Mitochondrial control of inflammation

Numerous mitochondrial constituents and metabolic products can function as damage-associated molecular patterns (DAMPs) and promote inflammation when released into the cytosol or extracellular milieu. Several safeguards are normally in place to prevent mitochondria from eliciting detrimental inflammatory reactions, including the autophagic disposal of permeabilized mitochondria. However, when the homeostatic capacity of such systems is exceeded or when such systems are defective, inflammatory reactions elicited by mitochondria can become pathogenic and contribute to the aetiology of human disorders linked to autoreactivity. In addition, inefficient inflammatory pathways induced by mitochondrial DAMPs can be pathogenic as they enable the establishment or progression of infectious and neoplastic disorders. Here we discuss the molecular mechanisms through which mitochondria control inflammatory responses, the cellular pathways that are in place to control mitochondria-driven inflammation and the pathological consequences of dysregulated inflammatory reactions elicited by mitochondrial DAMPs.Lorenzo Galluzzi and colleagues discuss the molecular mechanisms through which mitochondrial dysfunction elicits inflammatory reactions, the cellular pathways that are in place to control them and how the dysregulation of these systems contributes to pathology.

Gerhard Richter : life and work : in painting, thinking is painting
One of the world's most revered visual artists, Gerhard Richter embraces many concepts in his work and continually thwarts categorization. In this expansive and authoritative overview, Armin Zweite leads readers through every phase of Richter's celebrated career including his early artistic education in East Germany and his later prolific output in West Germany: the black and white photo paintings, the brilliantly conceived color charts and lush, inscrutable gray paintings, installations with glass and mirrors, his landscapes, portraits, and still lifes, and his monumental abstract paintings, which broke records at auction. Necroptosis, pyroptosis and apoptosis: an intricate game of cell death

Cell death is a fundamental physiological process in all living organisms. Its roles extend from embryonic development, organ maintenance, and aging to the coordination of immune responses and autoimmunity. In recent years, our understanding of the mechanisms orchestrating cellular death and its consequences on immunity and homeostasis has increased substantially. Different modalities of what has become known as ‘programmed cell death’ have been described, and some key players in these processes have been identified. We have learned more about the intricacies that fine tune the activity of common players and ultimately shape the different types of cell death. These studies have highlighted the complex mechanisms tipping the balance between different cell fates. Here, we summarize the latest discoveries in the three most well understood modalities of cell death, namely, apoptosis, necroptosis, and pyroptosis, highlighting common and unique pathways and their effect on the surrounding cells and the organism as a whole.

كان القرن العشرين إيذانا ببدء نهضة أدبية كبرى من أبرز مظاهرها الشعر، وقد اضطلع شاعران من أبناء مصر بتلك النهضة ؛ فكان (أحمد شوقي) أميرا للشعر والشعراء، واستطاع (حافظ إبراهيم) أن يتبوأ موقعا متميزا بين شعراء عصره حتى لقب بـ \"شاعر النيل\"، وكلا الشاعرين قد أحييا الشعر العربي، وردا إليه نشاطه ونضرته. وقد تصدى (طه حسين) لنقد شعرهما، وإنزاله المنزلة التي يستحقها، وعلى الرغم من أن علاقته مع (أحمد شوقي) لم تكن جيدة، إلا أنه حاول إلتزام الحيدة والإنصاف ؛ فجاء كتابه نقدا أمينا لهذين العلمين من أعلام الأدب العربي، راصدا الأخطاء التي وقعا فيها، وإن لم ينكر مكانتهما ودورهما في الشعر العربي.
Molecular mechanisms of gasdermin D pore-forming activity

The regulated disruption of the plasma membrane, which can promote cell death, cytokine secretion or both is central to organismal health. The protein gasdermin D (GSDMD) is a key player in this process. GSDMD forms membrane pores that can promote cytolysis and the release of interleukin-1 family cytokines into the extracellular space. Recent discoveries have revealed biochemical and cell biological mechanisms that control GSDMD pore-forming activity and its diverse downstream immunological effects. Here, we review these multifaceted regulatory activities, including mechanisms of GSDMD activation by proteolytic cleavage, dynamics of pore assembly, regulation of GSDMD activities by posttranslational modifications, membrane repair and the interplay of GSDMD and mitochondria. We also address recent insights into the evolution of the gasdermin family and their activities in species across the kingdoms of life. In doing so, we hope to condense recent progress and inform future studies in this rapidly moving field in immunology.
Devant and Kagan review the biochemical and cell biological mechanisms that control gasdermin D pore-forming activity and its diverse downstream immunological effects.

Programmed cell death and the immune system

In this Timeline article, Shigekazu Nagata and Masato Tanaka highlight some of the key discoveries that have shaped the field of programmed cell death over the past 50 years and explain their relevance for the immune system.
More than 50 years ago, cells were observed to die during insect development via a process that was named 'programmed cell death'. Later, a similar cell death process was found to occur in humans, and the process was renamed 'apoptosis'. In the 1990s, a number of apoptosis-regulating molecules were identified, and apoptosis was found to have essential roles in the immune system. In this Timeline article, we highlight the key events that have demonstrated the importance of programmed cell death processes, including apoptosis and programmed necrosis, in the immune system.

Saudi Arabia under Ibn Saud : economic and financial foundations of the state
At its founding in 1932, the Kingdom of Saudi Arabia was characterized by tribal warfare, political instability, chronic financial shortages and economic crises. As a desert chieftain, Abd al-Aziz Ibn Saud, the ruler and king until 1953, had the skills, the cunning and the power to control the tribes and bring peace to this realm. But financial and economic matters were not his forte and these he left mostly to a single individual, Abdullah al-Sulayman al-Hamdan. He was entrusted with nearly all of the country's early financial dealings and administrative development. This book examines the role of the Ministry of Finance and its minister, Abdullah al-Sulayman, in holding the country together financially and administratively until the promise of substantial oil income was realized a few years after the end of World War II.
Molecular mechanisms of cell death: recommendations of the Nomenclature Committee on Cell Death 2018

Over the past decade, the Nomenclature Committee on Cell Death (NCCD) has formulated guidelines for the definition and interpretation of cell death from morphological, biochemical, and functional perspectives. Since the field continues to expand and novel mechanisms that orchestrate multiple cell death pathways are unveiled, we propose an updated classification of cell death subroutines focusing on mechanistic and essential (as opposed to correlative and dispensable) aspects of the process. As we provide molecularly oriented definitions of terms including intrinsic apoptosis, extrinsic apoptosis, mitochondrial permeability transition (MPT)-driven necrosis, necroptosis, ferroptosis, pyroptosis, parthanatos, entotic cell death, NETotic cell death, lysosome-dependent cell death, autophagy-dependent cell death, immunogenic cell death, cellular senescence, and mitotic catastrophe, we discuss the utility of neologisms that refer to highly specialized instances of these processes. The mission of the NCCD is to provide a widely accepted nomenclature on cell death in support of the continued development of the field.
