A hybrid temporal convolutional attention model for water filter remaining useful life prediction
Accurate prediction of the remaining useful life of water filter cartridges is critical for ensuring water quality in both domestic and industrial applications. Conventional methods based on physical or chemical measurements are often time-consuming, costly, and reliant on expert knowledge. While data-driven time series models present a viable alternative, they frequently fail to capture long-range dependencies in complex, high-dimensional sensor data, limiting their feature extraction capability. To address these bottlenecks, this paper proposes a novel triple-fusion architecture that integrates Temporal Convolutional Networks, a Gated Attention Mechanism, and an LSTM module. The primary contributions are threefold: (1) Employing dilated causal convolutions to mitigate gradient vanishing and enable ultra-deep temporal feature extraction; (2) Introducing a gated attention mechanism during feature decoding to adaptively focus on critical time steps by dynamically weighting features, thereby enhancing modeling sensitivity; (3) Designing an HTCA-LSTM cell to improve the capture of long-term dependencies and high-dimensional complexity of key features. Experimental results demonstrate the model’s superior performance across prediction horizons of 96, 172, and 720 steps, with average reductions in MAE and MSE of 6.84–11.51%, and , respectively. This validates its efficacy in modeling long-term dependencies and extracting complex features, showcasing significant generalization ability and application potential for RUL prediction.

A switchable and signal-amplified aptasensor based on metal organic frameworks as the quencher for turn-on detection of T-2 mycotoxin

A simple and low-cost fluorescence aptasensor was developed for rapid and sensitive signal amplification detection of T-2 mycotoxin (T-2). Dual-terminal-fluorescein amidite (FAM)–labeled aptamer (D-aptamer) acted as a recognition element and signal indicator. The metal organic frameworks (MOFs) of N, N′-bis(2-hydroxyethyl)dithiooxamidato copper (II) (H2dtoaCu) were as the quencher. The D-aptamer was initially adsorbed to the surface of H2dtoaCu, leading to efficient quenching of the aptasensor. Upon addition of T-2, the D-aptamer underwent a conformation change to form the T-2/T-2 aptamer complex, which induced the signaling probe to be released from the H2dtoaCu surface. Thus, the fluorescence intensity (FL) of the D-aptamer was recovered. Versus the single-terminal-FAM-labeled aptamer (S-aptamer), the D-aptamer showed a lower detection limit of 0.39 ng/mL. The aptasensor was also successfully applied to detect T-2 in corn and wheat samples with good recoveries.

Morphine promotes microglial activation by upregulating the EGFR/ERK signaling pathway

Although they represent the cornerstone of analgesic therapy, opioids, such as morphine, are limited in efficacy by drug tolerance, hyperalgesia and other side effects. Activation of microglia and the consequent production of proinflammatory cytokines play a key pathogenic role in morphine tolerance, but the exact mechanisms are not well understood. This study aimed to investigate the regulatory mechanism of epidermal growth factor receptor (EGFR) on microglial activation induced by morphine in mouse microglial BV-2 cells. In this research, BV-2 cells were stimulated with morphine or pretreated with AG1478 (an inhibitor of EGFR). Expression levels of cluster of differentiation molecule 11b (CD11b), EGFR, and phospho-EGFR were detected by immunofluorescence staining. Cell signaling was assayed by Western blot. The migration ability of BV-2 cells was tested by Transwell assay. The production of interleukin-1beta (IL-1β) and tumor necrosis factor-alpha (TNF-α) in the cell supernatant was determined by ELISA. We observed that the expression of CD11b induced by morphine was increased in a dose- and time- dependent manner in BV-2 cells. Phosphorylation levels of EGFR and ERK1/2, migration of BV-2 cells, and production of IL-1β and TNFα were markedly enhanced by morphine treatment. The activation, migration, and production of proinflammatory cytokines in BV-2 cells were inhibited by blocking the EGFR signaling pathway with AG1478. The present study demonstrated that the EGFR/ERK signaling pathway may represent a novel pharmacological strategy to suppress morphine tolerance through attenuation of microglial activation.

Objective: To elucidate the chemical profile of Xanthocerais lignum’s extracts of different polarities and their impact on rheumatoid arthritis (RA), we identified anti-RA markers and predicted their action mechanisms. Methods: A collagen-induced arthritis rat model was established, and UPLC-Q-Exactive Orbitrap MS technology was employed to analyze and identify the chemical constituents within the alcohol extract of Xanthocerais lignum and its various extraction fractions, as well as their translocation into the bloodstream. Serum spectrum–effect correlation analysis was utilized to elucidate the pharmacodynamic material basis of Xanthocerais lignum against RA and to screen for Q-Markers. Finally, the potential anti-RA mechanisms of the Q-Markers were predicted through compound-target interaction data and validated using molecular docking techniques. Results: We identified 71 compounds, with flavan-3-ols and flavanones as key components. Of these, 36 were detected in the bloodstream, including 17 original and 19 metabolized forms. Proanthocyanidin A2, dihydroquercetin, catechin, and epicatechin (plus glucuronides) showed potential anti-RA activity. These compounds, acting as Q-Markers, may modulate ERK, NF-κB, HIF-1α, and VEGF in the HIF-1 pathway. Conclusions: This research clarifies Xanthocerais lignum’s pharmacodynamic material basis against RA, identifies 4 Q-Markers, and offers insights into their mechanisms, aiding quality assessment and lead compound development for RA treatment.

MoSDT1 triggers defense response through modulating phosphorylated proteins in rice

Key messageMoSDT1, a rice blast fungus transcription factor, is as an inducer to activate defense response through mainly mediating phosphorylated proteins in rice.Pathogen effector proteins play a dual role in infecting the host or triggering a defense response. Our previous research found a Magnaporthe oryzae effector, MoSDT1, which could activate the rice defense response when it was overexpressed in rice. However, we still know little about the mechanisms on how MoSDT1 in vivo or in vitro influences the resistance ability of rice. Our results showed that decreased ROS and increased lignin contents appeared along with significant upregulation of defense-related genes, raffinose synthesis gene, and phenylalanine ammonialyase gene. Moreover, we revealed that the contents of lignin were increased, which was in accordance with the upregulation of its precursor phenylalanine gene despite the fact that the glutamate-/thiamine-responsive genes were inhibited in MoSDT1 transgenic rice, and these indicated that MoSDT1 triggered the defense system of rice in vivo. Interestingly, in vitro studies, we further found that MoSDT1 induced the defense system by ROS synthesis, callose deposition, PR gene expression and SA/JA synthesis/signal genes using the purified prokaryotic expression system in rice plants. In addition, this defense response was confirmed to be activated by the zinc finger domain of MoSDT1 via prokaryotic expression of MoSDT1 truncated mutants in rice plants. To elucidate the regulative effects of MoSDT1 on protein phosphorylation in rice, phosphoproteome analysis was performed in both MoSDT1-transgenic and wild type rice. We found that MoSDT1 specifically up-regulated the expression levels of a few phosphorylated proteins, which were involved in multiple functions, such as biotic/abiotic stress and growth. In addition, the motifs in these specific proteins ranked the top among the top-five conserved motifs in the MoSDT1-transgenic rice. MoSDT1 played a crucial role in enhancing rice resistance by modulating several genes and signaling pathways.

Thyroid autoimmunity and spontaneous cervicocranial artery dissection: an exploratory segment-specific case-control study

IntroductionImmune-mediated mechanisms are thought to contribute to the local vascular inflammation associated with spontaneous cervicocranial arterial dissection (SCCAD). However, evidence linking thyroid autoimmunity to SCCAD remains conflicting. Furthermore, the potential correlation between thyroid autoimmunity and the anatomical distribution of dissections has not been evaluated.MethodsThis retrospective, hospital-based case-control study was conducted from June 2021 to May 2025, enrolling 164 patients with SCCAD and 164 healthy controls. Multivariate logistic regression analyses, and patient-level sensitivity analyses were employed to examine the impact of thyroid autoantibodies on SCCAD and their anatomical distributions. Additionally, propensity score matching, stratified analyses and interaction tests were performed to assess the robustness of the association between thyroid autoimmunity and SCCAD.ResultsMultivariate analyses showed that, compared with healthy controls, SCCAD had significantly higher rates of thyroid autoimmunity positivity (OR 6.715; 95% CI: 1.589–28.377; P = 0.010) and elevated TPOAb levels (OR 5.999; 95% CI: 1.379–26.096; P = 0.017). In segment-specific analyses, elevated TPOAb levels were positively associated with V4 segment involvement (OR 7.663, 95% CI: 1.443–40.680, P = 0.017) and inversely associated with V3 segment involvement (OR 0.065, 95% CI: 0.005–0.816, P = 0.034). The robustness of these segment-specific associations was further supported by patient-level sensitivity analyses (V3: OR 0.043, 95% CI: 0.003–0.669, P = 0.025; V4: OR 10.452, 95% CI: 1.143–95.584, P = 0.038).DiscussionIn conclusion, thyroid autoimmunity positivity and elevated TPOAb were independently associated with SCCAD, and TPOAb demonstrated a previously unrecognized segment-specific association with vertebral artery dissections—positive at V4 and inverse at V3.

Quercetin Protects Blood–Brain Barrier Integrity via the PI3K/Akt/Erk Signaling Pathway in a Mouse Model of Meningitis Induced by Glaesserella parasuis

Glaesserella parasuis (G. parasuis) causes serious inflammation and meningitis in piglets. Quercetin has anti-inflammatory and anti-bacterial activities; however, whether quercetin can alleviate brain inflammation and provide protective effects during G. parasuis infection has not been studied. Here, we established a mouse model of G. parasuis infection in vivo and in vitro to investigate transcriptome changes in the mouse cerebrum and determine the protective effects of quercetin on brain inflammation and blood–brain barrier (BBB) integrity during G. parasuis infection. The results showed that G. parasuis induced brain inflammation, destroyed BBB integrity, and suppressed PI3K/Akt/Erk signaling-pathway activation in mice. Quercetin decreased the expression of inflammatory cytokines (Il-18, Il-6, Il-8, and Tnf-α) and BBB-permeability marker genes (Mmp9, Vegf, Ang-2, and Et-1), increased the expression of angiogenetic genes (Sema4D and PlexinB1), reduced G. parasuis-induced tight junction disruption, and reactivated G. parasuis-induced suppression of the PI3K/Akt/Erk signaling pathway in vitro. Thus, we concluded that quercetin may protect BBB integrity via the PI3K/Akt/Erk signaling pathway during G. parasuis infection. This was the first attempt to explore the protective effects of quercetin on brain inflammation and BBB integrity in a G. parasuis-infected mouse model. Our findings indicated that quercetin is a promising natural agent for the prevention and treatment of G. parasuis infection.

Comparative study of the effects of baicalin and probenecid on microRNA expression profiles in porcine aortic vascular endothelial cells infected by Glaesserella parasuis

Background
Glaesserella parasuis
elicits severe inflammatory responses and vascular damage, thus resulting in high mortality and morbidity in pigs; consequently, early diagnosis and treatment are critical to controlling economic losses. MicroRNAs (miRNAs) have been demonstrated to be involved in vascular endothelial inflammation. Baicalin is an effective Chinese medicinal herb with anti-microbial, anti-inflammatory, and anti-oxidant activity. Probenecid has activity toward multiple mammalian biological processes. Herein, we compared the effects of baicalin and probenecid on the miRNA expression profiles of porcine aortic vascular endothelial cells (PAVECs) infected with
G. parasuis
.
Results
We identified 277 known miRNAs and 540 novel miRNAs. Twelve miRNAs were significantly differentially expressed in PAVECs after
G. parasuis
infection. Both baicalin and probenecid affected the miRNA expression profiles in
G. parasuis
-infected PAVECs but showed different modulation patterns. Ssc-miR-27b-5p and ssc-miR-1842 were the top differentially expressed miRNAs (DEmiRNAs) in baicalin group comparing to control group. Ssc-miR-9851-3p and ssc-miR-1296-5p were the top DEmiRNAs in probenecid group. And Ssc-miR-127, ssc-miR-1842, and ssc-miR-9810-3p were the top DEmiRNAs between the baicalin group and probenecid group, as validated by qRT-PCR. The target genes of DEmiRNAs between various groups were subjected to KEGG and GO enrichment analyses. Hematopoietic cell lineage, insulin resistance, and AMPK signaling pathway were the top significantly enriched pathways associated with the target genes of DEmiRNAs in
G. parasuis
-infected PAVECs pretreated with baicalin; in contrast, B cell receptor, T cell receptor, and HIF-1 signaling pathways predominated in
G. parasuis
-infected PAVECs treated with probenecid. We additionally constructed co-expression and protein–protein interaction networks based on the differentially expressed target genes of miR-127, miR-1842, and miR-9810-3p.
Conclusion
Our findings suggested that baicalin and probenecid regulated miRNAs associated with vascular inflammation and damage, but showed different modulation patterns. This report provided the first comparison of the effects of baicalin and probenecid on
G. parasuis
-infected PAVECs, and might aid in the development of novel biomarkers and therapeutic targets to control
G. parasuis
infection.

Overexpression of Abscisic Acid Biosynthesis Gene OsNCED3 Enhances Survival Rate and Tolerance to Alkaline Stress in Rice Seedlings

Alkaline stress with high pH levels could significantly influence plant growth and survival. The enzyme 9-cis-epoxycarotenoid dioxygenase (NCED) serves as a critical bottleneck in the biosynthesis of abscisic acid (ABA), making it essential for regulating stress tolerance. Here, we show that OsNCED3-overexpressing rice lines have increased ABA content by up to 50.90% and improved transcription levels of numerous genes involved in stress responses that significantly enhance seedling survival rates. Overexpression of OsNCED3 increased the dry weight contents of the total chlorophyll, proline, soluble sugar, starch, and the activities of antioxidant enzymes of rice seedlings, while reducing the contents of O2·−, H2O2, and malondialdehyde under hydroponic alkaline stress conditions simulated by 10, 15, and 20 mmol L−1 of Na2CO3. Additionally, the OsNCED3-overexpressing rice lines exhibited a notable increase in the expression of OsNCED3; ABA response-related genes OsSalT and OsWsi18; ion homeostasis-related genes OsAKT1, OsHKT1;5, OsSOS1, and OsNHX5; and ROS scavenging-related genes OsCu/Zn-SOD, OsFe-SOD, OsPOX1, OsCATA, OsCATB, and OsAPX1 in rice seedling leaves. The results of these findings suggest that overexpression of OsNCED3 upregulates endogenous ABA levels and the expression of stress response genes, which represents an innovative molecular approach for enhancing the alkaline tolerance of rice seedlings.

Poly(N-Isopropylacrylamide)-Functional Silicon Nanocrystals for Thermosensitive Fluorescence Cellar Imaging

Silicon nanocrystals (Si NCs) have received surging interest as a type of quantum dot (QD) due to the availability of silicon in nature, tunable fluorescence emission properties and excellent biocompatibility. More importantly, compared with many group II–VI and III–V based QDs, they have low toxicity. Here, thermoresponsive poly(N-isopropylacrylamide) (PNIPAAm)-functional Si NCs were firstly prepared for thermoresponsive detection of cancer cells. Si NCs were prepared under normal pressure with excellent water solubility. Then folic acid was bonded to the silicon nanocrystals through the reaction of amino and carboxyl groups for specific recognition of cancer cells. The folic-acid-modified silicon crystals (Si NCs-FA) could be modified by a one-pot copolymerization process into PNIPAAm nanospheres during the monomer polymerization process (i.e., Si NCs-FA-PNIPAAm) just by controlling the temperature below the lower critical solution temperature (LCST) and above the LCST. The results showed that the Si-FA-PNIAAm nanospheres exhibited not only reversible temperature-responsive on-off fluorescence properties, but also can be used as temperature indicators in cancer cells.
