Intermediate soil acidification induces highest nitrous oxide emissions
2024
Global potent greenhouse gas nitrous oxide (N
2
O) emissions from soil are accelerating, with increases in the proportion of reactive nitrogen emitted as N
2
O, i.e., N
2
O emission factor (EF). Yet, the primary controls and underlying mechanisms of EFs remain unresolved. Based on two independent but complementary global syntheses, and three field studies determining effects of acidity on N
2
O EFs and soil denitrifying microorganisms, we show that soil pH predominantly controls N
2
O EFs and emissions by affecting the denitrifier community composition. Analysis of 5438 paired data points of N
2
O emission fluxes revealed a hump-shaped relationship between soil pH and EFs, with the highest EFs occurring in moderately acidic soils that favored N
2
O-producing over N
2
O-consuming microorganisms, and induced high N
2
O emissions. Our results illustrate that soil pH has a unimodal relationship with soil denitrifiers and EFs, and the net N
2
O emission depends on both the N
2
O/(N
2
O + N
2
) ratio and overall denitrification rate. These findings can inform strategies to predict and mitigate soil N
2
O emissions under future nitrogen input scenarios.
Intermediate soil acidification alters the denitrifier community composition and induces high nitrous oxide (N
2
O) emissions, which contributes to the observed acceleration of N
2
O emissions from global soils

Dynamic co-catalysis of Au single atoms and nanoporous Au for methane pyrolysis
Nanocatalysts and single-atom catalysts are both vital for heterogeneous catalysis. They are recognized as two different categories of catalysts. Nevertheless, recent theoretical works have indicated that Au nanoparticles/clusters release Au single atoms in CO oxidation, and they co-catalyze the oxidation. However, to date, neither experimental evidence for the co-catalysis nor direct observations on any heterogeneous catalysis process of single-atom catalysts are reported. Here, the dynamic process of nanoporous Au to catalyze methane pyrolysis is monitored by in situ transmission electron microscopy with high spatial–temporal resolutions. It demonstrates that nanoporous Au surfaces partially disintegrate, releasing Au single atoms. As demonstrated by DFT calculation, the single atoms could co-catalyze the reaction with nanoporous Au. Moreover, the single atoms dynamically aggregate into nanoparticles, which re-disintegrate back to single atoms. This work manifests that under certain conditions, the heterogeneous catalysis processes of nanocatalysts and single-atom catalysts are not independent, where their dynamic co-catalysis exists.
Nanocatalysts and single‐atom catalysts are generally considered as two categories with distinct performances. Here, in situ TEM study of catalytic methane pyrolysis over nanoporous Au reveals a highly dynamic process where co‐catalysis exists among various catalyst forms.

Background
Our study aim was to evaluate the therapeutic efficacy and mechanisms of miR-133-overexpressing mesenchymal stem cells (MSCs) on acute myocardial infarction.
Methods
Rat MSCs were isolated and purified by whole bone marrow adherent culturing. After transfection with the agomir or antagomir of miR-133, MSCs were collected for assay of cell vitality, apoptosis, and cell cycle progression. At the same time, exosomes were isolated from the supernatant to analyze the paracrine miR-133. For in-vivo studies, constitutive activation of miR-133 in MSCs was achieved by lentivirus-mediated miR-133 overexpression. A rat myocardial infarction model was created by ligating the left anterior descending coronary artery, while control MSCs (vector-MSCs) or miR-133-overexpressed MSCs (miR-133-MSCs) were injected into the zone around the myocardial infarction. Subsequently, myocardial function was evaluated by echocardiography on days 7 and 28 post infarction. Finally the infarcted hearts were collected on days 7 and 28 for myocardial infarct size measurement and detection of
snail 1
expression.
Results
Hypoxia-induced apoptosis of MSCs obviously reduced, along with enhanced expression of total poly ADP-ribose polymerase protein, after miR-133 agomir transfection, while the apoptosis rate increased in MSCs transfected with miR-133 antagomir. However, no change in cell viability and cell-cycle distribution was observed in control, miR-133-overexpressed, and miR-133-interfered MSCs. Importantly, rats transplanted with miR-133-MSCs displayed more improved cardiac function after acute myocardial infarction, compared with those that received vector-MSC injection. Further studies indicated that cardiac expression of
snail 1
was significantly repressed by adjacent miR-133-overexpressing MSCs, and both the inflammatory level and the infarct size decreased in miR-133-MSC-injected rat hearts.
Conclusions
miR-133-MSCs obviously improved cardiac function in a rat model of myocardial infarction. Transplantation of miR-133-overexpressing MSCs provides an effective strategy for cardiac repair and modulation of cardiac-related diseases.

Combination of Fruit and Vegetable Storage and Fresh-Keeping with Postharvest Heat Treatment
2022
Hot water treatment can effectively improve the storage quality and shelf life of harvested vegetables. The essence of this method is the heat transfer process in which heat is transferred from the high-temperature treatment medium to the low-temperature fruit and vegetable tissues. However, most of the current research projects only focus on the effect of heat treatment on the biological aspects of fruits and vegetables. From the perspective of fruit and vegetable storage and preservation, there are few internal mechanisms that heat treatment affects biological preservation, and most of the research projects only focus on continuous heat treatment. This article aims to study the combination of storage and preservation of fruits and vegetables with postharvest processing technology. To this end, this article proposes the combined use of heat treatment technology through the analysis and improvement of data acquisition problems in the heat treatment process to make the data obtained more reliable and, at the same time, design experiments to explore the improved heat treatment technology. The experimental results in this paper show that the improved storage and preservation of fruits and vegetables combined with postharvest heat treatment technology improves the preservation of fruits and vegetables by 37%. In addition, the overall taste of fruits and vegetables during the preservation process has not been greatly lost, only reduced by 7%, which can be well applied to actual fruit and vegetable processing.

Azilsartan attenuates lipopolysaccharide-induced acute lung injury via the Nrf2/HO-1 signaling pathway
2022
Acute lung injury (ALI) is a severe complication of sepsis and hemorrhagic shock with high morbidity. In the present study, the protective effect of Azilsartan on lipopolysaccharide (LPS)-induced ALI in mice was investigated to explore the potential therapeutic property of Azilsartan for the treatment of ALI. LPS was used to induce an ALI model in mice. Hematoxylin–eosin (HE) staining sections were then evaluated for the pathological state of lung tissues. Bronchoalveolar lavage fluid (BALF) protein concentration, wet/dry weight ratios of lung tissues, and pulmonary myeloperoxidase (MPO) activity were detected to determine the degree of pulmonary injury. The number of total cells, macrophages, and neutrophils in BALF were counted using a hemocytometer to illustrate the inflammatory cell infiltration. The lung function was monitored using a spirometer. The concentrations of interleukin-1β (IL-1β), monocyte chemoattractant protein-1 (MCP-1), and interleukin-8 (IL-8) were determined using enzyme-linked immunosorbent assay (ELISA). Oxidative stress was evaluated by the superoxide dismutase (SOD) activity, glutathione (GSH), and malondialdehyde (MDA) concentrations in the lung tissue. The expressions of nuclear erythroid 2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) were determined using Western blot analysis. Azilsartan therapy alleviated LPS-induced lung tissue damage, increased BALF protein concentration, lung wet to dry weight ratio, MPO activity, and macrophage and neutrophils infiltration. Also, Azilsartan ameliorated the production of inflammatory factors (IL-1β, MCP-1, and IL-8). Azilsartan ameliorated LPS-impaired lung SOD activity, the GSH concentration, and the MDA concentration. Mechanistically, Azilsartan activated the LPS-impaired Nrf2/HO-1 signaling pathway. Azilsartan therapy attenuates LPS-induced ALI via the Nrf2/HO-1 signaling pathway.

Improved exfoliation of surface-functionalized graphene oxide by epoxy monomer and enhanced mechanical properties of epoxy nanocomposites
2022
The surface functionalization of graphene oxide (GO) provides an efficient way to improve the dispersion of GO in matrix and the interfacial properties of nanocomposites. In this contribution, GO grafted by bifunctional, trifunctional and tetrafunctional epoxy monomers (E51, AFG90, AG80) was designed and three epoxy-functionalized GO (EFGO) samples were acquired and utilized as fillers for epoxy nanocomposites. The chemical, structural and morphological characteristics of EFGO samples were studied. The exfoliation and dispersion state of EFGO in epoxy matrix were characterized. Mechanical properties of the nanocomposites at room temperature (RT), liquid nitrogen temperature (LNT) and after thermal cycling were comprehensively investigated. Results showed that the interlayer distance of EFGO was enlarged with the increase in functionality degree of the grafted epoxy monomers. AG80-functionalized GO (AGGO) with the largest interlayer distance showed the best exfoliation and dispersion in the nanocomposites, while E51-functionalized GO (EGO) showed the worst. Correspondingly, AGGO/epoxy nanocomposites exhibited the highest mechanical properties at RT, LNT and after thermal cycling. The toughening mechanisms of EFGO for epoxy resin were disclosed to better understand the role of epoxy monomers grafted to GO surfaces in the nanocomposites.

Risk Scores in Venous Thromboembolism Guidelines of ESC, ACCP, and ASH: An Updated Review
2024
Venous thromboembolism (VTE) is associated with high morbidity and mortality. Risk scores associated with VTE have been widely used in clinical practice. Among numerous scores published, those included in guidelines are usually typical risk scores which have been extensively validated and globally recognized. This review provides an updated overview of the risk scores associated with VTE endorsed by 3 guidelines which are highly recognized in the field of VTE including the European Society of Cardiology, American College of Chest Physicians, and American Society of Hematology, focusing on the development, modification, validation, and comparison of these scores, to provide a comprehensive and updated understanding of all the classic risk scores associated with VTE to medical readers including but not limited to cardiologists, pulmonologists, hematologists, intensivists, physicians, surgeons, and researchers. Although each score recommended by these guidelines was more or less validated, there may still be room for further improvement. It may still be necessary to seek simpler, more practical, and more universally applicable VTE-related risk scores in the future.

Selective Adsorption-Based Separation of Flue Gas and Natural Gas in Zirconium Metal-Organic Frameworks Nanocrystals
2019
Carbon capture from flue gas and natural gas offers a green path to construct a net-zero emissions economic system. Selective adsorption-based gas separation by employing metal-organic frameworks (MOFs) is regarded as a promising technology due to the advantages of simple processing, easy regeneration and high efficiency. We synthesized two Zirconium MOFs (UiO-66 and UiO-66-NH2) nanocrystals for selective capture and further removal of CO2 from flue gas and natural gas. In particular, UiO-66-NH2 nanocrystals have a smaller grain size, a large amount of defects, and pending –NH2 groups inside their pores which display effective CO2 selective adsorption abilities over CH4 and N2 with the theoretical separation factors of 20 and 7. This breakthrough experiment further verified the selective adsorption-based separation process of natural gas and flue gas. In one further step, we used the Monte Carlo simulation to investigate the optimized adsorption sites and energy of CO2, N2 and CH4 molecules in the gas mixture. The significantly large adsorption energy of CO2 (0.32 eV) over N2 (0.19 eV) and N2 (0.2 eV) may help us to reveal the selective adsorption mechanism.

A fast-linear mixed model for genome-wide haplotype association analysis: application to agronomic traits in maize
2020
Background
Haplotypes combine the effects of several single nucleotide polymorphisms (SNPs) with high linkage disequilibrium, which benefit the genome-wide association analysis (GWAS). In the haplotype association analysis, both haplotype alleles and blocks are tested. Haplotype alleles can be inferred with the same statistics as SNPs in the linear mixed model, while blocks require the formulation of unified statistics to fit different genetic units, such as SNPs, haplotypes, and copy number variations.
Results
Based on the FaST-LMM, the fastLmPure function in the R/RcppArmadillo package has been introduced to speed up genome-wide regression scans by a re-weighted least square estimation. When large or highly significant blocks are tested based on EMMAX, the genome-wide haplotype association analysis takes only one to two rounds of genome-wide regression scans. With a genomic dataset of 541,595 SNPs from 513 maize inbred lines, 90,770 haplotype blocks were constructed across the whole genome, and three types of markers (SNPs, haplotype alleles, and haplotype blocks) were genome-widely associated with 17 agronomic traits in maize using the software developed here.
Conclusions
Two SNPs were identified for LNAE, four haplotype alleles for TMAL, LNAE, CD, and DTH, and only three blocks reached the significant level for TMAL, CD, and KNPR. Compared to the R/lm function, the computational time was reduced by ~ 10–15 times.

Shikonin, a small-molecule natural product which inhibits the activity of pyruvate kinase M2 (PKM2), has been studied as an anti-cancer drug candidate in human cancer models. Here, our results demonstrate that shikonin is able to sensitize human breast cancer cells to chemotherapy by paclitaxel (taxol). Human breast adenocarcinoma MBA-MD-231 cells, which have higher levels of PKM2 expression and activity compared with MCF-7 cells, were selected to study further. The concentrations of shikonin and taxol were first selected at which they did not significantly induce cytotoxicity when treated alone, whereas the combination induced apoptosis. Surprisingly, PKM2 activity was decreased by shikonin, but not by the combination treatment. To identify the potential targets of this combination, human phospho-kinase antibody array analysis was performed and results indicated that the combination treatment inhibited the activation of ERK, Akt, and p70S6 kinases, which are known to contribute to breast cancer progression. Finally, how the combination affects breast cancer cell growth in vivo was tested using a xenograft tumor model. The results indicated that shikonin plus taxol prolonged animal survival and reduced tumor size than the vehicle treatment group. In summary, our results suggest that shikonin has a potential as an adjuvant for breast cancer therapy.
