Analysis of metabolites change from reflorescence buds of ‘Cuiguan’ pear (Pyrus pyrifolia) based on LC-MS/MS

Developed buds of pears ( Pyrus pyrifolia ) remain in a dormant state and do not break dormancy until the following spring. However, climatic factors and leaf diseases trigger defoliation in late summer and early autumn, which typically results in the bud paradormancy release and subsequent sprouting. This not only depletes tree nutrients but also diminishes the quantity of flowers and the yield of fruit in the following summer. In this study, metabolic changes in ‘Cuiguan’ pear buds were investigated following premature leaf fall. A total of 1,533 metabolites were annotated, with the majority being downregulated. Sugar levels decreased during the release of paradormancy, likely to provide energy for subsequent growth. Concurrently, most amino acids were consumed post-defoliation, with only a few showing increased trends. Furthermore, the observed low levels of phenylpropanoid-related metabolites in flower buds may contribute to premature senescence. This metabolic profile contributes to our understanding of the biological mechanism of paradormancy release from defoliation, and clarifies the metabolic changes during this process.

Concentrations and Exposure Evaluation of Metals in Diverse Food Items from Chengdu, China
A total of 520 food samples belonging to 29 food types and 63 drinking water were collected in Chengdu market of China in 2014 to investigate the concentrations of 11 metals, and to assess the related exposure to the local consumers by estimating the hazard quotient and carcinogenic risk (CR). The results showed that metals concentrations in drinking water were below the limit values suggested by the Ministry of Health of the People’s Republic of China, and FAO/WHO (Food and Agriculture Organization of the United Nations/World Health Organization). While As, Cd, and Cr were found at concentrations higher than the limit values in some of the foodstuffs. Children in Chengdu intake more metals compared to adults, with the same order of Mn > Zn > Cu > Sr > Cr > Ni > As > Cd > Pb > Co > Sb. Among all of the diverse food, rice, flour, and fish and seafood were the primary sources to intake metals for Chengdu residents. Residents in Chengdu are subjected to both carcinogenic and non-carcinogenic risks based on the calculated HI and CR values, especially for children. Finally, total daily metals intakes for both children and adults were calculated based on the current study and our previous studies, including consumption of food and drinking water and intake of outdoor and indoor dust. Dietary exposure is the predominant exposure route to metals for Chengdu residents, accounting for more than 75.8% of the total daily metals intakes for both children and adults.

Significant delay and decreased chance of treatment for acute ischemic stroke patients on remote outer islets of China compared with the main Island: The Putuo study

Introduction: Data from acute ischemic stroke patients throughout 2021 from one district of an archipelago city of China were collected and analyzed retrospectively to determine the management difference due to time lags from onset of symptoms to the arrival at the stroke center (FMCT) of two regions: main island (MI) and outer islets (OIs).
Methods: All patients information from 1 January to 31 December 2021 was retrieved through the electronic medical records system of the only stroke center in MI. After screening and exclusion, each patient's medical record was reviewed by two neurologists separately. Before OI patients were allocated to a group, their residential addresses at onset of the stroke were confirmed by telephone. Comparisons were analyzed between the two regions for gender, age, pre-stroke risk factors and peri-admission management parameters.
Results: A total of 326 patients met the inclusion criteria: 300 from the MI group and 26 for the OI group. Intergroup comparisons for gender, age and most of the risk factors showed no significant difference. FMCT were shown to be significantly distinct ('p'<0.001). Hospitalization expenses also showed significant difference. The odds ratio of the definite treatment IV thrombolysis was 0.131 (OI group to MI group range: 0.017-0.987, 'p'=0.021).
Conclusion: The diagnosis and treatment of acute ischemic stroke patients from OIs was significantly postponed compared to those from MI. Therefore, new effective and efficient solutions are urgently needed.

Evaluation of a Multilevel Program to Improve Clinician Adherence to Management Guidelines for Acute Ischemic Stroke

Promotion of clinician adherence to stroke guidelines can improve stroke outcomes.
To investigate the outcomes of a multilevel system program on clinician adherence to guidelines for treatment of patients with acute ischemic stroke (AIS).
This quality improvement study used a prospective interrupted time series (ITS) and difference-in-difference (DID) design, from August 1, 2018, to January 31, 2020, divided into preprogram term and short and long postprogram terms; each term had 6 months. Data were collected during hospitalization and at discharge with an automated medical record data capture system in 58 public hospitals in Zhejiang province, China. Data were analyzed from August 2018 to January 2020.
The multilevel system program included a modularized standard template for medical records, centrally supported continuing education, continuous monitoring and feedback, and collaborative workshops.
The primary outcome was adherence to 12 key performance indicators (KPIs), expressed as (1) percentage of patient-applicable KPIs achieved in each participant and (2) percentage of participants among whom all applicable KPIs were achieved (dichotomous all-or-none measure). The secondary outcome was severe disability or death (modified Rankin Scale 5-6) at discharge.
Among 45 091 patients (mean [SD] age, 69 [12] years; 18 347 female [40.7%]), 28 721 from 30 hospitals received the program and 16 370 from 28 hospitals continued routine care. In adjusted DID analysis, the program was associated with an increase in the absolute percentage of KPIs achieved per patient (6.46%; 95% CI, 5.49% to 7.43%), absolute rate of all-or-none success (8.29%; 95% CI, 6.99% to 9.60%), and decreased rate of severe disability or death at discharge (-1.68%; 95% CI, -2.99% to -0.38%). The ITS result showed the program was associated with an increase in KPIs achieved per patient per week (slope change in short-term period, 0.36%; 95% CI, 0.20% to 0.52%; level change in long-term period, (9.64%; 95% CI, 4.58% to 14.69%) and in all-or-none success (slope change in short-term period 0.34%; 95% CI, 0.23% to 0.46%; level change in long-term period 5.89%; 95% CI, 0.19% to 11.59%).
The centrally supported program was associated with increases in clinician adherence to guidelines and reduced the proportion of severely disabled or deceased patients with AIS at discharge, providing support for its wider implementation.

The digital economy (DE) has become a major breakthrough in promoting industrial upgrading and an important engine for high-quality economic growth. However, most studies have neglected the important driving effect of regional economic and social (RES) development on DE. In this paper, we discuss the mechanism of RES development promoting the development of DE, and establish a demand-driven regional DE development model to express the general idea. With the help of spatial analysis toolbox in ArcGIS software, the spatial development characteristics of DE in the Yangtze River Delta City Cluster (YRDCC) is explored. We find the imbalance of spatial development is very significant in YRDCC, no matter at the provincial level or city level. Quantitative analysis reveals that less than 1% likelihood that the imbalanced or clustered pattern of DE development in YRDCC could be the result of random chance. Geographically weighted regression (GWR) analysis with publicly available dataset of YRDCC indicates RES development significantly promotes the development of DE.

Background
Cardiovascular diseases are currently the leading cause of death in humans. The high mortality of cardiac diseases is associated with myocardial ischemia and reperfusion (I/R). Recent studies have reported that microRNAs (miRNAs) play important roles in cell apoptosis. However, it is not known yet whether miR-141-3p contributes to the regulation of cardiomyocyte apoptosis. It has been well established that in vitro hypoxia/reoxygenation (H/R) model can follow in vivo myocardial I/R injury. This study aimed to investigate the effects of miR-141-3p and CHD8 on cardiomyocyte apoptosis following H/R.
Results
We found that H/R remarkably reduces the expression of miR-141-3p but enhances CHD8 expression both in mRNA and protein in H9c2 cardiomyocytes. We also found either overexpression of miR-141-3p by transfection of miR-141-3p mimics or inhibition of CHD8 by transfection of small interfering RNA (siRNA) significantly decrease cardiomyocyte apoptosis induced by H/R. Moreover, miR-141-3p interacts with CHD8. Furthermore, miR-141-3p and CHD8 reduce the expression of p21.
Conclusion
MiR-141-3p and CHD8 play critical roles in cardiomyocyte apoptosis induced by H/R. These studies suggest that miR-141-3p and CHD8 mediated cardiomyocyte apoptosis may offer a novel therapeutic strategy against myocardial I/R injury-induced cardiovascular diseases.

Critical role of Tim-3 mediated autophagy in chronic stress induced immunosuppression

Background
Psychological and physical stress can either enhance or suppress immune functions depending on a variety of factors such as duration and severity of stressful situation. Chronic stress exerts a significantly suppressive effect on immune functions. However, the mechanisms responsible for this phenomenon remain to be elucidated. Autophagy plays an essential role in modulating cellular homeostasis and immune responses. However, it is not known yet whether autophagy contributes to chronic stress-induced immunosuppression. T cell immunoglobulin and mucin domain 3 (Tim-3) has shown immune-suppressive effects and obviously positive regulation on cell apoptosis. Tim-3 combines with Tim-3 ligand galectin-9 to modulate apoptosis. However, its impact on autophagy and chronic stress-induced immunosuppression is not yet identified.
Results
We found remarkably higher autophagy level in the spleens of mice that were subjected to chronic restraint stress compared with the control group. We also found that inhibition of autophagy by the autophagy inhibitor 3-methyladenine (3-MA) significantly attenuated chronic stress-induced alterations of pro-inflammatory and anti-inflammatory cytokine levels. We further elucidated that 3-MA dramatically inhibited the reduction of lymphocyte numbers. Moreover, chronic stress dramatically enhanced the expression of Tim-3 and galectin-9. Inhibition of Tim-3 by small interfering RNA against Tim-3 significantly decreased the level of autophagy and immune suppression in isolated primary splenocytes from stressed mice. In addition, α-lactose, a blocker for the interaction of Tim-3 and galectin-9, also decreased the autophagy level and immune suppression.
Conclusion
Chronic stress induces autophagy, resulting with suppression of immune system. Tim-3 and galectin-9 play a crucial regulatory role in chronic stress-induced autophagy. These studies suggest that Tim-3 mediated autophagy may offer a novel therapeutic strategy against the deleterious effects of chronic stress on the immune system.

Preparation and Characterization of Chinese Leek Extract Incorporated Cellulose Composite Films

This study aimed to prepare microcrystalline cellulose (MCC) films with good mechanical properties via plasticization using a Chinese leek (CL, Allium tuberosum ) extract. The microstructure, crystal structure, mechanical properties, barrier ability, and thermal properties of the films were investigated. The chemical structure analysis of CL extract showed the existence of cellulose, lignin, and low-molecular-weight substances, such as polysaccharides, pectins, and waxes, which could act as plasticizers to enhance the properties of MCC:CL biocomposite films. The results of scanning electron microscopy and atomic force microscopy analyses indicated the good compatibility between MCC and CL extract. When the volume ratio of MCC:CL was 7:3, the MCC:CL biocomposite film exhibited the best comprehensive performance in terms of water vapor permeability (2.11 × 10 –10 g/m·s·Pa), elongation at break (13.2 ± 1.8%), and tensile strength (24.7 ± 2.5 MPa). The results of a UV absorption analysis demonstrated that the addition of CL extract improved the UV-shielding performance of the films. Therefore, this work not only proposes a facile method to prepare MCC films with excellent mechanical properties via plasticization using CL extract but also broadens the potential applications of MCC films in the packaging area.
