Research on Rock Physics Modeling Methods for Fractured Shale Reservoirs

The Sichuan Basin is a significant region for exploration and development of shale gas in China, and it is essential to clarify the impact of deep shale gas reservoir parameters on cost-effective development at scale to ensure national energy security. Rock physics modeling is a significant means of communicating the physical and elastic parameters of rocks. A rock physics modeling method applicable to fractured shale gas reservoirs is proposed for the current situation of complex fluid relationships in shale gas reservoirs and unclear characteristics of gas identification seismic response. In this paper, based on the Self Consistent Approximation (SCA) model and the differential effective medium (DEM) model, the anisotropic source of shale is used as a starting point to add bound water, kerogen, clay, and brittle minerals, the Schoenberg linear slip theory is used to add fracture disturbance effects, and then the Brown–Korringa model is used to perform fluid replacement under anisotropic conditions. Finally a rock physics model applicable to fractured shale gas reservoirs is obtained, and the established rock physics model is used for analysis of elastic parameters, Thomsen parameters, and fracture weakness parameters. Rock physics tests were performed on shale in southern Sichuan as an example. The experimental results show that the model established by the process can accurately invert the longitudinal and transverse wave velocities of the shale, which can provide a conceptual basis for the study of fractured shale gas reservoirs.

Brittleness is an important factor that indicates shale properties, as well as fracturability, and it can be well-represented using the elasticity parameter Eρ. Seismic inversion allows direct access to the Eρ ofbrittleness parameters. Seismic inversion is a typical ill-posed problem that has an enormous multiplicity of solutions. In order to invert reservoir brittleness parameters more stably and reliably, a direct inversion method for determining brittleness parameters based on a reweighted Lp-norm is proposed, and the reweighted Lp method is introduced to brittleness parameter inversion for the first time. The alternating direction method of multipliers (ADMM) is used to establish the inversion structure and to optimize the objective function in blocks, which effectively improves the convergence speed. We first introduce a reweighted Lp method and establish a pre-stack inversion objective function based on the reweighted Lp method. Then, theoretical simulation data are applied to compare the inversion outcomes of the new method with those of the traditional method, and the effect of the method in this manuscript is verified. Finally, the feasibility of this method is further verified using actual data for experimental analysis. Through an analysis of the experimental results, we find that this method can be well-applied to seismic pre-stack inversion calculation and provides a new direct inversion method for the determination of brittleness parameters for exploration geophysics.

A high proportion of renewable energy access makes the net load of the power system volatile and uncertain, increasing the demand for the ramping capacity of the power system. Traditional electricity spot markets compensate for the power imbalances caused by an insufficient ramping capacity through traditional flexibility services such as ancillary services and interconnection power. However, conventional flexibility services may lead to frequency deviations in the power system, increased response costs, spikes in electricity prices, and dramatic price volatility in the traditional spot market. To solve the above problems, this paper proposes an FRP and convex electricity spot market joint clearing (FCESMJC) market mechanism. The FCESMJC model can more accurately represent the relationship between electrical power output and the price of electricity and reduces the number of spikes in electricity prices. In addition, a novel FRP pricing method is proposed to compensate FRP market participants for their FRP costs more reasonably. Additionally, the difference in system performance is provided by comparing the energy prices, pricing method, clearing prices, and system costs in the FCESMJC method and the traditional electricity spot market. The FCESMJC system reduces the total system cost by 18.6% compared with the electricity spot market. Numerical experiments are simulated on the IEEE 14-bus test system to validate the superiority of the proposed model.

Integrative analysis of metabolomics and proteomics reveals amino acid metabolism disorder in sepsis

Background
Sepsis is defined as a systemic inflammatory response to microbial infections with multiple organ dysfunction. This study analysed untargeted metabolomics combined with proteomics of serum from patients with sepsis to reveal the underlying pathological mechanisms involved in sepsis.
Methods
A total of 63 patients with sepsis and 43 normal controls were enrolled from a prospective multicentre cohort. The biological functions of the metabolome were assessed by coexpression network analysis. A molecular network based on metabolomics and proteomics data was constructed to investigate the key molecules.
Results
Untargeted metabolomics analysis revealed widespread dysregulation of amino acid metabolism, which regulates inflammation and immunity, in patients with sepsis. Seventy-three differentially expressed metabolites (|log

fold change| > 1.5, adjusted

value < 0.05 and variable importance in the projection (VIP) > 1.5) that could predict sepsis were identified. External validation of the hub metabolites was consistent with the derivation results (area under the receiver operating characteristic curve (AUROC): 0.81–0.96/0.62–1.00). The pentose phosphate pathway was found to be related to sepsis-associated encephalopathy. Phenylalanine metabolism was associated with sepsis-associated acute kidney injury. The key molecular alterations of the multiomics network in sepsis compared to normal controls implicate acute inflammatory response, platelet degranulation, myeloid cell activation involved in immune response and phenylalanine, tyrosine and tryptophan biosynthesis, and arginine biosynthesis.
Conclusions
Integrated analysis of untargeted metabolomics and proteomics revealed characteristic metabolite and protein alterations in sepsis, which were mainly involved in inflammation-related pathways and amino acid metabolism. This study depicted the pathological characteristics and pathways involved in sepsis and potential therapeutic targets.

ARHGEF15 in Sertoli cells contributes to germ cell development and testicular immune privilege

Sertoli cells (SCs), the only somatic cells in the seminiferous tubules, facilitate the maintenance of testicular immune privilege through the formation of the blood-testis barrier (BTB) and the expression of immunoregulatory factors. Rho guanosine exchange factor 15 (ARHGEF15) is a member of the guanosine exchange factors, which are involved in cell migration, cell polarity, and cell cycle progression via activation of Rho GTPases. This study investigated the functional role of ARHGEF15 in SCs during spermatogenesis using SC–specific Arhgef15 knockout mice. The results revealed that Arhgef15 deficiency in SCs affected the localization of SC nuclei, disrupted BTB integrity, and led to premature shedding of germ cells. In Arhgef15flox/flox/Amh-Cre+ mice, the ultrastructure of the round spermatids was impaired, accompanied by acrosome degeneration, acrosomal vesicle shedding, and atrophic nuclei. Consequently, the percentage of abnormal sperm in the Arhgef15flox/flox/AmhCre+ epididymis was markedly elevated. RNA-sequencing analysis revealed that most of the differentially expressed genes in SCs of Arhgef15flox/flox/Amh-Cre+ mice were associated with immunity. Further study revealed that the sera of Arhgef15flox/flox/Amh-Cre+ mice showed immunoreactivity against testicular lysate of wild-type mice, indicating the production of antibodies against testicular autoantigens in Arhgef15flox/flox/Amh-Cre+ mice. In conclusion, the specific deletion of Arhgef15 in SCs of mice leads to sperm abnormality, probably by disrupting the testicular immune homeostasis. BackgroundLymph node (LN) involvement is a critical prognostic factor in patients with gallbladder carcinoma (GBC). Controversy exists regarding optimal categorization of nodal metastasis status, including anatomical location of positive nodes (AJCC 7th N staging), number of metastatic lymph nodes (NMLN), log odds of metastatic LNs (LODDS), and lymph node ratio (LNR).MethodsPatients who underwent curative-intent resection for GBC from six Chinese tertiary hospitals between 2008 and 2013 were analyzed retrospectively. The relative discriminative abilities of the different LN staging systems were assessed by different models including the tree-augmented naïve Bayesian (TAN) model, Cox proportional hazards regression model, and binary logistic regression model.ResultsA total of 226 patients were involved in this cohort. Based on the TAN model and composite importance measures, the most important factor affecting the prognosis in the different LN staging systems was NMLN. Among the four TAN models which were built with 4 metastatic LN markers and baseline variables, the accuracy of the NMLN-based prognostic model was 88.15%, higher than 7th N staging (86.44%), LNR (87.34%), and LODDS (85.19%). The Cox model based on NMLN (C-index: 0.763, AIC: 1371.62) had a higher fitness than the others (7th N staging C-index: 0.756, AIC: 1375.51; LNR C-index: 0.759, AIC: 1378.82; LODDS C-index 0.748, AIC: 1390.99). The AUCs of different staging binary logistic regression models were NMLN (0.872), LNR (0.872), 7th N staging (0.869) and LODDS (0.856), respectively.ConclusionsNMLN was the optimal LN staging system in evaluating prognosis of GBC.

S100A4 promotes the progression of lipopolysaccharide-induced acute epididymitis in mice

S100A4 has been suggested to be a critical regulator of tumor metastasis and is implicated in the progression of inflammation. The aim of this study is to investigate the expression and possible role of S100A4 in epididymitis. Using a mouse model of epididymitis induced by the injection of lipopolysaccharide (LPS) in the deferent duct, we found that LPS administration induced an upregulation of S100a4 transcription (P < 0.05) and a recruitment of S100A4 positive cells in the epididymal interstitium of wild type (WT) mice. Co-immunofluorescence showed that S100A4 was mainly expressed by granulocytes, CD4 lymphocytes, and macrophages. Deficiency of S100A4 reduced epididymal pathological reaction and the mRNA levels of the pro-inflammatory cytokines IL-1β and TNF-α (P < 0.01), suggesting that S100A4 promotes the progression of epididymitis. Furthermore, S100A4 deficiency alleviated the decline of sperm motility and rectified the abnormal expression of sperm membrane protein AMAD3, which suggested that in the progression of epididymitis, S100A4 aggravates the damage to sperm vitality. In addition, both Ki-67 marked cell proliferation and transferase-mediated dUTP-biotin nick end labeling detected cell apoptosis were reduced in S100a4–/– mice compared with WT mice after LPS treatment, indicating that S100A4 promotes both cell proliferation and cell apoptosis in epididymitis. Overall, these results demonstrate that S100A4 promotes the progression of LPS-induced epididymitis and facilitates a decline in sperm vitality, and its function may be related to the process of cell proliferation and apoptosis during inflammation. Summary sentence S100A4, which was increased in LPS-induced mouse epididymitis, contributes to the development of epididymitis.

Evaluating the Losses and Recovery of GPP in the Subtropical Mangrove Forest Directly Attacked by Tropical Cyclone: Case Study in Hainan Island

The gross primary production (GPP) of the mangrove ecosystem determines the upper limit of the scale of its “blue carbon” sink. Tropical cyclones (TCs) are among the most important extreme events that threaten the subtropical mangrove ecosystem and have a serious impact on mangrove ecosystem GPP. However, there are somewhat insufficient scientific findings on regional-scale mangrove ecosystem GPP responding to large-scale weather events such as TCs. Therefore, we selected the subtropical Hainan Island mangrove ecosystem, where more than two TCs pass through per year, as the research area; selected direct-attack TCs as the research object; and took the mangrove vegetation photosynthesis light-use efficiency model established based on the eddy covariance observation data as the tool to evaluate the loss and recovery of mangrove ecosystem GPP after TCs attacked at a regional scale. We found that the TC impacted the mangrove ecosystem GPP through the photosynthetic area and rate, and the recovery of the rate occurred prior to the recovery of the area; the loss of mangrove ecosystem GPP is inversely proportional to the distance to the center of the TC and the distance to the coastline; and the canopy height, diameter at breast height, and aspect where the tree stands significantly influence the response of the mangrove ecosystem GPP to TCs. However, the response varies for different mangrove community compositions, soil conditions, and planting densities as well as different frequencies and intensities of TCs, and they should be analyzed in detail. This study is expected to provide technical and data support for the protection of blue carbon in a subtropical island mangrove ecosystem in response to extreme events and post-disaster recovery.

Astaxanthin Alleviates the Decline of Sperm Quality Caused by Heat Stress in Mice via Reducing Oxidative Stress

Heat stress impairs spermatogenesis primarily through increased testicular oxidative stress. Astaxanthin, a potent antioxidant, has shown beneficial effects in sperm cryopreservation. However, its potential to mitigate testicular damage induced by elevated temperatures remains uninvestigated. In this study, male mice were administered astaxanthin (10 mg/kg/d, 50 mg/kg/d, and 100 mg/kg/d) via gavage for 21 days, with heat exposure occurring during the final 14 days. Samples were collected after the last treatment or following a recovery period. Spermatozoa in the cauda epididymis were assessed using computer-aided sperm analysis (CASA) or Diff-Quik staining, and serum testosterone levels and oxidative stress markers in both serum and testis were quantified via enzyme-linked immunosorbent assay (ELISA). Heat treatment resulted in significant reductions in sperm motility, viability, and morphological integrity. However, daily supplementation of astaxanthin at 50 mg/kg and 100 mg/kg effectively alleviated these heat-induced impairments. Furthermore, astaxanthin at 50 mg/kg/d notably improved testis weight, testis index, and serum testosterone levels under heat stress. Its antioxidant capacity was validated by significant restoration of total antioxidant capacity (T-AOC), superoxide dismutase (SOD), and reduction in malondialdehyde (MDA) levels in both testis and serum. In conclusion, this study highlights the protective effects of astaxanthin against heat-induced testicular and sperm damage by reducing oxidative stress, supporting its potential use as a nutritional or nutraceutical supplement to promote male reproductive health, particularly in the context of summer subfertility in farm animals.

S100a4 promotes the development of lipopolysaccharide-induced mouse endometritis

S100A4 is suggested to be a critical regulator of tumor metastasis, and implicated in progression of inflammation. The aim of this study is to investigate the expression and possible role of S100A4 in endometritis. Using amousemodel of endometritis induced by local injection of lipopolysaccharide (LPS), we found that infection induced recruitment of S100A4-positive cells in the endometrium of wild-type mice. Deficiency of S100A4 reduced uterine pathological reaction and mRNA expression of proinflammatory cytokine IL-1β and TNF-α (P < 0.01), suggesting S100A4 promoted the progression of endometritis. To further explore the potential mechanism, we examined the cellular proliferation and apoptosis in the endometrium. Western blot and immunohistochemical results showed that cell apoptosis in uterus during endometritis, marked by cleaved-Caspase 3 protein, was significantly cut down in S100a4−/− mice; cell proliferation, which was indicated by Ki-67, was also significantly decreased in the inflamed endometrial stroma of S100a4−/− mice. Overall, these results demonstrate that S100A4 promotes the development of LPS-induced endometritis, and it may be related to the process of cell proliferation and apoptosis during the inflammation. Summary Sentence S100A4, which was increased in LPS-infected mouse endometrium, contributes to the development of endometritis.
