In vitro conservation of Amburana cearensis (Fabaceae)
Slow-growth tissue culture allows in vitro conservation of plant genetic resources and is a complementary technique to conventional preservation methods. The objective of this study was to evaluate the effect of the culture medium supplemented with sucrose, mannitol and sorbitol osmotic agents on the induction of in vitro slow growth of A. cearensis, seeking to establish alternative techniques for ex situ conservation of the species. Plants with age of 10 days were inoculated in woody plant medium (WPM) supplemented with different concentrations of sucrose (87.64, 131.46, 175.28 or 262.92 mM) combined with mannitol or sorbitol at varying concentrations (0.0, 43.8, 65.73, 87.64 or 131.46 mM), solidified with 0.7% agar. The following parameters were evaluated: survival percentage, number of senescent leaves, aerial part length, root length, aerial part dry mass, root dry mass; and number of shoots. The conservation of A. cearensis was viable for up to 300 days in WPM supplemented with 262.92 mM of sucrose or the combination of 87.64 mM of sucrose with 43.80 mM of mannitol, because besides achieving a survival percentage greater than 55.0%, the plants conserved in these media presented normal development and the best appearances, with predominance of uniform green leaves during the storage process.
Morpho-anatomical characterization, gene expression and protein cell wall modifications associated with natural finger drop in bananas
Banana (Musa spp.) is one of the main fruits consumed worldwide. However, finger drop, is a physiological disorder that causes many postharvest problems, which eventually reduces market value and consumer acceptance. Therefore, the objective of the study was to evaluate the anatomical changes that occur in the pedicel rupture area (drop zone) of bananas diploids (BB França) and tetraploid (BRS Pioneira) in three ripening stages. The levels of gene expression involved in the natural ripening process and in the development of finger drop, was also investigated. The accumulation of their mRNAs and those of expansin (EXP1), pectate lyase (PEL1) and xyloglucan endotransglucosylase/hydrolase protein (XTH4) genes already isolated from bananas were measured by quantitative polymerase chain reaction in three ripening stages. BB França presented a higher resistance to finger drop due to the presence of some specific morphoanatomical characteristics, such as larger parenchymal cells and greater deposition of lignin. In contrast, there was degeneration of the pedicel parenchymal tissue of the BRS Pioneira genotype, forming large empty spaces during the ripening of the fruits, mainly in stage 6, which contributed to the finger drop. The diploid BB França  is a strong candidate for use in  banana breeding programs aimed at fruit drop resistance. This will certainly improve the quality of banana varieties. Moreover, PEL1 proved to be an excellent candidate gene for functional studies of finger drop in bananas.
Ex situ conservation of the endangered “sempre-viva” species Comanthera mucugensis
The “sempre-viva” species Comanthera mucugensis is endemic to the municipality of Mucugê, Bahia, Brazil, where it was widely exploited through extractivism to be commercialized as an ornamental, causing a drastic reduction of its population, so that it is now classified as endangered. Although its main area of occurrence is now protected for being within Chapada Diamantina National Park, the continued risks of inclement conditions and anthropic actions make it necessary to develop alternative methods for ex situ conservation of the species, such as in vitro conservation. Therefore, the objective of this study was to test the effect of different concentrations of salts and sucrose on the in vitro conservation of Comanthera mucugensis. Two salts concentrations of the medium MS (½ and ¼) and two sucrose levels (7.5 and 15.0 g L-1) were tested, and the experimental design was completely randomized with four treatments. After 365 days, the survival, growth and regeneration of the conserved plants were analyzed, achieving up to 100% survival, reduced shoot growth and maintenance of regenerative capacity. Reduction of the concentration of salts and sucrose in the culture medium is indicated to conserve the plants in vitro for a period of one year.
IN VITRO SEED GERMINATION AND PLANT GROWTH OF “CABEÇA-DE-FRADE” (CACTACEAE)
ABSTRACT The genus Melocactus (“cabeça-de-frade”) comprises 32 species in Brazil, of which M. glaucescens and M. paucispinus are threatened with extinction. The present work evaluated the effects of different concentrations of Murashige & Skoog (MS, MS/2 and MS/4) culture medium and sucrose (15 g L-1 and 30 g L-1) on in vitro seed germination and plant growth of M. glaucescens and the efficiency of sterilization with sodium hypochlorite (NaOCl) on in vitro seed germination and plant growth of M. glaucescens when using seeds and M. glaucescens and M. paucispinus when using apical segment of cladode. In M. glaucescens, the final germination at the different MS and sucrose concentrations varied between 53.5 and 68.1% and the best results for in vitro growth were observed with the lowest mineral salt (MS/2 and MS/4) and sucrose (15 g L-1) concentrations, with lengths of the aerial portion of 9.70 and 10.76 mm, respectively. There was no difference in seed germination and plant growth in chemical and autoclave medium. It is concluded that the use of chemical sterilization with NaOCl at low concentrations of salts (MS/2 and MS/4) and sucrose (15 g L-1) are quite advantageous for producing ornamental plants germinated in vitro and/or apical segment of cladode of M. glaucescens andM. paucispinus, representing a reduction of costs for in vitro cultivation of this species. RESUMO O gênero Melocactus (cabeça-de-frade) possui 32 espécies no Brasil, das quais M. glaucescens e M. paucispinus estão ameaçadas de extinção. O objetivo deste trabalho foi analisar o efeito de diferentes concentrações de sais do meio Murashige e Skoog (MS, MS/2 e MS/4) e sacarose (15 g L-1 e 30 g L-1) na germinação de sementes e crescimento de plantas in vitro de M. glaucescens e a eficiência da esterilização do meio de cultura com hipoclorito de sódio (NaOCl) na germinação de sementes e crescimento de plantas in vitro de M. glaucescens ao utilizar sementes, M. glaucescens e M. paucispinus ao utilizar o segmento apical do cladódio. Para M. glaucescens a germinação final nas diferentes concentrações de MS e sacarose, variou de 53,5 a 68.1% e os melhores resultados para o crescimento in vitro foram observados nas menores concentrações de sais (MS/2 e MS/4) e de sacarose (15 g L-1) com valores de comprimento da parte aérea de 9,70 e 10,76 mm, respectivamente. Não houve diferença na germinação das sementes e crescimento das plantas em meio químico e autoclavado. Conclui-se que, o uso da esterilização química com NaOCl em baixas concentrações de sais (MS/2 e MS/4) e sacarose (15 g L-1) são as mais indicadas para produção de plantas ornamentais germinadas in vitro e /ou segmento apical do cladódio de M. glaucescens e M. paucispinus, o que representa uma redução de custo no cultivo in vitro dessas espécies.
Micropropagation of Chapada Diamantina ornamental bromeliad
Sincoraea mucugensis (Wand. & A.A. Conc.) LOUZADA & WAND, an endangered bromeliad, is confined to the central region of the Chapada Diamantina, in the municipality of Mucugê, Brazil. From various researches, it is evident that for the propagation of this species, the in vitro technique is a feasible option. However, due to the low multiplication rates reported in various papers, this study aimed to establish a micropropagation protocol of direct organogenesis for S. mucugensis. First, the inoculation of the stem explants was done in MS ½ culture medium which contained different levels of BAP (0.00; 6.66; 8.88; 11.10; 13.20 µM) and NAA (0.00; 2.60; 5.20 µM). These shoots were then subjected to a couple of distinct rooting periods (of 30- and 60-day duration) using activated charcoal; finally, these microplants were transferred to a greenhouse for acclimatization, and covered with transparent plastic cups, as a water loss prevention test method. All the data were submitted to the analysis of variance (ANOVA), and the means were subjected to regression analysis or compared using the Tukey test. The findings revealed that the S. mucugensis stem explants raised in the NAA-rich medium (6.42 to 7.43 shoots/explants) showed high multiplication rates; the shoot rooting was done for 30 days using activated charcoal with the medium. Acclimatization, which was performed by directly exposing the microplants to the ex vitro environment, showed 95% survival rate. RESUMO: Sincoraea mucugensis (Wand. & A.A. Conc.) LOUZADA & WAND é uma bromélia vulnerável de ocorrência restrita ao município de Mucugê, Chapada Diamantina. Estudos indicam que a cultura de tecidos é uma alternativa viável para a propagação in vitro desta espécie. Contudo, em função das baixas taxas de multiplicação obtidas em estudos anteriores, objetivou-se estabelecer um protocolo de micropropagação via organogênese direta para S. mucugensis. Explantes caulinares foram inoculados em meio de cultura MS ½ contendo diferentes concentrações de BAP (0,00; 6,66; 8,88; 11,10; 13,20 µM) e ANA (0,00; 2,60; 5,20 µM). Os brotos obtidos foram submetidos a diferentes períodos de enraizamento (30 e 60 dias) com carvão ativado; posteriormente as microplantas foram aclimatizadas em casa de vegetação, testando-se o efeito da cobertura com copos plásticos transparentes como estratégias contra perda de água. Os dados foram submetidos à análise de variância (ANOVA) e as médias analisadas por regressão ou comparadas pelo Teste de Tukey. Os resultados demonstram que altas taxas de multiplicação de S. mucugensis são geradas a partir de explantes caulinares cultivados em meio contendo ANA (6,42 a 7,43 brotos/explantes); o enraizamento dos brotos é realizado por 30 dias em meio com carvão ativado e a aclimatização é feita com exposição direta das microplantas ao ambiente ex vitro com 95% de sobrevivência.
Resposta morfogênica de embriões zigóticos de Erythrina velutina Willd: (Leguminosae) cultivados in vitro
O cultivo in vitro de embriões zigóticos é uma técnica promissora para se avançar no estudo do desenvolvimento embrionário e da quebra da dormência de sementes. Diante do exposto, objetivou-se, com este trabalho, avaliar o efeito dos reguladores vegetais 6-benzilaminopurina (BAP) e ácido naftalenoacético (ANA) no potencial morfogenético, in vitro, de embriões zigóticos de mulungu. Embriões zigóticos maduros, oriundos de sementes foram utilizados inteiros, ou seccionados em plúmula, região intermediária e radícula, sendo posteriormente inoculados em meio de cultura WPM, suplementado com combinações de BAP (0,0; 2,0; 4,0; 8,0; 12,0 e 16,0 µM) e ANA (0,0; 1,0 e 2,0 µM), acrescido de 87,64 mM de sacarose e solidificado com 0,7% de ágar. Após 30 dias, avaliaram-se a percentagem de regeneração dos embriões e ápice plumular, o número de brotos, o número de folhas, o comprimento da parte aérea dos brotos, o número de raízes e a percentagem de formação de calos oriundos da região intermediária e da radícula. É possível a regeneração in vitro de mulungu, a partir dos explantes plúmula e embriões zigóticos inteiros, cultivados em meio de cultura WPM, suplementado com 4,0 µM de BAP. Regiões intermediárias e da radícula promoveram a formação de calos compactos (96,06%), na combinação de 10,63 µM BAP e 2,0 µM de ANA.
In vitro establishment of Comanthera curralensis, “sempre viva” native of Chapada Diamantina - Bahia
The goal of the present study was to evaluate the germination, initial growth, and in vitro co-cultivation of Comanthera curralensis Moldenke, a \"sempre viva\" native of the Chapada Diamantina state of Bahia. Full strength (MS) and half-strength MS (MS1/2) growth media supplemented with two different sucrose concentrations (15 and 30g L-1) were tested for germination and initial plant growth. Three different plant densities were tested by in vitro culture (8, 10 and 12 plants per container). MS1/2 medium with 15g L-1 sucrose resulted in a higher percentage of germination and plant growth for the in vitro establishment of C. curralensis. The use of 12 plants per container is indicated for cost reduction in C. curralensis in vitro production. RESUMO: Este trabalho teve como objetivo avaliar a germinação, o crescimento inicial e o co-cultivo in vitro de Comanthera curralensis Moldenke, uma \"sempre viva\" nativa da Chapada Diamantina-BA. Para germinação e crescimento inicial, foram testados os meios de cultura MS completo e MS1/2 suplementados com duas concentrações de sacarose (15 e 30gL-1); no cultivo in vitro, foram testadas três quantidades de plantas por recipiente (8,10 e 12). A utilização do meio MS1/2 com 15gL-1 de sacarose proporcionou maiores porcentagem de germinação crescimento das plantas no estabelecimento in vitro de C. curralensis , e o uso de 12 plantas por recipiente é indicado para a redução de custos na produção in vitro da espécie.
Modulation of culture medium on the ex situ conservation of Neoregelia mucugensis Leme (Bromeliaceae)
ABSTRACT Bromeliads are the target of predatory extractivism and consequently many species are included in the red list of threatened species, such as those belonging to the genus Neoregelia. Although Neoregelia mucugensis has not been evaluated for the degree of threat, its exploitation is exclusively extractive and its occurrence in Chapada Diamantina-BA is subject to the action of fires that affect the region annually. In this context, applying techniques aimed at protecting this genetic resource is fundamental for both the maintenance of its natural populations and the ex situ conservation of this genetic material. Plant tissue culture techniques have been successfully applied for the conservation of several bromeliad species. One of the methods used is slow growth, which consists in reducing plant metabolism and consequently decelerating its growth, which allows the maintenance of in vitro plant collections without the need for subculture. In this context, the objective of this study was to test the reduction of salts in the culture medium and the addition of osmoregulators on the induction of slow growth of N. mucugensis. Plants were subjected to treatments composed of different concentrations of MS medium and mannitol for a period of 12 months, when then analyses were conducted to evaluate growth, chlorophyll content and regeneration capacity of shoots in vitro. It was found that the treatment containing MS ½ and 7.8 g.L-1 of mannitol is indicated for in vitro conservation of N. mucugensis with maintenance of the regenerative capacity of its tissues. RESUMO As bromélias são alvo de extrativismo predatório e com isso muitas estão inseridas na lista vermelha de espécies ameaçadas, a exemplo das que pertencem ao gênero Neoregelia. Apesar da Neoregelia mucugensis não ter sido avaliada quanto ao grau de ameaça, sua exploração é exclusivamente extrativista e sua ocorrência na Chapada Diamantina-BA está sujeita à ação das queimadas que afetam a região anualmente. Neste contexto, a aplicação de técnicas que visem proteger este recurso genético é fundamental tanto para manutenção de suas populações naturais quanto para conservação ex situ deste material genético. Técnicas de cultura de tecidos vegetais têm sido aplicadas com êxito para conservação de diversas espécies de bromélias. Um dos métodos utilizados é o crescimento lento que consiste na redução do metabolismo da planta e consequentemente na desaceleração do seu crescimento, o que possibilita a manutenção das coleções de plantas in vitro sem necessidade de subcultivo. Neste sentido, o objetivo deste trabalho foi testar a redução de sais do meio de cultura e a adição de osmorregulador na indução do crescimento lento de N. mucugensis. Plantas foram submetidas a tratamentos compostos por diferentes concentrações do meio MS e de manitol pelo período de 12 meses, quando então foram realizadas as análises de crescimento, teor de clorofila e capacidade de regeneração de brotos in vitro. Constatou-se que o tratamento contendo MS ½ e 7,8 g.L-1 de manitol é indicado para a conservação in vitro de N. mucugensis com manutenção da capacidade regenerativa dos seus tecidos.
Effect of cytokinins on in vitro development of autotrophism and acclimatization of Annona glabra L
The role of cytokinins in the differentiation of the photosynthetic apparatus in micropropagated plants and their effect on the plant's ability to transition from a heterotrophic to an autotrophic condition during acclimatization was investigated. Annona glabra L. shoots were cultured on woody plant medium supplemented with sucrose and different cytokinins to evaluate leaf tissue for chloroplast development, chloroplast numbers, photosynthetic pigmentation, total photosynthetic potential, and soluble sugar content. Plants were transferred to the rooting medium in the presence or absence of sucrose and then acclimatized. Kinetin and benzyladenine (BAP) stimulated chloroplast differentiation. Inclusion of zeatin in the medium induced the formation of greater numbers of chloroplasts in the leaves, while plants cultivated in the presence of only kinetin and BAP demonstrated greater chlorophyll a and carotenoid content. The use of kinetin and BAP during in vitro culture promoted accumulation of dry matter during the acclimatization phase, especially in plants rooted under autotrophic conditions (without sucrose). Kinetin and BAP promoted development of more leaf area and greater plant survival rates in plant acclimatization on both autotrophic and heterotrophic media. The inhibitory effects of thidiazuron on the differentiation of chloroplasts, accumulation of chlorophyll a, and photosynthetic potential were examined.
Induction, biochemical trait and phytochemical screening of calluses of Amburana cearensis (Allemão) A.C. Smith
Amburana cearensis is an arboreal legume of the Fabaceae family, with high phytotherapic and medicinal potential due the presence of secondary metabolites. The objective of this study was to evaluate the effect of 2,4-dichlorophenoxyacetic acid (2,4-D) and 4-amino-2,5,6- trichloro-2-pyridinecarboxylic acid (picloram) on the in vitro induction of callogenesis of A. cearensis and analyze the biochemical and phytochemical potential of these calluses. For callus induction, leaf and cotyledon segments were used as explants, which were inoculated in woody plant medium (WPM) supplemented with different concentrations of 2,4-D (0, 5, 10, 20, 40 μM) or picloram (0, 5, 10, 20, 40, 80 μM). The callus growth curve was estimated based on fresh weight, measured at 7-day intervals until 28 days after inoculation. The calluses were analyzed by biochemical tests to quantify the reducing sugars and total proteins. Phytochemical screening and high-performance liquid chromatography were performed to establish the phytochemical profile of extracts from calluses. The concentrations of 21.94 μM and 26.46 μM of 2,4-D induced the greatest formation of compact and friable calluses from the leaf and cotyledon segments, respectively. The growth curve had two distinct phases (lag and exponential) for both types of calluses evaluated. The maximum levels of reducing sugars and total proteins in the calluses from leaf and cotyledon segments were obtained on the day of inoculation and after 28 days of cultivation, respectively. The results of the phytochemical analysis identified the presence of coumarin in all the extracts evaluated, this secondary metabolite has high pharmacological potential.
