Introduction: Renal cancer is one of the most common cancers in the world, but the effect of therapies on advanced renal cancer has not improved for decades. Ferroptosis is an emerging type of programmed cell death and has been proved to play a vital role in many kinds of cancers. However, the mechanisms of ferroptosis regulated by long noncoding RNA (lncRNA) in the context of renal cancer was still unknown. Methods: We used bioinformation analysis to identify SLC16A1-AS1 as a survival-related lncRNA in renal cancer. The expression levels of SLC16A1-AS1 and microRNA-143-3p (miR-143-3p) were detected by quantitative reverse transcription–polymerase chain reaction. Cell counting kit-8 assay, 5-bromo-2′-deoxyuridine proliferation assay, and colony-formation assay were performed to evaluate cell viability and proliferation. Wound-healing assay and transwell assay were used to examine cell invasive and migration capacity. Dual-luciferase reporter assay and RNA-binding protein immunoprecipitation were used to identify the interaction among SLC16A1-AS1, miR-143-3p, and the target protein solute carrier family 7 membrane 11 (SLC7A11). Reduced glutathione and glutathione and lipid peroxidation measurements were carried out to evaluate the level of ferroptosis, and the expression levels of ferroptosis-related proteins were analyzed by western blot. Results: Our study revealed that SLC16A1-AS1 has high expression and was associated with overall survival in renal cancer. Knockdown SLC16A1-AS1 inhibited cell viability, proliferation, and migration of renal cancer cells. Furthermore, it was demonstrated that SLC16A1-AS1 served as a sponge of miR-143-3p, and knockdown SLC16A1-AS1 significantly increased the enrichment of miR-143-3p. And then, SLC7A11 was identified as the target protein of miR-143-3p, and overexpression miR-143-3p remarkably inhibited the expression of SLC7A11. Moreover, knockdown SLC16A1-AS1 could aggravate this effect. Finally, through inhibiting SLC7A11 expression, silencing SLC16A1-AS1 induced ferroptosis via increasing miR-143-3p. Conclusion: The present results suggest that silencing lncRNA SLC16A1-AS1 can induce ferroptosis through miR-143-3p/SLC7A11 signaling in renal cancer. Our study provided a novel view into the pathogenesis and treatment strategy of RCC.

Effectiveness of electroacupuncture for the treatment of refractory overactive bladder disease in women: study protocol for a randomised controlled clinical trial

IntroductionOveractive bladder (OAB) is a common condition, affecting approximately one-fifth to one-sixth of the world’s population. The American Urological Association and the Society of Urodynamics, Female Pelvic Medicine and Genitourinary Reconstruction guidelines recommend sacral neuromodulation (SNM) as third-line treatment. Although SNM can relieve the symptoms of refractory OAB, several factors, including high cost and the high incidence of adverse effects, limit its large-scale clinical use. Moreover, patient acceptance is low. Acupuncture is an alternative approach for treating OAB that is minimally invasive and has few side effects. Clinical studies have shown that it can reduce the number of episodes of urination and urgent urination within 24 hours and improve the quality of life. However, clinical evidence on the effectiveness of electroacupuncture for treating refractory OAB is limited.The primary objective of this studyIs to assess the short-term effectiveness of electroacupuncture for improving the symptoms and quality of life of women with refractory OAB and to observe its long-term effects. A secondary objective is to assess whether electroacupuncture is more effective than conventional acupuncture for treating patients with refractory OAB.Methods and analysisThe study is designed as a two-arm, parallel, participant-blinded randomised controlled trial (RCT). A total of 60 women between the ages of 18 years and 60 years who are experiencing refractory OAB will be randomly assigned to two groups: one receiving electroacupuncture and the other receiving conventional acupuncture. Each participant will undergo a treatment regimen lasting 4 weeks. During the initial 2 weeks, participants will receive five sessions of either electroacupuncture or conventional acupuncture per week, followed by three sessions per week during the subsequent 2 weeks. In total, each individual will receive 16 treatment sessions throughout the course of the study. The primary outcome measure will be the change in OAB Symptom Score (OABSS) over time between groups. The secondary outcome measures will include the OABSS, the Patient-Perceived Bladder Symptom Grading Scale, the Overactive Bladder Questionnaire, the Self-Rating Anxiety Scale and the Self-Rating Depression Scale during the treatment and follow-up periods. Blinding and safety will also be assessed. Two-sided p values <0.05 will be considered statistically significant.Ethics and disseminationThe study protocol was approved by the Institutional Review Board of Beijing Fengtai Hospital of Integrated Traditional and Western Medicine on 5 February 2024 (Approval No. 2024020101). The results will be disseminated through presentations at scientific conferences and peer-reviewed publications.Trial registration numberChiCTR2400081448.

Ferroptosis is involved in the anti-tumor effect of lycorine in renal cell carcinoma cells

Renal cell carcinoma (RCC) is a most common malignant tumor in the genitourinary system. Studies have shown that Lycorine has promising anticancer activities with minor side effects. However, the effect of lycorine on the proliferation of RCC cells and its underlying anti-tumor mechanism have not yet been fully elucidated. The human renal cancer cell lines 786-O, A498 and Caki-1 were cultured and treated with different concentrations of lycorine or ferrostatin-1, a ferroptosis inhibitor. Cell viability and colony formation assays were used to measure cell proliferation. The 5-, 12- and 15-HETE hydroxyeicosatetraenoic acid (HETE) and MDA levels, as well as the reduced to oxidized glutathione (GHS/GSSG) ratio, were analyzed. Western blot analysis was used to detect the expression of glutathione peroxidase 4 (GPX4) and acyl-CoA synthetase long chain family member 4 (ACSL4), which are key markers of ferroptosis. Transmission electron microscopy was used to observe the morphological features associated with ferroptosis. Lycorine was found to inhibit the proliferation of RCC cells. After lycorine treatment, the expression levels of GPX4 in RCC cells decreased, whereas those of ACSL4 increased. Lycorine induced the expression of 5-HETE, 12-HETE, 15-HETE and MDA in RCC cells, and reduced the GSH/GSSG ratio. In addition, ferrostatin-1 could prevent lycorine-induced ferroptosis in RCC cells.

A novel compound heterozygous PCDH15 variants is associated with arRP in a Chinese pedigree

Retinitis pigmentosa (RP) is a heterogeneous group of inherited retinal diseases. However, it is still not well understand about the relationship between

variants and RP.
Methods
In this study, we enrolled a Chinese autosomal recessive retinitis pigmentosa (arRP) pedigree and identified the causative gene in the proband by targeted whole exome sequencing (WES). The variants were validated in the family members by Sanger sequencing and co-segregation analysis.
Results
Novel compound heterozygous, Frame shift variants of the
PCDH15
gene, NM_001384140.1:c.4368 − 2147_4368-2131del and NM_001384140.1:c exon19:c.2505del: p. T836Lfs*6 were identified in the arRP pedigree, which co-segregated with the clinical RP phenotypes. The PCDH15 protein is highly conserved among species.
Conclusion
This is the first study to identify novel compound heterozygous variants c.4368 − 2147_4368-2131del and c.2505del(p.T836Lfs*6) in the
PCDH15
gene which might be disease-causing variants, and extending the variant spectra. All above findings may be contribute to genetic counseling, molecular diagnosis and clinical management of arRP disease.

Colorectal Cancer Migration and Invasion Initiated by microRNA-106a

MicroRNAs have been implicated in the regulation of several cellular signaling pathways of colorectal cancer (CRC) cells. Although emerging evidence proves that microRNA (miR)-106a is expressed highly in primary tumor and stool samples of CRC patients; whether or not miR-106a mediates cancer metastasis is unknown. We show here that miR-106a is highly expressed in metastatic CRC cells, and regulates cancer cell migration and invasion positively in vitro and in vivo. These phenotypes do not involve confounding influences on cancer cell proliferation. MiR-106a inhibits the expression of transforming growth factor-β receptor 2 (TGFBR2), leading to increased CRC cell migration and invasion. Importantly, miR-106a expression levels in primary CRCs are correlated with clinical cancer progression. These observations indicate that miR-106a inhibits the anti-metastatic target directly and results in CRC cell migration and invasion.

Compressive performance of innovative reinforced pillars in closed/abandoned mines

Pillar is closely related to the stability and reliability of underground spaces in closed/abandoned mines. The present research introduced a new technique to strengthen square cement mortar columns via fiber-reinforced polymer (FRP) strips to verify the strengthening effect of FRP on pillars. Compared to a fully wrapped FRP jacket, the advantages of FRP strip are cost-effective and easy-to-construct. A series of compression tests as well as theoretical analysis were carried out to explore the mechanical behavior of square cement mortar specimens partially strengthened with FRP strips. The results verified the effectiveness of FRP strips in enhancing the stress and strain of cement mortar. Different from unconfined cement mortar specimens, these FRP-strengthened cement mortar specimens are featured with the double-peaked behaviors, mainly attributed to the stress state transformation from a one-dimensional to a three-dimensional stress state. It also indicated that the enhancement of stress increased with the FRP strip width. Moreover, the brittle-ductile transition ductile failure characteristics were also observed in FRP-confined cement mortar specimens. The ultimate ductility of the cement mortar specimen decreases gradually with the growth of the FRP strip width. The main contribution of this research is to enrich the strengthening techniques for residual pillars.

Variable Structure Guidance Law for Supersonic Missile to Maneuver in Large Airspace

The large airspace varying trajectory is modified to large airspace spiral maneuver trajectory, in order to improve the penetration effect of a supersonic anti-warship missile against enemy long-distance ship-to-air missile (LDSAM). In the descent phase the missile may suffer the interception of LDSAM, so the descent phase is modified to spiral maneuver descent trajectory. A variable structure guidance law is designed firstly. On the basis of it, a variable structure guidance law with spiral maneuver is proposed, in order to realize the large airspace spiral maneuver trajectory. The simulation results show that the proposed guidance law can guide the supersonic anti-warship missile to realize the large airspace spiral maneuver trajectory. The entire missiles performance indexes satisfy the flight requirement.

Dynamic Anti-Windup Design for Missile Overload Control System

The former research on the overload control system of a supersonic missile didn’t consider actuator saturation. In order to reduce the windup effect caused by actuator saturation, we developed a dynamic anti-windup compensator. The overload control system design adopted a two-step approach. Firstly a sliding mode controller was designed ignoring the saturation. Secondly a dynamic anti-windup compensator was designed. It added to the control system to reduce the windup effect. It disengaged the actuator fast from saturation phase after the actuator entered saturation. Simulation results showed that the dynamic anti-windup compensator reduced 52% of saturation time of the control system. Hence the performance of the overload control system is improved effectively.
