In the Different Models, Same Initial Conditions (DIMOSIC) project, forecasts from different global medium-range forecast models have been created based on the same initial conditions. The dataset consists of 10-day deterministic forecasts from seven models and includes 122 forecast dates spanning one calendar year. All forecasts are initialized from the same ECMWF operational analyses to minimize the differences due to initialization. The models are run at or near their respective operational resolutions to explore similarities and differences between operational global forecast models. The main aims of this study are 1) to evaluate the forecast skill and how it depends on model formulation, 2) to assess systematic differences and errors at short lead times, 3) to compare multimodel ensemble spread to model uncertainty schemes, and 4) to identify models that generate similar solutions. Our results show that all models in this study are capable of producing high-quality forecasts given a high-quality analysis. But at the same time, we find a large variety in model biases, both in terms of temperature errors and precipitation. We are able to identify models whose forecasts are more similar to each other than they are to those of other systems, due to the use of similar model physics packages. However, in terms of multimodel ensemble spread, our results also demonstrate that forecast sensitivities to different model formulations still are substantial. We therefore believe that the diversity in model design that stems from parallel development efforts at global modeling centers around the world remains valuable for future progress in the numerical weather prediction community.

Macrophages regulate lung ILC2 activation via Pla2g5-dependent mechanisms

Group V phospholipase A2 (Pla2g5) is a lipid-generating enzyme necessary for macrophage effector functions in pulmonary inflammation. However, the lipid mediators involved and their cellular targets have not been identified. Mice lacking Pla2g5 showed markedly reduced lung ILC2 activation and eosinophilia following repetitive Alternaria Alternata inhalation. While Pla2g5-null mice had Wt levels of immediate IL-33 release after one Alternaria dose, they failed to upregulate IL-33 in macrophages following repeated Alternaria administration. Unexpectedly, while adoptive transfer of bone marrow-derived (BM)-macrophages restored ILC2 activation and eosinophilia in Alternaria-exposed Pla2g5-null mice, exogenous IL-33 did not. Conversely, transfers of Pla2g5-null BM-macrophages reduced inflammation in Alternaria-exposed Wt mice. Mass spectrometry analysis of free fatty acids (FFAs) demonstrated significantly reduced FFAs (including linoleic acid (LA) and oleic acid (OA)) in lung and BM-macrophages lacking Pla2g5. Exogenous administration of LA or LA+OA to Wt mice sharply potentiated IL-33-induced lung eosinophilia and ILC2 expansion in vitro and in vivo. In contrast, OA potentiated IL-33-induced inflammation and ILC2 expansion in Pla2g5-null mice, but LA was inactive both in vivo and in vitro. Notably, Pla2g5-null ILC2s showed significantly reduced expression of the FFA-receptor-1 compared to Wt ILC2s. Thus, macrophage-associated Pla2g5 contributes significantly to type-2 immunity through regulation of IL-33 induction and FFA-driven ILC2 activation.

Climate change alters the optimal wind-dependent flight routes of an avian migrant

Migratory birds can be adversely affected by climate change as they encounter its geographically uneven impacts in various stages of their life cycle. While a wealth of research is devoted to the impacts of climate change on distribution range and phenology of migratory birds, the indirect effects of climate change on optimal migratory routes and flyways, through changes in air movements, are poorly understood. Here, we predict the influence of climate change on the migratory route of a long-distant migrant using an ensemble of correlative modelling approaches, and present and future atmospheric data obtained from a regional climate model. We show that changes in wind conditions by mid-century will result in a slight shift and reduction in the suitable areas for migration of the study species, the Oriental honey-buzzard, over a critical section of its autumn journey, followed by a complete loss of this section of the traditional route by late century. Our results highlight the need for investigating the consequences of climate change-induced disturbance in wind support for long-distance migratory birds, particularly species that depend on the wind to cross ecological barriers, and those that will be exposed to longer journeys due to future range shifts.

Platelet-rich plasma (PRP) and bone marrow aspirate concentrate (BMAC) are orthobiologic therapies considered as an alternative to the current therapies for muscle, bone and cartilage. Different formulations of biomaterials have been used as carriers for PRP and BMAC in order to increase regenerative processes. The most common biomaterials utilized in conjunction with PRP and BMAC clinical trials are organic scaffolds and natural or synthetic polymers. This review will cover the combinatorial strategies of biomaterial carriers with PRP and BMAC for musculoskeletal conditions (MsCs) repair and regeneration in clinical trials. The main objective is to review the therapeutic use of PRP and BMAC as a treatment option for muscle, bone and cartilage injuries.

Strong maternal fidelity and natal philopatry shape genetic structure in North Pacific humpback whales
We quantified the relative influence of maternal fidelity to feeding grounds and natal fidelity to breeding grounds on the population structure of humpback whales Megaptera novaeangliae based on an ocean-wide survey of mitochondrial (mt) DNA diversity in the North Pacific. For 2193 biopsy samples collected from whales in 10 feeding regions and 8 breeding regions during the winter and summer of 2004 to 2006, we first used microsatellite genotyping (average, 9.5 loci) to identify replicate samples. From sequences of the mtDNA control region (500 bp) we identified 28 unique haplotypes from 30 variable sites. Haplotype frequencies differed markedly among feeding regions (overall F
ST = 0.121, ΦST = 0.178, p < 0.0001), supporting previous evidence of strong maternal fidelity. Haplotype frequencies also differed markedly among breeding regions (overall F
ST = 0.093, ΦST = 0.106, p < 0.0001), providing evidence of strong natal fidelity. Although sex-biased dispersal was not evident, differentiation of microsatellite allele frequencies was weak compared to differentiation of mtDNA haplotypes, suggesting male-biased gene flow. Feeding and breeding regions showed significant differences in haplotype frequencies, even for regions known to be strongly connected by patterns of individual migration. Thus, the influence of migratory fidelity seems to operate somewhat independently on feeding and breeding grounds over an evolutionary time scale. This results in a complex population structure and the potential to define multiple units to conserve in either seasonal habitat.

SUPPORTING CARERS OF COMMUNITY-DWELLING FRAIL OLDER ADULTS: HIDDEN AGENDA IN LONG-TERM CARE SYSTEM IN JAPAN

Abstract
The establishment of “the Community-based integrated care” is emphasized in the 2017 Long-Term Care Insurance (LTCI) Reform but the role of family carers in the system remains unclear. The purpose of this presentation is to explore how the issue of carers is treated in LTC policies in Japanese political, economic and cultural contexts from Carers’ perspectives. We found that the 2016 Child Care and Family Care Leave Act Reform, the policies to prevent giving up jobs due to caring roles, the 2015 National Dementia Strategy and the LTCI reform have not fully addressed the issue of family carers. The results of our survey on the Community-based integrated care support centers also indicated carers faced various difficulties despite the use of LTCI services. We will argue the societal risk in the increasing burden to carers for the frail older adults and the paradox of “socialization of care” in Japanese LTC model.

Monocyte chemoattractant protein-1 (MCP-1)/chemokine (C-C motif) ligand (CCL) 2 (CCL2) secreted from white adipose tissue (WAT) in obesity has been reported to contribute to tissue macrophage accumulation and insulin resistance by inducing a chronic inflammatory state. MCP-1 has been shown to be elevated in the fatty liver of lipoatrophic A-ZIP-transgenic (A-ZIP-Tg) mice. Treatment of these mice with the CC chemokine receptor (CCR) 2 antagonist has been shown to ameliorate the hyperglycaemia, hyperinsulinaemia and hepatomegaly, in conjunction with reducing liver inflammation. However, since CCR2 antagonists can block not only MCP-1 but also MCP-2 (CCL8) and MCP-3 (CCL7), it remains unclear whether MCP-1 secreted from the liver could contribute to hyperglycaemia, hyperinsulinaemia and hepatomegaly in conjunction with liver inflammation, as well as to the M1 and M2 states of macrophage polarisation.
Methods
To address these issues, we analysed the effects of targeted disruption of MCP-1 in A-ZIP-Tg mice.
Results
MCP-1 deficiency alone or per se resulted in a significant amelioration of insulin resistance in A-ZIP-Tg mice, which was associated with a suppression of extracellular signal-regulated protein kinase (ERK)-1/2 and p38 mitogen-activated protein kinase (p38MAPK) phosphorylation in liver. Although MCP-1 deficiency did not reduce the expression of macrophage markers, it increased the expression of the genes encoding M2 macrophage markers such as
Arg1
and
Chi3l3
, as well as significantly reducing the triacylglycerol content of livers from A-ZIP-Tg mice.
Conclusions/ interpretation
Our data clearly indicated that MCP-1 deficiency improved insulin resistance and hepatic steatosis in A-ZIP-Tg mice and was associated with switching macrophage polarisation and suppressing ERK-1/2 and p38MAPK phosphorylation.

Delimitation of a QTL region controlling cold tolerance at booting stage of a cultivar, ‘Lijiangxintuanheigu’, in rice, Oryza sativa L

Low temperature at the booting stage of rice causes male sterility resulting in severe yield loss. Cold tolerance has long been an important objective in rice breeding. We identified a quantitative trait locus (QTL) for cold tolerance on the long arm of chromosome 3 from the cold-tolerant breeding line ‘Ukei 840’ by using F2 and BC1F2 populations from crosses between ‘Ukei 840’ and ‘Hitomebore’. The cold tolerance of ‘Ukei 840’ is derived from the Chinese cultivar ‘Lijiangxintuanheigu’. The effect of this QTL on cold tolerance was confirmed by developing ‘Hitomebore’ chromosome segment substitution lines having ‘Lijiangxintuanheigu’ alleles on chromosome 3. By producing recombinants in chromosome 3, the QTL region for cold tolerance was delimited to the region of about 1.2-Mb region between RM3719 and RM7000. All lines heterozygous for the QTL showed seed fertilities as low as that of ‘Hitomebore’, suggesting that the ‘Lijiangxintuanheigu’ allele for cold tolerance in the QTL region is recessive. Determination of a 1.2-Mb nucleotide sequence of ‘Ukei 840’ and comparison with the published genomic sequence of ‘Nipponbare’ showed 254 SNPs, of which 11 were in coding regions of genes, seven in five genes being non-synonymous. SNPs were detected in the 5-kb upstream regions of 89 genes, but no differences of gene expression levels were detected between alleles of these genes. Although further delimitation is required to identify the gene responsible for cold tolerance of ‘Lijiangxintuanheigu’, SNP markers developed here will be useful for marker-assisted selection in a breeding program using ‘Lijiangxintuanheigu’ as a donor of cold tolerance.

Characterization of the T'-Phase Interface in Al-Zn-Mg Alloy Along the Zone Direction of Al-Matrix

In this study, we observed the T'-phase for the first time along the zone axis of the Al matrix. The orientation relationship, shape, misfit value, and interfacial conditions between the T'-phase and the Al matrix were investigated using highresolution transmission electron microscopy (HR-TEM). The T'-phase exhibited a fine cubic morphology, and we clarified each facet by analyzing diffraction patterns and fast Fourier transform (FFT) images. Additionally, we identified the (11–1)Al //(001)T' , (11–0)Al //(1–00)T' interface between the T'-phase and the Al matrix along the [1–1 – 2–]Al //[010]T' zone direction.
