The middle ground : Indians, empires, and republics in the Great Lakes region, 1650-1815
\"An acclaimed book and widely acknowledged classic, The Middle Ground steps outside the simple stories of Indian-white relations - stories of conquest and assimilation and stories of cultural persistence. It is, instead, about a search for accommodation and common meaning. High-Throughput Sequencing of the Zebrafish Antibody Repertoire

Despite tremendous progress in understanding the nature of the immune system, the full diversity of an organism's antibody repertoire is unknown. We used high-throughput sequencing of the variable domain of the antibody heavy chain from 14 zebrafish to analyze VDJ usage and antibody sequence. Zebrafish were found to use between 50 and 86% of all possible VDJ combinations and shared a similar frequency distribution, with some correlation of VDJ patterns between individuals. Zebrafish antibodies retained a few thousand unique heavy chains that also exhibited a shared frequency distribution. We found evidence of convergence, in which different individuals made the same antibody. This approach provides insight into the breadth of the expressed antibody repertoire and immunological diversity at the level of an individual organism.

Railroaded : the transcontinentals and the making of modern America
This work is a history of the transcontinental railroads and how they transformed America in the decades after the Civil War. The transcontinental railroads of the late nineteenth century were the first corporate behemoths. Their attempts to generate profits from proliferating debt sparked devastating panics in the U.S. economy. Their dependence on public largess drew them into the corridors of power, initiating new forms of corruption. Their operations rearranged space and time, and remade the landscape of the West. As wheel and rail, car and coal, they opened new worlds of work and ways of life. Their discriminatory rates sparked broad opposition and a new antimonopoly politics. With characteristic originality, range, and authority, Richard White shows the transcontinentals to be pivotal actors in the making of modern America. But the triumphal myths of the golden spike, robber barons larger than life, and an innovative capitalism all die here. Instead we have a new vision of the Gilded Age, often darkly funny, that shows history to be rooted in failure as well as success.
Sex Differences in Tuberculosis Burden and Notifications in Low- and Middle-Income Countries: A Systematic Review and Meta-analysis

Tuberculosis (TB) case notification rates are usually higher in men than in women, but notification data are insufficient to measure sex differences in disease burden. This review set out to systematically investigate whether sex ratios in case notifications reflect differences in disease prevalence and to identify gaps in access to and/or utilisation of diagnostic services.
In accordance with the published protocol (CRD42015022163), TB prevalence surveys in nationally representative and sub-national adult populations (age ≥ 15 y) in low- and middle-income countries published between 1 January 1993 and 15 March 2016 were identified through searches of PubMed, Embase, Global Health, and the Cochrane Database of Systematic Reviews; review of abstracts; and correspondence with the World Health Organization. Random-effects meta-analyses examined male-to-female (M:F) ratios in TB prevalence and prevalence-to-notification (P:N) ratios for smear-positive TB. Meta-regression was done to identify factors associated with higher M:F ratios in prevalence and higher P:N ratios. Eighty-three publications describing 88 surveys with over 3.1 million participants in 28 countries were identified (36 surveys in Africa, three in the Americas, four in the Eastern Mediterranean, 28 in South-East Asia and 17 in the Western Pacific). Fifty-six surveys reported in 53 publications were included in quantitative analyses. Overall random-effects weighted M:F prevalence ratios were 2.21 (95% CI 1.92-2.54; 56 surveys) for bacteriologically positive TB and 2.51 (95% CI 2.07-3.04; 40 surveys) for smear-positive TB. M:F prevalence ratios were highest in South-East Asia and in surveys that did not require self-report of signs/symptoms in initial screening procedures. The summary random-effects weighted M:F ratio for P:N ratios was 1.55 (95% CI 1.25-1.91; 34 surveys). We intended to stratify the analyses by age, HIV status, and rural or urban setting; however, few studies reported such data.
TB prevalence is significantly higher among men than women in low- and middle-income countries, with strong evidence that men are disadvantaged in seeking and/or accessing TB care in many settings. Global strategies and national TB programmes should recognise men as an underserved high-risk group and improve men's access to diagnostic and screening services to reduce the overall burden of TB more effectively and ensure gender equity in TB care.

Spatially resolved transcriptomics reveals the architecture of the tumor-microenvironment interface

During tumor progression, cancer cells come into contact with various non-tumor cell types, but it is unclear how tumors adapt to these new environments. Here, we integrate spatially resolved transcriptomics, single-cell RNA-seq, and single-nucleus RNA-seq to characterize tumor-microenvironment interactions at the tumor boundary. Using a zebrafish model of melanoma, we identify a distinct “interface” cell state where the tumor contacts neighboring tissues. This interface is composed of specialized tumor and microenvironment cells that upregulate a common set of cilia genes, and cilia proteins are enriched only where the tumor contacts the microenvironment. Cilia gene expression is regulated by ETS-family transcription factors, which normally act to suppress cilia genes outside of the interface. A cilia-enriched interface is conserved in human patient samples, suggesting it is a conserved feature of human melanoma. Our results demonstrate the power of spatially resolved transcriptomics in uncovering mechanisms that allow tumors to adapt to new environments.

Emergence of blueschists on Earth linked to secular changes in oceanic crust composition

An absence in the ancient geological record of blueschist metamorphic rocks has been taken as evidence against early subduction. Thermodynamic calculations now suggest that blueschist rocks could not have formed on a younger, hotter Earth.
The oldest blueschists—metamorphic rocks formed during subduction—are of Neoproterozoic age

, and 0.7–0.8 billion years old. Yet, subduction of oceanic crust to mantle depths is thought to have occurred since the Hadean, over 4 billion years ago

. Blueschists typically form under cold geothermal gradients of less than 400 °C GPa
−1
, so their absence in the ancient rock record is typically attributed to hotter pre-Neoproterozoic mantle prohibiting such low-temperature metamorphism; however, modern analogues of Archaean subduction suggest that blueschist-facies metamorphic conditions are attainable at the slab surface

. Here we show that the absence of blueschists in the ancient geological record can be attributed to the changing composition of oceanic crust throughout Earth history, which is a consequence of secular cooling of the mantle since the Archaean

. Oceanic crust formed on the hot, early Earth would have been rich in magnesium oxide (MgO). We use phase equilibria calculations to show that blueschists do not form in high-MgO rocks under subduction-related geothermal gradients. Instead, the subduction of MgO-rich oceanic crust would have created greenschist-like rocks—metamorphic rocks formed today at low temperatures and pressures. These ancient metamorphic products can hold about 20% more water than younger metamorphosed oceanic crust, implying that the global hydrologic cycle was more efficient in the deep geological past than today.

Who killed Jane Stanford? : a gilded age tale of murder, deceit, spirits and the birth of a university
\"A premier historian penetrates the fog of corruption and cover-up still surrounding the murder of a Stanford University founder to establish who did it, how, and why. In 1885 Jane and Leland Stanford cofounded a university to honor their recently deceased young son. After her husband's death in 1893, Jane Stanford, a devoted spiritualist who expected the university to inculcate her values, steered Stanford into eccentricity and public controversy for more than a decade. In 1905 she was murdered in Hawaii, a victim, according to the Honolulu coroner's jury, of strychnine poisoning. A zebrafish melanoma model reveals emergence of neural crest identity during melanoma initiation

In cancer biology, a tumor begins from a single cell within a group of precancerous cells that share genetic mutations. Kaufman et al. used a zebrafish melanoma model to visualize cancer initiation (see the Perspective by Boumahdi and Blanpain). They used a fluorescent reporter that specifically lit up neural crest progenitors that are only present during embryogenesis or during adult melanoma tumor formation. The appearance of this tumor correlated with a set of gene regulatory elements, called super-enhancers, whose identification and manipulation may prove beneficial in detecting and preventing melanoma initiation. Science , this issue p. 10.1126/science.aad2197 ; see also p. 453 Melanocytes with oncogenic or tumor suppressor mutations revert to expressing the crestin gene early in melanoma formation. [Also see Perspective by Boumahdi and Blanpain ] The “cancerized field” concept posits that cancer-prone cells in a given tissue share an oncogenic mutation, but only discreet clones within the field initiate tumors. Most benign nevi carry oncogenic BRAF V600E mutations but rarely become melanoma. The zebrafish crestin gene is expressed embryonically in neural crest progenitors (NCPs) and specifically reexpressed in melanoma. Live imaging of transgenic zebrafish crestin reporters shows that within a cancerized field ( BRAF V600E - mutant; p53- deficient), a single melanocyte reactivates the NCP state, revealing a fate change at melanoma initiation in this model. NCP transcription factors, including sox10 , regulate crestin expression. Forced sox10 overexpression in melanocytes accelerated melanoma formation, which is consistent with activation of NCP genes and super-enhancers leading to melanoma. Our work highlights NCP state reemergence as a key event in melanoma initiation.
