Single-cell RNA sequencing reveals intrahepatic and peripheral immune characteristics related to disease phases in HBV-infected patients

ObjectiveA comprehensive immune landscape for HBV infection is pivotal to achieve HBV cure.DesignWe performed single-cell RNA sequencing of 2 43 000 cells from 46 paired liver and blood samples of 23 individuals, including six immune tolerant, 5 immune active (IA), 3 acute recovery (AR), 3 chronic resolved and 6 HBV-free healthy controls (HCs). Flow cytometry and histological assays were applied in a second HBV cohort for validation.ResultsBoth IA and AR were characterised by high levels of intrahepatic exhausted CD8+ T (Tex) cells. In IA, Tex cells were mainly derived from liver-resident GZMK+ effector memory T cells and self-expansion. By contrast, peripheral CX3CR1+ effector T cells and GZMK+ effector memory T cells were the main source of Tex cells in AR. In IA but not AR, significant cell–cell interactions were observed between Tex cells and regulatory CD4+ T cells, as well as between Tex and FCGR3A+ macrophages. Such interactions were potentially mediated through human leukocyte antigen class I molecules together with their receptors CANX and LILRBs, respectively, contributing to the dysfunction of antiviral immune responses. By contrast, CX3CR1+GNLY+ central memory CD8+ T cells were concurrently expanded in both liver and blood of AR, providing a potential surrogate marker for viral resolution. In clinic, intrahepatic Tex cells were positively correlated with serum alanine aminotransferase levels and histological grading scores.ConclusionOur study dissects the coordinated immune responses for different HBV infection phases and provides a rich resource for fully understanding immunopathogenesis and developing effective therapeutic strategies.

This paper presents a new topology optimization approach based on the so-called Moving Morphable Components (MMC) solution framework. The proposed method improves several weaknesses of the previous approach (e.g., Guo et al. in J Appl Mech 81:081009,
2014a
) in the sense that it can not only allow for components with variable thicknesses but also enhance the numerical solution efficiency substantially. This is achieved by constructing the topological description functions of the components appropriately, and utilizing the ersatz material model through projecting the topological description functions of the components. Numerical examples demonstrate the effectiveness of the proposed approach. In order to help readers understand the essential features of this approach, a 188 line Matlab implementation of this approach is also provided.

Global, regional and national epidemiology of allergic disorders in children from 1990 to 2019: findings from the Global Burden of Disease study 2019

ObjectiveThis modelling study aimed to estimate the burden for allergic diseases in children during a period of 30 years.DesignPopulation-based observational study.Main outcomes and measuresThe data on the incidence, mortality and disability-adjusted life years (DALYs) for childhood allergic diseases, such as atopic dermatitis (AD) and asthma, were retrieved from the Global Burden of Disease study 2019 online database. This data set spans various groups, including different regions, ages, genders and Socio-Demographic Indices (SDI), covering the period from 1990 to 2019.ResultsIn 2019, there were approximately 81 million children with asthma and 5.6 million children with AD worldwide. The global incidence of asthma in children was 20 million. Age-standardised incidence rates showed a decrease of 4.17% for asthma, from 1075.14 (95% uncertainty intervals (UI), 724.63 to 1504.93) per 100 000 population in 1990 to 1030.33 (95% UI, 683.66 to 1449.53) in 2019. Similarly, the rates for AD decreased by 5.46%, from 594.05 (95% UI, 547.98 to 642.88) per 100 000 population in 1990 to 561.61 (95% UI, 519.03 to 608.29) in 2019. The incidence of both asthma and AD was highest in children under 5 years of age, gradually decreasing with age. Interestingly, an increase in SDI was associated with a rise in the incidence of both conditions. However, the mortality rate and DALYs for asthma showed a contrasting trend.ConclusionsOver the past three decades, there has been a worldwide increase in new asthma and AD cases, even though mortality rates have significantly declined. However, the prevalence of these allergic diseases among children varies considerably across regions, countries and age groups. This variation highlights the need for precise prevalence assessments. These assessments are vital in formulating effective strategies for prevention and treatment.

A novel conceptual design approach for autonomous underwater helicopter based on multidisciplinary collaborative optimization

Autonomous underwater helicopters (AUHs) are complex electromechanical systems consisting of multiple interconnected sub-disciplines, posing a significant challenge for traditional sequential design strategies in managing their interactions. To address this challenge, a multidisciplinary collaborative optimization (MCO) strategy is proposed, integrating structural, hydrodynamic, and energy disciplines to enhance AUH cruising range. Additionally, a novel approximation model is introduced, combining the Logistic-Tent chaos sparrow search algorithm (LTC-SSA) with the backpropagation neural network (BPNN) algorithm. This method aims to replace costly computational fluid dynamics (CFD) and structural mechanics calculations, thus reducing AUH optimization costs. The laminate design of the composite pressure hull follows classic lamination theory (CLT) and the Tsai-Wu failure criterion. Results demonstrate that the multidisciplinary co-design strategy significantly reduces optimization time costs of AUH, from several months to several days. Furthermore, the proposed LTC-SSA-BPNN algorithm accurately evaluates sailing resistance and Tsai-Wu failure index, with maximum errors of 1.64% and 9.32% respectively. Additionally, the composite pressure shell achieves a 30.4% weight reduction and a 4.1% reduction in sailing resistance while maintaining structural stability. Moreover, the maximum cruising range increases by 34.4%. This strategy offers theoretical support for AUH conceptual design and promises performance enhancement in practical engineering applications.

Developing MXenes from Wireless Communication to Electromagnetic Attenuation

HighlightsThe industrial application and foundational research of MXenes at gigahertz frequency are systematically reviewed.The design principles of “lightweight, wide, and strong” are specifically highlighted.Current challenges and future directions for MXenes in wireless communication and electromagnetic attenuation are outlined.There is an urgent global need for wireless communication utilizing materials that can provide simultaneous flexibility and high conductivity. Avoiding the harmful effects of electromagnetic (EM) radiation from wireless communication is a persistent research hot spot. Two-dimensional (2D) materials are the preferred choice as wireless communication and EM attenuation materials as they are lightweight with high aspect ratios and possess distinguished electronic properties. MXenes, as a novel family of 2D materials, have shown excellent properties in various fields, owing to their excellent electrical conductivity, mechanical stability, high flexibility, and ease of processability. To date, research on the utility of MXenes for wireless communication has been actively pursued. Moreover, MXenes have become the leading materials for EM attenuation. Herein, we systematically review the recent advances in MXene-based materials with different structural designs for wireless communication, electromagnetic interference (EMI) shielding, and EM wave absorption. The relationship governing the structural design and the effectiveness for wireless communication, EMI shielding, and EM wave absorption is clearly revealed. Furthermore, our review mainly focuses on future challenges and guidelines for designing MXene-based materials for industrial application and foundational research.

A double machine learning model for measuring the impact of the Made in China 2025 strategy on green economic growth

The transformation and upgrading of China’s manufacturing industry is supported by smart and green manufacturing, which have great potential to empower the nation’s green development. This study examines the impact of the Made in China 2025 industrial policy on urban green economic growth. This study applies the super-slacks-based measure model to measure cities’ green economic growth, using the double machine learning model, which overcomes the limitations of the linear setting of traditional causal inference models and maintains estimation accuracy under high-dimensional control variables, to conduct an empirical analysis based on panel data of 281 Chinese cities from 2006 to 2021. The results reveal that the Made in China 2025 strategy significantly drives urban green economic growth, and this finding holds after a series of robustness tests. A mechanism analysis indicates that the Made in China 2025 strategy promotes green economic growth through green technology progress, optimizing energy consumption structure, upgrading industrial structure, and strengthening environmental supervision. In addition, the policy has a stronger driving effect for cities with high manufacturing concentration, industrial intelligence, and digital finance development. This study provides valuable theoretical insights and policy implications for government planning to promote high-quality development through industrial policy.

ClearSight-RS: A YOLOv5-Based Network with Dynamic Enhancement for Remote Sensing Small Target Detection

Small target detection in remote sensing images faces challenges due to complex backgrounds, weak features, and large scale differences. This paper proposes an improved YOLOv5-based network, termed ClearSight-RS, with the full name “Clear and Accurate Small-target Insight for Remote Sensing”. As the name implies, the network is dedicated to achieving clear feature perception and accurate target localization for small targets in remote sensing images. The improvements focus on three aspects: integrating an improved Dynamic Snake Convolution (DSConv) module into the backbone network to strengthen the extraction of small target boundaries and geometric features, as well as the expression of weak textures; embedding a Bi-Level Routing Attention (BRA) module in the Neck part to enhance target focusing and suppress background interference; and optimizing the detection head by retaining only shallow high-resolution feature layers for prediction, reducing feature loss and redundant computations. Experimental results show that, based on the VEDAI dataset, ClearSight-RS achieves the highest mAP for all 8 vehicle categories; based on the NWPU VHR-10 dataset, its overall mAP reaches 93.8%, significantly outperforming algorithms such as Faster RCNN and YOLOv5l; based on the DOTA dataset, the capability of the proposed BRA module in suppressing background interference and capturing small target features is demonstrated. The network balances accuracy and efficiency, performing prominently in detecting vehicles and multi-category small targets in complex backgrounds, verifying its effectiveness.

infection in the resistant genotype A204 and the susceptible genotype A198 at 0, 24 and 48 hours post-inoculation (hpi).

More genes and metabolites were induced at 24 hpi in A204 than A198, suggesting the prompt activation of defense responses in the resistant genotype, which can attribute two proteases, subtilisin-like protease and xylem cysteine proteinase 1, involved in pathogen recognition and signal transduction in A204.
