningococcal antibodies. All adverse events occurred within 30 days after vaccination of each dose, and serious adverse events occurred within six months after full-course vaccination were collected for safety evaluation. This study was registered at the China drug trial registration with the identifier CTR 20182031. After 30 days of full-course vaccination, 92.78 % (95 % CI: 85.70 %-100.00 %) showed the immune response against all serogroups in both high-dose and low-dose groups by rabbit serum bactericidal antibody assay (rSBA) and the geometric mean titer (GMT) of all serogroups showed a high level (74.6–505.8, 95 % CI: 56.4–615.7). However, no significant difference between different dose groups was observed (P > 0.05). The common local and systemic adverse events in both groups were redness (3 %-7%), and fever (26 %-65 %), respectively. In addition, the grade 3 adverse event related to the vaccine was fever (1.67 %-12.50 %). No serious adverse event was reported to be associate with the vaccination. The serogroups ACYW meningococcal polysaccharide conjugate vaccine was safe and effective in the population aged 3–35 months. The vaccine efficacy and safety of the 20-μg dose group were not less than that of the 40-μg dose group.
Efficacy and safety of a pentavalent live human-bovine reassortant rotavirus vaccine (RV5) in healthy Chinese infants: A randomized, double-blind, placebo-controlled trial
•A randomized, double-blind, placebo-controlled trial was conducted in healthy Chinese infants.•Efficacy and safety of a pentavalent rotavirus vaccine (RV5) were assessed.•Efficacy against rotavirus gastroenteritis of any-severity caused by any serotype was 69.3%.•Efficacy against severe rotavirus gastroenteritis caused by any serotype was 78.9%.•RV5 was efficacious and generally well-tolerated in Chinese infants. A randomized, double-blind, placebo-controlled multicenter trial was conducted in healthy Chinese infants to assess the efficacy and safety of a pentavalent live human-bovine reassortant rotavirus vaccine (RotaTeq™, RV5) against rotavirus gastroenteritis (RVGE). 4040 participants aged 6–12weeks were enrolled and randomly assigned to either 3 oral doses of RV5 (n=2020) or placebo (n=2020), administered ∼4weeks apart. The participants also received OPV and DTaP in a concomitant or staggered fashion. The primary objective was to evaluate vaccine efficacy (VE) against naturally-occurring RVGE at least 14days following the third dose. Key secondary objectives included: VE against naturally-occurring severe RVGE and VE against severe and any-severity RVGE caused by rotavirus serotypes contained in the vaccine, occurring at least 14days after the third dose. All adverse events (AEs) were collected for 30days following each dose. Serious AEs (SAEs) and intussusception cases were collected during the entire study. (ClinicalTrials.gov registry: NCT02062385). VE against RVGE of any-severity caused by any serotype was 69.3% (95% CI: 54.5, 79.7). The secondary efficacy analysis showed an efficacy of: 78.9% (95% CI: 59.1, 90.1) against severe RVGE caused by any serotype; 69.9% (95% CI: 55.2, 80.3) and 78.9% (95% CI: 59.1, 90.1) against any-severity and severe RVGE caused by serotypes contained in the vaccine, respectively. Within 30days following any vaccination, 53.5% (1079/2015) and 53.3% (1077/2019) of participants reported at least one AE, and 5.8% (116/2015) and 5.7% (116/2019) reported SAEs in the vaccine and placebo groups, respectively. No SAEs were considered vaccine-related in recipients of RV5. Two intussusception cases were reported in recipients of RV5 who recovered after receiving treatment. Neither was considered vaccine-related. In Chinese infants, RV5 was efficacious against any-severity and severe RVGE caused by any serotype and generally well-tolerated with respect to AEs.
Poliovirus shedding after sequential immunization of Sabin-strain inactivated polio vaccines and oral attenuated polio vaccines
The identification of poliovirus in stool samples post-Oral Poliovirus Vaccine (OPV) immunization is essential for implementing post-eradication monitoring of poliomyelitis. This research presents the inaugural assessment of the virus shedding rate and the genetic diversity of OPV shedding strains across different Inactivated Poliovirus Vaccine (IPV)-OPV sequential immunization schedules. Our findings revealed that the shedding rate of different serotypes in each sequential immunization groups peaked within 7 days following the initial administration of OPV, and then gradually decreased. Prior vaccination with OPV reduced the rate and shortened the duration of virus shedding. Additionally, we observed a slight increase in the shedding rate of type3 after the removal of type2 from trivalent oral poliovirus vaccine (tOPV). The comprehensive analysis of the whole-genome high-throughput sequencing results for the shedding strain revealed that the variation sites among samples from different sequential immunization groups were distributed throughout the entire genome. The mutation frequencies within the 5’NCR, 2 C, 3 A, 3 C, and 3D regions were elevated of type1 and type3, while type2 had higher frequencies within the 5’NCR, VP1 and 3D regions. Consequently, it is imperative to expedite the transition from OPV to IPV, and to discontinue OPV as soon as wild poliovirus strains and vaccine-derived poliovirus (VDPVs) are eliminated.
Research Progress on Separation and Extraction Technologies of Lignin
Lignin, a complex natural three-dimensional aromatic polymer, is prone to condensation during the separation process, owing to the diverse properties of its basic structural units, linkage types, and spatial configurations. These inherent structural complexities present significant challenges for its efficient isolation and precise transformation. Current separation techniques primarily include physical, chemical (such as acid hydrolysis, alkaline dissolution, organic solvents, and ionic liquids), and biological methods. Each approach offers distinct advantages and limitations in terms of yield, purity, cost, and impact on lignin structure. Studies have indicated that ionic liquids and organic solvent methods demonstrate considerable application potential owing to their mild reaction conditions and high selectivity. Future research should focus on developing green, efficient, and low-cost separation technologies, while also enhancing detailed structural characterization and targeted lignin conversion to facilitate its large-scale utilization in the production of value-added materials and chemicals.
Overcoming the Limitations of Transition-Metal Catalysis in the Chemoenzymatic Dynamic Kinetic Resolution (DKR) of Atropisomeric Bisnaphthols
Chemoenzymatic dynamic kinetic resolution (DKR), combining a metal racemization catalyst with an enzyme, has emerged as an elegant solution to transform racemic substrates into enantiopure products, while compatibility of dual catalysis is the key issue. Conventional solutions have utilized presynthesized metal complexes with a fixed and bulky ligand to protect the metal from the enzyme system; however, this has been generally limited to anionic ligands. Herein, we report our strategy to solve the compatibility issue by employing a reliable ligand that firmly coordinates in situ to the metal. Such a reliable ligand offers π* orbitals, allowing additional metal-to-ligand d−π* back-donation, which can significantly enhance coordination effects between the ligand and metal. Therefore, we developed an efficient DKR method to access chiral BINOLs from racemic derivatives under dual copper and enzyme catalysis. In cooperation with lipase LPL-311-Celite, the DKR of BINOLs was successfully realized with a copper catalyst via in situ coordination of BCP (L8) to CuCl. A series of functionalized C 2- and C 1-symmetric chiral biaryls could be synthesized in high yields with good enantioselectivity. The racemization mechanism was proposed to involve a radical-anion intermediate, which allows the axial rotation with a dramatic decrease of the rotation barrier.
Research on Logistics Service Assessment for Smart City: A Users’ Review Sentiment Analysis Approach
The innovative development of logistics has become a powerful starting point and strong support for the construction of smart cities. An accurate evaluation of logistics service quality can promote intelligent transformation, and upgrading logistics enterprises can improve the urban public service infrastructure. In this study, we propose a logistics service quality evaluation model based on a combination of sentiment analysis technology and a traditional index evaluation system. With the help of sentiment analysis technology, the model focuses on extracting the sentiment characteristics of logistics service quality from user reviews and analyzing user attitudes from different aspects. We designed a new logistics service quality evaluation index system by improving the SERVQUAL model. The system uses sentiment analysis technology to explore evaluation content through feature extraction and builds relations between the evaluation content and indices. Additionally, we use sentiment orientation analysis with different indices to comprehensively evaluate service quality. The experimental analysis shows that the proposed model and algorithm have high accuracy.
Emulsification and Performance Measurement of Bio-oil with Diesel
To overcome the drawbacks of pyrolytic bio-oil, such as high acidity, viscidity, corrosivity, oxygen content and low heating value, a novel method of emulsifying bio-oil with diesel was investigated in this study. The effects of various factors on the stability of emulsified oil were examined. The optimal emulsification conditions of diesel and bio-oil are by using an emulsifier (mixture of Span 80 and Tween 80) dosage of 6% by mass and co-emulsifier (2-octanol) dosage of 4% by the mass of emulsifier, hydrophile lipophilic balance value of 7, shear velocity of 15,000 rpm, emulsifying time of 5 min, and emulsifying temperature at 40 °C. The stability time of emulsified oil is up to 384 h under the above optimal conditions. Some fuel properties were also measured to characterize the emulsified oil. The results show that the density, viscosity, corrosivity and heating value of emulsified oil at the optimal conditions are close to those of diesel oil. In addition, the mechanism model for the emulsion of bio-oil and diesel was also proposed. Graphic Abstract
The influence of load type on electromagnetic wave radiation during the opening of circuit breaker
Radiated electromagnetic waves during the opening of the circuit breaker can be used to evaluate the arc extinguishing performance of the circuit breaker. Current research shows that the duration of electromagnetic waves is the main feature. However, the current research does not consider the influence of the type of load on the electromagnetic wave radiation during the opening process. In this paper, an experimental research platform is established, and the influence of the load type on the electromagnetic wave radiation during the opening process is studied by testing the resistance, inductance and capacitance loads. The research results show that the arcing time and visible arcing times of breaking capacitive loads are significantly greater than resistive and inductive loads.
