Rectal Distension Increased the Rectoanal Gradient in Patients with Normal Rectal Sensory Function
BackgroundFrequent observation of abnormal manometric patterns consistent with dyssynergia in healthy volunteers has warranted the need for reassessment of the current methods to enhance the diagnostic value of anorectal manometry in functional defecatory disorders. Whether rectal distention at simulated evacuation will affect anorectal pressure profile and increase rectoanal gradient is not known.MethodsOne hundred and eight consecutive patients with chronic constipation, 93 females, median age 53 years (interquartile range: 40–65), were studied. Simulated evacuation was performed firstly with empty balloon and subsequently after balloon distention to 50 and 100 ml. Anorectal pressures were compared. We also performed subgroup analysis in relation to outcome of balloon expulsion test (BET). In addition, we studied the effect of rectal distension on the rectoanal pressure gradient with respect to rectal sensory function.ResultsRectal balloon distension at simulated evacuation improved rectoanal gradient and decreased the rate of dyssynergia during high-resolution anorectal manometry. In subgroup analysis, the increase in rectoanal gradient and correction of dyssynergia with rectal distension was limited to the patients who had normal BET and normal rectal sensory function. Rate of anal relaxation, residual anal pressures, and rectoanal gradient were significantly different between patients with and without normal BET at 50 ml of rectal distension. Rectoanal gradient recorded only after rectal distension, along with BMI and maximum tolerable volumes, could predict BET results independently in patients with chronic constipation.ConclusionsRectal distension during simulated evacuation will affect the anorectal pressure profile. Increase in rectoanal gradient and correction of dyssynergia was only significant in patients with normal rectal sensory function and normal BET.

Preparing for Climate Change: Forestry and Assisted Migration
Although plants have moved across the landscape in response to changing climate for millennia, projections of contemporary climate change suggest that forest tree species and populations will need to migrate faster than their natural ability. Therefore, climate change adaptation strategies, such as assisted migration, have gained attention since 2007. Effective implementation of assisted migration can only occur if target transfer guidelines are developed because our current seed transfer guidelines, established to guide the movement of plant materials, lack inherent spatial and temporal dynamics associated with climate change. This limitation restrains reforestation practitioners from making decisions about assisted migration. Lack of operating procedures, uncertainties about future climate conditions, risks associated with moving plants outside their current ranges, and existing policies have hampered formal actions in forest management and conservation. We review the current thinking on assisted migration of forest tree species and provide information that could facilitate implementation.

Skill-driven recommendations for job transition pathways
Job security can never be taken for granted, especially in times of rapid, widespread and unexpected social and economic change. These changes can force workers to transition to new jobs. This may be because new technologies emerge or production is moved abroad. Perhaps it is a global crisis, such as COVID-19, which shutters industries and displaces labor en masse . Regardless of the impetus, people are faced with the challenge of moving between jobs to find new work. Successful transitions typically occur when workers leverage their existing skills in the new occupation. Here, we propose a novel method to measure the similarity between occupations using their underlying skills. We then build a recommender system for identifying optimal transition pathways between occupations using job advertisements (ads) data and a longitudinal household survey. Our results show that not only can we accurately predict occupational transitions (Accuracy = 76%), but we account for the asymmetric difficulties of moving between jobs (it is easier to move in one direction than the other). We also build an early warning indicator for new technology adoption (showcasing Artificial Intelligence), a major driver of rising job transitions. By using real-time data, our systems can respond to labor demand shifts as they occur (such as those caused by COVID-19). They can be leveraged by policy-makers, educators, and job seekers who are forced to confront the often distressing challenges of finding new jobs.

An effective protocol to isolate and mechanically test silk fibers spun by Osmia lignaria Say (Hymenoptera: Megachilidae) fifth instar larvae
Silk, a remarkable protein-based fiber spun by various arthropod lineages, has been prized for millennia, with the cocoon silk of domesticated silkworms and spiders being the most utilized and extensively studied. There is limited information on how silk can be used to investigate biology, development, and health in other silk-producing species, particularly for solitary bees such as Osmia lignaria Say (Hymenoptera: Megachilidae). Osmia lignaria , an increasingly managed solitary pollinator, produces silk cocoons during the fifth instar larval stage. We have developed a minimally invasive protocol to isolate and mechanically test O. lignaria silk fibers using a 3-D printed well plate system for rearing and two specific isolation techniques. Our protocol allows for collecting individual fibers directly from silk-spinning larvae between silk initiation and cocoon formation without preventing subsequent cocoon development, enabling silk characterization as part of larger rearing and developmental studies. For this study, isolated fibers were mounted on C-cards, facilitating diameter measurement using a microscope and mechanical testing with an MTS Synergie 100 tensile testing instrument. We successfully isolated and tested the mechanical properties of naturally spun silk from O. lignaria , with 20 fibers isolated and mechanically tested from seven larvae. Further examination of isolated silk can reveal physical, molecular, chemical, and morphological characteristics, advancing our understanding of bee silk properties and their role in bee biology, evolution, and nutritional status. This protocol provides a practical tool for researchers to isolate and study silk from silk-producing bee species.
