Comparative Evaluation of Intestinal Absorption and Functional Value of Iron Dietary Supplements and Drug with Different Delivery Systems
Iron is a fundament micronutrient, whose homeostasis is strictly regulated. Iron deficiency anemia is among the most widespread nutritional deficiencies and its therapy, based on dietary supplement and drugs, may lead to severe side effects. With the aim of improving iron bioavailability while reducing iron oral therapy side effects, novel dietary supplements based on innovative technologies—microencapsulation, liposomes, sucrosomes—have been produced and marketed. In the present work, six iron dietary supplements for different therapeutic targets were compared in terms of bioaccessibility, bioavailability, and safety by using an integrated in vitro approach. For general-purpose iron supplements, ME + VitC (microencapsulated) showed a fast, burst intestinal iron absorption kinetic, which maintained iron bioavailability and ferritin expression constant over time. SS + VitC (sucrosomes), on the other side, showed a slower, time-dependent iron absorption and ferritin expression trend. ME + Folate (microencapsulated) showed a behavior similar to that of ME + VitC, albeit with a lower bioavailability. Among pediatric iron supplements, a time-dependent bioavailability increase was observed for LS (liposome), while PIC (polydextrose-iron complex) bioavailability is severely limited by its poor bioaccessibility. Finally, except for SS + VitC, no adverse effects on intestinal mucosa vitality and barrier integrity were observed. Considering obtained results and the different therapeutic targets, microencapsulation-based formulations are endowed with better performance compared to the other formulations. Furthermore, performances of microencapsulated products were obtained with a lower iron daily dose, limiting the potential onset of side effects.
Intestinal Absorption Study of a Granular Form of Ferric Pyrophosphate
Iron deficiency is one of the most prevalent nutritional disorders worldwide. The standard treatment involves iron supplementation, but this task is challenging because of poor solubility and organoleptic issues. Moreover, the need to increase iron bioavailability represents a challenge for treating iron-related disorders. In this study, gastroresistance and iron intestinal absorption of an innovative granular formulation composed of ferric pyrophosphate, modified starch and phospholipids branded as Ferro Fosfosoma® was investigated. Gastroresistant properties were studied using standard protocols, and a bioaccessible fraction was obtained by exposing a food supplement to in vitro digestion. This fraction was used for investigating iron absorption in Caco-2 and human follicle-associated intestinal epithelium (FAE) models. Ferro Fosfosoma® showed an improved resistance to gastric digestion and higher intestinal absorption than ferric pyrophosphate salt used as a control in both models. In the FAE model, Ferro Fosfosoma® induces larger iron absorption than in the Caco-2 monolayer, most likely due to the transcytosis ability of M cells. The larger iron absorption in the Ferro Fosfosoma®-treated FAE model corresponds to higher ferritin level, proving physiological iron handling that was once delivered by granular formulation. Finally, the formulation did not induce any alterations in viability and barrier integrity. To conclude, Ferro Fosfosoma® favors iron absorption and ferritin expression, while preserving any adverse effects.
Integrative microRNAome analysis of skeletal muscle of Colossoma macropomum (tambaqui), Piaractus mesopotamicus (pacu), and the hybrid tambacu, based on next-generation sequencing data
Background Colossoma macropomum (tambaqui) and Piaractus mesopotamicus (pacu) are good fish species for aquaculture. The tambacu, individuals originating from the induced hybridization of the female tambaqui with the male pacu, present rapid growth and robustness, characteristics which have made the tambacu a good choice for Brazilian fish farms. Here, we used small RNA sequencing to examine global miRNA expression in the genotypes pacu (PC), tambaqui (TQ), and hybrid tambacu (TC), (Juveniles, n  = 5 per genotype), to better understand the relationship between tambacu and its parental species, and also to clarify the mechanisms involved in tambacu muscle growth and maintenance based on miRNAs expression. Results Regarding differentially expressed (DE) miRNAs between the three genotypes, we observed 8 upregulated and 7 downregulated miRNAs considering TC vs. PC; 14 miRNAs were upregulated and 10 were downregulated considering TC vs. TQ, and 15 miRNAs upregulated and 9 were downregulated considering PC vs. TQ. The majority of the miRNAs showed specific regulation for each genotype pair, and no miRNA were shared between the 3 genotype pairs, in both up- and down-regulated miRNAs. Considering only the miRNAs with validated target genes, we observed the miRNAs miR-144-3p, miR-138-5p, miR-206-3p, and miR-499-5p. GO enrichment analysis showed that the main target genes for these miRNAs were grouped in pathways related to oxygen homeostasis, blood vessel modulation, and oxidative metabolism. Conclusions Our global miRNA analysis provided interesting DE miRNAs in the skeletal muscle of pacu, tambaqui, and the hybrid tambacu. In addition, in the hybrid tambacu, we identified some miRNAs controlling important molecular muscle markers that could be relevant for the farming maximization.
Biocontrol traits of Bacillus licheniformis GL174, a culturable endophyte of Vitis vinifera cv. Glera
Background Bacillus licheniformis GL174 is a culturable endophytic strain isolated from Vitis vinifera cultivar Glera, the grapevine mainly cultivated for the Prosecco wine production. This strain was previously demonstrated to possess some specific plant growth promoting traits but its endophytic attitude and its role in biocontrol was only partially explored. In this study, the potential biocontrol action of the strain was investigated in vitro and in vivo and, by genome sequence analyses, putative functions involved in biocontrol and plant-bacteria interaction were assessed. Results Firstly, to confirm the endophytic behavior of the strain, its ability to colonize grapevine tissues was demonstrated and its biocontrol properties were analyzed. Antagonism test results showed that the strain could reduce and inhibit the mycelium growth of diverse plant pathogens in vitro and in vivo. The strain was demonstrated to produce different molecules of the lipopeptide class; moreover, its genome was sequenced, and analysis of the sequences revealed the presence of many protein-coding genes involved in the biocontrol process, such as transporters, plant-cell lytic enzymes, siderophores and other secondary metabolites. Conclusions This step-by-step analysis shows that Bacillus licheniformis GL174 may be a good biocontrol agent candidate, and describes some distinguished traits and possible key elements involved in this process. The use of this strain could potentially help grapevine plants to cope with pathogen attacks and reduce the amount of chemicals used in the vineyard.
The Development and Validation of an LC-MS/MS Method for the Determination of Glyphosate, AMPA, and Glufosinate in Honey Following FMOC-Cl Derivatization: Application to Italian Samples
Glyphosate-based herbicides are among the most widely used pesticides worldwide, but data on their occurrence in food products, particularly honey, remain limited. The analytical determination of glyphosate, its primary metabolite aminomethylphosphonic acid (AMPA), and glufosinate ammonium is technically challenging due to their high polarity and distinctive physicochemical properties, requiring the development of dedicated single-residue analytic methods. In this study, honey samples were prepared by derivatizing the target analytes with 9-fluorenylmethyl chloroformate (FMOC-Cl), followed by solid-phase extraction (SPE) using hydrophilic–lipophilic balanced (HLB) cartridges to improve matrix clean-up and enhance analytical sensitivity. Quantification was performed by liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). The optimized method was validated according to the SANTE/11312/2021 guidelines, with all parameters, including sensitivity, linearity, mean recovery (accuracy), precision (RSDr), and limit of quantification (LOQ), meeting the required performance criteria. The validated method was applied to 126 honey samples of various botanical origins, representative of Italian production. The results indicated a frequent detection of glyphosate residues, although concentrations were generally low and remain below levels of regulatory concern.
Prognostic Potential of Immune Inflammatory Biomarkers in Breast Cancer Patients Treated with Neoadjuvant Chemotherapy
Immune inflammatory biomarkers are easily obtained and inexpensive blood-based parameters that recently showed prognostic and predictive value in many solid tumors. In this study, we aimed to investigate the role of these biomarkers in predicting distant relapse in breast cancer patients treated with neoadjuvant chemotherapy (NACT). All breast cancer patients who referred to our Breast Unit and underwent NACT were retrospectively reviewed. The pre-treatment neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR), monocyte-to-lymphocyte ratio (MLR), and pan-immune-inflammation value (PIV) were calculated from complete blood counts. The primary outcome was 5-year distant-metastasis-free survival (DMFS). In receiver operating characteristic analyses, the optimal cutoff values for the NLR, PLR, MLR, and PIV were determined at 2.25, 152.46, 0.25, and 438.68, respectively. High levels of the MLR, but not the NLR, PLR, or PIV, were associated with improved 5-year DMSF in the study population using both univariate (HR 0.52, p = 0.03) and multivariate analyses (HR, 0.44; p = 0.02). Our study showed that the MLR was a significant independent parameter affecting DMFS in breast cancer patients undergoing NACT. Prospective studies are required to confirm this finding and to define reliable cutoff values, thus leading the way for the clinical application of this biomarker.

Enhancing Urban Wastewater Treatment through Isolated Chlorella Strain-Based Phytoremediation in Centrate Stream: An Analysis of Algae Morpho-Physiology and Nutrients Removal Efficiency
The release of inadequately treated urban wastewater is the main cause of environmental pollution of aquatic ecosystems. Among efficient and environmentally friendly technologies to improve the remediation process, those based on microalgae represent an attractive alternative due to the potential of microalgae to remove nitrogen (N) and phosphorus (P) from wastewaters. In this work, microalgae were isolated from the centrate stream of an urban wastewater treatment plant and a native Chlorella-like species was selected for studies on nutrient removal from centrate streams. Comparative experiments were set up using 100% centrate and BG11 synthetic medium, modified with the same N and P as the effluent. Since microalgal growth in 100% effluent was inhibited, cultivation of microalgae was performed by mixing tap-freshwater with centrate at increasing percentages (50%, 60%, 70%, and 80%). While algal biomass and nutrient removal was little affected by the differently diluted effluent, morpho-physiological parameters (FV/FM ratio, carotenoids, chloroplast ultrastructure) showed that cell stress increased with increasing amounts of centrate. However, the production of an algal biomass enriched in carotenoids and P, together with N and P abatement in the effluent, supports promising microalgae applications that combine centrate remediation with the production of compounds of biotechnological interest; for example, for organic agriculture.
Retrospective and Prospective Surveillance and Clinical Presentation of Parvovirus B19 in Veneto, Italy, 2024
The recent increase in parvovirus B19 (B19V) infections across Europe has underscored the need for enhanced surveillance. In Italy, B19V surveillance is not mandated nationally. This ambispective (retrospective and prospective) study aimed to assess the epidemiological and clinical characteristics of the 2024 apparent surge in the Veneto Region by implementing mandatory disease notification starting in May 2024 and collecting clinical data on confirmed cases. During the surveillance period, 3156 B19V cases were reported, with 2.9% (93/3156) requiring hospitalisation (95% CI 2.4–3.5%). Children under 1 year of age exhibited the highest hospitalisation rate (29.0%), followed by adults aged 18–40 (17.5%). Infections disproportionately affected children aged 1–5 and 6–11, and the Granger causality test showed that an increase in cases among the 1–5-year-olds was predictive of subsequent rises in other age groups (p = 0.0086). Severe complications, including myocarditis, pericarditis, and miscarriage, were observed, and one death occurred following cardiac and neurological complications in a patient without prior risk factors. The presence of concurrent pathologies, particularly haematological disorders, was associated with increased disease severity. These findings highlight the importance of early warning when cases begin rising among preschool-aged children and underscore the need for improved B19V awareness, particularly in high-risk populations. Future efforts should evaluate the feasibility of implementing a systematic, potentially EU-wide, surveillance for B19V.
Usability and feasibility of INFORMA platform for computerized cognitive stimulation in mild cognitive impairment and early dementia: an 8-week multicenter study
Background Mild cognitive impairment (MCI) and dementia represent significant global health challenges requiring innovative intervention approaches. Computerized cognitive training (CCT) offers promising alternatives to traditional face-to-face interventions, providing flexibility and personalized care for individuals with cognitive disorders. Aims This study evaluated the usability of INFORMA, a novel software platform based on Cognitive Activation Therapy (CAT), for delivering telerehabilitation to participants with MCI and mild-to-moderate dementia. Methods A prospective, non-interventional multicenter study enrolled 241 participants (MMSE 20–30, CDR 0.5-1) from 12 Italian Centers for Cognitive Disorders and Dementia. Participants engaged in an 8-week tablet-based telerehabilitation program with weekly supervision. Usability was assessed using the System Usability Scale (SUS), Task Completion Rate (TCR), and Relative Task Completion Time (R-TCT), supplemented by participant and caregiver questionnaires. Results Of 219 completing participants (mean age 73.5 years), the mean SUS score was 82.0 (95% CI: 80.3–83.8), with 84% exceeding the usability threshold (SUS score > 68). Mean TCR reached 97.8%, while 70.3% demonstrated improved R-TCT. Higher SUS scores correlated with education and MMSE scores but decreased with age. Satisfaction ratings exceeded 87% for participants and 95% for caregivers. Discussion INFORMA demonstrated high usability across all educational and cognitive levels, with the hybrid model combining automated exercises and professional supervision being well-received. Conclusions INFORMA showed significant promise as a user-friendly computerized cognitive stimulation tool, supporting its potential efficacy for MCI and dementia interventions and warranting further therapeutic efficacy studies.
