mRNA vaccines encoding fusion proteins of monkeypox virus antigens protect mice from vaccinia virus challenge
The recent outbreaks of mpox have raised concerns over the need for effective vaccines. However, the current approved vaccines have either been associated with safety concerns or are in limited supply. mRNA vaccines, which have shown high efficacy and safety against SARS-CoV-2 infection, are a promising alternative. In this study, three mRNA vaccines are developed that encode monkeypox virus ( MPXV ) proteins A35R and M1R, including A35R extracellular domain -M1R fusions (VGPox 1 and VGPox 2) and a mixture of encapsulated full-length mRNAs for A35R and M1R (VGPox 3). All three vaccines induce early anti-A35R antibodies in female Balb/c mice, but only VGPox 1 and 2 generate detectable levels of anti-M1R antibodies at day 7 after vaccination. However, all three mRNA vaccine groups completely protect mice from a lethal dose of vaccinia virus ( VACV ) challenge. A single dose of VGPox 1, 2, and 3 provide protection against the lethal viral challenge within 7 days post-vaccination. Long-term immunity and protection were also observed in all three candidates. Additionally, VGPox 2 provided better passive protection. These results suggest that the VGPox series vaccines enhance immunogenicity and can be a viable alternative to current whole-virus vaccines to defend against mpox. The authors report mRNA vaccines encoding a fusion protein of MPXV A35R extracellular domain and full-length M1R and observe improved anti-M1R antibody response. The vaccines show enhanced active and passive protection in female mice challenged with a lethal dose of vaccinia virus.
How does the development of the digital economy in RCEP member countries affect China’s cross-border e-commerce exports?
As a significant new mode of trade export in the digital economy era, cross-border e-commerce injects new momentum into trade cooperation among the Regional Comprehensive Economic Partnership (RCEP) member countries. This paper utilizes multi-dimensional panel data constructed from World Bank databases and UNCTAD databases from 2012 to 2021 to analyze the impact mechanism of RCEP member countries’ digital economy development on China’s cross-border e-commerce export through direct and indirect channels and conducts empirical tests on it. The research results show that, in terms of direct impact, the development of the digital economy in RCEP member countries has promoted China’s cross-border e-commerce export, and its impact is heterogeneous. Regarding indirect impact, improving the quality of RCEP member countries’ transportation infrastructure and institutional quality is a moderating factor promoting China’s cross-border e-commerce export. This study provides important insights for deepening the development of the digital economy in RCEP countries and promoting China’s cross-border e-commerce export.
Evaluation of Osmia excavata (Hymenoptera: Megachilidae) sensitivity to high-temperature stress
The population of Osmia excavata , an important pollinator in commercial orchards, has been in serious decline over recent years. To evaluate the risk of high-temperature stress on O. excavata , we evaluated the high-temperature tolerance and potential physiological and biochemical responses of O. excavata after a series of high-temperature stresses. The results showed that the mortality rates of O. excavata increased gradually with increases in temperature and duration of stress ( R 2 = 0.88–0.99; p < 0.05). The larvae of O. excavata were more sensitive to temperature stress than adults, and the median lethal time (LT 50 ) value of the former was smaller than the latter in the acute response test. By comparing the results of acute and chronic responses of O. excavata to high-temperature stress, we found that the LT 50 values of natural cocoon-break adults at slightly higher temperatures (35°C and 40°C) were smaller than those of artificial cocoon-break adults, but the LT 50 values were similar under extreme high-temperature stress (45°C). Furthermore, the acute and chronic responses on the fat content of adult Osmia obtained by artificial and natural cocoon-break methods were significantly different ( F = 5.03; p < 0.05). Additionally, the mortalities of the young larvae and artificial cocoon-break adults were both significantly and positively correlated with trehalose content ( r = 0.78–0.82, p < 0.05). However, the mortality of the natural cocoon-break adults was negatively related to the acetylcholinesterase activity ( r = −0.93, p < 0.001). Overall, these results suggested that O. excavata has a low tolerance to high-temperature stress and provide evidence of causes that could be contributing to the population decline of O. excavata.
Cation controlled rotation in anionic pillar5arenes and its application for fluorescence switch
Controlling molecular motion is one of hot topics in the field of chemistry. Molecular rotors have wide applications in building nanomachines and functional materials, due to their controllable rotations. Hence, the development of novel rotor systems, controlled by external stimuli, is desirable. Pillar[n]arenes, a class of macrocycles, have a unique planar chirality, in which two stable conformational isomers p R and p S would interconvert by oxygen-through-the-annulus rotations of their hydroquinone rings. We observe the differential kinetic traits of planar chirality transformation in sodium carboxylate pillar[5]arene ( WP5-Na ) and ammonium carboxylate pillar[5]arene ( WP5-NH 4 ), which inspire us to construct a promising rotary platform in anionic pillar[5]arenes ( WP5 ) skeletons. Herein, we demonstrate the non-negligible effect of counter cations on rotational barriers of hydroquinone rings in WP5 , which enables a cation grease/brake rotor system. Applications of this tunable rotor system as fluorescence switch and anti-counterfeiting ink are further explored. Controlling molecular motion is challenging. Here, the authors report the effect of counter cations on the rotational barriers of hydroquinone rings in a molecular rotor based on anionic pillar[5]arenes.
Hyperband-Optimized CNN-BiLSTM with Attention Mechanism for Corporate Financial Distress Prediction
In the context of new quality productive forces, enterprises must leverage technological innovation and intelligent management to enhance financial risk resilience. This article proposes a financial distress prediction model based on deep learning, combined with a CNN, BiLSTM, and attention mechanism, using SMOTE for sample imbalance and Hyperband for hyperparameter optimization. Among four CNN-BiLSTM-AT model structures and seven mainstream models (CNN, BiLSTM, CNN-BiLSTM, CNN-AT, BiLSTM-AT, CNN-GRU, and Transformer), the 1CNN-1BiLSTM-AT model achieved the highest validation accuracy and relatively faster training speed. We conducted 100 repeated experiments using data from two companies, with validation on 2025 data, confirming the model’s stability and effectiveness in real-world scenarios. This article lays a solid empirical foundation for further optimization of financial distress warning models.
Urolithin A Attenuates Hyperuricemic Nephropathy in Fructose-Fed Mice by Impairing STING-NLRP3 Axis-Mediated Inflammatory Response via Restoration of Parkin-Dependent Mitophagy
Urolithin A (UroA) is one of the primary intestinal metabolites of ellagitannins, showing translational potential as a nutritional intervention in humans. Mounting evidence suggests that fructose consumption contributes to the progression of chronic kidney disease (CKD) that manifests in hyperuricemic nephropathy, renal inflammation, and tubulointerstitial injury. Here, we investigated the efficacy of UroA in alleviating fructose-induced hyperuricemic nephropathy in mice. Uric acid-exposed human kidney-2 (HK-2) cells were utilized for in vitro mechanism validation. Histopathological staining, immunoblotting, and transmission electron microscope were performed for the mechanistic investigations. Our results revealed that UroA ameliorated fructose-induced hyperuricemic nephropathy in mice. The histopathologic assessment showed that UroA attenuated tubular hypertrophy and dilation, glomerular basement membrane thickening, and collagen deposition in the kidney of fructose-fed mice. Mechanistically, UroA treatment impaired STING-NLRP3 activation, resulting in reduced production of proinflammatory cytokines IL-1β, IL-6, and TNF-α. Notably, UroA exhibited a scavenging effect against reactive oxygen species (ROS) and restored fructose-impaired PINK1/Parkin-mediated mitophagy in nephropathic mice. Furthermore, the inhibitory effect of UroA in STING-NLRP3 activation was impaired after Parkin gene silencing in HK-2 cells. Together, this study suggests that UroA alleviates fructose-induced hyperuricemic nephropathy by promoting Parkin-dependent mitophagy, thereby suppressing STING-NLRP3 axis-mediated inflammatory response. Thus, dietary supplementation with UroA or ellagitannins-rich foods may serve as a promising intervention to prevent CKD progression.
The first mitochondrial genome of Meroplius fukuharai (Diptera: Sepsidae)
is an important sanitary and ecological resource insect. We sequenced and annotated the mitogenome of which is the first representative of the genus with nearly complete mitochondrial data. This mitogenome is 14,803 bp long, which consists of 22 transfer RNA genes, 13 protein-coding genes (PCGs), and two ribosomal RNA genes. All genes have a conservational arrangement with other published species of Sepsidae. Our results also supported the monophyly of Sepsidae, and the genus is more closely related to genus , , and .
The identification of grain size genes by RapMap reveals directional selection during rice domestication
Cloning quantitative trait locus (QTL) is time consuming and laborious, which hinders the understanding of natural variation and genetic diversity. Here, we introduce RapMap, a method for rapid multi-QTL mapping by employing F 2 gradient populations (F 2 GPs) constructed by minor-phenotypic-difference accessions. The co-segregation standard of the single-locus genetic models ensures simultaneous integration of a three-in-one framework in RapMap i.e. detecting a real QTL, confirming its effect, and obtaining its near-isogenic line-like line (NIL-LL). We demonstrate the feasibility of RapMap by cloning eight rice grain-size genes using 15 F 2 GPs in three years. These genes explain a total of 75% of grain shape variation. Allele frequency analysis of these genes using a large germplasm collection reveals directional selection of the slender and long grains in indica rice domestication. In addition, major grain-size genes have been strongly selected during rice domestication. We think application of RapMap in crops will accelerate gene discovery and genomic breeding. Cloning quantitative trait loci (QTL) in crops is often slow and laborious. Here, the authors describe RapMap, a method to rapidly clone multiple QTL based on F 2 gradient populations coupled with a co-segregation standard, and show how it can be used to identify genes controlling grain size in rice.
New Fusarin Derivatives from the Marine Algicolous Fungus Penicillium steckii SCSIO41040
Five new fusarin derivatives, steckfusarins A–E (1–5), and two known natural products (6, 7), were isolated and identified from the marine algicolous fungus Penicillium steckii SCSIO 41040. The new compounds, including absolute configurations, were determined by spectroscopic analyses and calculated electronic circular dichroism (ECD). All new compounds were evaluated for their antioxidant, antibacterial, antifungal, antiviral, cytotoxic, anti-inflammatory, antioxidant, cholesterol-lowering, acetyl cholinesterase (AChE) enzyme and 6-phosphofructo-2-kinase (PFKFB3) and phosphatidylinositol-3-kinase (PI3K) inhibitory activities. The biological evaluation results revealed that compound 1 exhibited radical scavenging activity against 2,2-diphenyl-1-picrylhydrazylhydrate (DPPH), with an IC50 value of 74.5 µg/mL. In addition, compound 1 also showed weak anti-inflammatory activity at a concentration of 20 µM.
Detection of Key Organs in Tomato Based on Deep Migration Learning in a Complex Background
In the current natural environment, due to the complexity of the background and the high similarity of the color between immature green tomatoes and the plant, the occlusion of the key organs (flower and fruit) by the leaves and stems will lead to low recognition rates and poor generalizations of the detection model. Therefore, an improved tomato organ detection method based on convolutional neural network (CNN) has been proposed in this paper. Based on the original Faster R-CNN algorithm, Resnet-50 with residual blocks was used to replace the traditional vgg16 feature extraction network, and a K-means clustering method was used to adjust more appropriate anchor sizes than manual setting, to improve detection accuracy. The test results showed that the mean average precision (mAP) was significantly improved compared with the traditional Faster R-CNN model. The training model can be transplanted to the embedded system, which lays a theoretical foundation for the development of a precise targeting pesticide application system and an automatic picking device.
