Pharmacokinetics and drug delivery systems for puerarin, a bioactive flavone from traditional Chinese medicine

Pueraria lobata (Willd.) Ohwi is a medicinal and edible homologous plant with a long history in China. Puerarin, the main component isolated from the root of Pueraria lobata, possesses a wide range of pharmacological properties. Daidzein and glucuronides are the main metabolites of puerarin and are excreted in the urine and feces. As active substrates of P-gp, multidrug resistance-associated protein and multiple metabolic enzymes, the pharmacokinetics of puerarin can be influenced by different pathological conditions and drug-drug interactions. Due to the poor water-solubility and liposolubility, the applications of puerarin are limited. So far, only puerarin injections and eye drops are on the market. Recent years, researches on improving the bioavailability of puerarin are developing rapidly, various nanotechnologies and preparation technologies including microemulsions and SMEDDS, dendrimers, nanoparticles and nanocrystals have been researched to improve the bioavailability of puerarin. In order to achieve biocompatibility and desired activity, more effective quality evaluations of nanocarriers are required. In this review, we summarize the pharmacokinetics and drug delivery systems of puerarin up to date.

Formation and growth mechanism of thin Cu6Sn5 films in Sn/Cu and Sn-0.1AlN/Cu structures using laser heating

Purpose
The purpose of this study is the formation and growth of nanoscale intermetallic compounds (IMCs) when laser is used as a heat source to form solder joints.
Design/methodology/approach
This study investigates the Sn/Cu and Sn-0.1AlN/Cu structure using laser soldering under different laser power: (200, 225 and 250 W) and heating time: (2, 3 and 4 s).
Findings
The results show clearly that the formation of nano-Cu6Sn5 films is feasible in the laser heating (200 W and 2 s) with Sn/Cu and Sn-0.1AlN/Cu system. The nano-Cu6Sn5 films with thickness of 500 nm and grains with 700 nm are generally parallel to the Cu surface with Sn-0.1AlN. Both IMC films thickness of Sn/Cu and Sn-0.1AlN/Cu solder joints gradually increased from 524.2 to 2025.8 nm as the laser heating time and the laser power extended. Nevertheless, doping AlN nanoparticles can slow down the growth rate of Cu6Sn5 films in Sn solder joints due to its adsorption.
Originality/value
The formation of nano-Cu6Sn5 films using laser heating can provide a new method for nanofilm development to realize the metallurgical interconnection in electronic packaging.

Genomic regions under selection in the feralization of the dingoes
2020
Dingoes are wild canids living in Australia, originating from domestic dogs. They have lived isolated from both the wild and the domestic ancestor, making them a unique model for studying feralization. Here, we sequence the genomes of 10 dingoes and 2 New Guinea Singing Dogs. Phylogenetic and demographic analyses show that dingoes originate from dogs in southern East Asia, which migrated via Island Southeast Asia to reach Australia around 8300 years ago, and subsequently diverged into a genetically distinct population. Selection analysis identifies 50 positively selected genes enriched in digestion and metabolism, indicating a diet change during feralization of dingoes. Thirteen of these genes have shifted allele frequencies compared to dogs but not compared to wolves. Functional assays show that an A-to-G mutation in ARHGEF7 decreases the endogenous expression, suggesting behavioral adaptations related to the transitions in environment. Our results indicate that the feralization of the dingo induced positive selection on genomic regions correlated to neurodevelopment, metabolism and reproduction, in adaptation to a wild environment.

Chemical composition, antibacterial activity and action mechanism of different extracts from hawthorn (Crataegus pinnatifida Bge.)

Present study was designed to compared the total flavonoids and polyphenols contents and antibacterial activity of hawthorn extracts with different polarities as well as the underlying antibacterial mechanisms. The results showed that among all hawthorn extracts, methanol and ethanol extracts (ME and EE) exhibited high levels of total flavonoids and polyphenols contents, followed by acetone, ethyl acetate, trichloromethane and petroleum ether extracts. ME exhibited the strongest antibacterial activity against tested bacteria, especially
Staphylococcus aureus
with a 1.25 μg/mL of the minimum inhibitory concentration (MIC) and minimum bactericide concentration (MBC). Further analysis revealed that the main phenolic compounds from ME were epicatechin (281.6 mg/100 g DW), procyanidin B2 (243.5 mg/100 g DW), chlorogenic acid (84.2 mg/100 g DW) and quercetin (78.4 mg/100 g DW). The action mechanism of ME against
S. aureus
could be ascribed to ME damaging cell wall and cell membrane integrity, inhibiting intracellular enzyme activity, increasing reactive oxygen species (ROS), also changing expression of associated genes and then inducing apoptosis of
S. aureus
. In addition, the antimicrobial activity of ME against
S. aureus
has also been demonstrated to be efficient in the food matrix (whole milk).

A Pattern-Recognition-Based Ensemble Data Imputation Framework for Sensors from Building Energy Systems

Building operation data are important for monitoring, analysis, modeling, and control of building energy systems. However, missing data is one of the major data quality issues, making data imputation techniques become increasingly important. There are two key research gaps for missing sensor data imputation in buildings: the lack of customized and automated imputation methodology, and the difficulty of the validation of data imputation methods. In this paper, a framework is developed to address these two gaps. First, a validation data generation module is developed based on pattern recognition to create a validation dataset to quantify the performance of data imputation methods. Second, a pool of data imputation methods is tested under the validation dataset to find an optimal single imputation method for each sensor, which is termed as an ensemble method. The method can reflect the specific mechanism and randomness of missing data from each sensor. The effectiveness of the framework is demonstrated by 18 sensors from a real campus building. The overall accuracy of data imputation for those sensors improves by 18.2% on average compared with the best single data imputation method.

The Pan-STARRS1 z > 5.6 Quasar Survey. II. Discovery of 55 Quasars at 5.6 < z < 6.5

The identification of bright quasars at z ≳ 6 enables detailed studies of supermassive black holes, massive galaxies, structure formation, and the state of the intergalactic medium within the first billion years after the Big Bang. We present the spectroscopic confirmation of 55 quasars at redshifts 5.6 < z < 6.5 and UV magnitudes −24.5 < M 1450 < −28.5 identified in the optical Pan-STARRS1 and near-IR VIKING surveys (48 and 7, respectively). Five of these quasars have independently been discovered in other studies. The quasar sample shows an extensive range of physical properties, including 17 objects with weak emission lines, 10 broad absorption line quasars, and 5 objects with strong radio emission (radio-loud quasars). There are also a few notable sources in the sample, including a blazar candidate at z = 6.23, a likely gravitationally lensed quasar at z = 6.41, and a z = 5.84 quasar in the outskirts of the nearby (D ∼ 3 Mpc) spiral galaxy M81. The blazar candidate remains undetected in NOEMA observations of the [C ii] and underlying emission, implying a star formation rate <30–70 M ⊙ yr−1. A significant fraction of the quasars presented here lies at the foundation of the first measurement of the z ∼ 6 quasar luminosity function from Pan-STARRS1 (introduced in a companion paper). These quasars will enable further studies of the high-redshift quasar population with current and future facilities.

Understanding how gut flora influences gut-brain communications has been the subject of significant research over the past decade. The broadening of the term “microbiota-gut-brain axis” from “gut-brain axis” underscores a bidirectional communication system between the gut and the brain. The microbiota-gut-brain axis involves metabolic, endocrine, neural, and immune pathways which are crucial for the maintenance of brain homeostasis. Alterations in the composition of gut microbiota are associated with multiple neuropsychiatric disorders. Although a causal relationship between gut dysbiosis and neural dysfunction remains elusive, emerging evidence indicates that gut dysbiosis may promote amyloid-beta aggregation, neuroinflammation, oxidative stress, and insulin resistance in the pathogenesis of Alzheimer’s disease (AD). Illustration of the mechanisms underlying the regulation by gut microbiota may pave the way for developing novel therapeutic strategies for AD. In this narrative review, we provide an overview of gut microbiota and their dysregulation in the pathogenesis of AD. Novel insights into the modification of gut microbiota composition as a preventive or therapeutic approach for AD are highlighted.
