Morphological alterations of peridroplet mitochondria in human liver biopsy

Mitochondrial dysfunction and the accumulation of lipid droplets (LD) contribute to the pathogenesis of liver diseases. Mitochondria bound to LD, termed peridroplet mitochondria (PDM), form a subpopulation with distinct functions compared to cytoplasmic mitochondria (CM). In this first in vivo human liver study, we aimed to investigate the morphological differences between PDM and CM and to assess their associations with clinical parameters. Our analysis of mitochondrial ultrastructure using transmission electron microscopy images of human liver biopsies showed that CM were significantly smaller, more spherical, and solid, whereas PDM were larger and more elongated. Overall, PDM exhibited more uniform morphology, while CM displayed disease-specific morphological alterations. CM were associated with serum liver enzyme levels and high-density lipoprotein cholesterol, suggesting sensitivity to liver stress and a potential role in liver cholesterol transport. In contrast, PDM were associated with serum triglyceride levels, indicating a role in lipid metabolism. Total PDM and LD counts showed a positive correlation, reinforcing their close functional relationship. These findings show that PDM and CM represent distinct mitochondrial subpopulations with unique morphologies and differing associations with hepatic pathophysiological pathways, which highlights the significance of LD interactions in contributing to mitochondrial heterogeneity.

Artificial Intelligence vs. Human Cognition: A Comparative Analysis of ChatGPT and Candidates Sitting the European Board of Ophthalmology Diploma Examination

Background/Objectives: This paper aims to assess ChatGPT’s performance in answering European Board of Ophthalmology Diploma (EBOD) examination papers and to compare these results to pass benchmarks and candidate results. Methods: This cross-sectional study used a sample of past exam papers from 2012, 2013, 2020–2023 EBOD examinations. This study analyzed ChatGPT’s responses to 440 multiple choice questions (MCQs), each containing five true/false statements (2200 statements in total) and 48 single best answer (SBA) questions. Results: ChatGPT, for MCQs, scored on average 64.39%. ChatGPT’s strongest metric performance for MCQs was precision (68.76%). ChatGPT performed best at answering pathology MCQs (Grubbs test p < 0.05). Optics and refraction had the lowest-scoring MCQ performance across all metrics. ChatGPT-3.5 Turbo performed worse than human candidates and ChatGPT-4o on easy questions (75% vs. 100% accuracy) but outperformed humans and ChatGPT-4o on challenging questions (50% vs. 28% accuracy). ChatGPT’s SBA performance averaged 28.43%, with the highest score and strongest performance in precision (29.36%). Pathology SBA questions were consistently the lowest-scoring topic across most metrics. ChatGPT demonstrated a nonsignificant tendency to select option 1 more frequently (p = 0.19). When answering SBAs, human candidates scored higher than ChatGPT in all metric areas measured. Conclusions: ChatGPT performed stronger for true/false questions, scoring a pass mark in most instances. Performance was poorer for SBA questions, suggesting that ChatGPT’s ability in information retrieval is better than that in knowledge integration. ChatGPT could become a valuable tool in ophthalmic education, allowing exam boards to test their exam papers to ensure they are pitched at the right level, marking open-ended questions and providing detailed feedback.

THE UKRAINE CRISIS AND SHIFT IN US FOREIGN POLICY

War in Ukraine and the Russian annexation of Crimea are the events that changed the US policy towards Russia. The events in Ukraine forced the United States to take a closer look at Eastern and Central Europe. The United States’ policy during the Ukrainian crisis has been limited to sanctions and strong statements so far because in Ukraine there is an asymmetry of interests. Ukraine is much more important to Russia than to the United States. The United States may be willing to support the democratic and western aspiration of Ukrainians but will not risk a major conflict with Russia over it. However the crisis in Ukraine is not only about Ukraine or Russia. It is also about US credibility around the world. Both friends and foes are watching closely the American reaction to the situation. That is why the United States has increased its military presence in those NATO countries that share borders with Russia.

Linguistic measures of chemical diversity and the “keywords” of molecular collections

Computerized linguistic analyses have proven of immense value in comparing and searching through large text collections (“corpora”), including those deposited on the Internet – indeed, it would nowadays be hard to imagine browsing the Web without, for instance, search algorithms extracting most appropriate keywords from documents. This paper describes how such corpus-linguistic concepts can be extended to chemistry based on characteristic “chemical words” that span more than traditional functional groups and, instead, look at common structural fragments molecules share. Using these words, it is possible to quantify the diversity of chemical collections/databases in new ways and to define molecular “keywords” by which such collections are best characterized and annotated.

Neural Models for Imputation of Missing Ozone Data in Air-Quality Datasets

Ozone is one of the pollutants with most negative effects on human health and in general on the biosphere. Many data-acquisition networks collect data about ozone values in both urban and background areas. Usually, these data are incomplete or corrupt and the imputation of the missing values is a priority in order to obtain complete datasets, solving the uncertainty and vagueness of existing problems to manage complexity. In the present paper, multiple-regression techniques and Artificial Neural Network models are applied to approximate the absent ozone values from five explanatory variables containing air-quality information. To compare the different imputation methods, real-life data from six data-acquisition stations from the region of Castilla y León (Spain) are gathered in different ways and then analyzed. The results obtained in the estimation of the missing values by applying these techniques and models are compared, analyzing the possible causes of the given response.

PurposeThis paper aims to verify how the intellectual property (IP) box affects firms’ effective tax rate, growth and innovation activity outcomes related to intellectual property rights.Design/methodology/approachImplementing the innovation box regimes into the tax system intends to encourage firms to engage in more innovative activities. In UK, Italy and Poland, the IP box tax relief was introduced in 2013, 2015 and 2019, respectively. In return, companies may reduce their tax rate to increase their investment and innovativeness. With a panel model approach – system GMM and DiD with multiple time periods – it analyses data from the Orbis database for 2011–2019 of 673 firms from the gaming industry in 11 countries and hand-collected data on intellectual property rights protection. The authors study public and private companies from the gaming sector in leading European markets and all three countries that protect intellectual property rights of software (Japan, South Korea, the USA).FindingsRecent reforms enable gaming companies to use preferential tax treatment for IP-related income and significantly impact a firm’s revenue growth.Practical implicationsNevertheless, European gaming firms require time to leap the gap to the growth and innovativeness of countries that protect software.Originality/valueThe authors show that the IP box stimulates gaming firms to protect IP via wordmarks, figurative marks, trademarks and software patents that bring effects in five years. Despite the critics against IP box, the authors prove its lagged efficiency, especially in profitable and larger firms.

Oxidative stress plays an important role in the pathophysiology of many human disorders, while antioxidants prevent the development of various adverse symptoms. Diosmin is a natural flavonoid applied in vascular system disorders, especially in chronic venous insufficiency (CVI), and it plays a significant part in the alleviation of CVI symptoms. Due to antioxidant activity, it also has the ability to scavenge the oxygen free radicals and hence decreases the level of oxidative stress biomarkers, such as prostaglandins and their precursors—isoprostanes. In the study, the influence of diosmin treatment on the level of isoprostanes in plasma samples of patients suffering from CVI was examined. The qualitative analysis was performed using high-performance liquid chromatography with spectrometry detection (LC-MS). The statistically significant decrease of isoprostane content after 3 months of treatment was observed within the studied group; however, the most significant changes were observed in patients who smoke.

Mitochondria, as “power house of the cell”, are crucial players in cell pathophysiology. Beyond adenosine triphosphate (ATP) production, they take part in a generation of reactive oxygen species (ROS), regulation of cell signaling and cell death. Dysregulation of mitochondrial dynamics may lead to cancers and neurodegeneration; however, the fusion/fission cycle allows mitochondria to adapt to metabolic needs of the cell. There are multiple data suggesting that disturbed mitochondrial homeostasis can lead to Parkinson’s disease (PD) development. 2-methoxyestradiol (2-ME), metabolite of 17β-estradiol (E2) and potential anticancer agent, was demonstrated to inhibit cell growth of hippocampal HT22 cells by means of nitric oxide synthase (NOS) production and oxidative stress at both pharmacologically and also physiologically relevant concentrations. Moreover, 2-ME was suggested to inhibit mitochondrial biogenesis and to be a dynamic regulator. This review is a comprehensive discussion, from both scientific and clinical point of view, about the influence of 2-ME on mitochondria and its plausible role as a modulator of neuron survival.

The purpose of this study was to examine the application of the coordinated cis-[Cr(C2O4)(pm)(OH)2]+ cation where pm denotes pyridoxamine, as a specific sensing ion for the detection of hydrogen peroxide (H2O2). The proposed method for H2O2 detection includes two key steps. The first step is based on the nonenzymatic decarboxylation of pyruvate upon reaction with H2O2, while the second step is based on the interaction of cis-[Cr(C2O4)(pm)(OH2)2]+ with the CO2 released in the previous step. Using this method H2O2 generated during glutamate-induced oxidative stress was detected in HT22 hippocampal cells. The coordination ion cis-[Cr(C2O4)(pm)(OH2)2]+ and the spectrophotometric stopped-flow technique were applied to determine the CO2 concentration in cell lysates, supernatants and cell-free culture medium. Prior to CO2 assessment pyruvate was added to all samples studied. Pyruvate reacts with H2O2 with 1:1 stoichiometry, and consequently the amount of CO2 released in this reaction is equivalent to the amount of H2O2.

CCR: A combined cleaning and resampling algorithm for imbalanced data classification

Imbalanced data classification is one of the most widespread challenges in contemporary pattern recognition. Varying levels of imbalance may be observed in most real datasets, affecting the performance of classification algorithms. Particularly, high levels of imbalance make serious difficulties, often requiring the use of specially designed methods. In such cases the most important issue is often to properly detect minority examples, but at the same time the performance on the majority class cannot be neglected. In this paper we describe a novel resampling technique focused on proper detection of minority examples in a two-class imbalanced data task. The proposed method combines cleaning the decision border around minority objects with guided synthetic oversampling. Results of the conducted experimental study indicate that the proposed algorithm usually outperforms the conventional oversampling approaches, especially when the detection of minority examples is considered.
