Tolerance and pharmacokinetics of single-dose intravenous hemoporfin in healthy volunteers
Aim: To investigate the safety, tolerability and pharmacokinetics of intravenous hemoporfin, a novel photosensitive drug for the treatment of port-wine stain (PWS), in healthy Chinese volunteers following single-dose administration. Methods: Thirty-six healthy Chinese subjects were enrolled. The subjects were administered hemoporfin (2.5, 5, 7.5 or 10 mg/kg) via single-dose intravenous infusion. Pharmacokinetics of the drug were studied in the groups with doses of 2.5, 5 and 7.5 mg/kg, and tolerability was studied in all the 4 groups. Safety and tolerance were evaluated by monitoring adverse events and laboratory parameters, and pharmacokinetics were assessed by determining hemoporfin content with a validated high-performance liquid chromatography with fluorescence detection (HPLC/FLD) method. Results: Mild and transient adverse events occurred in the trial (n=10), but none were serious, and no subjects were withdrawn from the trial. The gastrointestinal tract adverse events, such as nausea, stomach upset, abdominal pain and vomiting, were observed in the groups with doses of 7.5 and 10 mg/kg. Increased alanine aminotransferase (ALT) concentration was found in 3 subjects, and increased alkaline phosphatase (ALP) concentration in one subject. The ha~f-life of hemoporfin for doses of 2.5, 5, and 7.5 mg/kg was 1.26 h, 1.31 h, and 1.70 h, respectively. Cmax and AUC increased with dose for intravenous single-dose administration of hemoporfin in the 2.5, 5, and 7.5 mg/kg groups. Urinary excretion of hemoporfin within 12 h was less than 0.2%. Conclusion: Hemoporfin is safe and well-tolerated in healthy Chinese volunteers at a single intravenous dose of up to 10 mg/kg. It was rapidly cleared from the blood and had a short half-life, which insures a short light-avoidance period.
Development of space end-effector with capabilities of misalignment tolerance and soft capture based on tendon-sheath transmission system
The essential requirements of the end-effector of large space manipulator are capabilities of misalignment tolerance and soft capture. According to these requirements, an end-effector prototype combining the tendon-sheath transmission system with steel cable snaring mechanism was manufactured. An analysis method based on the coordinate transformation and the projection of key points of the mechanical interface was proposed, and it was a guideline of the end-effector design. Furthermore, the tendon-sheath transmission system was employed in the capture subassembly to reduce the inertia of the capture mechanism and enlarge the capture space. The capabilities of misalignment tolerance and soft capture were validated through the dynamic simulation in ADAMS software. The results of the capture simulation and experiment show that the end-effector has outstanding capabilities of misalignment tolerance and soft capture. The translation misalignments in radial directions are ±100 mm, and angular misalignments about pitch and yaw are ±15°.
Current status and challenges of using geometric tolerance information in intelligent manufacturing systems
Recent development in computer-based manufacturing and inspection has necessitated extended knowledge and usage of geometric tolerances as carriers of design intent. The aim of applying geometrical tolerances in design is to provide function-oriented precise description of part geometry where the conventional size tolerance system fails to address. In view of the current development of computer-aided systems, applying geometric tolerances opens a new research front. This article examines the challenges in applying geometric tolerance information to carry the design intent to other downstream manufacturing processes and intelligently integrate the whole system. Based on the observed practical capabilities and literature studies, it is concluded that the current computer-aided design (CAD) systems cannot effectively provide the appropriate use of geometric tolerances. This article highlights the existing challenges and proposes a scheme of algorithm development for appropriate use of tolerance symbols and conditions at the design specification stage. This, in the long run, enables the CAD model to carry the design intent and opens a window of opportunity for intelligently integrating manufacturing systems.
Tolerance or Hospitality?
Facing the development of pluralistic cultures and the conflict of varied civilizations, we should examine political theory frameworks, particularly as concerns the construction of world order. On the problem of \"tolerance or hospitality,\" tolerance has been the target of opposition. Jacques Derrida deconstructs this concept and supports Immanuel Kant's notion of hospitality instead of tolerance. However, Jürgen Habermas advocates reconstructing the concept of tolerance, although he does note its limits. We should regard hospitality as the fundamental spirit of international relations and take the notion of \"Seeking Sameness and Respecting Diversity\" as a principle for handling relations between different cultures.
Stratified model for estimating fatigue crack growth rate of metallic materials
The curve of relationship between fatigue crack growth rate and the stress strength factor amplitude represented an important fatigue property in designing of damage tolerance limits and predicting life of metallic component parts. In order to have a more reasonable use of testing data, samples from population were stratified suggested by the stratified random sample model (SRAM). The data in each stratum corresponded to the same experiment conditions. A suitable weight was assigned to each stratified sample according to the actual working states of the pressure vessel, so that the estimation of fatigue crack growth rate equation was more accurate for practice. An empirical study shows that the SRAM estimation by using fatigue crack growth rate data from different stoves is obviously better than the estimation from simple random sample model.
