Lnc RNA HOTAIR functions as a competing endogenous RNA to regulate HER2 expression by sponging miR-331-3p in gastric cancer
Background Accumulating evidence indicates that the long non-coding RNA HOTAIR plays a critical role in cancer progression and metastasis. However, the overall biological role and clinical significance of HOTAIR in gastric carcinogenesis remains largely unknown. Methods HOTAIR expression was measured in 78 paired cancerous and noncancerous tissue samples by real-time PCR. The effects of HOTAIR on gastric cancer cells were studied by overexpression and RNA interference approaches in vitro and in vivo. Insights of the mechanism of competitive endogenous RNAs (ceRNAs) were gained from bioinformatic analysis, luciferase assays and RNA binding protein immunoprecipitation (RIP). The positive HOTAIR/HER2 interaction was identified and verified by immunohistochemistry assay and bivariate correlation analysis. Results HOTAIR upregulation was associated with larger tumor size, advanced pathological stage and extensive metastasis, and also correlated with shorter overall survival of gastric cancer patients. Furthermore, HOTAIR overexpression promoted the proliferation, migration and invasion of gastric carcinoma cells, while HOTAIR depletion inhibited both cell invasion and cell viability, and induced growth arrest in vitro and in vivo. In particular, HOTAIR may act as a ceRNA, effectively becoming a sink for miR-331-3p, thereby modulating the derepression of HER2 and imposing an additional level of post-transcriptional regulation. Finally, the positive HOTAIR/HER2 correlation was significantly associated with advanced gastric cancers. Conclusions HOTAIR overexpression represents a biomarker of poor prognosis in gastric cancer, and may confer malignant phenotype to tumor cells. The ceRNA regulatory network involving HOTAIR and the positive interaction between HOTAIR and HER2 may contribute to a better understanding of gastric cancer pathogenesis and facilitate the development of lncRNA-directed diagnostics and therapeutics against this disease.
Long noncoding RNA PVT1 indicates a poor prognosis of gastric cancer and promotes cell proliferation through epigenetically regulating p15 and p16
Background Mounting evidence indicates that long noncoding RNAs (lncRNAs) could play a pivotal role in cancer biology. However, the overall biological role and clinical significance of PVT1 in gastric carcinogenesis remains largely unknown. Methods Expression of PVT1 was analyzed in 80 GC tissues and cell lines by qRT-PCR. The effect of PVT1 on proliferation was evaluated by MTT and colony formation assays, and cell apoptosis was evaluated by Flow-cytometric analysis. GC cells transfected with sh PVT1 were injected into nude mice to study the effect of PVT1 on tumorigenesis in vivo. RIP was performed to confirm the interaction between PVT1 and EZH2. ChIP was used to study the promoter region of related genes. Results The higher expression of PVT1 was significantly correlated with deeper invasion depth and advanced TNM stage. Multivariate analyses revealed that PVT1 expression served as an independent predictor for overall survival (p = 0.031). Further experiments demonstrated that PVT1 knockdown significantly inhibited the proliferation both in vitro and in vivo. Importantly, we also showed that PVT1 played a key role in G1 arrest. Moreover, we further confirmed that PVT1 was associated with enhancer of zeste homolog 2 (EZH2) and that this association was required for the repression of p15 and p16. To our knowledge, this is the first report showed that the role and the mechanism of PVT1 in the progression of gastric cancer. Conclusions Together, these results suggest that lncRNA PVT1 may serve as a candidate prognostic biomarker and target for new therapies in human gastric cancer.
Stronger policy required to substantially reduce deaths from PM2.5 pollution in China
Air pollution kills nearly 1 million people per year in China. In response, the Chinese government implemented the Air Pollution Prevention and Control Action Plan (APPCAP) from 2013 to 2017 which had a significant impact on reducing PM 2.5 concentration. However, the health benefits of the APPCAP are not well understood. Here we examine the spatiotemporal dynamics of annual deaths attributable to PM 2.5 pollution (DAPP) in China and the contribution from the APPCAP using decomposition analysis. Despite a 36.1% increase in DAPP from 2000 to 2017, The APPCAP-induced improvement in air quality achieved substantial health benefits, with the DAPP in 2017 reduced by 64 thousand (6.8%) compared to 2013. However, the policy is unlikely to result in further major reductions in DAPP and more ambitious policies are required to reduce the health impacts of air pollution by 2030 and meet the United Nation’s Sustainable Development Goal 3. Chinese government has implemented the air pollution control measure-the Air Pollution Prevention and Control Action Plan in 2013, whose effects have not been fully studied. Here the authors show that from 2013 to 2017, the plan has achieved substantial public health benefits.
MicroRNA-196a promotes non-small cell lung cancer cell proliferation and invasion through targeting HOXA5
Background MicroRNAs (miRNAs) are short, non-coding RNAs (~22 nt) that play important roles in the pathogenesis of human diseases by negatively regulating gene expression. Although miR-196a has been implicated in several other cancers, its role in non-small cell lung cancer (NSCLC) is unknown. The aim of the present study was to examine the expression pattern of miR-196a in NSCLC and its clinical significance, as well as its biological role in tumor progression. Methods Expression of miR-196a was analyzed in 34 NSCLC tissues and five NSCLC cell lines by quantitative reverse-transcription polymerase chain reaction (qRT-PCR). The effect of DNA methylation on miR-196a expression was investigated by 5-aza-2-deoxy-cytidine treatment and bisulfite sequencing. The effect of miR-196a on proliferation was evaluated by MTT and colony formation assays, and cell migration and invasion were evaluated by transwell assays. Analysis of target protein expression was determined by western blotting. Luciferase reporter plasmids were constructed to confirm the action of miR-196a on downstream target genes, including HOXA5 . Differences between the results were tested for significance using Student’s t-test (two-tailed). Results miR-196a was highly expressed both in NSCLC samples and cell lines compared with their corresponding normal counterparts, and the expression of miR-196a may be affected by DNA demethylation. Higher expression of miR-196a in NSCLC tissues was associated with a higher clinical stage, and also correlated with NSCLC lymph-node metastasis. In vitro functional assays demonstrated that modulation of miR-196a expression affected NSCLC cell proliferation, migration and invasion. Our analysis showed that miR-196a suppressed the expression of HOXA5 both at the mRNA and protein levels, and luciferase assays confirmed that miR-196a directly bound to the 3’untranslated region of HOXA5. Knockdown of HOXA5 expression in A549 cells using RNAi was shown to promote NSCLC cell proliferation, migration and invasion. Finally, we observed an inverse correlation between HOXA5 and miR-196a expression in NSCLC tissues. Conclusions Our findings indicate that miR-196a is significantly up-regulated in NSCLC tissues, and regulates NSCLC cell proliferation, migration and invasion, partially via the down-regulation of HOXA5. Thus, miR-196a may represent a potential therapeutic target for NSCLC intervention.
Street Geometry Factors Influencing Outdoor Pedestrian Thermal Comfort in a Historic District
As China’s urbanization progresses, the urban heat island (UHI) effect has become more pronounced, impacting the health of residents and the activity intentions of visitors within historic urban areas. This study focuses on the historic district of the Three Lanes and Seven Alleys Tourist Area (SFQX) in Fuzhou, where simulations were conducted on four representative streets across various times during a typical summer meteorological day. Typological methods were employed to simplify neighborhood modeling, and Phoenics software was utilized to simulate the neighborhood’s wind environment and the outdoor pedestrian thermal comfort index. Aspect ratio (AR), sky view factor (SVF), air velocity (Va), and universal thermal climate index (UTCI) values at specific locations were collected for statistical analysis. The findings reveal that: (1) the N–S orientation exhibits more significant correlations between Va, the UTCI, and street geometry compared to the E–W orientation; (2) the relationship between SVF and the UTCI fluctuates with time; (3) areas with higher AR values, such as medium and deep canyons, offer better thermal comfort for outdoor pedestrians; and (4) at 8:00, the UTCI and wind speed show minimal correlations with street geometry and direction, being predominantly influenced by objective climatic factors. These insights are expected to significantly inform the geometric design and planning of streets in Fuzhou’s historic districts, aiming to create more comfortable outdoor environments for inhabitants and visitors alike.
Decreased expression of long noncoding RNA MEG3 affects cell proliferation and predicts a poor prognosis in patients with colorectal cancer
Colorectal cancer (CRC) remains an important public health problem in the world. Long noncoding RNA (lncRNA) is an RNA molecular that is longer than 200 nucleotides and cannot be translated into a protein. Recent studies have shown that lncRNAs play important roles in carcinogenesis and cancer metastasis. The aim of this study was to evaluate the expression and biological role of lncRNA maternally expressed gene 3 (MEG3) in colorectal cancer. Quantitative real-time-PCR (qRT-PCR) was performed to investigate the expression of MEG3 in tumor tissues and corresponding nontumor colorectal tissues from 62 patients. The lower expression of MEG3 was remarkably correlated with low histological grade, deep tumor invasion, and advanced tumor node metastasis (TNM) stage. Multivariate analyses revealed that MEG3 expression served as an independent predictor for overall survival. Further experiments revealed that overexpressed MEG3 significantly inhibited CRC cell proliferation both in vitro and in vivo. In conclusion, our study demonstrated that MEG3 is involved in the development and progression of colorectal cancer by regulating cell proliferation and shows that MEG3 may be a potential diagnostic and prognostic target in patients with colorectal cancer.
Transcriptome sequencing and metabolite analysis for revealing the blue flower formation in waterlily
Background Waterlily ( Nymphaea spp.), a perennial herbaceous aquatic plant, is divided into two ecological groups: hardy waterlily and tropical waterlily. Although the hardy waterlily has no attractive blue flower cultivar, its adaptability is stronger than tropical waterlily because it can survive a cold winter. Thus, breeding hardy waterlily with real blue flowers has become an important target for breeders. Molecular breeding may be a useful way. However, molecular studies on waterlily are limited due to the lack of sequence data. Results In this study, six cDNA libraries generated from the petals of two different coloring stages of blue tropical waterlily cultivar Nymphaea ‘King of Siam’ were sequenced using the Illumina HiSeq™ 2500 platform. Each library produced no less than 5.65 Gb clean reads. Subsequently, de novo assembly generated 112,485 unigenes, including 26,206 unigenes annotated to seven public protein databases. Then, 127 unigenes could be identified as putative homologues of color-related genes in other species, including 28 up-regulated and 5 down-regulated unigenes. In petals, 16 flavonoids (4 anthocyanins and 12 flavonols) were detected in different contents during the color development due to the different expression levels of color-related genes, and four flavonols were detected in waterlily for the first time. Furthermore, UA3GT s were selected as the most important candidates involved in the flavonoid metabolic pathway, UA3GT s induced blue petal color formation in Nymphaea ‘King of Siam’. Conclusions This study will improve our understanding of the molecular mechanism of blue flowers in waterlily and provide the basis for molecular breeding of blue hardy waterlily cultivars.
How Social Media Data Mirror Spatio-Temporal Behavioral Patterns of Tourists in Urban Forests: A Case Study of Kushan Scenic Area in Fuzhou, China
Exploring the spatial distribution of tourist attractions and comprehending the spatio-temporal behaviors of tourists within tourist attractions can provide local planning agencies, destination marketing organizations, and government departments with essential evidence for decision-making processes. This study examines the spatio-temporal behavior patterns of tourists in the Kushan Scenic Area by analyzing GPS trajectory data acquired from social media platforms. The investigation primarily utilizes three research methodologies: grid analysis, Markov chain, and K-means clustering. The grid analysis results reveal three spatial distribution patterns within the scenic area, while the outcomes from the Markov chain and K-means clustering delineate six tourist movement patterns, along with three choices regarding travel time. This finding holds significant practical implications for enhancing the attractiveness of scenic areas, optimizing spatial layout, and improving tourists’ experiences.
Pu-erh Tea Extract Ameliorates Ovariectomy-Induced Osteoporosis in Rats and Suppresses Osteoclastogenesis In Vitro
Tea drinking is associated with positive effects on bone health and may protect against osteoporosis, especially in elderly women. Pu-erh tea has many beneficial effects on human health; however, whether Pu-erh tea has anti-osteoporotic potential remains unclear. Thus, we investigated the effects of Pu-erh tea extract (PTE) on ovariectomy-induced osteoporosis in rats and on osteoclastogenesis . Female Wistar rats were divided into six groups: the sham, model, and Xian-Ling-Gu-Bao capsule (XLGB) groups, and the low-, medium-, and high-dose PTE groups. Ovariectomized (OVX) rats were used as an animal model of osteoporosis. The animals were intragastrically administered distilled water, XLGB, or different concentrations of PTE for 13 weeks. Body weight, blood biochemical indicators, relative organ coefficients, femoral bone mineral density (BMD), bone biomechanical properties, and bone microarchitecture were examined and analyzed. Additionally, the effects of PTE on osteoclastic activities were investigated using the RAW 264.7 cell line as an osteoclast differentiation model. The effects of PTE on osteoclast differentiation and the expression of osteoclast-specific genes and proteins were determined. PTE reduced OVX-induced body weight gain after 6 weeks of treatment, and the high-dose exerted a significant effect. High-dose PTE significantly ameliorated OVX-induced estradiol (E ) deficiency. PTE treatment maintained calcium and phosphorus homeostasis and improved other blood biochemical parameters to various degrees. In addition, PTE treatment improved organ coefficients of the femur, uterus, and vagina and improved femoral BMD and bone biomechanical properties. PTE treatment strikingly ameliorated bone microarchitecture. Moreover, in the studies, osteoclast differentiation using the differentiation cell model was significantly inhibited by PTE without cytotoxic effects. Additionally, PTE efficaciously suppressed the expression of key osteoclast-specific genes and proteins. PTE can ameliorate ovariectomy-induced osteoporosis in rats and suppress osteoclastogenesis .
Mini-mint ice cubes for early postoperative thirst relief in orthopedic patients undergoing general anesthesia: a randomized controlled trial
Postoperative thirst affects 70% of surgical patients, causing discomfort and potential complications, especially in orthopedic patients. Current relief strategies remain suboptimal. This trial evaluated the efficacy and safety of mini-mint ice cubes (1 cm³, 20% mint) for early postoperative thirst relief. 282 patients were randomized to three groups: mini-mint ice cubes, room temperature water or absolute fasting. Primary outcome was thirst intensity at PACU discharge, measured by Numerical Rating Scale. Secondary outcomes included PACU stay, oropharyngeal discomfort, satisfaction, 24-hour postoperative quality of recovery (QoR-15), postoperative nausea and vomiting (PONV), delirium and adverse events. Mini-mint group had lower thirst (median [IQR]: 3 [1–5]) vs. water (4 [2–5], median difference: -1, 95% CI: -2 to 0; P  = 0.004); vs. absolute fasting (7 [5–9], median difference: -4, 95% CI: -5 to -3; P  < 0.001), shorter PACU stay (35 [30–43] min) vs. water (40 [33–45] min, P  = 0.014); vs. absolute fasting (40 [35–55] min, P  < 0.001), less oropharyngeal discomfort (12.9% vs. absolute fasting 27.2%, P  = 0.015), higher satisfaction (4 [4–5]) vs. water (4 [4–5], P  = 0.003) and absolute fasting (3 [2–4], P  < 0.001), higher QoR-15 (124 [119–130]) vs. water (119 [114–125], P  < 0.001) and absolute fasting (117 [111–123], P  < 0.001). PONV, delirium, and hypoxemia showed no intergroup differences; cough occurred only in the water group (6.4%). Mini-mint ice cubes safely and effectively alleviate early postoperative thirst in orthopedic patients under general anesthesia, conferring clinically meaningful benefits compared to absolute fasting. Compared to room temperature water, the improvement is modest with uncertain clinical relevance. Clinical trial registration number: ClinicalTrials.gov (ChiCTR2400089335, 6/9/2024).
